The role of iron, omega-3 Fatty acids, and vitamins in heart failure.

The high prevalence of iron deficiency in heart failure (HF), its easy detection, and its rapid treatment effects with intravenous compounds including, among other things, improved New York Heart Association class, quality of life, and exercise capacity, may offer a major new addition to the treatment of HF. Although more research is required in HF, iron deficiency has been recognized as a disease for over a century and there is no question that its correction is desirable for improving the health and the quality of life of the iron-deficient patient. Iron deficiency with or without an associa

CYP-eicosanoids--a new link between omega-3 fatty acids and cardiac disease?

Fish oil omega-3 fatty acids such as eicosapentaenoic acid (EPA) and docosahexaenoic acid (DHA) protect against arrhythmia and sudden cardiac death by largely unknown mechanisms. Recent in vitro and in vivo studies demonstrate that arachidonic acid (AA) metabolizing cytochrome P450-(CYP) enzymes accept EPA and DHA as efficient alternative substrates. Dietary EPA/DHA supplementation causes a profound shift of the cardiac CYP-eicosanoid profile from AA- to EPA- and DHA-derived epoxy- and hydroxy-metabolites. CYP2J2 and other CYP epoxygenases preferentially epoxidize the ω-3 double bond of EPA an

Greasing the wheels of managing overweight and obesity with omega-3 fatty acids.

The epidemic of overweight and obesity around the world and in the US is a major public health challenge, with 1.5 billion overweight and obese adults worldwide, and 68% of US adults and 31% of US children and adolescents overweight or obese. Obesity leads to serious health consequences, including an increased risk of type 2 diabetes mellitus and heart disease. Current preventive and medical treatments include lifestyle modification, medication, and bariatric surgery in extreme cases; however, they are either not very efficacious or are very expensive. Obesity is a complex condition involving

Omega-3 fatty acids improve postprandial lipaemia in patients with nephrotic range proteinuria.

BACKGROUND: Patients with nephrotic range proteinuria have a marked increase in the risk of cardiovascular disease. Qualitative and quantitative changes in lipids and lipoproteins contribute to this increased risk with an abundance of atherogenic triglyceride (TG) rich apolipoprotein B containing lipoproteins. TG rich lipoproteins predominate postprandially and are associated with increased risk of coronary heart disease (CHD). Omega-3 fatty acids derived from fish oils have been shown to have beneficial effects on lipids and lipoproteins in patients without proteinuria. METHODS: 17 patients

Acne vulgaris, mental health and omega-3 fatty acids: a report of cases.

Acne vulgaris is a common skin condition, one that is associated with significant psychological disability. The psychological impairments in acne include higher rates of depression, anxiety, anger and suicidal thoughts. Despite a paucity of clinical research, patients with skin conditions and/or mental health disorders are frequent consumers of dietary supplements. An overlap may exist between nutrients that potentially have both anti-acne and mood regulating properties; examples include omega-3 fatty acids from fish oil, chromium, zinc and selenium. Here we report on five cases of acne treate

Clinical overview of Omacor: a concentrated formulation of omega-3 polyunsaturated fatty acids.

Omacor (omega-3-acid ethyl esters; Reliant Pharmaceuticals, Inc., Liberty Corner, NJ) is a highly purified, prescription omega-3 fatty acid formulation with high concentrations of eicosapentaenoic acid (EPA) (465 mg) and docosahexaenoic acid (DHA) (375 mg) in each 1-g capsule, along with 4 mg (6 IU) of vitamin E. At a typical dose of 4 capsules/day, Omacor significantly lowers plasma triglyceride levels either as monotherapy or in combination with 3-hydroxy-3-methylglutaryl coenzyme A (HMG-CoA) reductase inhibitors (statins) or fibrates. Omacor also modestly increases plasma levels of low-dens

Dietary omega-3 polyunsaturated fatty acids reduce IFN-gamma receptor expression in mice.

Enrichment of immune cells in vivo or in vitro with omega-3 polyunsaturated fatty acid (n-3 PUFA) has been reported to diminish their response to interferon-y (IFN-gamma). We hypothesized that the n-3 PUFA-induced hyporesponsiveness to IFN-gamma is mediated, in part, by a reduction in the number of IFN-gamma receptors (IFNGR) expressed on the surface of these cells. To test this hypothesis, we fed mice experimental diets containing low or high amounts of n-3 PUFA. Thioglycollate-elicited peritoneal macrophages (PEC) were collected and tested for binding and internalization of [125I]-labeled re

Effect of omega-3 fatty acids on glucose homeostasis: role of free fatty acid receptor 1.

Insulin resistance is a worldwide health problem. This study investigated the acute effects of eicosapentanoic acid (EPA) on glucose homeostasis focusing on the role of free fatty acid receptor 1 (FFAR1) and the chronic effects of fish oil omega-3 fatty acids on insulin resistance. Insulin resistance was induced by feeding mice high-fructose, high-fat diet (HFrHFD) for 16 weeks. In the first part, the acute effects of EPA alone and in combination with GW1100 and DC260126 (FFAR1 blockers) on glucose homeostasis and hepatic phosphatidyl-inositol 4,5-bisphosphate (PIP2) and diacylglycerol (DAG) w

Omega-3 fatty acids in vegetarian and vegan diets: Translating evidence into practice.

Vegetarian and vegan diets are consistently associated with reduced cardiometabolic risk, despite providing minimal amounts of long-chain omega-3 polyunsaturated fatty acids (n-3 PUFAs), key precursors of bioactive lipid mediators, raising questions about n-3 PUFA metabolism and requirements in plant-based populations. This review examines current evidence on the metabolic fate of alpha-linolenic acid (ALA), its conversion to eicosapentaenoic acid (EPA) and docosahexaenoic acid (DHA), and the implications for n-3 PUFA-derived signaling pathways. Although conversion efficiency is limited, it is

Effects of omega-3 fatty acids on skeletal muscle vascular health and metabolism.

PURPOSE OF REVIEW: This review aims to summarise recent findings on the effects of omega-3 fatty acids (FAs) on skeletal muscle vascular and metabolic health. We particularly focus on macrovascular (large blood vessel) outcomes, skeletal muscle microvascular (capillary) blood flow and skeletal muscle health and metabolism. RECENT FINDINGS: Recent meta-analysis and intervention studies suggest that omega-3 FAs improve macrovascular health outcomes in humans by enhancing endothelial function, reducing arterial stiffness and by increasing nitric oxide bioavailability. However, the benefits on ma

[Omega-3 polyunsaturated fatty acids for the treatment of oligoasthenozoospermia: Progress in research].

Oligoasthenozoospermia (OAS) is one of the most common types of male infertility, which, however, still lacks effective treatment. An increasing number of studies have shown the potential therapeutic value of omega-3 polyunsaturated fatty acid (ω-3 PUFA) in the treatment of OAS. This article presents an overview of the studies on the effects of ω-3 PUFA on fatty acid composition and metabolism, inflammatory response, and oxidative stress in OAS, hoping to provide some new ideas for the treatment of the disease.

Vitamin D and Omega-3 Polyunsaturated Fatty Acids in Type 1 Diabetes Modulation.

BACKGROUND: Type 1 diabetes (T1D) is a chronic autoimmune disease affecting people globally. Usually developed during childhood, T1D is characterized by the destruction of pancreatic β-cells due to immune cell attack and the establishment of an inflammatory process. OBJECTIVE: The study aimed to investigate the effects of vitamin D through its nuclear receptor and the ω-3 polyunsaturated fatty acids (PUFAs) through their lipid derivatives in T1D modulation. Both components exert anti-inflammatory activity and act directly on cells of the immune system, attenuating the destruction of insulin-p

Regulation of hepatic coenzyme Q biosynthesis by dietary omega-3 polyunsaturated fatty acids.

Dietary fats are important for human health, yet it is not fully understood how different fats affect various health problems. Although polyunsaturated fatty acids (PUFAs) are generally considered as highly oxidizable, those of the n-3 series can ameliorate the risk of many age-related disorders. Coenzyme Q (CoQ) is both an essential component of the mitochondrial electron transport chain and the only lipid-soluble antioxidant that animal cells can synthesize. Previous work has documented the protective antioxidant properties of CoQ against the autoxidation products of PUFAs. Here, we have exp

The use of omega-3 fatty acids in the treatment of oral diseases.

Omega-3 polyunsaturated fatty acids (ω-3 PUFAs) have been reported to exert important roles in the inflammatory response. There are many inflammatory diseases in dentistry which support the administration of ω-3 PUFAs as an adjunct therapy during the treatment of these diseases. The aim of this review was to evaluate the use of ω-3 PUFAs as an adjuvant therapy during the treatment of buccal diseases. The review showed that supplementation with ω-3 PUFAs was used for treatment of gingivitis, periodontal diseases, apical periodontitis, stomatitis, and orthodontic tooth movement. The results indi

Biochemical and therapeutic effects of Omega-3 fatty acids in sickle cell disease.

Sickle cell disease (SCD) is a hematologic disorder with complex pathophysiology that includes chronic hemolysis, vaso-occlusion and inflammation. Increased leukocyte-erythrocyte-endothelial interactions, due to upregulated expression of adhesion molecules and activated endothelium, are thought to play a primary role in initiation and progression of SCD vaso-occlusive crisis and end-organ damage. Several new pathophysiology-based therapeutic options for SCD are being developed, chiefly targeting the inflammatory pathways. Omega-3 fatty acids are polyunsaturated fatty acids that are known to ha
