Understanding the genetic basis of medication-related osteonecrosis of the jaw: A systematic review.

Medication-related osteonecrosis of the jaw (MRONJ) is an adverse event often associated with the use of antiresorptive drugs. This systematic review aims to identify genes and their polymorphisms associated with the risk of developing MRONJ in patients with oncological or skeletal-related diseases treated with antiresorptive drugs. A systematic literature review was conducted in accordance with PRISMA guidelines. Three electronic databases (PubMed, Scopus, and Web of Science) were searched for studies published up to December 2024. The search strategy included terms such as "MRONJ", "bone den

Heritable Risk and Protective Genetic Components of Glaucoma Medication Non-Adherence.

Glaucoma is the leading cause of irreversible blindness, affecting 76 million globally. It is characterized by irreversible damage to the optic nerve. Pharmacotherapy manages intraocular pressure (IOP) and slows disease progression. However, non-adherence to glaucoma medications remains problematic, with 41-71% of patients being non-adherent to their prescribed medication. Despite substantial investment in research, clinical effort, and patient education protocols, non-adherence remains high. Therefore, we aimed to determine if there is a substantive genetic component behind patients' glaucoma

Pearson Syndrome, A Medical Diagnosis Difficult to Sustain Without Genetic Testing.

BACKGROUND: The detection of sideroblastic anemia in a newborn may suggest developing Pearson syndrome. The prognosis of these patients is severe and death occurs in the first 3 years of life, so it is important to find new ways of diagnosis. Case Presentation: In the case of our patient the diagnosis was supported only at the age of 5 months, highlighting the difficulties of diagnosis at this age. CONCLUSIONS: The diagnosis of Pearson syndrome with neonatal onset is difficult to sustain or even impossible at that age. This diagnosis can be confirmed and supported during disease progression.

The case for a parental duty to use preimplantation genetic diagnosis for medical benefit.

This article explores the possibility that there is a parental duty to use preimplantation genetic diagnosis (PGD) for the medical benefit of future children. Using one genetic disorder as a paradigmatic example, we find that such a duty can be supported in some situations on both ethical and legal grounds. Our analysis shows that an ethical case in favor of this position can be made when potential parents are aware that a possible future child is at substantial risk of inheriting a serious genetic condition. We further argue that a legal case for a duty to use PGD for medical benefit can be m

Outcomes and genetic relatedness of carbapenem-resistant enterobacteriaceae at Detroit medical center.

BACKGROUND: Carbapenem-resistant Enterobacteriaceae (CRE) are rapidly emerging in hospitals in the United States and are posing a significant threat. To better understand the transmission dynamics and the acquisition of resistant strains, a thorough analysis of epidemiologic and molecular characteristics was performed. METHODS: CRE isolated at Detroit Medical Center were analyzed from September 2008 to September 2009. bla(KPC) genes were investigated by polymerase chain reaction (PCR), and repetitive extragenic palindromic PCR (rep-PCR) was used to determine genetic similarity among strains.

[Genetic markers in forensic medical examination of acute and chronic opioid intoxication].

The objective of the present study was to analyse cases of intoxication with opioid narcotics at different blood morphine levels, from low and very low (below 0.5 mg/l) to regular toxic ones (1-4 mg/I). It was shown that molecular genetic studies at postmortem morphine concentrations of up to 0.5 mg/l as a rule identify mutant alleles (CYP2D6*3*4; CYP2C19*2*3). Mutant alleles CYP2D6* and CYP2C19* are most frequently detected at postmortem blood morphine levels ranging from 1 to 4 mg/I. These findings make it possible to regard death cases at low morphine concentrations as due to genetic suscep

Assessing attitudes about genetic testing as a component of continuing medical education.

OBJECTIVE: To investigate the attitudes among mental health professionals regarding the use of genetic testing. METHODS: Psychiatrists and other mental health professionals (N=41) who were enrolled in a week-long course in psychiatric genomics completed questionnaires before and after the course designed to assess how diagnostic genetic tests should be used and the value of pharmacogenomic testing for clinical practice. RESULTS: Only 5% of the course participants knew their genotype for the CYP 2D6 and CYP 2C19 genes at the time they participated in the course. However, after completing the

Marfan syndrome--current medical and genetic knowledge: how to treat and when.

Marfan syndrome (MFS), first described 100 years ago, remains a clinical diagnosis. Two out of 3 major systems (ocular, cardiac, skeletal) must be classically affected, to avoid overdiagnosis. Diagnosis may be confirmed by linkage to the dominantly inherited gene MFS-1 on 15q21, or by discovering the family mutation. Either technique may be used for prenatal diagnosis. Modern medical and surgical management is prolonging life, by on average, 13 years, with postoperative 20 year survival rate 65%. Beta-blocker therapy slows aortic root dilatation, and elective surgery is offered at ascending ao

Integrating cases, enhancing interaction, and promoting learning through assessment: teaching reform and practical outcomes of medical genetics under the OBE concept.

In the new era, the teaching of Medical Genetics requires medical students to not only master solid knowledge, but also possess practical abilities in diagnosing and treating clinical genetic diseases. This paper systematically elaborates on the design and practical exploration of teaching reform in the Medical Genetics course under the Outcome-Based Education (OBE) concept. By constructing a structured teaching system closely aligned with clinical practice, building a teaching model centered on clinical cases, carrying out diverse teaching activities, developing a "boundless classroom", and e

Animal medical genetics: a historical perspective on more than 50 years of research into genetic disorders of animals at Massey University.

Over the last 50 years, there have been major advances in knowledge and technology regarding genetic diseases, and the subsequent ability to control them in a cost-effective manner. This review traces these advances through research into genetic diseases of animals at Massey University (Palmerston North, NZ), and briefly discusses the disorders investigated during that time, with additional detail for disorders of major importance such as bovine α-mannosidosis, ovine ceroid-lipofuscinosis, canine mucopolysaccharidosis IIIA and feline hyperchylomicronaemia. The overall research has made a signi

Diagnosis and management of glycogen storage disease type I: a practice guideline of the American College of Medical Genetics and Genomics.

PURPOSE: Glycogen storage disease type I (GSD I) is a rare disease of variable clinical severity that primarily affects the liver and kidney. It is caused by deficient activity of the glucose 6-phosphatase enzyme (GSD Ia) or a deficiency in the microsomal transport proteins for glucose 6-phosphate (GSD Ib), resulting in excessive accumulation of glycogen and fat in the liver, kidney, and intestinal mucosa. Patients with GSD I have a wide spectrum of clinical manifestations, including hepatomegaly, hypoglycemia, lactic acidemia, hyperlipidemia, hyperuricemia, and growth retardation. Individuals
