Plant histone variants: deposition mechanisms, functional diversification, and translational prospects.

Histone variants are non-allelic isoforms of canonical histones that assemble into nucleosomes via replication-independent pathways, reshaping chromatin biophysical properties and transcriptional control at specific genomic loci. Constrained by their sessile lifestyle, plants have evolved a rich set of histone variants, including lineage-specific innovations such as heterochromatin-stabilizing H2A.W, stress-inducible H1.3, gametophyte-restricted H2B.8, Arabidopsis H3.14, and Oryza-specific H4.V. This review systematically summarizes recent advances in plant histone variants from three dimensio

Current genetic approaches for the treatment of prion diseases.

Prion diseases are fatal neurodegenerative disorders caused by the misfolding of the host-encoded prion protein (PrP) into a pathogenic conformer (PrPSc). Despite decades of investigation, no therapy has proven effective, largely due to rapid disease progression and the absence of druggable intermediates. Recent molecular advances, however, have established PrP itself as a viable therapeutic substrate. Experimental ablation or suppression of Prnp in mice -the gene encoding PrP- confers complete resistance to prion infection in animal models, providing a strong genetic rationale for PrP- loweri

MeCP2 Dosage Control in Rett Syndrome: Non-Coding RNA-Based and Epigenetic Strategies for Safer Gene Therapy.

Rett syndrome (RTT) is a severe X-linked neurodevelopmental disorder that is caused in most cases by pathogenic variants in MECP2, the gene encoding methyl-CpG-binding protein 2 (MeCP2). Despite substantial progress in the development of gene therapy, restoring MECP2 expression remains challenging because MeCP2 is highly dosage-sensitive. Both deficiency and excessive expression of this protein are associated with severe neurological abnormalities. This makes simple viral vector-mediated replacement of MECP2 potentially unsafe and underscores the need for multilayered systems that control tran

Prevention of transgenerational transmission of disease susceptibility through perinatal intervention.

The observational findings of Barker's original epidemiological studies were generalized as the Barker hypothesis and extended as the Developmental Origins of Health and Disease (DOHaD) theory. Barker et al. proposed that low birthweight (LBW) was associated with the occurrence of various noncommunicable diseases (NCDs) later in life. In other words, LBW itself is associated with the development of NCDs. This led to the DOHaD theory which proposed that an organism may have a specific period of developmental plasticity that is highly sensitive to the factors in its environment, and that combina

Multiscale Convergence of Signaling, Epigenetic, and Environmental Networks in Mammalian Palatogenesis and Cleft Palate Pathogenesis.

Palatogenesis is tightly regulated at cellular and molecular levels. Cranial neural crest cells (CNCCs), multipotent stem cells that generate most palatal mesenchyme, self-renew, decide fate, and differentiate under cues from their stem cell niche. Orofacial clefts (OFC), among the most common congenital birth defects, arise when CNCC-mediated palate formation is disturbed. Here we review genes, epigenetic machineries, and environmental insults regulating secondary palate development, focusing on CNCC stem cell biology and translational perspectives in tissue engineering, gene therapy, and reg

Epitranscriptome: A Novel Regulatory Layer during Atherosclerosis Progression.

RNA modifications have recently gained great attention due to their extensive regulatory effects in a wide range of cellular networks and signaling pathways. In cardiovascular diseases (CVDs), several RNA changes, called "epitranscriptome" alterations, are found in all RNA molecules (tRNA, rRNA, mRNA, and ncRNAs). Unlike the epigenetic process, which influences the progression of atherosclerosis (AS), its transcriptional and post-transcriptional regulatory mechanisms are still unknown. Here, we described the main epitranscriptome signs to provide new insights into AS, including m6A, m5C, m1A,

Epigenetic Modulation of Immunity: Mechanisms, Implications, and Emerging Therapeutic Horizons; a Step Toward Epigenetics to Precision.

Epigenetic regulation plays a central role in immune cell development, specialization, and memory formation by dynamically modifying DNA, histones, and RNA. These processes enable adaptation to environmental cues, precise pathogen responses, and maintenance of immune tolerance, while their disruption contributes to autoimmune, inflammatory, and cancer pathogenesis. DNA methylation, histone modifications, and noncoding RNA regulation shape the lineage and activation states of T cells, B cells, macrophages, and natural killer (NK) cells, with specific alterations linked to diseases, such as syst

Recent progress in T-cell malignancies including adult T-cell leukemia-lymphoma.

T-cell lymphomas represent a heterogeneous group of lymphoid malignancies characterized by marked biological diversity and generally poor clinical outcomes. Recent updates to the fifth edition of the World Health Organization Classification and the 2022 International Consensus Classification have refined the disease entities based on transcription factor profiles, cytokine signatures, and molecular features. Comprehensive genomic and epigenomic analyses have revealed recurrent alterations affecting T-cell receptor signaling, epigenetic regulation, cell-cycle control, and immune pathways, there

NRSF regulation of OPRM1 through histone acetylation.

The Neuron Restrictive Silencer Factor (NRSF) is a master transcriptional regulator of neuronal differentiation. Dysregulation of NRSF in neurons contributes to multiple neurological disorder. Following peripheral nerve injury, NRSF is upregulated and has been implicated in the establishment and maintenance of neuropathic pain. While the molecular and genetic changes in nerves occur during neuropathic pain are numerous and complex, a key downstream consequence of NRSF activation is repression of the mu-opioid receptor gene (OPRM1), a critical mediator of nociception and opioid responsiveness.

Decoding heavy metal tolerance in rice: Nucleic acid-based technologies shaping global food security.

Global rice production is critically threatened by heavy metal contamination, particularly cadmium (Cd) and arsenic (As), which compromises yield, diminishes grain nutritional quality, and exposes billions of consumers to nephrotoxic and carcinogenic risks. Conventional remediation strategies (soil amendments, water management, phytoremediation) are prohibitively expensive, temporally protracted, and fundamentally reactive, while conventional breeding is constrained by linkage drag, polygenic trait architecture, and absence of natural alleles that restrict toxic metal uptake from essential min

The Tip60/Ep400 chromatin remodeling complex impacts basic cellular functions in cranial neural crest-derived tissue during early orofacial development.

The cranial neural crest plays a fundamental role in orofacial development and morphogenesis. Accordingly, mutations with impact on the cranial neural crest and its development lead to orofacial malformations such as cleft lip and palate. As a pluripotent and dynamic cell population, the cranial neural crest undergoes vast transcriptional and epigenomic alterations throughout the formation of facial structures pointing to an essential role of factors regulating chromatin state or transcription levels. Using CRISPR/Cas9-guided genome editing and conditional mutagenesis in the mouse, we here sho

The emerging role of epigenetic regulation in pain sensitization associated with headache disorders.

BACKGROUND: Headache disorders, including migraine, tension-type headache, and cluster headache, are among the most prevalent and disabling neurological conditions. Although genetic factors contribute to their pathogenesis, they fail to fully account for their clinical heterogeneity, episodic nature, and varied treatment responses. This review synthesizes current evidence on the role of epigenetic mechanisms—including DNA methylation, histone modifications, non-coding RNAs, and RNA modifications—in the pathophysiology of headache disorders. PRINCIPAL FINDINGS: Evidence from both human studies

Epigenetic Aspects and Prospects in Autoimmune Hepatitis.

The observed risk of autoimmune hepatitis exceeds its genetic risk, and epigenetic factors that alter gene expression without changing nucleotide sequence may help explain the disparity. Key objectives of this review are to describe the epigenetic modifications that affect gene expression, discuss how they can affect autoimmune hepatitis, and indicate prospects for improved management. Multiple hypo-methylated genes have been described in the CD4+ and CD19+ T lymphocytes of patients with autoimmune hepatitis, and the circulating micro-ribonucleic acids, miR-21 and miR-122, have correlated with

CRISPR/Cas9 therapeutics: a cure for cancer and other genetic diseases.

Cancer is caused by a series of alterations in genome and epigenome mostly resulting in activation of oncogenes or inactivation of cancer suppressor genes. Genetic engineering has become pivotal in the treatment of cancer and other genetic diseases, especially the formerly-niche use of clustered regularly interspaced short palindromic repeats (CRISPR) associated with Cas9. In defining its superior use, we have followed the recent advances that have been made in producing CRISPR/Cas9 as a therapy of choice. We also provide important genetic mutations where CRISPRs can be repurposed to create ad

Methylation Mesa define functional regulatory elements for targeted gene activation.

DNA methylation regulates transcription, yet the demethylation of canonical elements like promoter CpG islands exhibits inconsistent correlations with gene activation. We hypothesize that causal regulatory elements are defined by biophysical hypersensitivity. Profiling 24 whole-genome bisulfite sequencing samples across diverse human and murine models, we identify Methylation Mesa, narrow (~45-300 bp), structurally conserved epigenetic regulatory elements. Mesa show enrichment in 5' untranslated regions and associate with transcriptional activation significantly better than canonical promoters

RNLS hypomethylation as a potential epigenetic marker of lung cancer metastasis: an epigenome-wide association study.

BACKGROUND: Lung cancer is highly lethal because most patients have already developed metastases at diagnosis, yet the epigenetic mechanisms driving metastasis are unclear. METHODS: We performed an epigenome-wide association study on 42 lung cancer patients to identify differentially methylated positions (DMPs) in pulmonary tumour (PT), paratumor (PP), and normal (PN) tissues, and in peripheral blood. The impact of DMPs on gene expression and associated signalling pathways was examined. Functional validation included gene silencing in A549 and H1299 cells, analysis of the affected signalling

Hematoma-driven immuno-epigenetic remodeling after intracerebral hemorrhage: cell-type-specific mechanisms and therapeutic opportunities.

Intracerebral hemorrhage (ICH) is a life-threatening subtype of stroke characterized by the acute accumulation of blood within the brain parenchyma and progressive secondary brain injury. After ICH, dysregulated neuroinflammation drives a cascade of secondary injury processes that shape neurological deterioration and long-term recovery. However, the molecular mechanisms that determine the magnitude, temporal evolution, and resolution of immune-inflammatory responses after ICH remain incompletely understood, limiting the development of targeted therapeutic strategies. Accumulating evidence indi
