We describe a dot enzyme-linked immunosorbent assay (dot-ELISA) as a field applicable tool for the rapid diagnosis of schistosomiasis. This antibody capture assay is performed using 50 ng protein of crude Schistosoma mansoni egg antigen (SEA) in 1 microliter volumes per dot. Sera (1 microliter/dot) at 1:40 dilution were optimal. An enzyme-linked immunosorbent assay (ELISA) for the detection of Japanese encephalitis (JE) antibody is described, after 269 human sera were tested by both hemagglutination inhibition (HI) test and ELISA. In ELISA screening, the antigen concentration and the dilution of horseradish peroxidase conjugated goat anti-human Ig were determined by chequerboard titration as 10 micrograms/ml and 1:1000. The color produced by enzyme-substrate reaction was measured on a spectrophotometer. The results showed a good agreement with those obtained from a routine HI test.

The remainders of tuberculosis infection are still existing as a problem, as important and threatening as AIDS (acquired immune deficiency syndrome), especially in underdeveloped countries. The diagnosis of tuberculosis infection frequently creates problems which is related to clinical, radiological and bacteriological investigations and it requires a lot of time consumption. The ELISA procedure is a sensitive and reproducible technique and it doesn't require complex equipment. Recently, this technique provides hopeful results for the serodiagnosis of tuberculosis.
