Impact of pharmaceutical excipients on in vitro association of saquinavir to chylomicrons.

This study was performed to investigate the impact of pharmaceutical excipients commonly used for lymphatic transport on in vitro drug association with chylomicrons (CM). A CM association study was conducted using saquinavir solubilized in four different pharmaceutical excipients. We observed a linear relationship between saquinavir solubility and drug association, suggesting that the solubility of saquinavir in excipients is a key determinant for successful lymphatic delivery. Broadly, these results suggest that excipients with good solubilization properties may be advantageous for enhancing

Evaluation of dibutyrylchitin as new excipient for sustained drug release.

Dibutyrylchitin (DBC), a lipophilic chitin diester, has been synthesized from chitin and butyric anhydride with methanesulfonic acid as catalyst. Exhaustive esterification of free alcoholic groups of chitin was assessed by FT-IR and (1)H-NMR spectroscopy. High degree of alkyl substitution allowed DBC to acquire an almost completely lipophilic character. Tablets of paracetamol and metformin employing DBC as major excipient, in comparison with starch, microcrystalline cellulose, lactose and polyvinylpyrrolidone, were prepared and rates of drug release were checked by dissolution test assays. DBC

The role of excipients and package components in the photostability of liquid formulations.

A phenyl ether-based drug substance exhibits photochemical degradation in citrate buffers with both ultraviolet (300-450 nm range) and visible light (380-700 nm range) exposure, even though the drug molecule itself is non-light absorbing at wavelengths > 300 nm. The major contributors to the observed photosensitivity are the citrate buffer, parts per billion (ppb) levels of iron, oxygen, and light exposure level. Although a primary phenol photodegradate is generated, there are at least eight other species formed as well. The molecular weights and abundance of these species suggest that the pro

The interactions of water with cellulose- and starch-derived pharmaceutical excipients.

Water associated with polymeric pharmaceutical excipients derived from cellulose and starch can have a profound effect on the properties of the excipient and on the other ingredients making up a solid dosage form. Important questions which need to be addressed include How much water will be sorbed or desorbed at various relative humidities and temperatures? and What is the thermodynamic state of water associated with the solid as a function of moisture content? A critical review of the literature is presented to demonstrate the most likely answers to these questions. It appears that water exis

Analysis of nitrite in pharmaceutical excipients: A multi‑laboratory comparative study.

Rigorous control of N-nitrosamine impurities within drug products is essential due to their potential carcinogenicity. Nitrosamines are often formed through the reaction of vulnerable amines present in the drug substance with nitrite from excipients, where nitrite is often the limiting reagent. Therefore, it is important to understand the level of nitrite in excipients to assess the risk of nitrosamine formation in a drug product. For this purpose, the Lhasa Nitrites database was created to collate and curate nitrite data in excipients, reagents and solvents contributed by member companies wor

Ketoproof? A systematic classification of pharmaceutical excipients for ketogenic diet therapy.

INTRODUCTION: Ketogenic diet therapy (KDT) is an evidence-based treatment for refractory epilepsy, particularly in children. Strict adherence to carbohydrate restriction is essential to maintain ketosis, yet many medications contain excipients that can disrupt this balance. Pharmacists play a crucial role in evaluating and adjusting medication regimens for patients on KDT, but limited transparency regarding excipient content often makes this process inefficient and inconsistent. Although some regional databases exist, no systematic and comprehensive resources have been developed for broader us

Assessing the aquatic biodegradation potential of polymeric excipients for pharmaceutical formulation.

Polymeric excipients (PEx) are essential in drug formulation but raise environmental concerns upon wastewater release post-administration due to their potential detrimental effects to life-histories of freshwater vertebrates and invertebrates. Ten pharmaceutical polymeric compounds were assessed in a stepwise environmental biodegradation assessment according to standard OECD 301 guidelines to thoroughly evaluate biodegradability of these compounds. Polyvinyl alcohol (PVA), polyethylene glycol (PEG), chitosan, maize starch, and sodium starch glycolate (SSG) were found to be 'readily biodegradab

Investigation of water vapour sorption mechanism of starch-based pharmaceutical excipients.

Starch-based excipients are commonly used in oral solid dosage forms. The effect of particle size and pregelatinisation level of starch-based excipients on their water absorption behaviour have been evaluated. The results showed that starch-based excipients have type ii isotherms, indicating that the principal mechanism of sorption is the formation of monolayer coverage and multilayer water molecules (10-80 RH %). It was found that the particle size of starch-based excipients did not have any influence on the rate of water sorption, whereas the level of pregelatinisation changed the kinetics o

Biocompatibility investigation of different pharmaceutical excipients used in liquid dosage forms.

Aqueous pharmaceutical solutions provide prosperous living conditions for microbiological agents. In order to eliminate these microbes, we use preservatives which can harm human cells as well. Their cytotoxicity is concentration-dependent and the aim of our study was to find how other pharmaceutical excipients modify the cytotoxic attributes of preservatives. We tested the following compounds: methylparaben, benzalkonium chloride, polysorbate 20, Labrasol® and hydroxyethyl cellulose. The MTT tests indicated that surfactants increase the cytotoxicity while polymers may decrease it in some cases

Applications of various analytical techniques in quality control of pharmaceutical excipients.

Although excipients are pharmacologically inert substances, still they are integral and significant ingredients of a pharmaceutical formulation. Their amount and concentration level along with the active pharmaceutical ingredients is of utmost importance to decide the pharmacokinetics and stability of a dosage form. It is becoming progressively more important to illustrate the purity of excipients throughout the development and manufacture of pharmaceutical products as the safety and efficacy of the pharmaceutical formulation is largely affected by the quality of excipients. Therefore it is es

Inhibitory Effects of Commonly Used Excipients on P-Glycoprotein in Vitro.

Pharmaceutical excipients are no longer considered inert and have been shown to influence the activity of metabolic enzymes and transporters, resulting in altered pharmacokinetics of substrate drugs. In this study, the effect of 25 excipients commonly used in drug formulations were investigated for their effect on P-glycoprotein (P-gp) activity. The effect of excipients on P-gp were assessed by measuring the change in the cellular accumulation of a P-gp substrate, digoxin, in MDCK-MDR1 (Madin Darby canine kidney transfected with multidrug resistance 1 gene) cells. The cells were exposed to low

Effect of colloidal silica on rheological properties of common pharmaceutical excipients.

In pharmaceutical industry, the use of lubricants is mostly based on historical experiences or trial and error methods even these days. It may be demanding in terms of the material consumption and may result in sub-optimal drug composition. Powder rheology enables more accurate monitoring of the flow properties and because the measurements need only a small sample it is perfectly suitable for the rare or expensive substances. In this work, rheological properties of four common excipients (pregelatinized maize starch, microcrystalline cellulose, croscarmellose sodium and magnesium stearate) wer

Applications of Polymers as Pharmaceutical Excipients in Solid Oral Dosage Forms.

Over the last few decades, polymers have been extensively used as pharmaceutical excipients in drug delivery systems. Pharmaceutical polymers evolved from being simply used as gelatin shells comprising capsule to offering great formulation advantages including enabling controlled/slow release and specific targeting of drugs to the site(s) of action (the "magic bullets" concept), hence hold a significant clinical promise. Oral administration of solid dosage forms (e.g., tablets and capsules) is the most common and convenient route of drug administration. When formulating challenging molecules i

Recently Investigated Natural Gums and Mucilages as Pharmaceutical Excipients: An Overview.

Due to advances in drug delivery technology, currently, excipients are included in novel dosage forms to fulfil specific functions and in some cases they directly or indirectly influence the extent and/or rate of drug release and drug absorption. Recent trends towards use of plant based and natural products demand the replacement of synthetic additives with natural ones. Today, the whole world is increasingly interested in natural drugs and excipients. These natural materials have many advantages over synthetic ones as they are chemically inert, nontoxic, less expensive, biodegradable, and wid

Effects of pharmaceutical excipients on membrane permeability in rat small intestine.

Pharmaceutical excipients should not disturb the effects of drug therapy. In recent years, however, it has been reported that excipients induce some changes to the tight junction (TJ) and P-glycoprotein (P-gp), which can affect drug disposition. In this study, we examined the effects of 20 common pharmaceutical excipients from different classes on mucosal membrane and the differences of such effects among regions of the small intestine. We used the in vitro sac method in rat jejunum and ileum to study the effects of excipients on the membrane permeation of 5(6)-carboxyfluorescein (5-CF). 5-CF

Solid state compatibility studies with tablet excipients using non thermal methods.

Compatibility between two new active pharmaceutical ingredients (API) and several pharmaceutical excipients used in solid formulations has been investigated by FT-IR and HPLC following storage under two different conditions. Compatibility was investigated by storage at isothermal stress conditions for (i) 3days and subsequently analysed by FT-IR and (ii) 12weeks of storage and analysis by HPLC. For the majority of the examined excipients a large degradation measured by HPLC after 12weeks storage was also detected by FT-IR following storage at isothermal stress conditions for 3days, i.e. there

Regulations on excipients used in 3D printing of pediatric oral forms.

A promising solution to customize oral drug formulations for the pediatric population has been found in the use of 3D printing, in particular Fused Deposition Modeling (FDM) and Semi-Solid Extrusion (SSE). Although formulation development is currently limited to research studies, the rapid advances in 3D printing warn of the need for regulation. Indeed, even if the developed formulations include pharmaceutical excipients used to produce traditional oral forms such as tablets, the quantities of excipients used must be adapted to the process. Therefore, the aim of this literature review is to pr
