Preparation and characterization of biomimetic silk fibroin/chitosan composite nanofibers by electrospinning for osteoblasts culture.

In this study, we have successfully fabricated electrospun bead-free silk fibroin [SF]/chitosan [CS] composite nanofibers [NFs] covering the whole range of CS content (0%, 25%, 50%, 75%, and 100%). SF/CS spinning solutions were prepared in a mixed solvent system of trifluoroacetic acid [TFA] and dichloromethane. The morphology of the NFs was observed by scanning electron microscope, and the average fiber diameter ranges from 215 to 478 nm. Confocal laser scanning microscopy confirms the uniform distribution of SF and CS within the composite NFs. To increase biocompatibility and preserve nanost

Fabrication of silk fibroin blended P(LLA-CL) nanofibrous scaffolds for tissue engineering.

Electrospinning using natural proteins and synthetic polymers offers an attractive technique for producing fibrous scaffolds with potential for tissue regeneration and repair. Nanofibrous scaffolds of silk fibroin (SF) and poly(L-lactic acid-co-epsilon-caprolactone) (P(LLA-CL)) blends were fabricated using 1,1,1,3,3,3-hexafluoro-2-propanol as a solvent via electrospinning. The average nanofibrous diameter increased with increasing polymer concentration and decreasing the blend ratio of SF to P(LLA-CL). Characterizations of XPS and (13)C NMR clarified the presence of SF on their surfaces and no

Eudragit E100/silk fibroin hybrid nanoparticles for the controlled oral delivery of amphotericin B.

AIMS: Amphotericin B (AmB) is the first-line antifungal compound for severe fungal infections, yet it possesses high renal toxicity, low water solubility, and acidic instability in the stomach. Herein, we developed novel nanoparticles from silk fibroin with/without Eudragit E100 (FNP and FNP/EE) for the controlled oral delivery of AmB. METHODS AND RESULTS: The particles were formulated by two different methods of co-condensation and adsorption, which yielded comparable particle sizes of ~1500 nm (FNP-AmB) and ~800 nm (FNP/EE-AmB) and AmB entrapment efficiencies of >70%. AmB was rapidly adsorb

Shape-Memory Collagen/Silk-Fibroin Scaffold for Dura Sealing and Skull Base Regeneration.

When a skull base defect occurs, reconstruction is typically performed using a variety of autologous grafts, flaps, or synthetic materials. Failure to achieve appropriate management and closure can lead to serious consequences, such as cerebrospinal fluid (CSF) leakage and subsequent meningitis. Current grafts and synthetic patches often lack biological activity, mechanical integration, and sealing capacity. Here, we developed a multifunctional, injectable composite scaffold composed of collagen/silk-fibroin (S-F) and α-tricalcium phosphate (α-TCP), designed for simultaneous CSF sealing and ti

On the dynamic caddisfly silk H-fibroin gene: a population study in a net-spinning species.

Larvae of the caddisfly Arctopsyche grandis BANKS build protective structures and spin silken capture nets in flowing water. Caddisfly H-fibroin, the major protein component of its silk fibers, has a blocky structure with repeating units defined as beginning with a [(SX)nE]m region followed by a G-rich spacer. Previous observation of H-fibroin allelic variation in haploid-resolved individuals led us to investigate allelic variation within two geographically close but separated natural populations of A. grandis. The genomes of 18 individuals were sequenced, and 34 haploid-resolved H-fibroin seq

Silk Fibroin Biomimetic Membranes with Villus-Crypt Architecture for In Vitro Intestinal Epithelium Modeling.

Three-dimensional (3D) intestinal models require physiologically relevant microarchitectures and mechanically supportive matrices to accurately replicate epithelial behavior; however, most existing in vitro systems lack villus-crypt topography and do not provide the material cues needed to guide epithelial organization. In this work, we developed biomimetic silk fibroin (SF) membranes that reproduce the native villus-crypt structure using two complementary cross-linking and processing strategies: chemical cross-linking with BDDE to form soft hydrogels and ethanol-induced β-sheet formation to g

Polycationic silk fibroin chelating nano-hydroxyapatite promotes angiogenesis for burn wound healing.

The regeneration of deep burn wounds, which cause dermal tissue necrosis, remains a significant clinical challenge. Herein, we report a skin dressing fabricated by integrating nano-hydroxyapatite (nHA) with a polycationic silk fibroin (CSF) to promote burn wound healing via enhanced angiogenesis. The nHA was firmly incorporated into the CSF matrix through metal-phenolic coordination, improving the mechanical robustness and stability of the composite. The CSF-nHA dressing significantly promoted the proliferation of NIH-3T3 fibroblasts and enhanced the recruitment and migration of human umbilica

Heparin-grafted aligned chitosan/silk fibroin scaffolds with multifunctional bioactivities for cardiovascular applications.

Implantable cardiovascular biomaterials face critical clinical challenges, such as thrombosis, bacterial infection, and insufficient endothelial regeneration. In this study, aligned chitosan/silk fibroin (CS/SF) electrospun scaffolds were fabricated by heparin functionalization via parameter optimization, polydopamine-mediated surface modification, and controlled heparin grafting. A comprehensive characterization confirmed that the optimized scaffolds possessed well-aligned uniform nanofibers, desirable mechanical performance, adjustable hydrophilicity, and programmable degradation behavior. Metal-ion-coordinated silk fibroin nanogels for enhanced oral chemotherapy of colorectal cancer.

The efficacy of oral chemotherapy for colorectal cancer (CRC) is hampered by poor drug stability and absorption in the upper gastrointestinal tract, as well as inadequate targeting efficiency at tumor sites. To address these issues, we proposed a simple and biocompatible nanogel, which was co-assembled from chemotherapeutic drug 5-fluorouracil (5-FU), regenerated silk fibroin (SF) as a natural protein carrier, and metal ions (Ni2+/Cu2+). The obtained nanogel system exploited the coordination interactions among 5-FU, amino acid residues, and metal ions to form a multifunctional oral nano-drug s

Electrospun silk fibroin nanofibers enriched with metallic nanoparticles and phenolic acids for antimicrobial applications.

Current trends in the development of the next generation of advanced wound dressings are directed toward the use of natural polymeric materials with additional properties such as antimicrobial function, controlled permeability, renewability, easy preparation, wound conformability and biocompatibility. Herein, fibrous hybrid organic-inorganic membranes of silk fibroin (SF) were prepared by electrospinning and were enriched with metal nanoparticles (Ag and Cu NPs) and phenolic compounds: caffeic, rosmarinic, and ellagic acids (CA, RA, EA, respectively) by using inert gas condensation sputtering

Bioorthogonally Active Silk Fibroin Patch Facilitate Immunomodulatory Prodrug Activation for Tissue Repair.

Current silk fibroin (SF)-based tissue repair patches often exhibit limited immunomodulatory efficacy, exacerbating inflammatory responses and compromising functional tissue integration. To address this limitation, we developed a bioorthogonal prodrug activation strategy within an SF patch via tetrazine functionalization (SF-Tz). This platform enables the spatiotemporally controlled release of trans-cyclooctene (TCO)-caged dexamethasone (TCO-Dex) via inverse electron-demand Diels-Alder reactions. The SF-Tz platform was engineered through optimized carboxylation and tetrazine conjugation protoc

Engineered silk fibroin dendritic cell backpacks to boost antitumor immunity following radiotherapy.

Radiotherapy initiates an antitumor immune response by releasing tumor antigens; however, its efficacy is often limited by the subsequent quantitative and qualitative dysfunction of dendritic cells within the tumor microenvironment. This dysfunction leads to impaired antigen presentation and suboptimal T cell activation. To address this, we developed a biomaterial-based cellular engineering strategy using silk fibroin-based dendritic cell backpacks. Silk fibroin backpacks offer structural stability and aqueous processability, enabling the incorporation of CpG under mild conditions. The easy fu

Silk fibroin hollow microneedle system for sustained transdermal administration of liraglutide: development and characterization.

Liraglutide demonstrates a glucose-dependent hypoglycemic mechanism with significant therapeutic potential for type 2 diabetes mellitus. This study developed a hollow microneedle system featuring dual drug loading mechanisms cavity encapsulated and matrix embedded to achieve sustained release of liraglutide. The cavity structure avoids direct contact between the drug and the silk fibroin shell, as well as the binding state transition caused by hydrophobic interactions. The shell has piercing mechanical properties and provides swelling diffusion channels. The pharmacokinetic data in SD rats sho

Sustainable and standardized fabrication, recycling, and sterilization of salt-bed casted silk fibroin sponges.

Silk fibroin, as a highly versatile biomaterial, is increasingly applied in research, medicine and other sterile applications. The rising usage calls for a robust, standardized and reproducible silk scaffold production and a higher need for silk resources. In this study we standardized the silk fibroin sponges fabrication, set up a quality control protocol, introduced a recycling method to reduce the resources needed and analyzed a sterilization method for translation into preclinical applications. In overall, the development of a fast and efficient quality control protocol led to a reduction

A sequential drug delivery system based on silk fibroin scaffold for effective cartilage repair.

Endogenous repair of cartilage defects is a preferential strategy for cartilage repair, but always hindered by insufficient early-stage cells and incomplete cell differentiation at later stages. For in-situ cartilage regeneration, it is crucial to develop a sequential drug release system capable of recruiting endogenous bone marrow mesenchymal stem cells (BMSCs) and promoting their chondrogenic differentiation. Herein, based on our long-term and fruitful research on silk fibroin (SF) porous scaffolds, a cell-free sequential drug delivery SF scaffold was developed. BMSCs affinity peptide PFSSTK

Peptide-Laden Silk Fibroin Microneedles Modulate Inflammation and Promote Myocardial Repair In Vitro.

Myocardial infarction is a major cause of death and disability worldwide and poses a serious threat to global public health. Chronic ischemia and hypoxic environment induce excessive inflammatory response, leading to heart failure and even death. Therefore, inhibiting excessive inflammation and promoting tissue repair are important for myocardial repair. Based on this, natural polymeric Silk Fibroin Methacryloyl microneedles loaded with Tc peptide drugs (SilMA MNs) were fabricated for the MI therapy by photocuring and micro-molding techniques, which facilitate the protection of Tc peptides tha

Magnetosome-incorporated silk fibroin-based magnetic hydrogel for efficient tumor magnetic hyperthermia.

We developed a magnetosome-incorporated silk fibroin-based magnetic hydrogel (MSMH) for efficient tumor magnetic hyperthermia. The MSMH was fabricated by integrating biologically synthesized magnetosomes with glycidyl methacrylate-modified silk fibroin (SFGMA), enabling injectable deployment and rapid photo-crosslinking. The as-fabricated MSMH exhibits excellent magnetothermal conversion performance, achieving prominent temperature rises under alternating magnetic fields (AMF). MSMH can also serve as a T₂-weighted MRI contrast agent (r2 = 173.57 mM-1 s-1 at 0.5 T), enabling imaging-guided ther

Silk fibroin microneedles loaded with epigallocatechin gallate mitigate atrazine-induced testicular toxicity.

Atrazine (ATZ), a commonly used herbicide, disrupts male endocrine systems, impacting reproductive health. Epigallocatechin gallate (EGCG) possesses potent antioxidant properties and shows promise in alleviating testicular dysfunction induced by endocrine disruptors. However, its clinical utility is hindered by poor physicochemical stability and low oral bioavailability. Herein, we first developed a silk fibroin microneedles (MNs) patch loaded with EGCG, enabling EGCG to directly target the testes through transdermal administration. Our findings revealed that EGCG-loaded silk fibroin microneed

Studies on the potential risk of amyloidosis from exposure to cultured fibril from silk fibroin.

Amyloid A (AA) amyloidosis is induced by administering amyloid fibrils to animals under inflammatory conditions. Silk fibroin (SF), the main component of silk threads, forms amyloid-like fibrils and has been previously reported to induce AA amyloidosis in mice. In this study, SF was cultured in ethanol solution, and after confirming fibril formation through thioflavin T assay, Congo red assay, and observation under electron microscopy, cultured SF ethanol solutions were administered to mice via various routes to investigate the induction of target organs and amyloidosis. As a result, cultured

Hemostatic and Ultrasound-Controlled Bactericidal Silk Fibroin Hydrogel via Integrating a Perfluorocarbon Nanoemulsion.

Excessive blood loss and infections are the prominent risks accounting for mortality and disability associated with acute wounds. Consequently, wound dressings should encompass adequate adhesive, hemostatic, and bactericidal attributes, yet their development remains challenging. This investigation presented the benefits of incorporating a perfluorocarbon nanoemulsion (PPP NE) into a silk-fibroin (SF)-based hydrogel. By stimulating the β-sheet conformation of the SF chains, PPP NEs drastically shortened the gelation time while augmenting the elasticity, mechanical stability, and viscosity of th
