A novel ACTA2 variant in sudden fatal familial thoracic aortic dissection: literature review and genotype-phenotype expansion.

Thoracic aortic dissection (TAD) is a life-threatening emergency frequently linked to underlying genetic variants, particularly in familial cases. Molecular autopsy has become increasingly valuable in forensic practice for identifying occult heritable disorders when routine autopsy alone cannot fully clarify the cause of death. Here, we report a 17-year-old male who died suddenly due to pericardial tamponade caused by ruptured TAD. Gene testing identified a novel ACTA2 variant, c.155_162delinsGCACA. This indel leads to two amino acid substitutions (p.K52_D53delinsST) and a deletion (p.S54del),

Effect of Storage Time on DNA Obtained from Umbilical Cord Used in Forensic Identification.

BACKGROUND/OBJECTIVES: In the identification of human remains, their genetic profile can be compared with that obtained from biological samples of the person from whom they are suspected to have come. Our aim was to evaluate the effect of storage time on DNA obtained from umbilical cord that could be used as a reference in forensic identification. METHODS: Fifteen umbilical cords stored by the donors in their homes for a period of between 0.5 and 47 years were collected. Saliva samples were obtained for use as a reference. DNA extraction was performed using the PrepFiler Express BTA™ Forensic

Benefits and outcomes of a new multidisciplinary approach for the management and financing of sudden unexplained death cases in a forensic setting in Switzerland.

Sudden cardiac death (SCD) is an important public health issue. In young persons aged between 1 and 40 years, most SCDs are caused by potentially inherited cardiac diseases, often not detectable during conventional medico-legal investigations and therefore termed as sudden unexplained deaths (SUD). In this study, we describe the implementation, feasibility and importance of a standardized procedure to investigate SUD cases within the forensic framework at the Zurich Institute of Forensic Medicine in Switzerland. This new approach involves a multidisciplinary collaboration including forensic au

Drug-facilitated sexual assault followed by femicidal chloroform poisoning and suffocation: A case-report of criminal responsibility.

INTRODUCTION: Criminal responsibility evaluation represents one of the most controversial and debated issues in forensic psychiatry. Although clear procedures have been recommended, little research exists on decision-making process by forensic psychiatrists. We present a case assessing the criminal responsibility of a murderer who committed femicide as a result of chloroform poisoning and suffocation after a drug-facilitated sexual assault. MATERIALS AND METHODS: A.S., a 30-year-old female, was found dead in the home of S.P., a 50-year-old male. S.P. recounted killing A.S. by forced inhalatio

The Dynamics of Neuroinflammation in Traumatic Brain Injury: Molecular Markers Useful for Establishing the Post-Traumatic Interval in Forensic Practice.

In forensic pathology, accurately estimating the time since injury is essential. Current histological and imaging approaches commonly miss subtle temporal changes, especially in deaths occurring within hours of injury. This review discusses the timing of neuroinflammation after traumatic brain injury and emphasizes possible markers for estimating the time of injury in forensic cases. Promising markers include microglial activation (allograft inflammatory factor 1 and transmembrane protein 119, detectable within 10 min to 2 h), β-amyloid precursor protein accumulation (20-35 min), high-mobility

Sudden aortic death-proposal for a comprehensive diagnostic approach in forensic and in clinical pathology practice.

BACKGROUNDS: Aortic rupture or dissection as immediate cause of sudden death is encountered in forensic and clinical autopsy practice. Despite a common denominator of 'sudden aortic death' (SAD), we expect that in both settings the diagnostic workup, being either primarily legal or primarily disease related, differs substantially, which may affect the eventual diagnoses. METHODS: We retrospectively reviewed case records of deceased persons who fitted a diagnosis of SAD in the continuous autopsy cohorts in a forensic (Suisse) and a clinical setting (The Netherlands). Clinical characteristics,

A mitochondrial DNA metabarcoding system based on tNGS enables multiplex identification of vertebrate species in samples containing degraded and mixed DNA.

Next-generation sequencing (NGS) has facilitated the widespread use of DNA metabarcoding in fields such as biodiversity, population research, and food adulteration. To improve the identification of animal-derived DNA in samples containing degraded and mixed DNA, a DNA metabarcoding system targeting the mitochondrial Cytb, COI, 16 S rRNA, and 12 S rRNA genes using targeted NGS (tNGS) was developed for vertebrate species. In line with the International Society for Forensic Genetics (ISFG) recommendations for non-human DNA analysis, the forensic performance of the novel assay was systematically e

22q11 deletion syndrome and forensic research: can we go there?

Chromosome 22q11 deletion syndrome (22q11DS) encompasses velocardiofacial syndrome (VCFS), DiGeorge syndrome (DGS), and conotruncal anomaly face syndrome (CTAFS). The disorder may represent the interface between genetics and brain-behavior relationships. As there is a strong relationship between the genetic syndrome and schizophrenia, individuals with the disorder are likely to be disproportionately represented in the criminal justice system. The purpose of this article is to review the 22q11DS in the context of forensic research. The existing literature regarding the syndrome and its relation

Forensic STR and SNP Genotyping of Formalin-Fixed Skeleton Samples With Illumina's ForenSeq System.

Formalin fixatives are widely used in forensics to preserve tissues. However, extracting high-quality genomic DNA from formalin-fixed samples is challenging. Traditional short tandem repeat (STR) analysis using capillary electrophoresis (CE) for forensic DNA typing frequently results in failure. Massively parallel sequencing (MPS) can handle many samples and thousands of genetic markers, usually single-nucleotide polymorphisms (SNPs) and STRs, in a single test. Thus, it is useful for assessing highly deteriorated forensic evidence. Few studies have examined the effectiveness of STRs and SNPs g

Environmental DNA as a tool for detecting illegal wildlife trade.

The illegal wildlife trade presents significant challenges to wildlife conservation due to its large impact on population and species persistence. Forensic technology plays an important role in detecting and prosecuting such trade but has lagged human forensics where trace genetic evidence is important in conviction or exoneration. At present, most genetic applications in wildlife forensics focus on identification of taxa or populations via tissue samples or visible trace material (e.g., blood, hair or feathers). However, enforcement officials may encounter common household objects that are su

A multidisciplinary approach in a forensic case study of a skeletonized child homicide victim.

Child homicide represents a severe form of violent crime, and cases identified exclusively through skeletal remains are uncommon and pose particular forensic challenges, requiring a multidisciplinary investigative approach. We report a Romanian cold case involving a 5-year-old girl who disappeared in 2006 and whose remains were discovered in 2021 in a shallow grave beneath rose bushes on her father's property. Clothing analysis revealed nine perforations consistent with sharp-force trauma, subsequently confirmed through forensic reconstruction. The remains were completely skeletonized and oste

Population genetics of 30 insertion/deletion polymorphisms in Han Chinese population from Zhejiang Province.

Insertion/deletion (InDels) markers can serve as a useful supporting tool to short tandem repeat (STR) typing systems for human identification. The Qiagen DIPplex Investigator kit, which contains 30 biallelic autosomal InDels and amelogenin, has been developed for forensic use. To estimate the genetic diversity of the 30 markers in Han Chinese individuals living in Zhejiang and to further evaluate their applicability in forensic science, 246 unrelated Han Chinese from Zhejiang were genotyped at these loci. No significant departures from Hardy-Weinberg equilibrium were observed at these loci in

Forensic molecular pathology of violent deaths.

In forensic pathology, while classical morphology remains a core procedure to investigate deaths, a spectrum of ancillary procedures has been developed and incorporated to detail the pathology. Among them, postmortem biochemistry is important to investigate the systemic pathophysiological changes involved in the dying process that cannot be detected by morphology. In addition, recent advances in molecular biology have provided a procedure to investigate genetic bases of diseases that might present with sudden death, which is called 'molecular autopsy'. Meanwhile, the practical application of R

Morphological Variations of Coronoid Process in Dry Adult Human Mandibles: A Narrartive Review.

The coronoid process of the mandible is a significant anatomical structure integral to mastication and mandibular stability. It serves as an attachment site for the masseter and temporalis muscles, crucial for jaw movement and function. Beyond its functional roles, it holds therapeutic value in reconstructive craniomaxillofacial surgeries, where it serves as a versatile graft material for addressing osseous defects such as fractures, alveolar defects, and sinus augmentations. Anatomically, it exhibits diverse morphological variations, including triangular, hook-shaped, and rounded forms. These

Sudden Death of a Toddler With Rubinstein-Taybi Syndrome: Forensic Investigation and Pathophysiological Correlation.

Sudden death in childhood is a particularly demanding subject of forensic investigation, especially when a known genetic syndrome coexists. We present the case of the sudden death of a three-year-old female toddler diagnosed with Rubinstein-Taybi syndrome, who was found pulseless and apneic at home and pronounced dead shortly after being transferred to a children's hospital. A comprehensive forensic investigation was conducted, including an autopsy, histological examination, and toxicological analysis. The histopathological findings revealed extensive lesions of severe bronchopneumonia with ne

A 30-InDel Assay for Genetic Variation and Population Structure Analysis of Chinese Tujia Group.

In the present study, thirty autosomal insertion and deletion polymorphic loci were simultaneously amplified and genotyped in a multiplex system, and their allelic frequencies as well as several forensic parameters were obtained in a sample of 236 unrelated healthy Tujia individuals. All the loci were in Hardy-Weinberg equilibrium after applying a Bonferroni correction and all pair-wise loci showed no significant linkage disequilibrium. These loci were observed to be relatively informative and discriminating, quite efficient for forensic applications. Allelic frequencies of 30 loci were compar
