The rare TXNRD1_v3 ("v3") splice variant of human thioredoxin reductase 1 protein is targeted to membrane rafts by N-acylation and induces filopodia independently of its redox active site integrity.

The human selenoprotein thioredoxin reductase 1 (TrxR1), encoded by the TXNRD1 gene, is a key player in redox regulation. Alternative splicing generates several TrxR1 variants, one of which is v3 that carries an atypical N-terminal glutaredoxin domain. When overexpressed, v3 associates with membranes and triggers formation of filopodia. Here we found that membrane targeting of v3 is mediated by myristoylation and palmitoylation of its N-terminal MGC motif, through which v3 specifically targets membrane rafts. This was suggested by its localization in cholera toxin subunit B-stained membrane ar

Identification of in vivo induced protein antigens of Salmonella enterica serovar Typhi during human infection.

During infectious disease episodes, pathogens express distinct subsets of virulence factors which allow them to adapt to different environments. Hence, genes that are expressed or upregulated in vivo are implicated in pathogenesis. We used in vivo induced antigen technology (IVIAT) to identify antigens which are expressed during infection with Salmonella enterica serovar Typhi. We identified 7 in vivo induced (IVI) antigens, which included BcfD (a fimbrial structural subunit), GrxC (a glutaredoxin 3), SapB (an ABC-type transport system), T3663 (an ABC-type uncharacterized transport system), T3

Hereditary sideroblastic anemias: pathophysiology, diagnosis, and treatment.

Inherited sideroblastic anemia comprises several rare anemias due to heterogeneous genetic lesions, all characterized by the presence of ringed sideroblasts in the bone marrow. This morphological aspect reflects abnormal mitochondrial iron utilization by the erythroid precursors. The most common X-linked sideroblastic anemia (XLSA), due to mutations of the first enzyme of the heme synthetic pathway, delta-aminolevulinic acid synthase 2 (ALAS2), has linked heme deficiency to mitochondrial iron accumulation. The identification of other genes, such as adenosine triphosphate (ATP) binding cassette

Arabidopsis thaliana 3-mercaptopyruvate sulfurtransferases interact with and are protected by reducing systems.

The formation of a persulfide group (-SSH) on cysteine residues has gained attention as a reversible posttranslational modification contributing to protein regulation or protection. The widely distributed 3-mercaptopyruvate sulfurtransferases (MSTs) are implicated in the generation of persulfidated molecules and H2S biogenesis through transfer of a sulfane sulfur atom from a suitable donor to an acceptor. Arabidopsis has two MSTs, named STR1 and STR2, but they are poorly characterized. To learn more about these enzymes, we conducted a series of biochemical experiments including a variety of po

Grx attenuates diabetes cataract by activating AMPK/BNIP3/BNIP3L-mediated mitophagy in lens epithelial cells.

Glutaredoxin (Grx) plays an important antioxidant role in the pathogenesis of cataracts. Our previous research found that Grx knockout accelerated the occurrence of aging-related cataract (ARC) and was closely related to lens opacity, but the regulatory mechanism of Grx in diabetes cataract (DC) is still unclear. Here, we found that AMPK activity was significantly decreased in the anterior lens capsule of DC patients and high-glucose (HG) treated human lens epithelial cells HLEC-B3, and Grx protein expression and activity were compensatory increased. Moreover, Grx protected HLEC-B3 cells from

Causal Relationship between Mitochondrial Biological Function and Periodontitis: Evidence from a Mendelian Randomization Study.

A growing number of studies indicate that mitochondrial dysfunction serves as a pathological mechanism for periodontitis. Therefore, this two-sample Mendelian randomization (MR) study was carried out to explore the causal associations between mitochondrial biological function and periodontitis, because the specific nature of this causal relationship remains inconclusive in existing MR studies. Inverse variance weighting, Mendelian randomization-Egger, weighted mode, simple mode, and weighted median analyses were performed to assess the causal relationships between the exposure factors and peri

Regulation of the Dimerization and Activity of SARS-CoV-2 Main Protease through Reversible Glutathionylation of Cysteine 300.

Severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2), the causative agent for coronavirus disease 2019 (COVID-19), encodes two proteases required for replication. The main protease (Mpro), encoded as part of two polyproteins, pp1a and pp1ab, is responsible for 11 different cleavages of these viral polyproteins to produce mature proteins required for viral replication. Mpro is therefore an attractive target for therapeutic interventions. Certain proteins in cells under oxidative stress undergo modification of reactive cysteines. We show Mpro is susceptible to glutathionylation, leading

GLRX5-associated [Fe-S] cluster biogenesis disorder: further characterisation of the neurological phenotype and long-term outcome.

BACKGROUND: Identification and characterisation of monogenic causes of complex neurological phenotypes are important for genetic counselling and prognostication. Bi-allelic pathogenic variants in the gene encoding GLRX5, a protein involved in the early steps of Fe-S cluster biogenesis, are rare and cause two distinct phenotypes: isolated sideroblastic anemia and a neurological phenotype with variant non-ketotic hyperglycinemia. In this study, we analysed the evolution of clinical and MRI findings and long-term outcome of patients with GLRX5 mutations. METHODS: Four patients from three Austral

Note on the activation of the heme-stabilized translational inhibitor of reticulocyte lysates by oxidized glutathione.

The heme-controlled translational inhibitor (HCI) of reticulocyte lysates can be activated either by a lack of by heme or, in the presence of heme, by oxidized glutathione (GSSG) and various oxidative processes. The latter activation can be prevented or reversed by NADPH or NADPH generators, such as glucose-6-phosphate (G-6-P). Since reticulocyte lysates contain a very active GSSG reductase, it was conceivable that GSSG acts by draining lysate NADPH via the reaction GSSG + NADPH + H+ in equilibrium 2 GSH + NADP+. However, removal of lysate GSSG reductase by its corresponding antibody has no ef

ZmGRXCC13, a key factor in coping with drought and saline-alkali stress.

Maize (Zea mays L.) is a paramount global staple crop; however, its production is severely limited by diverse abiotic stresses, particularly drought and saline-alkali stress. Although glutaredoxins (GRXs) are known to regulate plant growth and stress responses, their specific functions and underlying molecular mechanisms in maize under drought and saline-alkali conditions remain largely elusive. In this study, we cloned the maize GRX family member ZmGRXCC13 (Zm00001d037757) and characterized its biological functions in abiotic stress tolerance. Quantitative real-time PCR (qRT-PCR) analyses rev

Endometrial pinopode biomarkers: Molecules and microRNAs.

Ultrastructural changes on the apical surface of the luminal epithelium of the uterus are known as pinopodes. Their morphology in species and in special species is associated with different results about size, duration, and percentage of surface area covered by pinopodes. The content of pinopodes is different in rodents and humans. In mice and rats pinopodes have many vacuoles and no organelle that extends to the actin stalk above the microvilli. Human pinopodes do not have a large vacuole and contain the golgi complex, a rough endoplasmic reticulum, secretory vesicles, and mitochondria that e

Pathophysiology and genetic mutations in congenital sideroblastic anemia.

Sideroblastic anemias are heterogeneous congenital and acquired disorders characterized by anemia and the presence of ringed sideroblasts in the bone marrow. Congenital sideroblastic anemia (CSA) is a rare disease caused by mutations of genes involved in heme biosynthesis, iron-sulfur [Fe-S] cluster biosynthesis, and mitochondrial protein synthesis. The most common form is X-linked sideroblastic anemia, due to mutations in the erythroid-specific δ-aminolevulinate synthase (ALAS2), which is the first enzyme of the heme biosynthesis pathway in erythroid cells. Other known etiologies include muta

The trypanothione system and the opportunities it offers to create drugs for the neglected kinetoplast diseases.

Parasitic trypanosomatids (Kinetoplastida) are the causative agents of devastating and hard-to-treat diseases such as African sleeping sickness, Chagas disease and various forms of Leishmaniasis. Altogether they affect > 30 Million patients, account for half a million fatalities p.a. and cause substantial economical problems in the Third World due to human morbidity and life stock losses. The design of efficacious and safe drugs is expected from inhibition of metabolic pathways that are unique and essential to the parasite and absent in the host. In this respect, the trypanothione system first

Saccharomyces cerevisiae isolates with extreme hydrogen sulfide production showed different oxidative stress resistances responses during wine fermentation by RNA sequencing analysis.

In the wine industry, off-odors occurring during fermentation and bottling caused by hydrogen sulfide (H2S) produced by Saccharomyces cerevisiae are still a major problem. Here, two native S. cerevisiae isolates possessing extreme H2S production capacities were isolated from 166 strains in Chinese vineyards. Significant differences were observed in RNA sequencing analyses between the two strains. The results showed that sulfur metabolism was highly repressed and genes involved in the NADPH-dependent thioredoxin and glutaredoxin oxidoreduction systems were highly expressed in the non-H2S produc

Oncogenic STAT5 signaling promotes oxidative stress in chronic myeloid leukemia cells by repressing antioxidant defenses.

STAT5 transcription factors are frequently activated in hematopoietic neoplasms and are targets of various tyrosine kinase oncogenes. Evidences for a crosstalk between STAT5 and reactive oxygen species (ROS) metabolism have recently emerged but mechanisms involved in STAT5-mediated regulation of ROS still remain elusive. We demonstrate that sustained activation of STAT5 induced by Bcr-Abl in chronic myeloid leukemia (CML) cells promotes ROS production by repressing expression of two antioxidant enzymes, catalase and glutaredoxin-1(Glrx1). Downregulation of catalase and Glrx1 expression was als

A recombinant thioredoxin-glutathione reductase from Fasciola hepatica induces a protective response in rabbits.

Antioxidant systems are fundamental components of host-parasite interactions, and often play a key role in parasite survival. Here, we report the cloning, heterologous expression, and characterization of a thioredoxin glutathione reductase (TGR) from Fasciola hepatica. The deduced polypeptide sequence of the cloned open reading frame (ORF) confirmed the experimental N-terminus previously determined for a native F. hepatica TGR showing thioredoxin reductase (TR) activity. The sequence revealed the presence of a fusion between a glutaredoxin (Grx) and a TR domain, similar to that previously repo

Spatiotemporal Redox Regulation of Cell-Cycle Progression: Dynamic Functions of Cellular Redox Regulatory Proteins.

Aerobic life revolves around the enigmatic role of oxygen, which sustains the life cycle by supporting energy production while simultaneously threatening its existence through the generation of reactive oxygen species (ROS). Intriguingly, ROS are not merely by-products of energy generation; cells actively invest in their production through dedicated pro-oxidant systems. A long-standing perception in the scientific community has been that cellular antioxidant systems primarily evolved as terminators of ROS to protect against oxidative damage. It appears paradoxical, however, that the complex, m

The conserved CDC motif in the yeast iron regulator Aft2 mediates iron-sulfur cluster exchange and protein-protein interactions with Grx3 and Bol2.

The Saccharomyces cerevisiae transcriptional activator Aft1 and its paralog Aft2 respond to iron deficiency by upregulating expression of proteins required for iron uptake at the plasma membrane, vacuolar iron transport, and mitochondrial iron metabolism, with the net result of mobilizing iron from extracellular sources and intracellular stores. Conversely, when iron levels are sufficient, Aft1 and Aft2 interact with the cytosolic glutaredoxins Grx3 and Grx4 and the BolA protein Bol2, which promote Aft1/2 dissociation from DNA and subsequent export from the nucleus. Previous studies unveiled t

Iron homeostasis proteins Grx4 and Fra2 control activity of the Schizosaccharomyces pombe iron repressor Fep1 by facilitating [2Fe-2S] cluster removal.

The Bol2 homolog Fra2 and monothiol glutaredoxin Grx4 together play essential roles in regulating iron homeostasis in Schizosaccharomyces pombe. In vivo studies indicate that Grx4 and Fra2 act as coinhibitory partners that inactivate the transcriptional repressor Fep1 in response to iron deficiency. In Saccharomyces cerevisiae, Bol2 is known to form a [2Fe-2S]-bridged heterodimer with the monothiol Grxs Grx3 and Grx4, with the cluster ligands provided by conserved residues in Grx3/4 and Bol2 as well as GSH. In this study, we characterized this analogous [2Fe-2S]-bridged Grx4-Fra2 complex in S.

Cysteine Reactivity Profiling Illuminates Monoclonal Antibody Disulfide Bond Reduction Mechanisms in Biopharmaceutical Process Intermediates.

Monoclonal antibodies (mAbs) are crucial biotherapeutics in the face of increasing global demand, but their production can be impacted by the reduction of disulfide bonds. This study presents a chemical proteomics workflow aimed at elucidating the mechanisms underlying disulfide bond reduction in mAbs produced from Chinese hamster ovary (CHO) cells. We employed iodoacetamide-desthiobiotin (IA-DTB) and the parallel accumulation and serial fragmentation combined with data-independent acquisition (diaPASEF) methodology for cysteine reactivity profiling and successfully quantified approximately 4,
