Rhodiola L. is a genus exhibiting rapid radiation and represents a typical case for studying plastid gene adaptation in species that spread from high altitudes to low altitudes. In this study, 23 samples of 18 Rhodiola species were collected from the Qinghai-Tibetan Plateau and five scattered alpine areas, and the plastid genomes (plastomes) of these species were sequenced, annotated, and compared between high-altitude and widely distributed groups. The plastomes of Rhodiola were found to be highly conserved in terms of gene size, content, and order but highly variable in several lineage-speci

Rehmannia chingii is an important medicinal plant with immense value in scientific research. However, its mitochondrial genome (mitogenome) has not yet been characterized. Herein, based on whole-genome Illumina short reads and PacBio HiFi reads, we obtained the complete mitogenome of R. chingii through a de novo assembly strategy. We carried out comparative genomic analyses and found that, in comparison with the plastid genome (plastome) showing a high degree of structural conservation, the R. chingii mitogenome structure is relatively complex, showing an intricate ring structure with 16 conne

Most apicomplexan parasites contain a plastid-derived organelle called the apicoplast, which originated through secondary endosymbiosis. As a result of this evolutionary trajectory, the non-photosynthetic apicoplast is surrounded by four membranes and contains many bacterial-like, druggable targets. It is widely accepted that asexual malaria parasites (Plasmodium falciparum) can thrive under antibiotic treatment if supplemented with high concentrations of isopentenyl pyrophosphate (IPP, 200 μM) and these IPP-rescued parasites are thought to lack the apicoplast and its 35 kb genome but possess

Katablepharids (or kathablepharids) are plastid-less, biflagellate protists widely distributed in marine and freshwater environments. Here, we report a new katablepharid species, Katablepharis ulleungensis sp. nov., isolated from a black sandy beach in Ulleung-do, South Korea. The stock strain was co-cultured with a small bicosoecid flagellate, serving as its prey. Cells were oblong, displayed two rows of Type I ejectisomes, and possessed two subapically inserted flagella with the posterior flagellum slightly longer than the anterior one. Rather than exhibiting linear or spiral locomotion, cel

Primula octopoda W.B.Ju & Q.Huang, a new species of Primula (Primulaceae) in sect. Aleuritia, is herein described and illustrated from Batang County, Sichuan Province, southwestern China. Morphologically, P. octopoda is most similar to P. stenocalyx, but it can be readily distinguished by its dwarf habit, the presence of stolons 2-7 cm in length ending in small rosettes occasionally bearing a single flower, elliptic to broadly obovate leaf blades, a very short or absent scape bearing one or two flowers, and a smaller, annulate corolla with a prominent yellow eye. The complete chloroplast genom

The apicoplast is a four-membrane plastid found in the apicomplexans, which harbors biosynthesis and organelle housekeeping activities in the matrix. However, the mechanism driving the flux of metabolites, in and out, remains unknown. Here, we used TurboID and genome engineering to identify apicoplast transporters in Toxoplasma gondii. Among the many novel transporters, we show that one pair of apicomplexan monocarboxylate transporters (AMTs) appears to have evolved from a putative host cell that engulfed a red alga. Protein depletion showed that AMT1 and AMT2 are critical for parasite growth.

The deadly malaria parasite, Plasmodium falciparum, contains a unique subcellular organelle termed the apicoplast, which is a clinically-proven antimalarial drug target. The apicoplast is a plastid with essential metabolic functions that evolved via secondary endosymbiosis. As an ancient endosymbiont, the apicoplast retained its own genome and it must be inherited by daughter cells during cell division. During the asexual replication of P. falciparum inside human red blood cells, both the parasite, and the apicoplast inside it, undergo massive morphological changes, including DNA replication a

Malaria is caused by the parasite Plasmodium falciparum, which contains an essential non-photosynthetic plastid called the apicoplast. A single DNA polymerase, apPOL, is targeted to the apicoplast, where it replicates and repairs the genome. apPOL has no direct orthologs in mammals and is considered a promising drug target for the treatment and/or prevention of malaria. We previously reported screening the Malaria Box to identify MMV666123 as an inhibitor of apPOL. Herein we extend our studies and report structure-activity relationships for MMV666123 and identify key structural motifs necessar

The human malaria parasite Plasmodium falciparum genome is among the most A + T rich, with low complexity regions (LCRs) inserted in coding sequences including those for proteins targeted to its essential relict plastid (apicoplast). Replication of the apicoplast genome (plDNA), mediated by the atypical multifunctional DNA polymerase PfPrex, would require additional enzymatic functions for lagging strand processing. We identified an apicoplast-targeted, [4Fe-4S]-containing, FEN/Exo (PfExo) with a long LCR insertion and detected its interaction with PfPrex. Distinct from other known exonuclease

The emergence and spread of Plasmodium falciparum parasites resistant to front-line antimalarial artemisinin-combination therapies (ACT) threatens to erase the considerable gains against the disease of the last decade. Here, we develop a large-scale phenotypic screening pipeline and use it to carry out a large-scale forward-genetic phenotype screen in P. falciparum to identify genes allowing parasites to survive febrile temperatures. Screening identifies more than 200 P. falciparum mutants with differential responses to increased temperature. These mutants are more likely to be sensitive to ar

Rubisco (ribulose 1,5-bisphosphate carboxylase/oxygenase) is the primary CO2-fixing enzyme on our planet. Its slow kinetics and poor discrimination between carbon dioxide and oxygen as substrates severely limit the efficiency of photosynthetic carbon fixation by plants. Attempts to improve Rubisco by genetic engineering have been hampered by the complexity of the Rubisco system, with a gene family for the enzyme's small subunit being encoded in the nuclear genome and the gene for the large subunit residing in the plastid (chloroplast) genome. Another nuclear gene family encodes the enzyme Rubi

BACKGROUND: In plant-derived animal feedstuffs, nearly 80 % of the total phosphorus content is stored as phytate. However, phytate is poorly digested by monogastric animals such as poultry, swine and fish, as they lack the hydrolytic enzyme phytase; hence it is regarded as a nutritionally inactive compound from a phosphate bioavailability point of view. In addition, it also chelates important dietary minerals and essential amino acids. Therefore, dietary supplementation with bioavailable phosphate and exogenous phytases are required to achieve optimal animal growth. In order to simplify the ob

BACKGROUND: Insulin-like growth factor-1 (IGF-1), in addition to having insulin-like effects, has boosting effects on all cells in human body. Most of the recombinant IGF-1 required for patients suffering from its deficiency is currently produced by bacterial and yeast systems. Plant systems, especially chloroplasts, have many benefits for producing human blood proteins. Production costs are low in these systems, and their side effects are less than other systems.
OBJECTIVES: In this study, the transfer and expression of mature IGF-1 protein cDNA in tobacco chloroplasts under the control of s

The order Ishigeales is an early-diverging lineage of brown algae that exhibits remarkable morphological diversity ranging from simple filamentous forms to robust terete and foliose thalli, which may reflect complex evolutionary histories in their organelle genomes. However, limited organellar genomic data for the Ishigeales, presently confined to Ishige okamurae, have hindered a comprehensive understanding of organelle genome dynamics in that order. To address this, we de novo assembled the complete plastomes and mitogenomes of the four Ishigeales species: Pilinia rimosa, Petroderma maculifor

BACKGROUND: Albinism in plants is characterized by lack of chlorophyll and results in photosynthesis impairment, abnormal plant development and premature death. These abnormalities are frequently encountered in interspecific crosses and tissue culture experiments. Analysis of albino mutant phenotypes with full or partial chlorophyll deficiency can shed light on genetic determinants and molecular mechanisms of albinism. Here we report analysis of RNA-seq transcription profiling of barley (Hordeum vulgare L.) near-isogenic lines, one of which is a carrier of mutant allele of the Alm gene for alb

Lichen photobionts exhibit significant physiological variability with important ecological implications. While this variability is relatively well studied at the interspecific level, it has rarely been examined at the intraspecific level. Here, we investigated the physiological responses to temperature and irradiance of two Trebouxia lynniae strains differing by a single nucleotide polymorphism in the ITS2 region. Both strains were isolated and characterized through biomass accumulation, chlorophyll fluorescence, pigment quantification, transmission electron microscopy, metabolomic profiling a

BACKGROUND: Cyclospora cayetanensis is an emerging coccidian parasite that causes endemic and epidemic diarrheal disease called cyclosporiasis, and this infection is associated with consumption of contaminated produce or water in developed and developing regions. Food-borne outbreaks of cyclosporiasis have occurred almost every year in the USA since the 1990s. Investigations of these outbreaks are currently hampered due to lack of molecular epidemiological tools for trace back analysis. The apicoplast of C. cayetanensis, a relict non-photosynthetic plastid with an independent genome, provides

Diatom genome sequences clearly reveal the presence of different systems for HCO3(-) uptake. Carbon-concentrating mechanisms (CCM) based on HCO3(-) transport and a plastid-localized carbonic anhydrase (CA, EC 4.2.1.1) appear to be more probable than the others because CAs have been identified in the genome of many diatoms. CAs are key enzymes involved in the acquisition of inorganic carbon for photosynthesis in phytoplankton, as they catalyze efficiently the interconversion between carbon dioxide and bicarbonate. Five genetically distinct classes of CAs exist, α-, β-, γ-, δ- and ζ and all of t

Mosla chinensis is a high-value medicinal plant with scarce genomic resources for precision breeding. Here, we present the first chromosome-scale genome of an alpine ecotype (Baiyunjian Reserve, 1,611 m above sea level) assembled via SMRT sequencing and Hi-C technologies. 93.46% of sequences were anchored to 9 pseudochromosomes with 99.99% accuracy, resulting in the assembly of the 444.26 Mb genome (contig N50 = 41.38 Mb; 98.7% BUSCO). High proportion of repetitive sequences (56.90%) and species-specific genome duplication revealed the species-specific adaptations of this ecotype. Multi-tissue

INTRODUCTION: Rhodiola is one of the few genera in Crassulaceae that includes both dioecious and hermaphroditic species. However, previous studies have mainly relied on representative individuals or limited plastid fragments, which may not fully resolve evolutionary relationships within the genus, especially lineage divergence associated with sexual-system differentiation.
METHODS: In this study, we analyzed the complete chloroplast genomes of 89 newly sequenced samples representing 35 Rhodiola species. A batch-processing pipeline was used to integrate raw-read processing, automated chloropla
