L-asparaginase, which hydrolyzes asparagine into aspartic acid and ammonia, is frequently used to treat acute lymphoblastic leukaemia in children. When combined with other chemotherapy drugs, the event-free survival rate is 90%. Due to immunogenicity and drug resistance, however, not all patients benefit from it, restricting the use of L-asparaginase therapy in other haematological cancers. To solve the problem of immunogenicity, several L-ASNase variants have emerged, such as Erwinia-ASNase and PEG-ASNase. However, even when Erwinia-ASNase is used as a substitute for E. coli-ASNase or PEG-ASN

The phylogenetic relationships of the type strains of 38 species from 15 genera of the family Enterobacteriaceae were investigated by comparative 16S rDNA analysis. Several sequences of strains from the genera Citrobacter, Erwinia, Pantoea, Proteus, Rahnella and Serratia, analysed in this study, have been analysed previously. However, as the sequences of this study differ slightly from the published ones, they were included in the analysis. Of the 23 enterobacterial genera included in an overview dendrogram of relatedness, members of the genera Xenorhabdus, Photorhabdus, Proteus and Plesiomona

Three out of 21 patients treated at the Children's Hospital of Eastern Switzerland for ALL or NHL with the respective BFM-90 protocols experienced thrombotic complications during Erwinia L-asparaginase therapy. We therefore investigated the development of the haemostatic imbalance in six children during the induction phase of both protocols. In the average, elevated thrombin generation was found simultaneously to an increase in fibrinogen, cross-linked fibrin degradation products and platelet counts between protocol days 28 to 35. This period thus seems to harbour an increased potential of hyp

In May 2023, characteristic symptoms of fire blight, suspected to be caused by Erwinia amylovora, were observed infected wild Cotoneaster plants in the secondary forest along the lower reaches of the Jilgilang River, Xinjiang, China. Symptomatic tissues were collected, and bacterial isolation and purification were performed. The bacterium was identified through morphological characterization of the colonies, pathogenicity assays, tobacco hypersensitivity tests, and specific polymerase chain reaction (PCR) amplification, followed by sequence analysis. The isolated bacterium exhibited morphologi

With the increasing importance of alternative pesticides for the control of bacterial pathogens in agricultural and clinical settings, the use of bacteriophage viruses (phages) to reduce bacterial growth and prevent disease is gaining in popularity. Phages have been shown to be highly effective in killing bacterial cells both in vitro and across plant and animal host systems, although many questions remain about the predictability of their success across more realistic ecological conditions and in light of natural strain variation of pathogens. Furthermore, as phage application becomes more co

BACKGROUND: The olive fruit fly Bactrocera oleae (Rossi) (OLF) is a major agricultural pest, whose control primarily relies on the use of chemical insecticides. Therefore, development of sustainable control strategies is highly desirable. The primary endosymbiotic bacterium of OLF, 'Candidatus Erwinia dacicola', is essential for successful larval development in unripe olive fruits. Therefore, targeting this endosymbiont with antimicrobial compounds may result in OLF fitness reduction and may exert control on natural populations of OLF.
RESULTS: Here, we evaluate the impact of compounds with a

L-asparaginase (L-ASP) is one of the cornerstones of the treatment of acute lymphoblastic leukemia and non-Hodgkin lymphoma. It is an enzyme of bacterial origin capable of transforming L-asparagine to aspartic acid. The extracellular depletion of L-asparagine inhibits protein synthesis in lymphoblasts, inducing their apoptosis. Numerous studies have demonstrated that treatment with L-ASP improves survival of patients, but there are clear differences in the characteristics of the three currently available formulations. This article reviews the dosage, activity and side effects of the two L-ASP

Asparaginase is an established chemotherapeutic agent for acute lymphoblastic leukemia (ALL) and lymphoblastic lymphoma (LL) treatment. Pegylated asparaginase (PEG-Asp) is first-line but may lead to allergic reactions in ∼30% of recipients, leading to substitution by Erwinia -derived asparaginase. Studies demonstrate mixed results on the impact of universal premedication on the incidence of infusion reactions. Our center adopted universal premedication with H1 and H2 antagonists and nadir serum asparaginase activity (SAA) level monitoring in 2019. We assessed the impact of universal premedicat

The olive fly Bactrocera oleae (Rossi) is one of the most destructive olive pests, threatening olive and olive oil production worldwide. Traditionally managed with synthetic insecticides, its control now requires alternative strategies due to regulatory restrictions. The disruption of the obligate association between the pest and its primary symbiont Candidatus Erwinia dacicola, Capuzzo et al. (2005) has been reported following the application of a Zn-Cu-citric acid bio-complex compound (Dentamet) to olives infested with newly laid eggs. In this study, the effects of Dentamet exposure on the c

Most damage associated with fire blight disease (Erwinia amylovora) is due to blossom blight, so less research has been dedicated to shoot infections. Current control of shoot trauma blight, associated with wind or hail damage, relies on copper and antibiotics, yet sustainable alternatives are needed. This study evaluated the efficacy of chitosan, rhamnolipids, and caprylic acid against shoot trauma blight. We demonstrated the antimicrobial activity of all three compounds in vitro and optimized a host inoculation method mimicking high-infection pressure conditions. Caprylic acid, with high ant

Phage populations of isolates from quince and pear affected with fire blight disease were studied using electron microscopy, restriction analysis and both agarose gel electrophoresis of particles and host range scoping method. The isolate from quince (pMA1) comprises at least three phage populations and two phage variants that can be detected on different bacterial indicators. After titration of this isolate on Erwinia amylovora the bacteriophage KEY of B1 morphotype with the genome size of 82.4 kb was identified. The isolate pMA1 also includes a unique phage population 4*, which can be identi

The treatment of the acute lymphoblastic leukemia in childhood includes frequent administration of L-asparaginase by intravenous route. L-asparaginase is an enzyme produced by E. coli and Erwinia chrysanthemi strains. Adverse reactions produced by L-asparaginase are numerous, and pancreatitis is being the most severe. Children with the acute lymphoblastic leukemia were followed up for 2 years. Hyperglycaemia and glycosuria were noted in 10% of them resulting in L-asparaginase cessation or replacement by less toxic agents. The acute pancreatitis was produced in 8% of the patients, and was treat

The reuse of water using effluents containing antibiotics from anthropogenic activities has been mainly linked to the development of antibiotic resistance. However, we report that the development of bacterial tolerance promotes plant growth. In the present study, we aimed to evaluate the efficiency of inoculation of a new antibiotic-degrading bacterium, Erwinia strain S9, in augmenting the tolerance of pea (Pisum sativum L.) plants to tetracycline (TET) (10 and 20 mg/L). Physiological parameters such as tissue elongation and biomass, as well as relative water content, were remarkably lower in

Recent studies suggest that besides the long-known farmer's lung antigen sources Saccharopolyspora rectivirgula (Micropolyspora faeni), Thermoactinomyces vulgaris, and Aspergillus fumigatus, additionally the mold Absidia (Lichtheimia) corymbifera as well as the bacteria Erwinia herbicola (Pantoea agglomerans) and Streptomyces albus may cause farmer's lung in Germany. In this study the sera of 64 farmers with a suspicion of farmer's lung were examined for the following further antigens: Wallemia sebi, Cladosporium herbarum, Aspergillus versicolor, and Eurotium amstelodami. Our results indicate

BACKGROUND: Although the most common path of infection for fire blight, a severe bacterial disease on apple, is via host plant flowers, quantitative trait loci (QTLs) for fire blight resistance to date have exclusively been mapped following shoot inoculation. It is not known whether the same mechanism underlies flower and shoot resistance.
RESULTS: We report the detection of a fire blight resistance QTL following independent artificial inoculation of flowers and shoots on two F1 segregating populations derived from crossing resistant Malus ×robusta 5 (Mr5) with susceptible 'Idared' and 'Royal

Glucocorticoids alone made Fibrinogen drop to 148 mg/dl, AT III and Protein C rise to 136% or even 179% respectively. After day 12, immediately following the start of therapy with Coli-Asparaginase (ASP), fibrinogen continued to drop to reach its lowest average value of 46 mg/dl on day 24. Anticoagulant factors like plasminogen (lowest average value: 36%), AT III (47%) and Protein C (93%) dropped abruptly. These alterations were reversed afte

The behaviour of 12 strains of gram-negative bacteria was analysed in media having a low content of nutrients. Sterile tap water, de-ionized water, aqua bidestillata and mineral salt solution were used. It was found that Serratia marcescens, Pseudomonas aeruginosa, Enterobacter, Klebsiella and Citrobacter are those with the most frequently observed growth. In the mineral salt solution survived also Providencia, E. coli, Erwinia, Hafnia, and Alkaligenes and reached max. colony numbers of some 100000/ml. When the salts used were added stepwise potassium phosphate and especially magnesium sulfate

Severe hyperlipidaemia with asparaginase therapy is rare. We report six cases, four of which developed significant problems with severe hyperlipidaemia during induction therapy for ALL and lymphoblastic lymphoma. The median triglyceride level was 22.3 mmol/L and the median cholesterol level was 12.3 mmol/L. None of the patients showed signs or symptoms of pancreatitis. Three children were re-exposed with Peg asparaginase, and one with Erwinia asparaginase, without recurrence of hyperlipidaemia or other symptoms. These cases highlight the dilemma in managing such rare cases of symptomatic hyper
