Importance of extracellular vesicle secretion at the blood-cerebrospinal fluid interface in the pathogenesis of Alzheimer's disease.

Increasing evidence indicates that extracellular vesicles (EVs) play an important role in the pathogenesis of Alzheimer's disease (AD). We previously reported that the blood-cerebrospinal fluid (CSF) interface, formed by the choroid plexus epithelial (CPE) cells, releases an increased amount of EVs into the CSF in response to peripheral inflammation. Here, we studied the importance of CP-mediated EV release in AD pathogenesis. We observed increased EV levels in the CSF of young transgenic APP/PS1 mice which correlated with high amyloid beta (Aβ) CSF levels at this age. The intracerebroventricu

Local Echo-Free Space in a Limb with Lymphedema Represents Extracellular Fluid in the Entire Limb.

BACKGROUND: We previously reported that local subcutaneous echo-free space (SEFS) in the leg was stirringly correlated with extracellular fluid (ECF) status in the same part of the leg when assessed using bioelectrical impedance analysis (BIA). In this study, we examined whether local SEFS represents the ECF status in the entire limb. METHODS AND RESULTS: In 51 legs and 40 arms with lymphedema (LE), subcutaneous tissue ultrasonography was performed to determine SEFS severity (range, 0-2). BIA was also performed to calculate the L-Dex® score (LDS), which is a BIA-derived parameter to represent

Chronic electrographic seizure reduces glutamine and elevates glutamate in the extracellular fluid of rat brain.

Effects of spontaneous seizures on extracellular glutamate and glutamine were studied in the kainate-induced rat model of epilepsy in the chronic phase. Extracellular fluid from the CA1-CA3 regions of the hippocampus was collected with a 2-mm microdialysis probe every 2 min for 5h. EEG seizures with no or mild behavioral components caused 2- to 6-fold elevation of extracellular glutamate. Concomitantly, extracellular glutamine decreased at t=5h to 48% of the initial value (n=6). The changes in extracellular glutamate and glutamine correlated with the frequency and magnitude of seizure activity

Drainage of brain extracellular fluid into blood and deep cervical lymph and its immunological significance.

Cerebral extracellular fluids drain from brain to blood across the arachnoid villi and to lymph along certain cranial nerves (primarily olfactory) and spinal nerve root ganglia. Quantification of the connection to lymph in rabbit, cat and sheep, using radiolabelled albumin as a marker of flow, indicates that a minimum of 14 to 47% of protein injected into different regions of brain or cerebrospinal fluid passes through lymph. The magnitude of the outflow to lymph is at variance with the general assumption that the absence of conventional lymphatics from the brain interrupts the afferent arm of

High-performance liquid chromatographic methods for base and nucleoside analysis in extracellular fluids and in cells.

High-performance liquid chromatography based methods for the study of the metabolism of purine and pyrimidine bases and nucleosides have been developed. These methods, using 200--50 microliter samples of extracellular fluids and employing isocratic separations, can measure a wide range of compounds. Hypoxanthine, xanthine and uridine concentrations in plasma from normal men are relatively stable. Species differences have been detected: concentrations of cytidine are higher in rat and mouse serum than in man, since the concentrations of uridine are similar; purine/pyrimidine ratios may be diffe

Molecule-Level Interpretable SERS Diagnosis of Prostate Cancer via Prostatic Fluid Metabolites and Extracellular Vesicles.

Prostate cancer (PCa) remains a major global health burden, yet current screening tools often lead to overdiagnosis due to low specificity, highlighting the urgent need for more precise diagnostic approaches. Prostatic fluid (PSF) represents a promising but underexplored biofluid with exceptional diagnostic potential due to its direct contact with the PCa microenvironment. Here, we employed molecule-level interpretable surface-enhanced Raman spectroscopy (SERS) to comprehensively investigate PCa-associated alterations in two PSF components including metabolites and small extracellular vesicles

Uterine fluid extracellular vesicle miR-223-3p as endometrial receptivity biomarker and regulator in implantation.

Endometrial receptivity is a pivotal determinant of successful embryo implantation, yet its regulation by uterine fluid (UF) extracellular vesicles (EVs) remains poorly understood. This study aimed to identify key UF EV miRNAs during the endometrial receptive period and observe the role in implantation. UF EVs were isolated from the same in vitro fertilization/intracytoplasmic sperm injection patients at two time points: the prereceptive (LH + 3/progesterone [P] + 1) and receptive (LH + 7/P + 5). MiRNA sequencing and validation identified miR-223-3p as the key miRNA. Bioinformatic analysis pre

Seminal fluid extracellular vesicles restore human sperm osmoregulation after cystic fibrosis transmembrane conductance regulator inhibition.

Cystic fibrosis transmembrane conductance regulator (CFTR) is an anion channel that promotes the exchange of chloride (Cl-) and bicarbonate (HCO3-), modulating the balance of ions and water in various tissues. CFTR is also expressed in the equatorial region of the spermatozoa's head. However, the modulation of ionic and water transport via CFTR in spermatozoa needs further investigation. To better understand how CFTR participates in the molecular mechanisms behind spermatozoa modulation of water transport, and its impact on sperm physiology, CFTR function of human spermatozoa was inhibited wit

Proteome profiling of cerebrospinal fluid-derived extracellular vesicles reveals potential biomarkers for drug-resistant epilepsy.

BACKGROUND: Epilepsy is one of the most common neurological disorders in humans and in dogs. Treatment currently focuses on alleviating symptoms, and a wide range of anti-seizure medications (ASMs) is available. Still, over one-third of patients have an inadequate response to ASM. The proteome of cerebrospinal fluid (CSF)-derived extracellular vesicles (EVs) offers a potential source of biomarkers for drug-resistant epilepsy (DRE). METHODS: We utilised a spontaneous canine epilepsy model to study the proteomic content of CSF-derived EVs as a source of biomarkers for DRE. We included 37 drug-n

Automated morphological classification and quantification of cerebrospinal fluid extracellular vesicles via AFM and machine learning.

Morphology of extracellular vesicles (EVs) from cerebrospinal fluid is an important property that could uncover brain-related conditions. However, native morphology could get distorted during imaging, such as with atomic force microscopy (AFM) in air, which enables relatively simple visualisation and automated morphology assessment. Therefore, we compared 24 different preparation methods for the same sample of EVs according to the resulting size, height, aspect ratio and shape distributions obtained from the AFM images. We defined 5 different shape categories (round, flat, concave, single-lobe

Normozoospermic seminal fluid extracellular vesicles could ameliorate adverse effects of freeze-thaw in oligoasthenoteratospermia men.

BACKGROUND: Cryopreservation can lead to sperm damage, which is why incorporating extracellular vesicles derived from normal semen can help protect against the damage caused by freezing. MATERIALS AND METHODS: Extracellular vesicles were obtained from normozoospermic semen samples (n = 15), and their effects on the cryopreservation process were studied in three groups: the control group (A, n = 6), the extracellular vesicles treated before freezing group (B, n = 6), and the extracellular vesicles treated after freezing group (C, n = 6). Exceptional stability of a Gd-free extracellular fluid MRI contrast agent based on manganese porphyrin.

Metal-ligand stability plays a central role in medicinal bioinorganic chemistry, as it impacts not only the efficacy but also the safety profiles of metal-based therapeutic and diagnostic agents. Gadolinium-based contrast agents (GBCAs) have been widely applied in clinical magnetic resonance imaging (MRI) scans for decades to enhance the detection of otherwise hardly visible diseases. However, evidence of metal dissociation from Gd chelates in vivo have raised safety concerns and led to the withdrawals of several clinically approved GBCAs in recent years. Here we report an exceptionally high s

A Method to Collect Cerebrospinal Fluid from Mouse Cisterna Magna to Determine Extracellular Tau Levels.

Despite being a cytoplasmic protein abundant in neurons, tau is detectable in various extracellular fluids. In addition to being passively released from dying/degenerating neurons, tau is also actively released from living neurons in a neuronal activity-dependent mechanism. In vivo, tau released from neurons first appears in brain interstitial fluid (ISF) and subsequently drains into cerebrospinal fluid (CSF) by glymphatic system. Changes in CSF tau levels alter during the course of AD pathogenesis and are considered to predict the disease-progression of AD. A method to collect CSF from variou

Extracellular fluid excess linked to reduced choroidal vascularity index in patients with chronic kidney disease.

Extracellular fluid (ECF) excess is common in patients with chronic kidney disease (CKD). This study (involving 284 patients with CKD) explored the association between choroidal vascularity index (CVI) and ECF excess. We categorised patients into three groups based on extracellular water/total body water: normal, mildly overhydrated, and severely overhydrated. The more severe ECF status was associated with a lower CVI after adjustment (B = - 0.902, p = 0.001). In non-diabetic patients, both vascular luminal (LA, p < 0.001) and stromal areas (SA, p = 0.003) were significantly reduced in patient

Amniotic fluid-derived small extracellular vesicles for predicting postnatal severe outcome of congenital diaphragmatic hernia.

Congenital diaphragmatic hernia (CDH) is a life-threatening condition with high morbidity and mortality rates. The survival rate of neonates with severe CDH is reportedly only 10%-15%. However, prenatal prediction of severe cases is difficult, and the discovery of new predictive markers is an urgent issue. In this study, we focused on microRNAs (miRNAs) in amniotic fluid-derived small EVs (AF-sEVs). We identified four miRNAs (hsa-miR-127-3p, hsa-miR-363-3p, hsa-miR-493-5p, and hsa-miR-615-3p) with AUC > 0.8 to classify good prognosis group and poor prognosis group in human study. The AUC for h

Visualizing neutrophil extracellular traps in septic equine synovial and peritoneal fluid samples using immunofluorescence microscopy.

Septic synovitis and peritonitis are routinely diagnosed in horses based on clinical examination findings and laboratory assessment of synoviocentesis and abdominocentesis samples, respectively. Diagnosis is difficult in some cases because of an overlap in laboratory results for septic and non-septic inflammation. Neutrophil extracellular trap (NET) formation is part of the innate immune response against pathogens. Identifying and quantifying NETs, which have not been explored in clinical samples from horses with septic synovitis and peritonitis, to our knowledge, may be helpful in detecting i

Studies on oxygen and extracellular fluid restrictions in cultured heart cells: high energy phosphate metabolism.

Although cultured heart cells are increasingly used for the study of cardiac metabolism, relatively little is known about their energy turnover. We studied the effects of anoxia with simultaneous restrictions of the volume of the extracellular medium ("ischaemia") on high energy phosphate catabolism in cells from neonatal rat ventricles, cultured for 5 days. The cells were incubated for up to 4 h in Ham-F10 medium either in the presence or in the absence of glucose. High energy phosphates in cell extracts and AMP catabolites in the incubation medium were measured by high pressure liquid chroma

Follicular-fluid extracellular vesicles support energy metabolism of bovine oocytes, improving blastocyst development and quality†.

Extracellular vesicles (EVs) from follicular fluid (FF) seem to play a significant role in communication within ovarian follicles in several species. The present study aimed to examine the supporting effect of FF-derived small EVs (FF-sEVs) during in vitro maturation (IVM) of bovine cumulus-oocyte complexes (COCs) under conditions of disturbed energy metabolism. Bovine COCs were matured in vitro with inhibitors targeting lipid metabolism (etomoxir) or glucose metabolism (iodoacetate combined with dehydroepiandrosterone), in the presence or absence of FF-sEVs. Following maturation, oocytes and
