Comparing the effects of static and variable loading in potentiation of the lower extremities through vertical jump.

BACKGROUND: Post-activation potentiation (PAP) can elicit increased force in a muscular contraction following a specific preparatory activity, though little research exists regarding how various loading methods in said activity impact the potentiation effect. This study seeks to determine if banded PAP activities are more effective than static loading in improving power production. METHODS: Participants in this randomized cross-over design study were 7 female collegiate volleyball players with a 6-month minimum training age and no significant injuries. The study took place across 3 sessions,

[The impairment of excitation-contraction (E-C) coupling in myasthenia gravis].

We have developed a new novel method to assess the function of excitation-contraction (E-C) coupling in patients with myasthenia gravis (MG). In our procedure, masseteric compound muscle action potential (CMAP) and mandibular movement-related potentials (MRP) were recorded simultaneously after stimulating the trigeminal motor nerve with a needle electrode. The E-C coupling time (ECCT) was calculated by the latency difference between CMAP and MRP. Bite force was measured using a pressure-sensitive sheet. Our serial studies demonstrate that masseteric E-C coupling is impaired in some MG patients

Endurance exercise alters the contractile responsiveness of rat heart to extracellular Na+ and Ca2+.

PURPOSE AND METHODS: The isovolumic contractile responsiveness of left ventricular (LV) myocardium to altered extracellular [Ca2+], [Na+], and pacing frequency was examined using perfused hearts (37 degrees C) isolated from sedentary (SED) and treadmill-trained (TR) adult female rats. RESULTS: The suppressive effect of reducing perfusate free [Ca2+] to 0.7 mM on LV developed pressure (delta LVP) was greater in the TR hearts compared with SED hearts (P < 0.05). When perfusate [Na+] was reduced to 120 mM ([Ca2+] = 0.7 mM), delta LVP augmentation was greatest in the TR hearts (P < 0.05). The neg

Comparative electrophysiological and mechanical actions of ATP-sensitive potassium channel openers in canine Purkinje fibers.

1. Electrophysiological and mechanical effects of ATP-dependent potassium channel openers on canine Purkinje fibers were examined. 2. Cromakalim and pinacidil (0.3-10 microM) and nicorandil (0.3-1 mM) shortened the action potential duration (APD), and reduced the contractile force, in a concentration-dependent manner. Other action potential parameters were unaffected. The effects were reversible. 3. Effects of the openers were antagonized by tetraethylammonium (a non-specific potassium channel blocker) and more potently by glibenclamide (a blocker of ATP-sensitive K+ channel). 4. The APD short

Functional and Molecular Characterisation of Heart Failure Progression in Mice and the Role of Myosin Regulatory Light Chains in the Recovery of Cardiac Muscle Function.

Heart failure (HF) as a result of myocardial infarction (MI) is a major cause of fatality worldwide. However, the cause of cardiac dysfunction succeeding MI has not been elucidated at a sarcomeric level. Thus, studying the alterations within the sarcomere is necessary to gain insights on the fundamental mechansims leading to HF and potentially uncover appropriate therapeutic targets. Since existing research portrays regulatory light chains (RLC) to be mediators of cardiac muscle contraction in both human and animal models, its role was further explored In this study, a detailed characterisatio

Augmentation of Ouabain-Induced Increase in Heart Muscle Contractility by Akt Inhibitor MK-2206.

Cardiac steroids (CSs), such as ouabain and digoxin, increase the force of contraction of heart muscle and are used for the treatment of congestive heart failure (CHF). However, their small therapeutic window limits their use. It is well established that Na+, K+-ATPase inhibition mediates CS-induced increase in heart contractility. Recently, the involvement of intracellular signal transduction was implicated in this effect. The aim of the present study was to test the hypothesis that combined treatment with ouabain and Akt inhibitor (MK-2206) augments ouabain-induced inotropy in mammalian mode

Potentiation of cortico-spinal output via targeted electrical stimulation of the motor thalamus.

Cerebral white matter lesions prevent cortico-spinal descending inputs from effectively activating spinal motoneurons, leading to loss of motor control. However, in most cases, the damage to cortico-spinal axons is incomplete offering a potential target for therapies aimed at improving volitional muscle activation. Here we hypothesize that, by engaging direct excitatory connections to cortico-spinal motoneurons, stimulation of the motor thalamus could facilitate activation of surviving cortico-spinal fibers thereby immediately potentiating motor output. To test this hypothesis, we identify opt

Lower-Extremity Joint Function During a Drop Jump: Relationships to Performance and Maximal Strength.

Haischer, MH, Ahn, N, and Kipp, K. Lower-extremity joint function during a drop jump: relationships to performance and maximal strength. J Strength Cond Res XX(X): 000-000, 2026-Humans modulate joint stiffness during movement based on stretch-shortening cycle (SSC) loading demands. In performance tasks such as jumping, the SSC helps maximize vertical impulse through potentiation of concentric force after eccentric loading. However, joints can serve different locomotor functions (i.e., strut, spring, motor, or damper) while coordinating to achieve an overarching goal. Furthermore, while strengt

CaMKIIα holoenzymes self-organize into chain-like mesoscale clusters.

Calcium- and calmodulin-dependent protein kinase II (CaMKII) is highly enriched in dendritic spines at concentrations comparable to those of cytoskeletal proteins and plays a central role in synaptic plasticity. During long-term potentiation, CaMKIIα further accumulates in spines. However, the mechanisms governing its higher-order organization remain poorly understood. Here, we use high-speed atomic force microscopy to visualize interholoenzyme interaction of CaMKIIα at mesoscopic scales (5 to 500 nanometers). Under freely diffusible conditions, CaMKIIα holoenzymes do not form stable clusters.

Effects of Acupuncture on Explosive Force Production by the Healthy Female Shoulder Joint.

BACKGROUND: Acupuncture is often used to treat chronic conditions, such as pain. In recent years, given the importance of the explosive forces generated by shoulder muscles for the completion of motor tasks, studies in which nerves were stimulated through acupuncture to increase the explosive forces were conducted. This study explored the effect of acupuncture on explosive force production by the muscles of the female shoulder joint. METHODS: Eighteen healthy women underwent shoulder adduction (Add), abduction (Abd), flexion (Flex), and extension (Ext) tests with an isokinetic measurement sys

Increase in post activation potentiation in females following a cycling warmup.

Post activation potentiation (PAP) is a phenomenon in which muscular force is acutely enhanced as a result of prior contractile activity. The net augmentation is dependent upon the intensity of the preceding conditioning contraction influencing calcium release and phosphorylation of the regulatory myosin light chain. This phenomenon has been recorded after various types of conditioning contractions, however the interaction of a warmup on PAP remains uncertain and whether this differs between males and females requires consideration. We investigated the effect of a cycling warmup on twitch cont

Phenolic compounds prevent the oligomerization of α-synuclein and reduce synaptic toxicity.

Lewy bodies, mainly composed of α-synuclein (αS), are pathological hallmarks of Parkinson's disease and dementia with Lewy bodies. Epidemiological studies showed that green tea consumption or habitual intake of phenolic compounds reduced Parkinson's disease risk. We previously reported that phenolic compounds inhibited αS fibrillation and destabilized preformed αS fibrils. Cumulative evidence suggests that low-order αS oligomers are neurotoxic and critical species in the pathogenesis of α-synucleinopathies. To develop disease modifying therapies for α-synucleinopathies, we examined effects of

Pyrazinamide Susceptibility Is Driven by Activation of the SigE-Dependent Cell Envelope Stress Response in Mycobacterium tuberculosis.

Pyrazinamide (PZA) plays a crucial role in first-line tuberculosis drug therapy. Unlike other antimicrobial agents, PZA is active against Mycobacterium tuberculosis only at low pH. The basis for this conditional drug susceptibility remains undefined. In this study, we utilized a genome-wide approach to interrogate potentiation of PZA action. We found that mutations in numerous genes involved in central metabolism as well as cell envelope maintenance and stress response are associated with PZA resistance. Further, we demonstrate that constitutive activation of the cell envelope stress response

Influence of type of muscle contraction, gender, and lifting experience on postactivation potentiation performance.

Postactivation potentiation (PAP) or enhanced contractile capabilities may be influenced by a number of factors. This study examined the influence of type of muscle contraction (isometric vs. dynamic), gender, and previous weightlifting experience on PAP as demonstrated by changes in jump height and power output. Thirty young men (n = 15) and women (n = 15), classified as either having previous weightlifting experience (n = 20) or not (n = 10), performed 3 different sets of countermovement jumps, with the first set used to determine baseline measures of jump height and power. The second set wa

The characterization of contractile and myoelectric activities in paralyzed tibialis anterior post electrically elicited muscle fatigue.

This study aimed to understand the myoelectric and mechanical characteristics of muscle recovering from electrically elicited fatigue. A modified Burke fatigue protocol was delivered to activate the tibialis anterior of 13 spinal cord injured subjects for 4 min. Before and after the fatigue protocol, a series of pulse trains was delivered to induce three twitches and a fused contraction at 0, 1, 3, and 5 min and then followed every 5 min for 60 min. The recovery processes of the ankle dorsiflexion torque and the evoked electromyography (EMG) parameters were analyzed and characterized by a firs

Refined Carbohydrate-Rich Diet Promotes Early Myocardial Dysfunction Associated with Adiponectin Reduction, Inflammation, and Impaired Ca2+ Handling in Female Rats.

Introduction: Diets rich in refined carbohydrates are associated with cardiometabolic dysfunction, but the mechanisms linking metabolic disturbances to myocardial impairment in females remain unclear. This study investigated the effects of short-term consumption of a refined carbohydrate-rich diet on papillary muscle contractility in female rats. Methods: Adult female Wistar rats were assigned to either a control diet (CT) or a high-carbohydrate diet (HCD) for 15 days. The HCD consisted of 45% sweetened condensed milk, 10% refined sugar, and 45% standard chow. Metabolic, inflammatory, function

Acute Effects of Dynamic Stretching on Muscle Flexibility and Performance: An Analysis of the Current Literature.

Stretching has long been used in many physical activities to increase range of motion (ROM) around a joint. Stretching also has other acute effects on the neuromuscular system. For instance, significant reductions in maximal voluntary strength, muscle power or evoked contractile properties have been recorded immediately after a single bout of static stretching, raising interest in other stretching modalities. Thus, the effects of dynamic stretching on subsequent muscular performance have been questioned. This review aimed to investigate performance and physiological alterations following dynam
