Genomic, Antimicrobial Resistance, and Public Health Insights into Enterococcus spp. from Australian Chickens.

Due to Australia's management of antimicrobial use in poultry, particularly the discontinued use of avoparcin for nearly 20 years, it is hypothesized that vancomycin-resistant enterococci associated with human disease are not derived from poultry isolates. This study evaluated antimicrobial resistance (AMR) of five enterococcal species isolated from Australian meat chickens, genomic features of Enterococcus faecium and Enterococcus faecalis, and the phylogenetic relationship of the poultry-derived E. faecium with isolates from human sepsis cases. All enterococcal isolates from chicken ceca wer

Copper deficiency and heart disease: molecular basis, recent advances and current concepts.

Copper is an essential trace element and has profound influence on cardiac myopathy and heart metabolism. Dietary Cu restriction in rats results in cardiomyopathy, and affects the integrity of the basal lamina of cardiac myocytes and capillaries. Decreased levels of delta subunits of ATP synthetase and nuclear encoded subunits of cytochrome oxidase system have been observed. Alteration in expression of glutathione peroxidase and catalase in heart and liver in Cu deficiency (Cu-) has been noted involving both transcriptional and post transcriptional mechanisms. A short description of two geneti

A Macroporous Magnesium Oxide-Templated Carbon Adsorbs Shiga Toxins and Type III Secretory Proteins in Enterohemorrhagic Escherichia coli, Which Attenuates Virulence.

Enterohemorrhagic Escherichia coli (EHEC) is one of the most common foodborne pathogens. However, no drug that prevents the severe complications caused by this bacterium has been approved yet. This study showed that a macroporous magnesium oxide (MgO)-templated carbon material (MgOC150) adsorbs Shiga toxins, and Type III secretory EspA/EspB proteins responsible for EHEC pathogenesis, and decreases the extracellular levels of these proteins. On the other hand, this material did not affect the growth of EHEC. Citrobacter rodentium traditionally used to estimate Type III secretion system-associat

Investigating gut microbiota and their metabolites as biomarkers for tacrolimus pharmacokinetic variability.

Tacrolimus (TAC), a cornerstone immunosuppressant in transplantation, presents a clinical challenge due to its narrow therapeutic index and substantial interindividual pharmacokinetic (PK) variability. This exploratory study investigated the association between gut microbiota composition, short-chain fatty acid (SCFA) metabolites, and TAC PK variability during the early post-kidney transplantation period. Based on prediction errors derived from a previously established population PK model, 36 transplant recipients were stratified into positive (n = 17) and negative (n = 19) deviation groups. M

Membrane-acting bacteriocin purified from a soil isolate Pediococcus pentosaceus LB44 shows broad host-range.

Bacteriocin LB44 was purified from cell-free supernatant (CFS) of Pediococcus pentosaceus LB44 using activity-guided chromatography techniques. It was stable up to 121 °C, pH 2.0-6.0, sensitive to proteinase K, papain and trypsin, and retained complete activity in the presence of organic solvents tested. The molecular weight of bacteriocin was ∼6 kDa and initial ten amino acid residues (GECGMCXECG) suggested a new compound. The loss in viable cell count and K+ ion efflux of target cells of Micrococcus luteus suggested bactericidal activity. The cell membrane of bacteriocin-treated cells was fo

The type I macrophage scavenger receptor binds to gram-positive bacteria and recognizes lipoteichoic acid.

Macrophage scavenger receptors exhibit unusually broad binding specificity for polyanionic ligands and have been implicated in atherosclerosis and various host defense functions. Using a radiolabeled, secreted form of the type I bovine macrophage scavenger receptor in an in vitro binding assay, we have found that this receptor binds to intact Gram-positive bacteria, including Streptococcus pyogenes, Streptococcus agalactiae, Staphylococcus aureus, Enterococcus hirae, and Listeria monocytogenes. Competition binding studies using purified lipoteichoic acid, an anionic polymer expressed on the su

New approaches to disinfection of thermolabile medical devices using an indirect method with cold atmospheric plasma-aerosol.

Cold atmospheric plasma-aerosol (CAP-A) offers a promising alternative to conventional sterilisation and disinfection methods, which are often unsuitable for thermolabile medical devices due to high temperatures, toxic chemicals or radiation. CAP-A efficiently inactivates microorganisms and viruses without compromising the material integrity. Given the ongoing risk of infection associated with ultrasound probes and other delicate diagnostic instruments, this study investigates whether an indirect CAP-A method can meet all requirements for effective and safe disinfection of thermolabile medical

Novel Multidrug-Resistant Enterococcal Mobile Linear Plasmid pELF1 Encoding vanA and vanM Gene Clusters From a Japanese Vancomycin-Resistant Enterococci Isolate.

Vancomycin-resistant enterococci are troublesome pathogens in clinical settings because of few treatment options. A VanA/VanM-type vancomycin-resistant Enterococcus faecium clinical isolate was identified in Japan. This strain, named AA708, harbored five plasmids, one of which migrated during agarose gel electrophoresis without S1 nuclease treatment, which is indicative of a linear topology. We named this plasmid pELF1. Whole genome sequencing (WGS) analysis of the AA708 strain revealed that the complete sequence of pELF1 was 143,316 bp long and harbored both the vanA and vanM gene clusters. F

Isolation and characterization of enterococci from poultry reveals high incidence of Enterococcus thailandicus in Victoria, Australia.

AIMS: Antibiotic resistance is a global health crisis. Roughly two-thirds of all antibiotics used are in production animals, which have the potential to impact the development of antibiotic resistance in bacterial pathogens of humans. There is little visibility on the extent of antibiotic resistance in the Australian food chain. This study sought to establish the incidence of antibiotic resistance among enterococci from poultry in Victoria. METHODS AND RESULTS: In 2016, poultry from a Victorian processing facility were swabbed immediately post-slaughter and cultured for Enterococcus species.

Sustainable Disruption of Microbial Biofilms: Functional and Antimicrobial Assessment of a Lipopeptide Biosurfactant.

Biofilm-associated infections pose significant challenges due to the increased antibiotic resistance of microorganisms within these structures. This study involved isolating and characterizing the biosurfactant produced by Bacillus subtilis RSL-2 using molasses to develop a sustainable antibiofilm agent. The biosurfactant was identified as a surfactin-like lipopeptide (molecular weight: 1037 g/mol) based on FTIR, 1H NMR, and HRMS analyses. It demonstrated a low critical micelle concentration (CMC) of 70 mg/L and markedly decreased surface and interfacial tensions to 24.46 and 0.46 mN/m, respec

Gut microbiome is associated with stereotypic behavior of the giant panda (Ailuropoda melanoleuca).

The giant panda (Ailuropoda melanoleuca) is a relict species endemic to China. Captive breeding has boosted its population. However, some captive individuals exhibit stereotypic behavior. Though extensive research showing that host behavior can be influenced by gut microbiota via the gut-brain axis, the gut microbiota related to stereotypic behavior of giant pandas has not been reported. To explore gut microbial composition and functional characteristics of stereotypically behaving (SB) giant pandas, the behavior of 15 captive giant pandas was observed, and their feces were analyzed using 16S

Antimicrobial Efficacy of a Taurolidine-Based Antimicrobial Compound on Contaminated Surfaces Simulated in a Standardized 4-Field Test.

Implantable medical devices, including cardiac electronic implants, joint prostheses, and breast implants, are essential to modern healthcare but remain susceptible to infection from microbial contamination during placement. Staphylococcus spp. and Candida albicans are the predominant pathogens, often causing severe complications, increased mortality, and substantial healthcare costs. With antibiotic resistance on the rise, intraoperative surface disinfection has emerged as a critical yet underutilized preventive strategy. Spectral Phasor Applied to Spectrally-Resolved Single Molecule Localization Microscopy.

Spectrally-resolved single-molecule localization microscopy (srSMLM) has emerged as a powerful tool for exploring the spectral properties of single emitters in localization microscopy. By simultaneously capturing the spatial positions and spectroscopic signatures of individual fluorescent molecules, srSMLM opens up the possibility of investigating an additional dimension in super-resolution imaging. However, appropriate and dedicated tools are required to fully capitalize on the spectral dimension. Here, we propose the application of the spectral phasor analysis as an effective method for summ

Assessment of the antioxidant, antimicrobial and antibiofilm activities of essential oils for potential application of active chitosan films in food preservation.

In the food industry, the development of microbial biofilms is a serious problem that leads to the contamination and deterioration of food products. To overcome that, our aim consists of searching for natural antimicrobial and non-toxic compounds (essential oils EOs), which might be used alone or adsorbed on natural biopolymer films (chitosan). In this work, the antioxidant activity of eight EOs was evaluated by DPPH radical-scavenging method while their antibacterial activity was determined by diffusion on agar and microdilution methods. Among all tested EOs, Eugenia caryophyllus, Cinnamomum

Development of Quality Control Ranges for Biocide Susceptibility Testing.

Every laboratory test needs validation by quality controls. For biocide susceptibility testing (BST), neither quality control (QC) strains nor QC ranges applicable to these strains are currently available. As QC strains, four well-defined laboratory reference strains (Staphylococcus aureus ATCC® 6538, Enterococcus hirae ATCC® 10541, Escherichia coli ATCC® 10536 and Pseudomonas aeruginosa ATCC® 15442), which have been used previously for biocide efficacy testing, were selected. In an interlaboratory trial with eleven participating laboratories, BST QC ranges should be developed for the aforemen

Elaboration and general evaluation of chitosan-based films containing terpene alcohols-rich essential oils.

Recently, the scientific community is interested in the synthesis of biodegradable and bioactive packaging to replace oil-based ones. Therefore, the present study aims to elaborate an active and biodegradable material using chitosan (CS-film) combined with pelargonium, tea tree, marjoram, and thyme essential oils (EOs), and then evaluate their different properties and biological activities. The obtained data showed an augmentation in CS-film thickness and opacity following the addition of EOs ranging from 17 ± 3 to 42 ± 2 μm and from 1.53 ± 0.04 to 2.67 ± 0.09, respectively. Furthermore, a sig

Screening of lactic acid bacteria from the milk of Sahiwal cows and characterization of their probiotic potential for preventing bovine mastitis.

Antibiotic therapy has been the most popular line of treatment for the control of mastitis worldwide during the last few decades. Alternative and sustainable treatments must be developed because pathogens are becoming more resistant to antibiotics, leading to the development and spread of antimicrobial resistance (AMR). The aim of the current investigation was to isolate lactic acid bacteria (LAB) with probiotic potential that can inhibit mastitis-causing pathogens to prevent bovine mastitis. Milk samples were collected from Sahiwal cows, and a total of 150 bacteria were isolated, of which 76
