The biochemical alteration of soil beneath a decomposing carcass.

The processes associated with cadaver decomposition in outdoor settings, particularly those that occur during the extended postmortem interval (>30 days) are poorly understood. Thus, few methods are currently available to accurately estimate the extended postmortem interval (PMI). Of these methods, a soils-based approach has the potential to address the postmortem interval between which entomology and anthropology are the most valuable. Although the validity of soil-based methods has previously been established, little work has been conducted to explain the processes that have been designated

Effect of bait decomposition on the attractiveness to species of Diptera of veterinary and forensic importance in a rainforest fragment in Brazil.

Insects associated with carrion can have parasitological importance as vectors of several pathogens and causal agents of myiasis to men and to domestic and wild animals. We tested the attractiveness of animal baits (chicken liver) at different stages of decomposition to necrophagous species of Diptera (Calliphoridae, Fanniidae, Muscidae, Phoridae and Sarcophagidae) in a rainforest fragment in Brazil. Five types of bait were used: fresh and decomposed at room temperature (26 °C) for 24, 48, 72 and 96 h. A positive correlation was detected between the time of decomposition and the abundance of C

Recovery & identification of human Y-STR DNA from immatures of chrysomya albiceps (Diptera: Calliphoridae). Simulation of sexual crime investigation involving victim corpse in state of decay.

The number of sexual crimes in Brazil, as in several other countries, is very high. In many of these crimes the women raped are murdered and their bodies are found days later, in an advanced state of decomposition, with intense cadaverous fauna. Forensic Entomology studies insects and other arthropods that can be used in the expert analysis of various types of crimes. Diptera, the order of insects that comprises the two-winged or true flies, represents one of the largest known groups of insects and is the principal source of cadaveric entomofauna. Members of its Calliphoridae family are observ

Pupal age estimation of forensically important Megaselia scalaris (Loew) (Diptera: Phoridae).

In addition to causing myiasis in humans and animals, Megaselia scalaris (Loew) has been reported as a forensically important fly. The determination of a minimum postmortem interval (PMI) often relies on the determination of the age of the larvae and pupae. The pupal stage represents about 50% of the immature development time and the pupal age may therefore serve as an important tool in entomological minimum PMI estimation. The present study focuses on the key developmental processes during metamorphosis of M. scalaris pupae at different constant temperatures (18, 21, 24, 27, 30, 33 and 36°C).

First record of the forensically important genus Protopiophila Duda (Diptera: Piophilidae) from South Korea and an integrative framework for forensic identification.

The family Piophilidae is of significant forensic importance due to its association with late-stage decomposition; however, the Korean piophilid fauna remains poorly studied, lacking comprehensive morphological and molecular data. This study reports the first record of the genus Protopiophila Duda from South Korea. Five male specimens were collected and identified as Protopiophila contecta (Walker) based on morphological characters. We provide the first detailed illustrations and descriptions of the previously poorly documented male internal genitalia and a new identification key to the South

Insect succession on a decomposing piglet carcass placed in a man-made freshwater pond in Malaysia.

This entomological study was conducted in a man-made freshwater pond in a palm oil plantation in Tanjung Sepat, Selangor from 23 July 2007 by using pig (Sus scrofa) as a carcass model. A 1.5 month old piglet (5 kg), which died of asphyxia after being accidentally crushed by its mother, was thrown into a pond. Observation was made for ten days; one visit per day and climatological data were recorded. On the first two days, the piglet carcass sunk to the bottom of the pond. The carcass floated to the surface on the third day but no fly activities were seen. The blow fly, Chrysomya megacephala an

An experimental investigation into the colonization of concealed cadavers by necrophagous blowflies.

We used seven baited boxes with different combinations of access holes and odor diffusion surfaces to study the arrival of necrophagous flies. During laboratory experiments, 30 gravid Lucilia sericata females were kept in a chamber with one of the boxes. The box with the largest odor diffusion surface (99 cm(2)) combined with the lowest accessibility (one 1 cm(2) entrance hole) was entered least (5 ± 3.7 flies per run). In contrast, the most frequently entered box (one 9 cm(2) entrance hole with no additional odor diffusion surface) caught a mean of 24.6 ± 3.4 flies per run. These results indi

Exogenous and endogenous microbiomes of wild-caught Phormia regina (Diptera: Calliphoridae) flies from a suburban farm by 16S rRNA gene sequencing.

The black blow fly, Phormia regina (Meigen) (Diptera: Calliphoridae) is one of the most abundant carrion flies in North America. Calliphorids are important in agriculture and animal production, veterinary sciences, forensics and medical entomology. While the role of flies in the epidemiology of human and animal diseases is an active area of research, little is known about the microorganisms associated with these insects. We examined the diversity of wild-caught black blow fly endogenous (internal body) and exogenous (external body) microbial communities using 16S rRNA gene sequencing. Overall,

Genetic diversity of nine necrophagous Sarcophagidae (Diptera) species in Portugal.

Flesh flies (Diptera: Sarcophagidae) are among the most important insect groups used in forensic investigations, particularly for post-mortem interval estimation and assessing possible corpse relocation. Their practical use, however, is limited by the high morphological similarity of closely related species and by the limited representation of regional populations in DNA reference libraries. This study assessed two mitochondrial markers, cytochrome c oxidase subunit I (COI) and NADH dehydrogenase subunit 4 (ND4), for the identification and population-level characterisation of nine necrophagous

The effect of insect excretions/secretions and decomposition fluid on DNA quantity and quality in human bloodstains.

The larval excretions/secretions (ES) of blowflies contain proteolytic enzymes and bacteria that assist with tissue breakdown. Decomposition fluid (DF) contains organic and inorganic waste products from cell death. This study investigated if human DNA recovery from blood was impacted by exposure to ES and DF over time. Lucilia sericata ES were collected daily from 50 larvae, and all available DF was collected from two fetal piglets left to decompose for 2 weeks. Daily for 3-5 days, 28 μL-30 μL of ES, DF, or a 1:1 mixture of the fluids was added to 30 μL of blood on cotton. Three bloodstains pe

Mitogenomic Insights Into the Ancient Split and Cryptic Diversity of the Poecilochirus carabi Complex (Acari: Mesostigmata).

Phoretic associations with insects are widespread in the Mesostigmata. These temporary symbiotic associations have been important in driving the evolutionary development of this taxon. The phoretic mite genus Poecilochirus, which is considered obligately associated with burying beetles, is of great value in forensic entomology. However, due to the morphological conservatism and the difficulty of obtaining adult specimens, the taxonomy of this genus has long been based on deutonymphal characters, and the true species diversity is far from resolved. Here we used next-generation sequencing to det

Evaluation of the influence of the antibiotic ciprofloxacin in the development of an Old World screwworm fly, Chrysomya putoria.

Chrysomya putoria (Wiedemann) (Diptera: Calliphoridae), an Old World screwworm fly, is a species with potential for maggot therapy practice and has been described in myiasis and forensic entomology studies. The objective of the present study was to assess the action of different ciprofloxacin concentrations on the growth and development of C. putoria. First instar maggots of the third generation were raised on 60 g of chicken gizzard homogenate in 65% agar diet and received ciprofloxacin chloridrate. Each concentration of the antibiotic tested (3.33 µg/mL, 6.66 µg/mL, and 13.33 µg/mL) and the

Ophthalmomyiasis Case Caused by Two Blow Fly (Diptera: Calliphoridae) Species in North America.

Ophthalmomyiasis is the result of fly larvae feeding on the tissues of the eye. Commonly associated with poor hygiene and open wounds, this condition is rare and often stigmatized. Treatment can be straightforward, and full recovery is common. Identifying the species responsible for ophthalmomyiasis is important for the medical, forensic, and entomological communities. Here, we present a case of ophthalmomyiasis where 30-40 blow fly (Diptera: Calliphoridae) larvae were removed from the eye of a human male. A representative subsample of five larvae was used for taxonomic identification via two

Expression of human cardiac-specific genes: a novel method for post-mortem interval estimation.

In legal medicine, the determination of post-mortem interval (PMI) is not only an important but also one of the most difficult aspects. Several methods are used to estimate PMI such as physicochemical, entomological, biochemical, metabolic, autolytic, and physical methods. These methods provide a wide range of PMI as they are affected by different factors. The approach behind the present study is to calculate an accurate PMI by using mRNA degradation and fold change expression (FCE) of cardiac-specific genes viz. N-terminal pro-B-type natriuretic peptide (NPPB) and cardiac troponin I (TNNI3).

A severe head louse (Pediculus humanus capitis) infestation: Timeline, pathogen detection, kdr-gene screening and morphological anomalies of lice.

Lice are assuming an increasing importance in forensic investigations, given their capacity to provide information about an individual's care. Head louse pediculosis is a frequent condition in school-age children and can be properly controlled using topical treatments combined with good personal hygiene. Prolonged and chronic infestations may result in more serious outcomes including severe iron deficiency anaemia. We conducted entomological and laboratory investigations of a head louse infestation in a 12-year-old girl who experienced severe anaemia and subsequent death. Numerous lice were fo

Unexpected results found in larvae samples from two postmortem forensic cases.

PURPOSE: In forensics, entomological specimens can be used as additional/alternative matrices to detect xenobiotics when human specimens are limited in their application. Despite some advantages over implementing putrefied human remains, most medico-legal laboratories do not include entomotoxicological procedures as routine analytical methods. We thus applied two authentic cases to evaluate necrophagous larvae's potential as complementary matrices for toxicological analysis after extensive postmortem decomposition. METHODS: Larvae and postmortem human samples, including hair, stomach contents

Recovery of male DNA acquired from carrion-feeding insects in a simulated sexual assault scenario.

When bodies of sexual assault victims are discovered in advanced stages of decomposition, the recovery of direct DNA evidence is often compromised, posing a challenge in criminal investigations. This study aimed to determine insect colonization and succession patterns on pig carcasses inoculated with semen and to assess the feasibility of recovering Y-chromosomal short tandem repeat (Y-STR) DNA from fly larvae, fly pupae, and beetles. Three female pigs were used as models to simulate sexual homicide scenarios, and an indoor control experiment using chicken livers mixed with 3 mL of semen was a

Postmortem artifacts made by ants and the effect of ant activity on decompositional rates.

Ants belong to the family Formicidae of the order Hymenoptera and they are one of the world's dominant insect groups. Ants can be present at all stages of carrion decomposition as they are typically observed shortly after death or during the early postmortem period, but even later once the fly maggots had left the body. Their role in the faunal succession varies from predator on the eggs and larvae of other insects (mainly Diptera), thus reducing significantly the rate of decomposition, to scavenger on the flesh or exudates from the corpse itself. As they can deeply affect the occurrence of ca

Preservation of Calliphora vicina (Diptera: Calliphoridae) pupae for use in post-mortem interval estimation.

Post-mortem interval (PMI) is frequently calculated using immature stages of carrion frequenting Calliphoridae (Diptera). This is based on identification to species level, followed by age estimation of the samples. These two processes depend on suitable preservation of insects for subsequent analyses, yet preservation methods for the pupal stage are poorly defined and inappropriate methods may result in discolouration or nucleic acid degradation. This study examined the effects of 21 common preservation methods on Calliphora vicina pupae of 4 and 7d old, assessing consequences of the various m
