Ability of prenylflavanones present in hops to induce apoptosis in a human Burkitt lymphoma cell line.

The identification of effective cancer preventive compounds from hops has become an important issue in public health-related research. We compared the antiproliferative and apoptosis-inducing effects of side chain variants of prenylflavanones, e. g., 8-prenylnaringenin (7) and 8-geranylnaringenin (10), which have been identified in hops (Humulus lupulus), and their synthetic variations 8-furanmethylnaringenin (8) and 8-cinnamylnaringenin (9). These were accessible by a Mitsunobu reaction and Claisen rearrangement. Flavanones 9 and 10 showed cytotoxic and apoptotic activities. Apoptosis was ind

Evaluation of Naringenin as a Promising Treatment Option for COPD Based on Literature Review and Network Pharmacology.

Chronic obstructive pulmonary disease (COPD) is a chronic respiratory disease characterized by incompletely reversible airflow limitation and seriously threatens the health of humans due to its high morbidity and mortality. Naringenin, as a natural flavanone, has shown various potential pharmacological activities against multiple pathological stages of COPD, but available studies are scattered and unsystematic. Thus, we combined literature review with network pharmacology analysis to evaluate the potential therapeutic effects of naringenin on COPD and predict its underlying mechanisms, expecti

Integration of Transcriptomics and Metabolomics Reveals Organ-Specific Biosynthesis and Accumulation of Pharmacologically Active Flavonoids in Rhododendron yedoense var. poukhanense.

Rhododendron yedoense var. poukhanense is an important medicinal plant, but still little is known about how its bioactive flavonoids are made in different organs. Comprehensive metabolomic profiling was performed using ultra-performance liquid chromatography-tandem mass spectrometry (UPLC-MS/MS), coupled with reference-based RNA sequencing (RNA-seq), to analyze root, stem, and leaf tissues from three independent biological replicates. Subsequently, we performed two-way orthogonal partial least squares (O2PLS) regression, canonical correlation analysis (CCA), and Pearson correlation analyses. A

Isosakuranetin ameliorates hypertension in rats induced by L-NAME.

BACKGROUND: Hypertension is a major global health problem that often develops without noticeable symptoms. Current treatments and complementary approaches focus on improving endothelial function and enhancing nitric oxide (NO) production to promote vasodilation and lower blood pressure. Isosakuranetin, a flavanone found in Chromolaena odorata leaves, has shown potential antihypertensive properties. This study aimed to investigate the effects of isosakuranetin on L-NG-Nitroarginine methyl ester (L-NAME)-induced hypertension, focusing on its ability to enhance NO production and reduce oxidative

Eriodictyol: a review of its pharmacological activities and molecular mechanisms related to ischemic stroke.

Ischemic stroke (IS) is characterized by a prominent mortality and disability rate, which has increased the burden on the global economy to a certain extent. Meanwhile, patients benefit little from the limited clinical strategies of intravenous alteplase and thrombectomy due to the limited therapeutic window. Given this, it is urgent to study new therapeutic methods to intervene in these patients. Eriodyctiol (ERD) is a major natural flavonoid, which widely exists in fruits, vegetables, and medicinal herbs, and has various pharmacological properties. It has been reported that ERD can maintain

The Potential Role of Naringin and Naringenin as Nutraceuticals Against Metabolic Syndrome.

Metabolic syndrome, an increasing problem in western society, is a cluster of conditions that affect cardiovascular health, lipid and glucose management, increasing the risk of heart diseases, stroke and diabetes. Bioactive flavonoids are a great resource of compounds with proven antiinflammatory activities. Naringin, a natural flavanone found in citrus fruits, and its aglycone have demonstrated to ameliorate obesity, dyslipidemia, and insulin resistance in animal models. The principal mechanisms by which these flavonoids exert their action involve AMPK and PPARα up-regulation and the down-reg

Nigratine as dual inhibitor of necroptosis and ferroptosis regulated cell death.

Nigratine (also known as 6E11), a flavanone derivative of a plant natural product, was characterized as highly specific non-ATP competitive inhibitor of RIPK1 kinase, one of the key components of necroptotic cell death signaling. We show here that nigratine inhibited both necroptosis (induced by Tumor Necrosis Factor-α) and ferroptosis (induced by the small molecules glutamate, erastin, RSL3 or cumene hydroperoxide) with EC50 in the µM range. Taken together, our data showed that nigratine is a dual inhibitor of necroptosis and ferroptosis cell death pathways. These findings open potential new

Naringenin promotes coronary vasodilation via inhibition of L-type calcium channels (CaV1.2): Evidence from tissue bath and molecular studies.

Naringenin, a citrus-derived flavanone, was investigated for its vasorelaxant mechanisms in goat coronary arteries, focusing on endothelial signaling and direct inhibition of L-type calcium channels (CaV1.2). In silico docking against a homology-modeled CaV1.2 channel revealed that naringenin exhibited the highest binding affinity (-8.5 kcal/mol) among ten related flavanones, forming stabilizing interactions within the channel's pore region. To further validate this interaction, molecular dynamics simulations demonstrated sustained complex stability, minimal structural deviation, and consisten

Dietary (Poly)phenol Intake, Inflammatory Biomarkers, and Mild Cognitive Impairment in Italian Adults.

BACKGROUND: Dietary (poly)phenols have been proposed to protect against age-related cognitive decline through antioxidant, anti-inflammatory, and neuroprotective mechanisms. However, evidence specifically addressing mild cognitive impairment (MCI) remains limited. This study investigated the association between habitual dietary (poly)phenol intake, cognitive status, and circulating inflammatory biomarkers in older adults. METHODS: In this cross-sectional study, Italian adults aged ≥65 years attending a memory clinic underwent standardized neuropsychological assessment for MCI diagnosis. Habit

Morutamins A-L from Morus alba L. Root Bark with Inhibitory Effects on Epidermal Growth Factor Receptor and Toll-Like Receptor-Mediated NF-κB/AP‑1 Activation.

Morus alba L. root bark (MAR, "Sang-Bai-Pi" in traditional Chinese medicine) has been traditionally used for the treatment of inflammatory disorders. Phytochemical investigation of MAR yielded 11 new flavanones and one new chalcone (1-12, morutamins A-L), along with 27 known compounds (13-39). Their structures and absolute configurations were elucidated by combining HRESIMS and NMR analyses with DP4 and DP4+ probability calculations and ECD data. Among these, morutamin L (12) exhibited potent anti-inflammatory and immunomodulatory activities by inhibiting epidermal growth factor receptor and t

An integrative multi-omics approach reveals metabolic mechanism of flavonoids during anaerobic fermentation of de'ang pickled tea.

Anaerobic fermentation (AF) is critical process for Yunnan De'ang pickled tea production. Therefore, widely targeted metabolomics and metagenomics were integrated to reveal the AF mechanism. Lactic acid bacteria (LAB) (e.g. Lactiplantibacillus plantarum, Lactobacillus vaccinostercus and Lactobacillus paracollinoides) and yeasts like Candida metapsilosis and Cyberlindnera fabianii dominated in the AF. Based on bacterial community succession and metabolites variation, the whole AF processes were divided into two phases, i.e., before and after four months. A total of 327 characteristic metabolite

A metabolomic approach to identifying chemical mediators of mammal-plant interactions.

Different folivorous marsupials select their food from different subgenera of Eucalyptus, but the choices cannot be explained by known antifeedants, such as formylated phloroglucinol compounds or tannins, or by nutritional quality. Eucalypts contain a wide variety of plant secondary metabolites so it is difficult to use traditional methods to identify the chemicals that determine food selection. Therefore, we used a metabolomic approach in which we employed (1)H nuclear magnetic resonance spectroscopy to compare chemical structures of representatives from the two subgenera and to identify chem

Inhibition of Endothelial Dysfunction by Dietary Flavonoids and Preventive Effects Against Cardiovascular Disease.

Cardiovascular disease (CVD), such as stroke, ischemic heart disease, and heart failure, accounts for many deaths, and its increasing incidence is a worldwide concern. Accumulating evidence suggests that the elevated risk of CVD caused by dysfunction of vascular endothelial cells and resultant arteriosclerosis can be mitigated by increased consumption of fruits and vegetables. These foods contain phytochemicals such as polyphenols and carotenoids, as well as dietary fiber. Flavonoids of the polyphenol class are found in vegetables, fruits, grains, bark, roots, stems, flowers, tea, and wine. Se

New natural product carbonic anhydrase inhibitors incorporating phenol moieties.

Carbonic anhydrases (CAs, EC 4.2.1.1) catalyze the fundamental reaction of CO2 hydration in all living organisms, being actively involved in the regulation of a plethora of patho/physiological conditions. They represent a typical example of enzyme convergent evolution, as six genetically unrelated families of such enzymes were described so far. The need to find selective CA inhibitors (CAIs) triggered the investigation of natural product libraries, which proved to be a valid source of agents with such an activity, as demonstrated for the phenols, polyamines and coumarins. Herein we report an i
