Fumonisin B1 (FB1) is a mycotoxin frequently found in agricultural commodities, and poses a considerable risk for human and animal health. The aim of this study was to investigate the toxic effect of FB1 in mice intestine. Male Kunming mice (n = 40) were treated with FB1 diet for 42 days. Histopathological and biochemical analyses, including ion concentrations, transcription of ATPase subunits and mRNA expression of cytochrome P450s (CYP450s) analyses were performed on duodenum, cecum and colon of mice. The results revealed that FB1 caused histological alterations, including partial shedding o

Male Wistar rats were intraperitoneally dosed with fumonisin B1 (FB1; 0, 20, 50 and 100 mg kg-1 dietary dose equivalent) for 5 & 10 days to assess dose- and time-dependent effects on renal and hepatic phosphatidylcholine (PC), phosphatidylinositol (PI) and phosphatidylethanolamine (PE) fatty acid (FA) profiles. Renal PC showed increasing FA saturation (SAT) after 5 days; after 10 days polyunsaturation (PUFA) decreased markedly (Σ n3 (total n3), Σ n6, PUFA, unsaturation index (UI) and average FA chain length (ACL)), mostly with linear dose response. In the PI FAs similar changes were observed,

Fu monisin B1 (FB1) is a mycotoxin commonly found in maize and maize-based products. Ingestion of FB1-contaminated causes a myriad of dose- and species-dependent toxic effects to human and animal health. In the present study we evaluated the effects of FB1 (8 mg/kg, i.p. for 4 days) on body weight and oxidative stress parameters in the liver, kidney and lung of C57BL/6 male mice. No changes in the organ-to-body weight ratio, organ-to-adrenal gland weight ratio or organ-to-brain weight ratio were found. On the other hand, FB1 exposure increased NPSH levels in liver and lungs whereas decreased F

The influences of dietary fumonisin B(1) (FB(1)), a metabolite of Fusarium verticillioides, on the onset of puberty, semen quality, fertility rates and testicular morphology in male rabbits (bucks) were studied. Forty male rabbits were randomly assigned and fed four diets containing 0.13, 5.0, 7.5 and 10.0 mg FB(1)/kg, constituting diets 1 (control), 2, 3 and 4 respectively, for a period of 175 days in a completely randomized design. During the last week of the feeding trial, two untreated female rabbits were mated to each of the four treated bucks per treatment to assess the fertility rate of

Fumonisins, mycotoxins produced by Fusarium moniliforme and a number of other fungi, cause neuronal degeneration, liver and renal toxicity, cancer, and other injury to animals. Recent work with rat hepatocytes (Wang, E., Norred, W. P., Bacon, C. W., Riley, R. T., and Merrill, A. H., Jr. (1991) J. Biol. Chem. 266, 14486-14490) found that fumonisins block sphingosine biosynthesis by inhibiting the conversion of sphinganine to dihydroceramides, which precedes introduction of the 4,5-trans-double bond of sphingosine. The current study utilized mouse cerebellar neurons in culture to evaluate how th

The purpose of the present study was to investigate the effects of phytic acid (IP6) on morphological and immunohistochemical parameters and oxidative stress response in intestinal explants of pigs exposed to fumonisin B₁ (FB₁) and/or deoxynivalenol (DON). The jejunal explants were exposed to the following treatments: vehicle, IP6 5 mM, DON 10 µM, FB₁ 70 µM, DON 10 µM + FB₁ 70 µM, DON 10 µM + IP6 5 mM, FB₁ 70 µM + IP6 5 mM, and DON 10 µM + FB₁ 70 µM + IP6 5 mM. The decrease in villus height and goblet cell density was more evident in DON and DON + FB₁ treatments. In addition, a significant inc

Fusarium ear rot, caused by Fusarium verticillioides, is one of the most common diseases of maize, causing yield and quality reductions and contamination of grain by fumonisins and other mycotoxins. Drought stress and various insects have been implicated as factors affecting disease severity. Field studies were conducted to evaluate the interactions and relative influences of drought stress, insect infestation, and planting date upon Fusarium ear rot severity and fumonisin B1 contamination. Three hybrids varying in partial resistance to Fusarium ear rot were sown on three planting dates and su

Fumonisins are mycotoxins found primarily in corn and corn products that are produced by Fusarium verticillioides, F. proliferatum, and several other Fusarium species. The toxicity of fumonisin B1 (FB) from culture material with and without activated carbon was evaluated using weanling piglets. Fifty-six weanling pigs were assigned to one of four treatments diets based on BW. The treatment diets were 1) control = corn-soybean basal diet with < 2 ppm FB; 2) AC = control + activated carbon at 1% of the diet, as fed; 3) FB = control + culture material (formulated to contain 30 ppm FB, as-fed basi

We have established an Arabidopsis protoplast model system to study plant cell death signaling. The fungal toxin fumonisin B1 (FB1) induces apoptosis-like programmed cell death (PCD) in wild-type protoplasts. FB1, however, only marginally affects the viability of protoplasts isolated from transgenic NahG plants, in which salicylic acid (SA) is metabolically degraded; from pad4-1 mutant plants, in which an SA amplification mechanism is thought to be impaired; or from jar1-1 or etr1-1 mutant plants, which are insensitive to jasmonate (JA) or ethylene (ET), respectively. FB1 susceptibility of wil

Fumonisins are mycotoxins produced by Fusarium moniliforme and other Fusarium species. They are commonly found in corn and corn-based foodstuffs. Fumonisins inhibit sphingolipid (SL) biosynthesis, alter cellular SL profiles, and thus may affect cell proliferation and differentiation, both of which are important processes for reproduction. However, there are few reports of the effects of F. moniliforme or fumonisins on mammalian reproduction or development. To study the reproductive effects of this fungus, diets formulated with culture material of toxic F. moniliforme strain MRC 826 (CM) to pro

Ochratoxin A (OTA), Zearalenone (ZEA), and Fumonisins (FB) are common contaminants of poultry feed associated with oxidative damage and potentially dangerous residues in products from exposed animals. We investigated the molecular effects in broilers of a short-term (10 days) dietary exposure to OTA (0.26 mg/kg), ZEA (2.9 mg/kg), or FB (60 mg/kg) on cytochrome P450 enzymes (CYP), drug transporters (DT) and the antioxidant defence system. OTA markedly decreased serum antioxidant capacity, while all mycotoxins depressed reduced glutathione content and increased lipid peroxidation in the liver, i

Poultry feed is often contaminated with fumonisins, deoxynivalenol, and zearalenone, which can result in oxidative damage, inflammation and change in lipid metabolism. Although sphingolipids play key roles in cells, only the effects of fumonisins on the sphingolipidome are well-documented. In chickens, fumonisins have been shown to increase the sphinganine to sphingosine ratio and the C22-24:C16 sphingolipid ratio, which has been proposed as a new biomarker of toxicity. In this study, we used UHPLC-MSMS targeted analysis to measure the effect of fusariotoxins on sphingolipids in the livers of

Field trials based on manual infestation of the Asian corn borer (ACB) (Ostrinia furnacalis [Guenée]) and Fusarium verticillioides (Nirenberg) atomization were conducted on four maize hybrids to investigate the relationship between ACB infestation and F. verticillioides infection, yield loss, and fumonisin contamination in maize. Analysis of fumonisins B1 and B2 was carried out using an LC-MS/MS system. In this study, manual ACB infestation significantly promoted F. verticillioides infection (both symptomatic and symptomless) and grain fumonisin levels. Ear rot incidence and severity, symptoml

SCOPE: Fumonisin B1 (FB1 ) is found in corn-based foods and is a possible risk factor for neural tube defects (NTDs). The mechanism(s) underlying NTD induction by FB1 in LM/Bc mice is not understood; however, evidence suggests disrupted folate transport is involved.
METHODS AND RESULTS: Female LM/Bc mice were fed folate-sufficient (control) or folate-deficient diet beginning 5 wk before mating, treated with 0 (vehicle), 2.5 or 10 mg/kg FB1 by intraperitoneal injection on embryonic days 7 (E7) and E8, and their fetuses examined on E16. Dose-dependent NTD induction was found in groups fed the c

Fumonisin B1 (FB1) induces apoptosis and decreases the cellular uptake of 5-methyltetrahydrofolate. Two folate transporters (folate receptor, FR, and Reduced Folate Carrier, hRFC1) are involved in the cell uptake of folate. We aimed to study whether FB1 modifies the expression of the FR and the hRFC1 and whether its apoptotic effect is influenced by folate. Incubation of HepG2 cells with FB1 induced apoptosis in concentration and time-dependent manner in complete medium (experimental control medium, ECM), as well as in folate-depleted medium (FDM). FDM increased the toxicity of FB1 as the cell

Fumonisin B1 (FB1) is a mycotoxin produced by Fusarium verticillioides, commonly present in corn and other cereals. Exposure to FB1 causes organ-specific diseases in various species, e.g., equine leukoencephalomalacia and porcine pulmonary edema; in mice the response is hepatotoxicity. We earlier reported that ceramide synthase inhibition by FB1, the initial biochemical effect of this mycotoxin, results in modulation of cytokine network in response to accumulated free sphingoid bases. In the current study we used NZB/NZW-F1 (NZBW) mice that have modified cytokine expression and develop lupus b

Consumption of corn or corn-based products contaminated with Fusarium moniliforme/fumonisins has been associated with a variety of animal and human diseases and is a major food/feed safety issue. This study focused on the clinical toxicity and performance parameters in growing swing exposed to low to moderate levels of pure fumonisin B1 (FB.) for 8 weeks. Male (castrated) and female pigs were fed diets containing 0,0.1,1.0, and 10 mg FB1/kg diet (ppm). Weight gains and feed consumption were measured weekly. Blood samples were collected throughout the study, and various clinical and hematologic

Maize-soybean succession dominates grain production in tropical Brazil, and rotation is often recommended to improve soil health. However, few field studies have followed both soil bacterial diversity and fumonisin-producing pathogens over several seasons in tropical no-till systems. We compared continuous maize, continuous soybean and maize-soybean rotation for three seasons (2016-2017 to 2018-2019). In a fourth season (2019-2020), maize was grown in all plots to test legacy effects. Bacterial communities were profiled by 16 S rRNA (V3-V4) amplicon sequencing. The fum1 gene of fumonisin-produ

Fumonisin B1 (FB1), a highly toxic secondary metabolite produced by Fusarium fungi, poses a significant threat to global food safety; therefore, sensitive, selective, and reliable analytical methods are urgently needed. In this work, we report a novel molecularly imprinted electrochemiluminescence (ECL) sensor for the quantitative determination of FB1. A Co/Zn bimetallic metal-organic framework@polyoxometalate (Co/Zn-MOF@POMs) nanocomposite was synthesized via a one-pot strategy and used to modify a glassy carbon electrode. The incorporated polyoxometalates (POMs) markedly improved the interfa

Fumonisin B1 (FB1), a worldwide contaminating mycotoxin, can cause global food issue. It has been reported that FB1 is related to chronic kidney disease of unknown etiology. However, the study of FB1-induced nephrotoxicity in vitro is very limited and the mechanism is unknown. Human renal tubule epithelial (HK-2) cells were used in this study. The results showed that FB1 exposure could decrease cell viability, induce cell apoptosis and up-regulate the expression of Kim-1, collagen I, α-SMA and TGF-β1. In addition, autophagy was activated after FB1 exposure, including the conversion of LC3 and
