Nervous system development related gene expression regulation in the zebrafish embryo after exposure to valproic acid and retinoic acid: A genome wide approach.

The evaluation of chemical and pharmaceutical safety for humans is moving from animal studies to New Approach Methodologies (NAM), reducing animal use and focusing on mechanism of action, whilst enhancing human relevance. In developmental toxicology, the mechanistic approach is facilitated by the assessment of predictive biomarkers, which allow mechanistic pathways perturbation monitoring at the basis of human hazard assessment. In our search for biomarkers of maldevelopment, we focused on chemically-induced perturbation of the retinoic acid signaling pathway (RA-SP), a major pathway implicate

Developmentally Regulated Expression and Activity of Sulphotransferases in the Rat Choroid Plexuses.

During development, entry of any substances from the circulation into the brain is tightly regulated by a series of blood-brain interfaces. Notably, the choroid plexuses, which form the blood-cerebrospinal fluid barrier, serve as a key interface for molecular exchange in early life. Control mechanisms within the choroid plexuses include efflux transporters and conjugating enzymes, such as glutathione S-transferases and UDP-glucuronosyltransferases, which have been shown to play key roles in safeguarding the developing brain. Sulphotransferases are another family of conjugating enzymes reported

Screening of upregulated genes in suckling mouse central nervous system during the disease stage of rabies virus infection.

The pathogenesis of hydrophobia remains unclear. The aim of this study was to identify the differentially upregulated genes that correlated with disease development in an experimental mouse model to provide better understanding of pathological mechanisms in rabies. The present work employed Clontech mouse array 1.2 II containing 1,176 gene transcripts. Suckling mice were intracerebrally infected with canine rabies virus. The gene expression profiles on day 2, 4 and 6 post inoculation were followed. The results show genes whose expression increased at least twofold above the control, mock-infec

A BAC transgenic reporter recapitulates in vivo regulation of human telomerase reverse transcriptase in development and tumorigenesis.

Telomerase is tightly regulated in humans relative to mice, owing to the differential regulation of TERT genes. To explore hTERT regulation in vivo, we engineered mice with a 160-kb transgenic bacterial artificial chromosome (BAC) spanning the hTERT locus with a Renilla luciferase (Rluc) cassette downstream of its promoter. Analysis of multiple founder lines revealed that the Rluc expression profile from the transgenic hTERT reporter locus reproduced that of the native hTERT gene in all tissues and organs examined, demonstrating that genetic sequence determined the species-specific development

[HOX genes and the limb development in the clinical praxis and in the experiment].

In human, congenital malformations of the limbs are ranked among the most prevalent of all congenital birth defects. Substantial portion of these defects has genetic origin. Increasing knowledge about the particular mutations responsible for limb malformations in human results in the increasing availability of DNA diagnostic procedures for confirmation of clinical diagnosis and family counselling. Increasing understanding of the underlying developmental processes revealed by studying limb defects both in human and animal models may offer better therapeutic options in the future. Taking microRNAs to heart.

MicroRNAs (miRNAs) are a class of highly conserved, small non-coding RNAs that regulate gene expression post-transcriptionally. The emerging field of miRNA biology has begun to reveal roles for these regulatory molecules in a wide range of biological processes. Dysregulated miRNA expression has been correlated to diseased hearts in human patients, whereas inhibiting the maturation of miRNAs conditionally in murine hearts has revealed that miRNAs are essential for cardiac development and function. Moreover, genetic studies have identified distinct roles for specific miRNAs during cardiogenesis,

Molecular basis of male (in)fertility.

It is obvious that the molecular basis of male (in)fertility is not a linear order of genetic events but the interaction of complex genetic networks functional in three main developmental pathways: male germ-line development, male gonad development, and male somatic development. Consequently, primary genetic switch signals should exist for linking the different gene networks and/or for starting them. There is some evidence that such switch signals are concentrated on the sex chromosomes. However multiple genes encoding gene products functional for male fertility exist also on other chromosomes

Transducin duplicates in the zebrafish retina and pineal complex: differential specialisation after the teleost tetraploidisation.

Gene duplications provide raw materials that can be selected for functional adaptations by evolutionary mechanisms. We describe here the results of 350 million years of evolution of three functionally related gene families: the alpha, beta and gamma subunits of transducins, the G protein involved in vision. Early vertebrate tetraploidisations resulted in separate transducin heterotrimers: gnat1/gnb1/gngt1 for rods, and gnat2/gnb3/gngt2 for cones. The teleost-specific tetraploidisation generated additional duplicates for gnb1, gnb3 and gngt2. We report here that the duplicates have undergone se

When half is not enough: gene expression and dosage in the 22q11 deletion syndrome.

The 22q11 Deletion Syndrome (22q11DS, also known as DiGeorge or Velo-Cardio-Facial Syndrome) has a variable constellation of phenotypes including life-threatening cardiac malformations, craniofacial, limb, and digit anomalies, a high incidence of learning, language, and behavioral disorders, and increased vulnerability for psychiatric diseases, including schizophrenia. There is still little clear understanding of how heterozygous microdeletion of approximately 30-50 genes on chromosome 22 leads to this diverse spectrum of phenotypes, especially in the brain. Three possibilities exist: 1) 22q11

Craniofacial developmental biology in the single-cell era.

The evolution of a unique craniofacial complex in vertebrates made possible new ways of breathing, eating, communicating and sensing the environment. The head and face develop through interactions of all three germ layers, the endoderm, ectoderm and mesoderm, as well as the so-called fourth germ layer, the cranial neural crest. Over a century of experimental embryology and genetics have revealed an incredible diversity of cell types derived from each germ layer, signaling pathways and genes that coordinate craniofacial development, and how changes to these underlie human disease and vertebrate

SELF-PRUNING Acts Synergistically with DIAGEOTROPICA to Guide Auxin Responses and Proper Growth Form.

The SELF PRUNING (SP) gene is a key regulator of growth habit in tomato (Solanum lycopersicum). It is an ortholog of TERMINAL FLOWER1, a phosphatidylethanolamine-binding protein with antiflorigenic activity in Arabidopsis (Arabidopsis thaliana). A spontaneous loss-of-function mutation (sp) has been bred into several industrial tomato cultivars, as it produces a suite of pleiotropic effects that are favorable for mechanical harvesting, including determinate growth habit, short plant stature, and simultaneous fruit ripening. However, the physiological basis for these phenotypic differences has n

Major changes in gene expression between strains of diapausing and non-diapause spruce budworm.

To survive harsh winter conditions, many insects enter a state of dormancy called diapause which allows them to withstand extreme temperatures and lack of food. When diapause is induced, insects enter a state of arrested development with a low metabolic rate. Because diapause and environmental conditions are closely linked, variation within and between species in diapause induction, depth, and duration is extremely common. Studies investigating the genetic underpinnings of diapause tend to focus on either different populations and/or environmental variation. We use the eastern spruce budworm (

Postnatal pulmonary artery development from transcript to tissue.

Many congenital conditions and surgical interventions perturb haemodynamics in the proximal pulmonary arteries during postnatal development, thereby altering gene expression and associated changes in vascular structure and function. Among these, pathological conditions include patent ductus arteriosus, pulmonary atresia and stenosis and hypoxemia-induced pulmonary hypertension while surgical interventions include the placement of a Blalock-Thomas-Taussig shunt and the Glenn and Fontan procedures. Despite the significant morbidity associated with these diverse conditions, little attention has b

Evolutionary perspectives on the development of the mammalian cerebellum: From cellular diversity to gene regulatory programs.

During mammalian evolution, the cerebellum has diversified in size and morphology and broadened its roles from motor control to higher-order cognitive and emotional processing. To understand how cerebellar form and function have evolved, it is critical to investigate the underlying molecular and developmental mechanisms. Recent advances in single-cell genomics now allow high-resolution tracking of cellular dynamics, gene expression, and regulatory programs in complex tissues. This presents an opportunity to integrate genome-wide data to the long tradition of research on development and evoluti

ETV2 functions as a pioneer factor to regulate and reprogram the endothelial lineage.

The vasculature is an essential organ for the delivery of blood and oxygen to all tissues of the body and is thus relevant to the treatment of ischaemic diseases, injury-induced regeneration and solid tumour growth. Previously, we demonstrated that ETV2 is an essential transcription factor for the development of cardiac, endothelial and haematopoietic lineages. Here we report that ETV2 functions as a pioneer factor that relaxes closed chromatin and regulates endothelial development. By comparing engineered embryonic stem cell differentiation and reprogramming models with multi-omics techniques

Making a head: Neural crest and ectodermal placodes in cranial sensory development.

During development of the vertebrate sensory system, many important components like the sense organs and cranial sensory ganglia arise within the head and neck. Two progenitor populations, the neural crest, and cranial ectodermal placodes, contribute to these developing vertebrate peripheral sensory structures. The interactions and contributions of these cell populations to the development of the lens, olfactory, otic, pituitary gland, and cranial ganglia are vital for appropriate peripheral nervous system development. Here, we review the origins of both neural crest and placode cells at the n

From "the Worm" to "the Worms" and Back Again: The Evolutionary Developmental Biology of Nematodes.

Since the earliest days of research on nematodes, scientists have noted the developmental and morphological variation that exists within and between species. As various cellular and developmental processes were revealed through intense focus on Caenorhabditis elegans, these comparative studies have expanded. Within the genus Caenorhabditis, they include characterization of intraspecific polymorphisms and comparisons of distinct species, all generally amenable to the same laboratory culture methods and supported by robust genomic and experimental tools. The C. elegans paradigm has also motivate

New insight into functional limb regeneration: A to Z approaches.

Limb/digit amputation is a common event in humans caused by trauma, medical illness, or surgery. Although the loss of a digit is not lethal, it affects quality of life and imposes high costs on amputees. In recent years, the increasing interest in limb regeneration has led to enhanced scientific knowledge. However, the limited ability to develop functional limb regeneration in the clinical setting suggests that a challenging issue remains in limb regeneration. Recently, the emergence of regenerative engineering is a promising field to address this challenge and close the gap between science an

De novo characterization of microRNAs in oriental fruit moth Grapholita molesta and selection of reference genes for normalization of microRNA expression.

MicroRNAs (miRNAs) are a group of endogenous non-coding small RNAs that have critical regulatory functions in almost all known biological processes at the post-transcriptional level in a variety of organisms. The oriental fruit moth Grapholita molesta is one of the most serious pests in orchards worldwide and threatens the production of Rosacea fruits. In this study, a de novo small RNA library constructed from mixed stages of G. molesta was sequenced through Illumina sequencing platform and a total of 536 mature miRNAs consisting of 291 conserved and 245 novel miRNAs were identified. The role of microRNAs in mammalian oocytes and embryos.

Advanced genomic analysis has revealed an enormous inventory of non-coding RNAs (ncRNAs), which are functionally important at transcriptional and post-transcriptional level for different cellular processes. Among the ncRNAs, microRNAs (miRNAs) have recently been highlighted extensively for their pivotal role in disease, fertility and development through post-transcriptional regulation of gene expression. The presence and spatio-temporal expression of miRNAs and miRNA processing machinery genes in oocytes and preimplantation embryos has evidenced the involvement of miRNAs for growth and maturat
