Prevalence, risk factors and genetic diversity of Chlamydia felis in cats.

INTRODUCTION: Chlamydia felis is the main chlamydial pathogen of cats and is associated with conjunctivitis and respiratory disease. This study aimed to estimate the prevalence of Chlamydiaceae and Chlamydia felis, to explore risk factors and predictors (age, sex, breed, origin and ocular signs) for infection using logistic regression, and to appraise genetic diversity via ompA sequencing and phylogenetic analysis. MATERIAL AND METHODS: Conjunctival swabs from 156 cats were examined using real-time PCR assays for Chlamydiaceae and C. felis. Logistic regression and Kaplan-Meier analysis evalua

Expression of IZUMO1 and JUNO in the gonads of domestic cats (Felis catus).

Because of the time-consuming nature of surgical neutering and the rapid rate of reproduction among domestic cats, it is crucial to investigate alternative, nonsurgical methods of contraception for this species. Sperm protein IZUMO1 and its oocyte receptor JUNO have been proposed as potential targets for nonsurgical contraceptives. This study aimed to demonstrate (1) the protein coding sequence of feline IZUMO1 and JUNO, (2) gene expression in specific organs by measuring mRNA levels in different visceral tissues, and (3) the expression of IZUMO1 and JUNO during sperm maturation and folliculog

Cardio-pulmonary parasites of the European wildcat (Felis silvestris) in Germany.

BACKGROUND: In the last years, research on feline cardio-pulmonary parasites has considerably increased in Europe. Not only domestic cats (Felis catus), but also European wildcats (Felis silvestris) can serve as definitive hosts for these nematodes. The F. silvestris population in Germany has been growing rapidly within the last decades; therefore, the assessment of its cardio-pulmonary parasite status is of importance to unravel whether the wildcat population serves as a substantial reservoir for these nematodes and might pose a health threat to domestic cats. METHODS: As part of a nature co

Evaluation of various sample sources for the cytologic diagnosis of Cytauxzoon felis.

BACKGROUND: Cytauxzoon felis is a life-threatening protozoan disease of cats. Identification of schizont-laden macrophages is a point-of-care diagnostic test for acute cytauxzoonosis. HYPOTHESIS/OBJECTIVES: The primary objective determined cytologic agreement between sample types to diagnose acute cytauxzoonosis. The secondary objective evaluated novices' ability to identify cytauxzoon organisms in blood films and tissue aspirates. ANIMALS: Thirty-eight cats with suspected acute cytauxzoonosis and 5 controls examined postmortem. METHODS: Cases were prospectively submitted and collected. Blo

Lack of evidence for perinatal transmission of Cytauxzoon felis in domestic cats.

Cytauxzoon felis is a hemoprotozoan parasite of cats capable of causing severe, often fatal disease during acute infection, but cats that survive the acute stage of disease become chronic carriers. These otherwise healthy carriers are capable of transmitting the infection to other cats via the bite of a vector tick. A variety of other hematoprotozoan parasites are capable of vertical transmission from mother to offspring. If this were possible for C. felis, it could be an important part of the explanation for the apparent emergence of this disease with an increased incidence in an expanding ge

Chlamydophila felis in cats--are the stray cats dangerous source of infection?

Chlamydophila felis is a causative agent of acute or chronic conjunctivitis, and pneumonia in cats. Natural transmission mostly occurs consequently to close contact with other infected cats, their aerosol and fomites. We have examined 93 cats with symptoms of acute or chronic conjunctivitis, from Košice region in Slovakia, during the period of 2 years. Conjunctival samples were obtained from 55 domestic cats (59.14%) and 38 stray cats (40.86%). Of the total number of 93 examined animals, 42 cats were positive, which represents 45.16% overall positivity. Out of the 42 positive cats, 25 cats wer

Rickettsia felis and Bartonella spp. in fleas from cats in Albania.

Fleas can serve as vectors for bacterial pathogens like Bartonella and Rickettsia species, which have been isolated worldwide. However, the knowledge of the epidemiology of vector-borne diseases in general and thus on flea-borne diseases in Albania is limited. Therefore, from 78 free-roaming cats in Tirana, Albania, fleas (371 Ctenocephalides felis and 5 Ctenocephalides canis) were collected to examine them for the presence of Rickettsia and Bartonella species. Ten of the 371 C. felis (2.7%) were positive for Rickettsia felis, and 24 (6.5%) for Bartonella spp. (B. henselae and B. clarridgeiae)

[Interhost migration behaviour of Ctenocephalides felis on cats and in their resting sites].

The cat flea, Ctenocephalides felis, maintains its life cycle indoors feeding on pets. However, it must be emphasized that only about 1-5% of the flea population is on their hosts, the remaining 95-99% (eggs, larvae and pupae) are spread around the habitat. The eggs and dried blood feces fall from the coat of the animal on its sleeping, resting or feeding sites. In the first study, 100 pupae of the cat flea were applied on the resting sites of 6 cats' (3 female, 3 male), each in a separate box. Five days later all 6 cats were examined for adult fleas. From the 600 applied pupae a total of 326

Transcription of the Rickettsia felis ompA gene in naturally infected fleas.

Rickettsia felis is maintained transovarially in Ctenocephalides felis fleas in a widespread geographic distribution and is transmitted to humans and animals, including opossums. This rickettsia is phylogenetically a member of the spotted fever group, most closely related to Rickettsia akari and R. australis. An unusual feature of this rickettsia is that the gene for the outer membrane protein A (OmpA) is interrupted by stop codons. To determine if this putatively dying gene is expressed, mRNA was extracted from laboratory-maintained, R. felis-infected cat fleas. Reverse transcriptase-polymera

Antigenic classification of Rickettsia felis by using monoclonal and polyclonal antibodies.

Rickettsia felis is a flea-transmitted rickettsia. There is a discrepancy between its reported phylogenic and phenotypic identifications. Following the first report of R. felis, it was considered by tests with serologic reagents to be closely related to another recognized flea-transmitted rickettia, R. typhi. Subsequently, it appeared to be more closely related to spotted fever group (SFG) rickettsiae by genetic analysis. In the present work, R. felis was studied by microimmunofluorescence (MIF) serologic typing and with monoclonal antibodies (MAbs). Mouse polyclonal antisera to R. felis cross

Development and evaluation of a PCR-based assay for detection of Haemobartonella felis in cats and differentiation of H. felis from related bacteria by restriction fragment length polymorphism analysis.

The 16S rRNA gene of Haemobartonella felis was amplified by using universal eubacterial primers and was subsequently cloned and sequenced. Based on this sequence data, we designed a set of H. felis-specific primers. These primers selectively amplified a 1,316-bp DNA fragment of the 16S rRNA gene of H. felis from each of four experimentally infected cats at peak parasitemia. No PCR product was amplified from purified DNA of Eperythrozoon suis, Mycoplasma genitalium, and Bartonella bacilliformis. Blood from the experimental cats prior to infection was negative for PCR products and was greatly di

Do commercial dog extracts cross-react with Felis domesticus allergen 1.

Background: Half of U.S. households own a dog despite 10% of individuals being sensitized to dog. Assessment and treatment options for dog allergy include the use of commercial dog extracts which have inconsistent performance, making diagnosing and managing dog allergy challenging. Contamination of dog extracts with other allergens has previously been reported. Objective: We sought to determine whether commercial dog extracts contain other aeroallergens. Methods: An extract purity and quantification study on acetone precipitated dog hair and dander extract (AP dog) was performed, 6 aeroallerge

Characterisation of Staphylococcus felis isolated from cats using whole genome sequencing.

This study used phenotypic tests and whole genome sequencing to characterise a collection of 37 clinical Staphylococcus felis isolates from cats. Samples were isolated from a range of diseases including feline lower urinary tract disease (n = 15), otitis externa (n = 13), and ocular disease (n = 2). Isolates were identified using MALDI-TOF MS and by BLASTn analysis of S. felis-specific 16S rRNA, rpoB and nuc genes in whole genome sequence-based contigs. Phenotypic antimicrobial resistance was determined using disk diffusion and broth microdilution. Coagulase activity was assessed using feline

Crystal structure and functional insight of HP0420-homolog from Helicobacter felis.

Helicobacter pylori infect more than half of the world's population and are considered a cause of peptic ulcer disease and gastric cancer. Recently, hypothetical gene HP0421 was identified in H. pylori as a cholesterol alpha-glucosyltransferase, which is required to synthesize cholesteryl glucosides, essential cell wall components of the bacteria. In the same gene-cluster, HP0420 was co-identified, whose function remains unknown. Here we report the crystal structure of HP0420-homolog of H. felis (HF0420) to gain insight into the function of HP0420. The crystal structure, combined with size-exc

Maxillary incisor root resorption after rapid palatal expansion in Felis catus.

Root resorption after rapid palatal expansion (RPE) treatment was found in anchored teeth but has not been studied on non-anchored incisors. This study evaluated root resorption, root tipping, and root proximity of maxillary incisors after RPE treatment. Fourteen cats were divided into treated (n = 10) and untreated (n = 4) groups. The RPE treatment consisted of active, retention, and relapse phases, lasting 25, 60 and 60 d, respectively. Standardized occlusal radiographs were taken to measure tipping and root proximity before and after each treatment phase. Maxillary incisors were analysed hi
