A closer look into the GL7 antigen: its spatio-temporally selective differential expression and localization in lymphoid cells and organs in human.

The GL7 epitope was originally described as part of a late lymphocyte activation antigen expressed in mouse and widely used since then as a marker of germinal center. Here we report on its differential expression by rat and human immune cells and lymphoid organs. Expression pattern of the GL7 epitope in rats is similar to that described earlier in mice, namely that GL7 antigen appears only on lymphocytes after 48h activation. In humans lymphocytes, but not the differentiated cells of myeloid origin, express this epitope. The GL7 epitope is up-regulated upon in vitro activation of primary T cel

An immunoinformatics-derived multi-epitope DNA vaccine targeting the Micropterus salmoides rhabdovirus G protein elicits protective immunity in largemouth bass.

Largemouth bass (Micropterus salmoides) is a commercially important freshwater aquaculture species. However, the sustainable development of this industry is severely threatened by Micropterus salmoides rhabdovirus (MSRV), a critical pathogen that causes acute mortality and substantial economic losses. In this study, five cytotoxic T lymphocyte (CTL) epitopes, three helper T lymphocyte (HTL) epitopes, and two B cell epitopes were identified from the MSRV G protein using immunoinformatics approaches. By serially linking these epitopes, a multi-epitope antigen (Multi-G) consisting of 149 amino ac

Identification of Candidate epitopes from nation-enriched sequences in the African swine fever virus genomes.

African swine fever virus (ASFV) is a highly lethal pathogen that causes African swine fever (ASF) and is found in Africa, Europe, and Asia. In addition, extensive regional genetic variation poses significant challenges for vaccine development. In this study, we aimed to identify amino acid sequence fragments that are commonly enriched within ASFV strains from each country and evaluate their potential as epitopes. Amino acid sequences of genomes were segmented into overlapping 9-, 12-, 15-, and 20-mer peptides; selected fragments enriched in countries with at least ten available genomes; and c

Design of multi-epitope vaccine candidate against Brucella type IV secretion system (T4SS).

Brucellosis is a common zoonosis, which is caused by Brucella infection, and Brucella often infects livestock, leading to abortion and infertility. At present, human brucellosis remains one of the major public health problems in China. According to previous research, most areas in northwest China, including Xinjiang, Tibet, and other regions, are severely affected by Brucella. Although there are vaccines against animal Brucellosis, the effect is often poor. In addition, there is no corresponding vaccine for human Brucellosis infection. Therefore, a new strategy for early prevention and treatme

An Approach for a Synthetic CTL Vaccine Design against Zika Flavivirus Using Class I and Class II Epitopes Identified by Computer Modeling.

The threat posed by severe congenital abnormalities related to Zika virus (ZKV) infection during pregnancy has turned development of a ZKV vaccine into an emergency. Recent work suggests that the cytotoxic T lymphocyte (CTL) response to infection is an important defense mechanism in response to ZKV. Here, we develop the rationale and strategy for a new approach to developing cytotoxic T lymphocyte (CTL) vaccines for ZKV flavivirus infection. The proposed approach is based on recent studies using a protein structure computer model for HIV epitope selection designed to select epitopes for CTL at

In silico screening and identification of CTL and HTL epitopes in the secreted virulence factors of Mycobacterium tuberculosis.

BACKGROUND: Mycobacterium tuberculosis (MTb) is a highly infectious pathogen and a global health threat due to its resilient cell wall and immune evasion strategies. Despite the availability of the antituberculosis Bacille Calmette-Guérin (BCG) vaccine, its efficacy varies (0%-80%) and gradually decreases over time. This study aimed to identify cytotoxic T-lymphocyte (CTL) and helper T-lymphocyte (HTL) epitopes in MTb secretory proteins using immunoinformatics tools. MATERIALS AND METHODS: The Protein Variability Server was used to identify highly conserved sequences, and epitope population c

Design of multi-epitope-based therapeutic vaccine candidates from HBc and HBx proteins of hepatitis B virus using reverse vaccinology and immunoinformatics approaches.

The major problem in cases of chronic hepatitis B (CHB) is the failure of the patient's immune response to eliminate the covalently closed circular DNA (cccDNA) minichromosome of hepatitis B virus (HBV). Epigenetic regulation involving the HBV core protein (HBc) and HBV X protein (HBx) influences the transcription and stability of the cccDNA minichromosome. The HBc and/or HBx-based therapeutic vaccines that have been developed cannot accommodate differences between HBV genotypes. This research aims to design a therapeutic vaccine candidate based on the multi-epitope of HBc and HBx using revers

[Study on the clonal characteristics of idiopathic thrombocytopenic purpura-review].

The autoantigenic epitopes carried by the platelet glycoprotein in patients with idiopathic thrombocytopenic purpura (ITP) have been observed to localize on the specific regions of glycoprotein. There is a limited number of autoantigenic epitopes on the respective glycoproteins. The clonal B-cell and/or T cell expansion have been detected in some patients with ITP, and the autoantibodies is clonally derived. This research is of clinical importance since identification of clonally derived autoantibodies not only may help to discover the pathogenesis of ITP but also lead to less toxic and more s

Determining if T cell antigens are naturally processed and presented on HLA class I molecules.

BACKGROUND: Determining T cell responses to naturally processed and presented antigens is a critical immune correlate to determine efficacy of an investigational immunotherapeutic in clinical trials. In most cases, minimal epitopes and HLA restriction elements are unknown. RESULTS: Here, we detail the experimental use of ex vivo expanded autologous B cells as antigen presenting cells to overcome the limitation of unknown HLA restriction, and the use of electroporated full length mRNA encoding full length parental proteins to ensure that any observed T cell responses are specific for antigens

Coordinate regulation of complex T cell populations responding to bacterial infection.

Bacterial infections activate complex T cell populations that differ in size and antigen specificity. We used tetramerized MHC class I molecules complexed with Listeria monocytogenes-derived epitopes to characterize four distinct CD8+ T lymphocyte populations during bacterial infection. Surprisingly, T cell populations differing in antigen specificity expand, contract, and enter the T cell memory compartment synchronously. Because the four L. monocytogenes epitopes are presented with different efficiencies and have distinct stabilities in infected cells, our findings suggest that these factors

Epitope-Based Vaccine Target Screening against Highly Pathogenic MERS-CoV: An In Silico Approach Applied to Emerging Infectious Diseases.

Middle East respiratory syndrome coronavirus (MERS-CoV) with pandemic potential is a major worldwide threat to public health. However, vaccine development for this pathogen lags behind as immunity associated with protection is currently largely unknown. In this study, an immunoinformatics-driven genome-wide screening strategy of vaccine targets was performed to thoroughly screen the vital and effective dominant immunogens against MERS-CoV. Conservancy and population coverage analysis of the epitopes were done by the Immune Epitope Database. The results showed that the nucleocapsid (N) protein

Antigen specificity of CD4 T cell response in the central nervous system of mice infected with mouse hepatitis virus.

Previously, we showed that the transmembrane (M) and surface (S) glycoproteins were recognized by splenic CD4 T lymphocytes harvested from mice infected intraperitoneally with mouse hepatitis virus, strain JHM (MHV-JHM), whereas only the S protein was recognized by splenocytes derived from mice with MHV-induced chronic demyelination. From these results, it could not be determined which proteins were recognized by T cells localized in the infected central nervous system (CNS). Herein, we show that CD4 T cells responding to both the M and S proteins can be detected in the CNS of mice with either

Toxoplasma gondii: Vaccination with a DNA vaccine encoding T- and B-cell epitopes of SAG1, GRA2, GRA7 and ROP16 elicits protection against acute toxoplasmosis in mice.

Toxoplasma gondii (T. gondii) is an obligate, intracellular, protozoan parasite that infects large variety of warm-blooded animals including humans, livestock, and marine mammals, and causes the disease toxoplasmosis. Although T. gondii infection rates differ significantly from country to country, it still has a high morbidity and mortality. In these circumstances, developing an effective vaccine against T. gondii is urgently needed for preventing and treating toxoplasmosis. The aim of this study was to construct a multi-epitopes DNA vaccine and evaluate the immune protective efficacy against

Developing a multiepitope vaccine for the prevention of SARS-CoV-2 and monkeypox virus co-infection: A reverse vaccinology analysis.

BACKGROUND: Severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) and monkeypox virus (MPXV) severely threaten human health; however, currently, no vaccine can prevent a co-infection with both viruses. METHODS: Five antigens were selected to predict dominant T and B cell epitopes screened for immunogenicity, antigenicity, toxicity, and sensitization. After screening, all antigens joined in the construction of a novel multiepitope vaccine. The physicochemical and immunological characteristics, and secondary and tertiary structures of the vaccine were predicted and analyzed using bio- an

Hierarchies of antigen-specific cytotoxic T-cell responses.

Studies carried out using either mice or humans have shown that cytotoxic T-lymphocyte (CTL) responses to many different pathogenic organisms often comprise CTL specific for multiple class I-restricted peptide epitopes. Differences in the magnitude of epitope-specific CTL responses appear to arise mainly from differences in the expression level of the corresponding class I/peptide complex on the surface of the antigen-presenting cell. The size of the CTL response may be limited by the frequency and possibly by the affinity of specific CTL precursors in the naive T-cell pool. Thus, both the eff

Recognition of adult T-cell leukemia/lymphoma cells by CD4+ helper T lymphocytes specific for human T-cell leukemia virus type I envelope protein.

PURPOSE: Human T-cell leukemia virus type I (HTLV-I) can cause an adult T-cell leukemia/lymphoma (ATLL). Because ATLL is a life-threatening lymphoproliferative disorder and is resistant to chemotherapy, the establishment and enhancement of T-cell immunity to HTLV-I through the development of therapeutic vaccines could be of value. Thus, the identification of HTLV-I epitopes for both CD8(+) and CD4(+) T cells should facilitate the development of effective vaccines. Although numerous HTLV-I epitopes for CTLs have been identified, few epitopes recognized by CD4(+) helper T cells against this viru

Design and immunoinformatics characterization of a novel multi-epitope vaccine rPRRSV-35N targeting NADC30-like and NADC34-like porcine reproductive and respiratory syndrome virus variants.

BACKGROUND: Porcine reproductive and respiratory syndrome (PRRS), caused by the genetically diverse porcine reproductive and respiratory syndrome virus (PRRSV), remains a major economic burden to the global swine industry. Current commercial vaccines, including modified live virus (MLV) and inactivated vaccines, have shown limited effectiveness against newly emerging PRRSV variants, particularly NADC30-like and NADC34-like strains. There is therefore a need for improved vaccine strategies targeting these circulating strains. In this study, we used immunoinformatics approaches to design a multi

An Immunoinformatic Approach for a Multi-Epitope Vaccine Against Toxoplasma gondii.

Infection with the intracellular apicomplexan parasite Toxoplasma gondii causes severe and often fatal clinical outcomes worldwide, especially in patients with immunodeficiency, diabetes and in pregnant women and infants. Despite approximately one-third of the global population being infected with T. gondii, there is currently no effective vaccine available for humans. The objective was to develop a potential vaccine candidate for T. gondii, which would incorporate the B- and T-lymphocyte epitopes derived from three immunogenic antigens of the parasite. Initially, the immunodominant epitopes p

Endogenous presentation of CD8+ T cell epitopes from Epstein-Barr virus-encoded nuclear antigen 1.

Epstein-Barr virus (EBV)-encoded nuclear antigen (EBNA)1 is thought to escape cytotoxic T lymphocyte (CTL) recognition through either self-inhibition of synthesis or by blockade of proteasomal degradation by the glycine-alanine repeat (GAr) domain. Here we show that EBNA1 has a remarkably varied cell type-dependent stability. However, these different degradation rates do not correspond to the level of major histocompatibility complex class I-restricted presentation of EBNA1 epitopes. In spite of the highly stable expression of EBNA1 in B cells, CTL epitopes derived from this protein are effici
