Reversible Charge-Mediated Capture and Release of Extracellular Vesicles Using Giant Unilamellar Vesicles.

Extracellular vesicles (EVs) play crucial roles in intercellular communication and represent promising therapeutic vehicles, yet current isolation and manipulation methods often irreversibly alter their properties. This study presents a charge-mediated approach for the reversible interaction and release of EVs using Giant Unilamellar Vesicles (GUVs) exploiting tailored surface charges. Negatively charged EVs can be selectively bound through electrostatic interactions with positively charged GUVs, forming stable aggregates. Critically, these interactions are reversible upon the introduction of

Gut microbiota-derived extracellular vesicles in Parkinson's disease.

The gut microbiota contributes to the pathophysiology of Parkinson's disease (PD), but the mechanism is not known. Extracellular vesicles (EVs), membrane-enclosed nanosized particles, contain various biomolecules, cross biological barriers, and reach sterile body compartments. We hypothesized that EVs released by the gut microbiota might influence PD pathophysiology. We isolated gut microbiota-derived EVs from the fecal samples of 20 patients with PD and 19 healthy controls. Patients with PD had higher concentrations of fecal EVs, and the mean particle size of EVs was larger. The 16S ribosomal

Food bioactives and extracellular vesicles: a narrative review.

Extracellular vesicles (EVs) are now recognized as key mediators of intercellular communication, transferring cargos such as proteins, lipids, and nucleic acids between cells and thereby influencing physiological and pathological processes. While the role of EVs has been extensively investigated in physiological and pathophysiological contexts, nutrition has only recently been recognized as an important modulator of endogenous EV biology. Emerging evidence indicates that nutrition and food bioactive compounds can influence EV biology through three distinct mechanisms: (i) modulation of the bio

Transcriptomic and functional analysis of fibroid extracellular vesicles.

Exosomes were isolated from cultures of fibroid explants and matched myometrial explants, and their RNA cargo was analyzed by next-generation sequencing to profile both long and short RNA species. Fibroid-derived exosomes (Fib-EXO) expressed the canonical extracellular vesicle markers CD81 and CD63, and their size distribution (30-200 nm) was consistent with exosomal vesicles. The RNA cargo of Fib-EXO generally reflected that of its tissue of origin, although selective enrichment of specific transcripts, such as piR-1398740 and piR-333378, suggested active loading mechanisms. Long RNA sequenci
