The role of hepcidin and haemojuvelin in the pathogenesis of iron disorders in patients with severe malnutrition.

INTRODUCTION AND OBJECTIVE: The clinical consequences of malnutrition are multi-directional and result in dysfunctions of the majority of internal organs and systems. The results of recent studies suggest that a significant role is played by malnutrition in pathophysiology of iron homeostasis disorders, but the underlying mechanism is unclear. The study describes the potential role of hepcidin and hemojuvelin in the pathogenesis of disorders of iron metabolism during malnutrition. STATE OF KNOWLEDGE: The participation of hepcidin in regulating iron homeostasis encompasses inhibiting the absor

Serum total sialic acid and regan isoenzyme levels in patients with lung cancer.

Serum total sialic acid (TSA), Regan isoenzyme (RI) (heat stable placenta like alkaline phosphatase) and TSA/Total protein (TP) ratio were estimated in 44 cases of Lung cancer using highly specific chemical methods to assess their efficacy as malignancy markers. Levels of the two markers were compared with 27 matched healthy controls. A significant increase (P less than 0.001) in both the bio-markers were compared with 27 matched healthy controls. A significant increase (P less than 0.001) in both the bio-markers was observed compared to controls. TSA/TP ratio was significantly higher (P less

ER entry pathway and glycosylation of GPI-anchored proteins are determined by N-terminal signal sequence and C-terminal GPI-attachment sequence.

Newly synthesized proteins in the secretory pathway, including glycosylphosphatidylinositol (GPI)-anchored proteins (GPI-APs), need to be correctly targeted and imported into the endoplasmic reticulum (ER) lumen. GPI-APs are synthesized in the cytosol as preproproteins, which contain an N-terminal signal sequence (SS), mature protein part, and C-terminal GPI-attachment sequence (GPI-AS), and translocated into the ER lumen where SS and GPI-AS are removed, generating mature GPI-APs. However, how various GPI-APs are translocated into the ER lumen in mammalian cells is unclear. Here, we investigat

Three-finger snake neurotoxins and Ly6 proteins targeting nicotinic acetylcholine receptors: pharmacological tools and endogenous modulators.

Snake venom neurotoxins and lymphocyte antigen 6 (Ly6) proteins, most of the latter being membrane tethered by a glycosylphosphatidylinositol (GPI) anchor, have a variety of biological activities, but their three-finger (3F) folding combines them in one Ly6/neurotoxin family. Subsets of two groups, represented by α-neurotoxins and Lynx1, respectively, interact with nicotinic acetylcholine receptors (nAChR) and, hence, are of therapeutic interest for the treatment of neurodegenerative diseases, pain, and cancer. Information on the mechanisms of action and 3D structure of the binding sites, whic

The in vitro effect of omentin-1 on the global proteome of granulosa cells from normal weight Large White and fat Meishan pigs.

Ovarian granulosa cells (Gc) play a vital role in follicle maturation and successful ovulation. Omentin-1 (ITLN1) is an adipokine involved in energy metabolism and insulin resistance; its expression has been demonstrated in the ovary and varies depending on the degree of pig fatness. However, its effect on the global proteome of Gc has not been previously investigated. It was hypothesized that ITLN1 affects the abundance of proteins involved in key processes occurring in Gc in pigs. Liquid chromatography-tandem mass spectrometry (LC-MS/MS) analysis of Gc identified 208 significantly differenti

Injectable Hydrogel-Based Delivery of Soluble Cripto Protein Enhances Repair After Skeletal Muscle Injury.

Injectable protein therapies offer a minimally invasive approach for enhancing skeletal muscle regeneration, yet their clinical translation has been limited by poor in vivo stability, rapid diffusion, and insufficient temporal bioavailability at the injury site. Here, we report a thermoresponsive, fully injectable Pluronic F127-fibrinogen (FF) hydrogel that enables temporally synchronized delivery of a soluble form of Cripto, a GPI-anchored co-receptor that promotes myogenic regeneration by counteracting myostatin signaling. Cripto is physically entrapped within the FF precursor and undergoes

Dynamic inter-glycan interactions regulate site-specific N-glycan maturation in human Fcγ receptor III.

Protein N-glycosylation generates diverse glycoforms that influence protein structure and function, yet the molecular rules governing site-specific glycan maturation remain incompletely understood. Accumulating evidence indicates that glycosylation outcomes are shaped by determinants embedded within glycoproteins themselves, beyond enzyme availability alone. Here, we investigate how neighboring glycans modulate site-specific N-glycan maturation in human Fcγ receptor III (FcγRIII). Building on previous glycoproteomic studies showing restricted processing of the N45 glycan in FcγRIII molecules,

ERC/Mesothelin Is Associated with the Formation of Microvilli on the Mesothelium and Has Limited Functional Relevance Under Physiological Conditions.

In adults, expressed in renal cancer (ERC)/mesothelin is exclusively expressed in the mesothelial cells lining the pleural, pericardial, and peritoneal cavities, yet its function under physiological conditions is unknown. To explore this, we studied ERC expression in wild-type (WT) mice at different developmental stages by immunohistochemistry and analyzed the ultrastructure of the mesothelium in WT and Erc-knockout (KO) mice via electron microscopy. Additionally, cardiopulmonary function in adult WT and Erc-KO mice was assessed using echocardiography and the forced oscillation technique (FOT)

An engineered T-cell engager with selectivity for high mesothelin-expressing cells and activity in the presence of soluble mesothelin.

Mesothelin (MSLN) is an attractive immuno-oncology target, but the development of MSLN-targeting therapies has been impeded by tumor shedding of soluble MSLN (sMSLN), on-target off-tumor activity, and an immunosuppressive tumor microenvironment. We sought to engineer an antibody-based, MSLN-targeted T-cell engager (αMSLN/αCD3) with enhanced ability to discriminate high MSLN-expressing tumors from normal tissue, and activity in the presence of sMSLN. We also studied the in vivo antitumor efficacy of this molecule (NM28-2746) alone and in combination with the multifunctional checkpoint inhibitor

Bone morphogenic proteins in iron homeostasis.

The bone morphogenetic protein (BMP)-SMAD signaling pathway plays a central role in regulating hepcidin, which is the master hormone governing systemic iron homeostasis. Hepcidin is produced by the liver and acts on the iron exporter ferroportin to control iron absorption from the diet and iron release from body stores, thereby providing adequate iron for red blood cell production, while limiting the toxic effects of excess iron. BMP6 and BMP2 ligands produced by liver endothelial cells bind to BMP receptors and the coreceptor hemojuvelin (HJV) on hepatocytes to activate SMAD1/5/8 signaling, w

Mmp17b is essential for proper neural crest cell migration in vivo.

The extracellular matrix plays a critical role in neural crest (NC) cell migration. In this study, we characterize the contribution of the novel GPI-linked matrix metalloproteinase (MMP) zebrafish mmp17b. Mmp17b is expressed post-gastrulation in the developing NC. Morpholino inactivation of mmp17b function, or chemical inhibition of MMP activity results in aberrant NC cell migration with minimal change in NC proliferation or apoptosis. Intriguingly, a GPI anchored protein with metalloproteinase inhibitor properties, Reversion-inducing-Cysteine-rich protein with Kazal motifs (RECK), which has p

Detection of the GPI-anchorless prion protein fragment PrP226* in human brain.

BACKGROUND: The accumulation of the misfolded forms of cellular prion protein, i.e. prions (PrPSc), in the brain is one of the crucial characteristics of fatal neurodegenerative disorders, called transmissible spongiform encephalopathies (TSEs). Cellular prion protein is normally linked to the cell surface by the glycosylphosphatidylinositol (GPI) anchor. There is accumulating evidence that the GPI-anchorless prion protein may act as an accelerator of formation and propagation of prions. In the TSE affected human brain we have previously discovered a novel GPI-anchorless prion protein fragment

Membrane traffic in polarized neurons in culture.

Fetal hippocampal neurons develop axons and dendrites in culture. To study how neurons form and maintain different plasma membrane domains, hippocampal neurons were infected with RNA viruses and the distribution of the viral glycoproteins was analyzed by light and electron microscopy. Infection of hippocampal cells with vesicular stomatitis virus (VSV) and fowl plague virus (FPV) resulted in the polarized distribution of the newly synthesized viral glycoproteins. The VSV glycoprotein appeared firstly in the Golgi apparatus and then in the dendrites. In contrast, the hemagglutinin of FPV, after

The role of GPI-anchored LY6/uPAR family proteins in connecting membrane microdomains with immune regulation and diseases.

Glycosylphosphatidylinositol (GPI)-anchored LY6/uPAR family proteins are structurally conserved yet functionally diverse regulators of immune signaling. Defined by a characteristic three-finger LY6/uPAR (LU) domain and preferential localization to lipid rafts, members such as LY6A, LY6C, LY6E, LY6G, CD59, PSCA, and uPAR orchestrate essential immune processes, including T and B cell activation, dendritic cell maturation, neutrophil and natural killer (NK) cell responses, and macrophage polarization. These proteins are increasingly implicated in the pathogenesis of cancer, infectious diseases, a

Decreased prion protein expression in human peripheral blood leucocytes from patients with paroxysmal nocturnal haemoglobinuria.

The cellular isoform of the prion protein (PrPC) is a cell surface glycoprotein attached to the outer leaflet of the plasma membrane by a glycosylphosphatidyl-inositol (GPI) anchor. PrPC is involved in the pathogenesis of prion diseases and has recently been shown to play a role in haemopoietic cell activation and proliferation. We have used the PrPC-specific monoclonal antibody (mAb) 3F4 in a flow cytometry approach to analyse the constitutive expression of PrPC on human peripheral blood (PB) cell populations from patients with paroxysmal nocturnal haemoglobinuria (PNH), which are characteriz

Red cell membrane polypeptides under normal conditions and in genetic disorders.

Broadly speaking, the red cell membrane is comprised of --a cholesterol-rich phospholipid bilayer that is studded by a large number of trans-bilayer proteins, --of glycosylphosphatidylinositol-anchored proteins (GPI-proteins) standing outside, and --an important protein assembly, the erythrocyte or membrane skeleton, that laminates the inner surface of the bilayer. Among the trans-bilayer proteins, one finds the anion exchanger, the glycophorins, the glucose transporter, a variety of cation transporters and pumps, and of course proteins carrying the epitopes of many blood groups. Among the GPI

Aberrantly expressed CEACAM6 is involved in the signaling leading to apoptosis of acute lymphoblastic leukemia cells.

OBJECTIVE: The aberrant expression of myeloid antigens on acute lymphoblastic leukemia (ALL) cells is a well-documented phenomenon. So far, there have been no reports of a functional consequence of this aberrant expression. The granulocytic marker carcinoembryonic antigen-related cell adhesion molecule 6 (CEACAM6, CD66c) is a GPI-anchored molecule that is reported to be the most frequently aberrantly expressed myeloid marker in ALL with a strong correlation with genotype. MATERIALS AND METHODS: We mimicked CEACAM6 signaling in ALL cells by cross-linking with anti-CEACAM6 antibody. Next, we me

Mesothelin in pancreatic cancer: clinical applications and analytical considerations.

One of the most prevalent and aggressive kinds of pancreatic cancer (PC) worldwide is pancreatic ductal adenocarcinoma (PDAC). It has no preliminary symptoms and poses significant challenges for diagnosis in the early stages. Due to late diagnosis, it shows resistance to standard treatment. Despite improvements in cancer care, the five-year data depict a low survival rate. Current status of PDAC demands exploration of effective, non-surgical biomarkers that can facilitate rapid diagnosis, prognosis, and monitoring of treatment, which has put mesothelin (MSLN) in the spotlight. MSLN is a transm

The role of adipokines in the pathogenesis of psoriasis - a focus on resistin, omentin-1 and vaspin.

BACKGROUND: Psoriasis is a chronic immune-mediated skin condition with several types of manifestation, including psoriatic arthritis. In recent years, studies have demonstrated multiple molecules and mechanisms that play important roles in the pathophysiology of psoriasis. Studies have been conducted to determine the role of adipokines, bioactive peptides secreted by the adipose tissue, in the pathogenesis of inflammatory diseases. These studies have shown that adipokines are dysregulated in psoriasis and their abnormal expression profile could contribute to the inflammatory mechanisms observe
