The role of SLC2A1 mutations in myoclonic astatic epilepsy and absence epilepsy, and the estimated frequency of GLUT1 deficiency syndrome.

The first mutations identified in SLC2A1, encoding the glucose transporter type 1 (GLUT1) protein of the blood-brain barrier, were associated with severe epileptic encephalopathy. Recently, dominant SLC2A1 mutations were found in rare autosomal dominant families with various forms of epilepsy including early onset absence epilepsy (EOAE), myoclonic astatic epilepsy (MAE), and genetic generalized epilepsy (GGE). Our study aimed to investigate the possible role of SLC2A1 in various forms of epilepsy including MAE and absence epilepsy with early onset. We also aimed to estimate the frequency of G

Investigating the Effects of NMDAR and LGI1 Antibodies on Absence Seizures: Insights from Genetic Absence Epilepsy Rats and Acute Pharmacological Model of Absence Seizures.

INTRODUCTION: Childhood Absence Epilepsy, a subtype of genetic generealised epilepsy, is characterised by sudden and brief episodes of impaired consciousness. The Leucine-rich glioma-inactivated protein 1 (LGI1) and N-methyl-D-aspartate receptor (NMDAR) are key proteins involved in regulating neuronal excitability. In conditions like anti-LGI1 encephalitis and anti-NMDAR encephalitis, autoantibodies target and disrupt these proteins, causing memory deficits, behavioural changes, sleep disturbances, and epileptic seizures. However, the roles of LGI1 and NMDAR dysfunction in the pathophysiology

Sleep Architecture and Microstructure in Childhood Absence Epilepsy: Clinical and Neurophysiological Perspectives.

UNLABELLED: Childhood absence epilepsy (CAE) is one of the most common epilepsy syndromes in childhood and has traditionally been regarded as a condition with a favorable neurological prognosis. However, increasing evidence suggests that CAE is associated with functional disturbances in neuronal networks that extend beyond seizure generation and may involve sleep and wakefulness regulation. METHODS: This narrative mini-review summarizes and critically discusses current clinical and neurophysiological evidence regarding alterations in sleep architecture and sleep electroencephalographic (EEG)

Long-term seizure remission in childhood absence epilepsy: might initial treatment matter?

OBJECTIVE: Examine the possible association between long-term seizure outcome in childhood absence epilepsy (CAE) and the initial treatment choice. METHODS: Children with CAE were prospectively recruited at initial diagnosis and followed in a community-based cohort study. Children presenting with convulsive seizures, significant imaging abnormalities, or who were followed <5 years were excluded. Early outcomes included success of initial medication, early remission, and pharmacoresistance. The primary long-term outcome was complete remission: ≥5 years both seizure free and medication free. Electroencephalographic differences between WAG/Rij and GAERS rat models of absence epilepsy.

The inbred Wistar Albino Glaxo Rats from Rijswijk (WAG/Rij) and the Genetic Absence Epilepsy Rats from Strasbourg (GAERS) are well-validated genetic models of absence epilepsy. Although they share similar characteristics including the spike-and-wave discharges (SWDs) in the EEG, some differences have been reported between both strains. This study aimed a systematic and detailed comparison of the SWD patterns of both strains in terms of the intensity, frequency and waveform morphology of the discharges by using exactly the same measurement and analysis techniques. The number, cumulative total d

[Functional state of cardiovascular system by progressive absences--epilepsy and its treatment].

Age-dependent increase of seizure activity at absence epilepsy exacerbates hemodynamic and autonomic regulation of heart rate. Cardiac dysfunction is accompanied by an increasing duration of intervals repolarization left ventricular QTc, which causes the risk of life-threatening arrhythmias, increases the threat of sudden cardiac death. Anticonvulsant drug therapy provides an opportunity to improve the functional state of the cardiovascular system, if not exceeded a certain level of seizure activity of the brain. This possibility remains as long as the progressive seizure activity isn't reache

Childhood absence epilepsy and febrile seizures: a family with a GABA(A) receptor mutation.

Although several genes for idiopathic epilepsies from families with simple Mendelian inheritance have been found, genes for the common idiopathic generalized epilepsies, where inheritance is complex, presently are elusive. We studied a large family with epilepsy where the two main phenotypes were childhood absence epilepsy (CAE) and febrile seizures (FS), which offered a special opportunity to identify epilepsy genes. A total of 35 family members had seizures over four generations. The phenotypes comprised typical CAE (eight individuals); FS alone (15), febrile seizures plus (FS(+)) (three); m

Prognostic significance of failure of the initial antiepileptic drug in children with absence epilepsy.

PURPOSE: In children with childhood absence epilepsy (CAE) and juvenile absence epilepsy (JAE), to determine the impact of failure of initial antiepileptic drug (AED) for lack of efficacy in eventual seizure control and long-term remission of epilepsy. METHODS: Centralized EEG records for the province of Nova Scotia allowed identification of all children seen with CAE or JAE between 1977 and 1985. Information regarding success or failure of initial AED in fully controlling seizures and long-term seizure control and remission of epilepsy was collected by patient questionnaire and chart review.

Genetic models of absence epilepsy, with emphasis on the WAG/Rij strain of rats.

In this review, the main characteristics of genetic models of absence epilepsy, in particular with respect to WAG/Rij rats, are presented. Genetic models are important and relevant, since evidence exists that these models mimic spontaneously occurring human epilepsy more than models in which epilepsy is artificially induced. Genetic models can be divided into models in which seizures are elicited and into those in which epilepsy appears without any sensory stimulation. The majority of genetic models show that absence type of epilepsy; during the last few years, we and others have noticed that

Subclinical auditory dysfunction in a genetic rat model of childhood absence epilepsy.

OBJECTIVE: Childhood absence epilepsy (CAE) is defined by cognitive, behavioral, and neurophysiological abnormalities, but its impact on auditory function remains underexplored. Using Genetic Absence Epilepsy Rats from Strasbourg (GAERS), this study aimed to examine both peripheral and central auditory pathways function to better understand the auditory consequences of CAE. METHODS: The experimental and control groups consisted of six-month-old male GAERS (n=6) and Wistar (n=6) rats. Auditory functions were assessed using distortion product otoacoustic emissions (DPOAE) and auditory brainstem

Lower myelin-water content of the frontal lobe in childhood absence epilepsy.

OBJECTIVE: The frontal lobe in childhood absence epilepsy (CAE) might be affected due to the suggested involvement of the frontal lobe during absence seizures and reports on attentional deficits. Previously, subtle white matter abnormalities have been reported in CAE. However, the impact of one of the most characteristic components of the white matter, the myelin content, remains underdetermined. Therefore, this study investigated whether the myelin content in frontal areas is adversely affected in CAE compared to controls. METHODS: Seventeen children with childhood absence epilepsy (mean age

Complex SCN8A DNA-abnormalities in an individual with therapy resistant absence epilepsy.

BACKGROUND: De novo SCN8A missense mutations have been identified as a rare dominant cause of epileptic encephalopathy. We described a person with epileptic encephalopathy associated with a mosaic deletion of the SCN8A gene. METHODS: Array comparative genome hybridization was used to identify chromosomal abnormalities. Next Generation Sequencing was used to screen for variants in known and candidate epilepsy genes. A single nucleotide polymorphism array was used to test whether the SCN8A variants were in cis or in trans. RESULTS: We identified a de novo mosaic deletion of exons 2-14 of SCN8A
