Hypoenergetic nutrition support in hospitalized obese patients: a simplified method for clinical application.

Nutrition support in obese hospitalized patients is controversial, with some practitioners advocating restricted energy or hypoenergetic feedings when patients are being actively treated for another disease. To eliminate the need for indirect calorimetry, this randomized, double-blind, prospective study was undertaken to determine whether obese hospitalized patients given a hypoenergetic parenteral regimen administered to provide 2 g protein x kg ideal body wt (IBW)(-1) x d(-1), could achieve nitrogen balance comparable with that of control subjects given isonitrogenous normoenergetic formula.

The formation, characteristic, and stability of oat protein-hyaluronic acid complexes on perilla oil emulsion and its application in plant-based mayonnaise.

This study explored the formation mechanism of the oat protein and hyaluronic acid complex, applying it to a stable in-phase highly emulsified system (HIPE) for protecting perilla oil and as a fat substitute in low-fat mayonnaise. Results showed that hyaluronic acid (0.25-2.0 wt%) increased electrostatic repulsion and induced structural changes in oat protein, including the elevated β-sheet content, reduced surface hydrophobicity, and quenched intrinsic fluorescence. Molecular docking confirmed hydrogen bonding and hydrophobic interactions between oat protein and hyaluronic acid. At 1.5 % hyal

The 12-LOX/12-HETE/GPR31 metabolic pathway promotes tumor-associated macrophage M2 polarization mediated pancreatic cancer development.

BACKGROUND: The 12-lipoxygenase (12-LOX)/12-hydroxyeicosatetraenoic acid (12-HETE) pathway is associated with various tumors. M2 macrophages in the tumor microenvironment promote tumorigenesis and progression. However, the role of the 12-LOX/12-HETE/G protein-coupled receptor 31 (GPR31) metabolic pathway and its relationship with M2 macrophages remains unclear in pancreatic cancer (PC). METHODS: The expression levels of 12-LOX, GPR31, and 12-HETE were detected in PC and mouse PC models using western blot and enzyme-linked immunosorbent assays (ELISA). In vivo and in vitro experiments were con

GWAS and network analysis of co-occurring nicotine and alcohol dependence identifies significantly associated alleles and network.

Alcohol dependence (AD) and nicotine dependence (ND) co-occur frequently (AD+ND). We integrated SNP-based, gene-based, and protein-protein interaction network analyses to identify shared risk genes or gene subnetworks for AD+ND in African Americans (AAs, N = 2,094) and European Americans (EAs, N = 1,207). The DSM-IV criterion counts for AD and ND were modeled as two dependent variables in a multivariate linear mixed model, and analyzed separately for the two populations. The most significant SNP was rs6579845 in EAs (p < 1.29 × 10-8 ) in GM2A, which encodes GM2 ganglioside activator, and is a

Endothelial dysfunction accelerates AKI-to-CKD transition by promoting β-catenin activation in macrophages.

Acute kidney injury (AKI) frequently progresses to chronic kidney disease (CKD), resulting in long-term renal dysfunction. Although traditional risk factors such as hypertension, diabetes, and aging contribute to this transition, endothelial dysfunction has emerged as a central mediator. In a murine model of severe ischemia-reperfusion injury (IRI), we observed persistent fibrosis with sustained activation of β-catenin signaling, especially when there is an endothelial nitric oxide synthase (eNOS) deficiency. Impaired nitric oxide (NO)-cyclic guanosine monophosphate (cGMP)-protein kinase G (PK

High-speed shearing assisted genipin crosslinking of soybean lipophilic protein: insights into conformational rearrangement and interfacial modulation.

Native soybean lipophilic protein (SLP) readily forms compact aggregates through strong hydrophobic interactions, impairing its water solubility and interfacial adsorption capacity. This study investigated the effects of high-speed shearing followed by genipin crosslinking on the structural and functional properties of SLP. Spectroscopic analyses revealed enhanced exposure of tryptophan and tyrosine residues, indicating that shearing induced conformational unfolding of SLP. Subsequent genipin crosslinking resulted in a clear blue shift of the amide II band, confirming the formation of covalent

Mucosal delivery of a respiratory syncytial virus CTL peptide with enterotoxin-based adjuvants elicits protective, immunopathogenic, and immunoregulatory antiviral CD8+ T cell responses.

In an effort to develop a safe and effective vaccine against respiratory syncytial virus (RSV), we used Escherichia coli heat-labile toxin (LT), and LTK63 (an LT mutant devoid of ADP-ribosyltransferase activity) to elicit murine CD8(+) CTL responses to an intranasally codelivered CTL peptide from the second matrix protein (M2) of RSV. M2(82-90)-specific CD8(+) T cells were detected by IFN-gamma enzyme-linked immunospot and (51)Cr release assay in local and systemic lymph nodes, and their induction was dependent on the use of a mucosal adjuvant. CTL elicited by peptide immunization afforded pro

Succinate drives macrophage inflammation by promoting GPR91 mitochondrial signaling in rheumatoid arthritis.

High succinate concentrations are implicated in rheumatoid arthritis (RA) and other inflammatory diseases through G protein-coupled receptor 91 (GPR91)-mediated signaling. Despite the therapeutic potential of targeting GPR91, conflicting reports on the receptor's inflammatory roles have hindered treatment development. Here, we report that the effects of succinate on GPR91 signaling are biphasic and concentration dependent. At physiological succinate concentration, membrane-localized GPR91 promotes M2 polarization through Gq-mediated activation of phospholipase C and intracellular calcium mobil

Bi-layered living cellular constructs promote recruitment and activation of an M2 macrophage phenotype.

BACKGROUND: Skin substitutes, also known as Cellular and/or Tissue-based Products (CTPs), and wound dressings often are designed to support the innate wound healing response of the patient. The mechanism of action for these products varies, with some products strictly acting as bacterial barriers while others can alter the wound microenvironment by providing additional extracellular matrix components or modulating the inflammatory response. Bi-layered living cellular constructs (BLCCs) are CTPs that mimic human skin and can heal itself in response to wounding in vitro. How could signals from B

Differential regulation of chemokine production by Fcgamma receptor engagement in human monocytes: association of CCL1 with a distinct form of M2 monocyte activation (M2b, Type 2).

CC chemokine ligand 1 (CCL1; I-309) is a CC chemokine that interacts with CC chemokine receptor 8, which is preferentially expressed in polarized T helper cell type 2 and Tc2 cells, in eosinophils, and in T regulatory cells. The present study, prompted by transcriptional profiling of human monocytes undergoing different forms of activation, was designed to characterize the production of CCL1 in monocytes compared with the production of other chemokines (CCL2, CCL22, and CCL18) differentially regulated by distinct activation signals. Lipopolysaccharide (LPS), interferon-gamma (IFN-gamma), inter

Long term therapeutic effects of icariin-loaded PLGA microspheres in an experimental model of optic nerve ischemia via modulation of CEBP-β/G-CSF/noncanonical NF-κB axis.

An ischemic insult at optic nerve (ON) is followed by detrimental neuroinflammation that results in progressive and long-lasting retinal ganglion cell (RGC) death and vision loss. Icariin was reported to be a safe and effective natural anti-inflammatory drug. Herein, we evaluated the long-term therapeutic effects of a single intravitreal injection of poly(lactide-co-glycolide) PLGA-icariin in a rat model of anterior ischemic optic neuropathy (rAION). Treatment with PLGA microspheres of icariin preserved the visual function and RGC density for 1 month in the rAION model. In addition, ON edema a

Dual modification of rice bran protein by enzymatic hydrolysis and tannic acid: Impacts on structural characteristics and emulsifying properties.

Rice bran protein (RBP) exhibits limited functional properties because of its compact structure, which reduces its interfacial adsorption capacity and consequently constrains its food applications. This study combined moderate enzymatic hydrolysis and non-covalent complexation with tannic acid (TA) to improve RBP functionality. Moderate hydrolysis combined with TA induced protein unfolding via hydrophobic forces and hydrogen bonding. This effectively enhanced the structural flexibility of RBP and improved its interfacial properties, significantly enhancing its emulsifying and foaming capacity.

Conformational Complexity and Dynamics in a Muscarinic Receptor Revealed by NMR Spectroscopy.

The M2 muscarinic acetylcholine receptor (M2R) is a prototypical GPCR that plays important roles in regulating heart rate and CNS functions. Crystal structures provide snapshots of the M2R in inactive and active states, but the allosteric link between the ligand binding pocket and cytoplasmic surface remains poorly understood. Here we used solution NMR to examine the structure and dynamics of the M2R labeled with 13CH3-ε-methionine upon binding to various orthosteric and allosteric ligands having a range of efficacy for both G protein activation and arrestin recruitment. We observed ligand-spe
