Periodontopathic bacteria and herpesviruses in chronic periodontitis.

Periodontal disease involves complex interactions of microorganisms and host defenses. This work investigated the associations between putative bacterial pathogens, herpesviruses and chronic periodontitis. Subgingival samples were collected from 40 periodontally healthy individuals and from 40 patients with chronic periodontitis with probing depths of < or =3 mm or > or =6 mm. Multiplex and nested polymerase chain reactions were used to identify bacterial pathogens and herpesviruses. Porphyromonas gingivalis, Tannerella forsythia, Epstein-Barr virus (EBV) type 1, cytomegalovirus (CMV), Aggrega

A cross-sectional study on the periodontal status and prevalence of red complex periodontal pathogens in a Japanese population.

This large-scale study cross-sectionally examined the periodontal status and prevalence of "red complex" bacteria (Porphyromonas gingivalis, Treponema denticola, and Tannerella forsythia) in Japanese adults. A total of 977 participants were enrolled in the study. Probing depth (PD), bleeding on probing (BOP), and bone crest level (BCL) were recorded, and the presence of red complex bacteria in the saliva was examined using polymerase chain reaction. The mean BCL value and the percentage of sites with a PD ≥4 mm or the presence of BOP were significantly higher in older participants. The detecti

Red-complex bacteria: immunological background leading to the development of head and neck cancers.

Oral microbiome imbalance is involved in the development of head and neck cancers (HNCs). There is a group of oral pathogens, such as Porphyromonas gingivalis, Tannerella forsythia, and Treponema denticola, that creates red-complex. Currently, the oral pathogens' role in the pathogenesis of periodontitis is well-described; nevertheless, data regarding the link between HNCs and periopathogens are still limited, especially considering T. forsythia and T. denticola. These microbes utilise various virulence factors to promote the carcinogenesis process, affecting the immunological background. This

Plasmin inhibition by bacterial serpin: Implications in gum disease.

Tannerella forsythia is a periodontopathogen that expresses miropin, a protease inhibitor in the serpin superfamily. In this study, we show that miropin is also a specific and efficient inhibitor of plasmin; thus, it represents the first proteinaceous plasmin inhibitor of prokaryotic origin described to date. Miropin inhibits plasmin through the formation of a stable covalent complex triggered by cleavage of the Lys368-Thr369 (P2-P1) reactive site bond with a stoichiometry of inhibition of 3.8 and an association rate constant (kass) of 3.3 × 105 M-1s-1. The inhibition of the fibrinolytic activ

Reduction of two red complex bacteria by sustained-release doxycycline and correlation to improvement in mean pocket depth.

OBJECTIVE: The objective was to evaluate the effects of an 8.5% sustained-release doxycycline-containing polymer formulation (SRDF) on deep pockets (pocket depth [PD] ≥ 7 mm) in chronic periodontitis. Total bacterial counts were used to estimate the number of viable bacteria present before treatment and for up to 6 months posttreatment. METHODS: All sites had PD ≥ 5 mm and bled on probing in 23 subjects who received treatment with SRDF. There was an average of 8.7 teeth or 23 sites for each subject. One deep pocket (≥ 7 mm) in each subject was selected for monitoring. This site was sampled pr
