Plasmalogens in the retina: from occurrence in retinal cell membranes to potential involvement in pathophysiology of retinal diseases.

Plasmalogens (Pls) represent a specific subclass of glycerophospholipids characterized by the presence of a vinyl-ether bond at the sn-1 position of glycerol. Pls are quantitatively important in membranes of neuronal tissues, including the brain and the retina, where they can represent until almost two-third of ethanolamine glycerophospholipids. They are considered as reservoirs of polyunsaturated fatty acids as several studies have shown that arachidonic and docosahexaenoic acids are preferentially esterified on Pls when compared to other glycerophospholipids. Reduced levels of Pls were obser

Chlorpromazine-induced increase in dipalmitoylphosphatidylserine surface area in monolayers at room temperature.

The Langmuir technique revealed that the surface area of acidic glycerophospholipids (dipalmitoylphosphatidylserine, -glycerol, and dipalmitoylphosphatidic acid) in monolayers increased dramatically when micromolar concentrations of the antipsychotic drug chlorpromazine (CPZ) were present in the subphase. Monolayers of neutral glycerophospholipids (dipalmitoylphosphatidylcholine and -ethanolamine) did not show such a large effect with CPZ. Compared to CPZ, millimolar concentrations of the monovalent cations Li+, K+, Na+, Rb+, and Cs+ did not appear to influence the dipalmitoylphosphatidylserin

Acute coronary syndrome is associated with a substantial change in the platelet lipidome.

AIMS: Platelets play a key role in the pathophysiology of coronary artery disease (CAD) and patients with enhanced platelet activation are at increased risk to develop adverse cardiovascular events. Beyond reliable cardiovascular risk factors such as dyslipoproteinaemia, significant changes of platelet lipids occur in patients with CAD. In this study, we investigate the platelet lipidome by untargeted liquid chromatography-mass spectrometry, highlighting significant changes between acute coronary syndrome (ACS) and chronic coronary syndrome (CCS) patients. Additionally, we classify the platele

LipidPedia: a comprehensive lipid knowledgebase.

MotivationLipids are divided into fatty acyls, glycerolipids, glycerophospholipids, sphingolipids, saccharolipids, sterols, prenol lipids and polyketides. Fatty acyls and glycerolipids are commonly used as energy storage, whereas glycerophospholipids, sphingolipids, sterols and saccharolipids are common used as components of cell membranes. Lipids in fatty acyls, glycerophospholipids, sphingolipids and sterols classes play important roles in signaling. Although more than 36 million lipids can be identified or computationally generated, no single lipid database provides comprehensive informatio

Influence of sample preparation on lipidomics analysis of polar lipids in adipose tissue.

The main limitations of lipidomics analysis are the chemical complexity of the lipids, the range of concentrations at which they exist, and the variety of samples usually analyzed. These limitations particularly affect the characterization of polar lipids owing to the interference of neutral lipids, essentially acylglycerides, which are at high concentration and suppress ionization of low concentrated lipids in mass spectrometry detection. The influence of sample preparation on lipidomics analysis of polar lipids in adipose tissue by LC-MS/MS was the aim of this research. Two common extractant

EPT1 (selenoprotein I) is critical for the neural development and maintenance of plasmalogen in humans.

Ethanolamine phosphotransferase (EPT)1, also known as selenoprotein 1 (SELENOI), is an enzyme that transfers phosphoethanolamine from cytidine diphosphate-ethanolamine to lipid acceptors to produce ethanolamine glycerophospholipids, such as diacyl-linked phosphatidylethanolamine (PE) and ether-linked plasmalogen [1-alkenyl-2-acyl-glycerophosphoethanolamine (plasmenyl-PE)]. However, to date there has been no analysis of the metabolomic consequences of the mutation of EPT1 on the concentration of ethanolamine glycerophospholipids in mammalian cells. We studied a patient with severe complicated h

Serum lipidomics reveals class-specific lipid remodelling and altered acyl chain composition in Parkinson's disease.

BACKGROUND: Lipid metabolism is increasingly implicated in Parkinson's Disease (PD) pathology, yet the specific lipid classes and acyl chain compositions involved, and their relationship with PD clinical symptoms, remain poorly characterised. OBJECTIVES: We aimed to identify serum lipid signatures that differentiate people with PD (PwPD) from healthy controls and to evaluate associations with levodopa-induced dyskinesia (LID). METHODS: Serum collected from 58 PwPD and 58 age- and sex-matched non-PD controls was analysed using targeted liquid chromatography - mass spectrometry (LC-MS). Group

Therapeutic Effects of Sulforaphane on Helicobacter pylori-Infected Mice: Insights from High-Coverage Metabolomics and Lipidomics Analyses of Serum and Liver.

Sulforaphane, a natural isothiocyanate predominantly found in cruciferous vegetables, has shown potential in preventing and treating Helicobacter pylori infection. However, the underlying metabolic mechanisms remain largely unclear. This study employed high-coverage metabolomics and lipidomics methods to comprehensively investigate the effects of sulforaphane on the serum and liver metabolic profiles of H. pylori-infected mice. Metabolomics and lipidomics analysis revealed that H. pylori infection disrupted multiple metabolic pathways, leading to perturbations in amino acids, fatty acids, bile

Analysis of glycerophospho- and sphingolipids by CE.

Glycerophospholipids are amphiphilic molecules possessing polar head groups with a glycerol backbone and nonpolar variable long-chain fatty acids. Numerous molecular species are found in a single class of glycerophospholipid, conferring to these lipids a high structural diversity. They are major components of biological membranes and participate in important activities involving cell signaling and substrate transport. Sphingolipids consist of long-chain bases linked by an amide bond to a fatty acid and via the terminal hydroxyl group to complex carbohydrate or phosphorus moieties, constituting

Alterations in secondary lipids are associated with neuroinflammation in the brain of Neu1-deficient mice.

Neu1 (lysosomal sialidase 1) is essential for removing sialic acid from oligosaccharides and glycoconjugates. Neu1 deficiency impairs lysosomal digestion, leading to sialidosis and sialoglycoprotein accumulation. It also increases lipids, including gangliosides GM3, GD3, GM4, and LM1, in the kidney, liver, and spleen. Neu1-/- mice display symptoms resembling Type II sialidosis, including enlarged spleen and liver, kidney issues, neurological problems, spinal defects, and oligosaccharide buildup. The study examined secondary lipid alterations and inflammation in the cortex and cerebellum of the

Structures, functions, and syntheses of glycero-glycophospholipids.

Biological membranes consist of integral and peripheral protein-associated lipid bilayers. Although constituent lipids vary among cells, membrane lipids are mainly classified as phospholipids, glycolipids, and sterols. Phospholipids are further divided into glycerophospholipids and sphingophospholipids, whereas glycolipids are further classified as glyceroglycolipids and sphingoglycolipids. Both glycerophospholipids and glyceroglycolipids contain diacylglycerol as the common backbone, but their head groups differ. Most glycerolipids have polar head groups containing phosphate esters or sugar m

An improved high-throughput lipid extraction method for the analysis of human brain lipids.

We have developed a protocol suitable for high-throughput lipidomic analysis of human brain samples. The traditional Folch extraction (using chloroform and glass-glass homogenization) was compared to a high-throughput method combining methyl-tert-butyl ether (MTBE) extraction with mechanical homogenization utilizing ceramic beads. This high-throughput method significantly reduced sample handling time and increased efficiency compared to glass-glass homogenizing. Furthermore, replacing chloroform with MTBE is safer (less carcinogenic/toxic), with lipids dissolving in the upper phase, allowing f

Analysis of metabolites of fungal balls in the paranasal sinuses.

Fungal ball sinusitis is characterized by complex fungus infections with non-invasive inflammation. But no research reported fungal ball composition and metabolic-related product types currently. 12 patients with chronic rhinosinusitis who underwent surgery and 9 healthy control were enrolled in this study. Samples from both groups were analyzed for high-throughput metabolites by UPLC-MS. OsiriX software was applied to perform imaging measurements on sinus CT. 2138 and 394 metabolites were screened from cationic and anionic modes. There was a significant difference in the abundance of glycerop

A lipidomic study: Nobiletin ameliorates hepatic steatosis through regulation of lipid alternation.

Hepatic lipidome has been given emphasis for years since hepatic steatosis is the most remarkable character of nonalcoholic fatty liver diseases, an increasingly serious health issue worldwide. Nobiletin (NOB), one of the citrus flavonoids, exerted outstanding effect on lipid metabolism disorder. However, the underlying mechanism of NOB exerting effect on hepatic lipid alternation remains unclear. In this study, the animal model was built by feeding APOE-/- mice with high fat diet (HFD). The results of Oil Red O-stained liver section and the biochemical assay of lipid parameters confirmed the

Sex-specific lipidomic profiling of the marine mussel Mytilus coruscus: A comparative analysis of male and female lipid metabolism.

Sex-dependent lipid metabolism plays a pivotal role in physiological adaptation and reproduction in marine bivalves, yet its molecular basis remains poorly understood. Here, UPLC-HRMS-based lipidomics was applied to characterize sex-specific lipid profiles in mussels. Females showed higher levels of glycerophospholipids, particularly phosphatidylcholine and phosphatidylethanolamine enriched with polyunsaturated fatty acids, whereas males contained more glycerolipids, especially saturated fatty acid-rich triacylglycerols. Principal component analysis and orthogonal partial least squares-discrim

Steaming reheating preserves the original flavor of prepared hand-grabbed mutton: Insights from lipidomics and flavor chemistry.

Reheating is critical for the quality of prepared dishes. This study investigated flavor variations of prepared hand-grabbed mutton (PHM) under three home reheating protocols (microwave, steaming and boiling) using electronic tongue, gas chromatography-mass spectrometry, sensory evaluation, and untargeted lipidomics. Compared with freshly cooked hand-grabbed mutton (FHM), boiling reduced the bitterness by 26.43%, microwave increased the saltiness by 4.1%, while steaming added four flavor compounds while preserving the original flavor of FHM. Further investigations were conducted to explore the
