P21 deficiency exhibits delayed endochondral ossification during fracture healing.

INTRODUCTION: Endochondral ossification is a complex biological phenomenon involving a variety of factors and cells. Cyclin-dependent kinase inhibitor 1 (p21) inhibits cell cycle progression and is affected by external stress. We recently reported that embryonic endochondral ossification is unaffected by endogenous p21 deficiency. In this study, we evaluated whether p21 expression affects endochondral ossification during fracture healing. METHODS: Tibial fractures were introduced into p21 knockout (p21-/-) (n = 24) and wild-type C57BL/6 (p21+/+) (n = 24) mice at age 10 weeks. Fracture healing

Irisin promotes fracture healing by improving osteogenesis and angiogenesis.

BACKGROUND: Osteogenesis and angiogenesis are important for bone fracture healing. Irisin is a muscle-derived monokine that is associated with bone formation. METHODS: To demonstrate the effect of irisin on bone fracture healing, closed mid-diaphyseal femur fractures were produced in 8-week-old C57BL/6 mice. Irisin was administrated intraperitoneally every other day after surgery, fracture healing was assessed by using X-rays. Bone morphometry of the fracture callus were assessed by using micro-computed tomography. Femurs of mice from each group were assessed by the three-point bending testin

Radiologic Assessment of Skull Fracture Healing in Young Children.

BACKGROUND: Skull fractures are commonly seen after both accidental and nonaccidental head injuries in young children. A history of recent trauma may be lacking in either an accidental or nonaccidental head injury event. Furthermore, skull fractures do not offer an indication of the stage of healing on radiologic studies because they do not heal with callus formation as seen with long bone fractures. Thus, a better understanding on the timing of skull fracture resolution may provide guidance on the medical evaluation for accidental or nonaccidental head injury. OBJECTIVE: The aim of the study

Breaking Barriers: Studying Fracture Healing in the BONES Program.

The Bioventus Observational Noninterventional EXOGEN Studies (BONES) Program includes 3 concurrent studies designed to estimate the incidence of fracture nonunions in patients treated with the EXOGEN Ultrasound Bone Healing System compared with those receiving standard fracture care. This article outlines the design and methodology within the fifth metatarsal fracture study; similar approaches are taken in the second and third BONES Program studies, which examine nonunions of the tibia and scaphoid. The BONES Program is an external comparator design and incorporates several unique, fit-for-pur

Enhancement of fracture healing after citrulline supplementation in mice.

Around 10 % of long bone fractures show inadequate bone healing resulting in non-union development. A deregulated arginine-citrulline-nitric oxide metabolism caused by a poor nutritional status of the patients is a risk factor for non-unions. Additionally, previous research in mice with a disrupted arginine to citrulline conversion showed delayed healing. The study hypothesis was that stimulating said metabolism could positively influence the healing process through promotion of collagen synthesis and angiogenesis. Adult wild-type mice underwent a femur osteotomy and plate-screw osteosynthesis

Mechanoregulation modeling of bone healing in realistic fracture geometries.

In bone fracture healing, new tissue gradually forms, ossifies, and eventually remodels itself to restore mechanical stiffness and strength across injury site. Mechanical strain at the fracture site has been implicated in controlling the process of healing and numerical mechanoregulation models with strain-based fuzzy logic rules have been applied to simulate bone healing for simple fracture geometries. However, many of these simplified models cannot capture in vivo observations such as delays in healing with torsional instability or differences in healing rate between different fracture types

Biological methods to enhance bone healing and fracture repair.

This article looks into normal physiological fracture healing with special emphasis on the diamond concept. A precise definition of nonunion of long bones is described. Most often inadequate fixation (too rigid or too loose) is the reason for nonunion in long bone fractures. Because a critical bone defect cannot be bridged, it may lead directly or indirectly (lack of fixation) to nonunion. Individual inadequate local biological characteristics are also often found to be the cause; poor soft tissue coverage as well as a lack of periosteum and muscle or fascia or skin defects can lead to comprom

[The Biological Knife II - Fracture Healing and Pharmacological Interactions].

The reasons for delayed fracture healing or non-union formation are multifactorial. Mechanical factors are well known to influence the process of fracture healing. However, there is a lack of knowledge about the biological conditions that have to be achieved for adequate bone healing. Treatment of non-unions is still demanding and we have only poor options to stimulate the bone healing process. Therefore, it is important to optimise the biological conditions to avoid non-union formation. Beside patient-related factors, pharmacological drugs are able to impede or stimulate fracture healing. Computational modeling of ultrasonic backscattering to evaluate fracture healing.

Fracture healing is a complex, regenerative procedure including several phases of recovery as the original mechanical and geometrical features of bone are gradually restored. Ultrasonic evaluation of bone pathologies such as osteoporosis and fracture healing has recently gained significant interest due to the non-invasive and non-radiating nature of the method. In this study, we present numerical simulations of ultrasonic backscattering in simple, two dimensional geometries of healing long bones to investigate the monitoring capacity of the acoustic pressure in the backward direction. The SERM raloxifene improves diaphyseal fracture healing in mice.

Although several studies reported that raloxifene treatment improves postmenopausal osteoporotic bone structure and reduces fracture risk, only a few animal and no human studies have examined its effects on the fracture healing process. Thus the aim of the present study was to determine, whether systemic application of the selective estrogen receptor modulator raloxifene promotes fracture healing compared to untreated control-, estrogen-deficient-, as well as estrogen-treated mice using a standardized femoral osteotomy model (n = 60 mice). Ten days after surgery, contact radiography and undeca

Calcium homeostasis influences radiological fracture healing in postmenopausal women.

INTRODUCTION: Recent studies suggest that calcium and 25-[OH]-cholecalciferol represent substantial co-factors in fracture healing. However, there still seems to be no sustainable consensus regarding the influence on fracture healing patterns. In this study, the influence of calcium and vitamin D levels on fracture callus formation was prospectively analysed using pQCT scan. METHODS: 94 postmenopausal females with distal radius fractures and consecutive surgery were included. Calcium, 25-[OH]-cholecalciferol, parathyroid hormone and bone-specific alkaline phosphatase levels were obtained prio

Demineralized bone matrix for fracture healing: fact or fiction?

Demineralized bone matrix (DBM) has been touted as an excellent grafting material; however, there are no Level I studies that use DBM alone in humans to back up this claim. DBM functions best in a healthy tissue bed but should be expected to have little impact in an anoxic or avascular tissue bed, a situation often encountered in traumatic orthopaedic pathologies. Moreover, there is some evidence of differential potencies of DBM preparations based on donor variability and the manufacturing process. DBM efficacy may also be related to its formulation and the various carriers used. The fact that

Periodontal healing after bonding treatment of vertical root fracture.

Vertical root fractures lead to advanced periodontal breakdown with deep periodontal pockets and vertical bone defects. The purpose of this study is to evaluate clinically the periodontal healing of root fracture treatment using adhesive resin cement. In 22 patients, 23 teeth with vertical root fractures were treated with 4-META/MMA-TBB resin cement. Eleven fractured roots were bonded through the root canal (group A) and 12 fractured roots were bonded extra-orally and replanted (group B). All teeth were then restored with full cast crowns (n=20) or coping (n=3). Mean probing depth was 6.6 mm a

Does helical plating for proximal humeral shaft fractures benefit bone healing? - an in silico analysis in fracture healing.

OBJECTIVES: Helical plating is an established alternative to straight plating for humeral shaft fractures in order to prevent iatrogenic radial nerve damage. However, a previous biomechanical investigation indicated differences in fracture healing for helical plating due to a potential shift of interfragmentary movements compared to straight plating. Therefore, fracture healing simulations were performed to assess any differences in bone healing of helical vs. straight plating. METHODS: A systematic workflow for fracture healing analytics was created, covering essential steps of bone modellin
