Phenotypic and genotypic analysis of benzimidazole resistance in reciprocal genetic crosses of Haemonchus contortus.

Haemonchus contortus is arguably one of the most economically important and ubiquitous parasites of livestock globally and commonly involved in cases of anthelmintic resistance. Here, we performed reciprocal genetic crosses using susceptible (MHco3(ISE)) and multiple anthelmintic resistant (MHco18(UGA2004)) H. contortus isolates. Resultant admixed populations were designated MHco3/18 or MHco18/3, where the lead isolate reflects the origin of the females. Three independent filial generations were generated for each cross, which were subjected to bioassays, molecular approaches and population ge

Parasitism by Oestrus ovis: influence of sheep breed and nematode infections.

Previous studies showed that Santa Ines (SI) hair sheep were more resistant to gastrointestinal nematode infections (GIN) than Ile de France (IF) sheep. The present experiment aimed to evaluate if that reported resistance difference against GIN also occurred against Oestrus ovis infestation and also to evaluate the influence of O. ovis infestation on the gastrointestinal nematodes (GIN) infections. SI (n=12) and IF (n=12) young male lambs were weaned at 2 months of age and moved to a paddock (0.3 ha) with Brachiaria decumbens grass, where they also received concentrate ration. The animals were

The effect of infection with the abomasal nematode, Haemonchus contortus, on the avoidance behaviour of sheep in a motivational-choice test.

The behaviour of immune and non-immune sheep infected with H. contortus and undergoing a variety of experimental treatments was investigated in a motivational-choice test, the arena test. This test evaluates motivational state in sheep by pitting the motivation of test animals to approach a small flock of sheep against the motivation to avoid a human decoy located directly in front of the small flock. Approach distance is decreased by infection in immune ewes but was unaffected by infection in non-immune weaner lambs in the present study. Experimental drug-treatments with the opiate-antagonist

An epidemiological study of helminthiasis in cattle in subtropical Argentina.

Nine farms were each visited regularly for a year in 1979 and 1980 in Salta Province in the subtropics of Argentina. A necropsy was done at each visit and faecal and blood samples were taken from other animals mostly aged between 6 and 18 months. Ninety necropsies were done and nearly 1,500 faecal samples collected. Gastro-intestinal helminthiasis was most general and severe in autumn when calves were about 9 months old and this was followed by a dramatic fall in numbers of worms by the end of winter. Helminths were found in other organs but were not considered important. At peak periods 20% o

In vivo and in vitro anthelmintic activity of quebracho-chestnut tannin extract against Haemonchus contortus in lambs.

Haemonchus contortus is considered the most pathogenic gastrointestinal nematode of sheep and, due to the increasing resistance to synthetic anthelmintics, it poses a serious challenge to small ruminant production systems. The aim of this work was to evaluate the in vivo and in vitro anthelmintic activity of quebracho-chestnut tannin extract against Haemonchus contortus in lambs. The in vitro assays (egg hatching inhibition and larval migration inhibition) were performed to determine the effective concentrations required to inhibit 50% of egg hatching and larval migration (EC50), which were 3.

Haemonchus contortus parasitic stages development and host immune responses in lambs of different sheep breeds.

Variable host resistance against Haemonchus contortus infection was extensively associated with sheep breed, but the mechanisms responsible for the enhanced resistance remains scarcely elucidated. The aim of the present study was compare the phenotypic profile of three breeds (Santa Inês, Texel and White Dorper) with the relative expression of immune-related genes in the abomasal mucosa of sheep breeds infected with H. contortus. Besides, these phenotypic profiles were compared with relative expression of immune related genes in the animal abomasum. Decreasing resistance against H. contortus i

Experimental hyper-progesteronemia increases the egg shedding and fertility of Haemonchus contortus in sheep.

Hyperprogesteronemia effects on the establishment and fertility of Haemonchus contortus were measured in an experimental infection in gonadectomized male lambs. Sixteen lambs were divided into two groups. Lambs from the progesterone (PG) group (n = 8) were implanted subcutaneously at week -2 with two devices (CIDR 330, Zoetis lab®) containing 600 mg of ovine progesterone. The second group (n = 8) was not implanted and served as the control. Lambs from both groups were inoculated intra-ruminally with 5000 L3 of H. contortus at week 0. All lambs were humanely euthanized at week seven p.i. All la

In-vitro anthelmintic activity and phytochemistry of Salvia sclarea extracts against Haemonchuscontortus.

Haemonchus contortus -the barber's pole worm poses a critical threat to ruminant health and productivity worldwide. This is due to its high host pathogenicity and poor susceptibility to chemical anthelmintics. To address this threat, sustainable and natural solutions are needed to control its infections. This study evaluated the anthelmintic efficacy of Salvia sclarea extracts against H. contortus in comparison to reference drug - the Albendazole. The extracts were obtained in ethyl acetate, ethanol, and petroleum ether, analyzed phytochemically and subjected to in-vitro testing at different c

Catalytically Active Recombinant Cysteine Proteases of Haemonchus contortus: Their Ability to Degrade Host Blood Proteins and Modulate Coagulation.

Haemonchus contortus is a blood-feeding gastrointestinal nematode that significantly impacts the health and productivity of small ruminants. The burden of parasitism and the escalating incidence of anthelmintic resistance necessitate alternative control methods. Here, we characterize the enzymatic activities of five mammalian cell-expressed recombinant H. contortus cysteine proteases (HcCPs), which include two cathepsin B-like proteins (HcCBP1 and HcCBP2) and three cysteine protease 1 proteins (HcCP1a, HcCP1b, and HcCP1c). We hypothesize that these enzymes degrade host blood proteins, thereby

Antiparasitic Activity of Silver Nanoparticles Synthesized from Artemisia cina Aqueous Extract Against Haemonchus contortus.

Parasitic infections, particularly those caused by Haemonchus contortus (H. contortus), severely impact livestock production, with growing resistance to commercial anthelmintics posing a major challenge. Green-synthesized metallic nanoparticles using Artemisia cina (A. cina), a plant with known anthelmintic and antioxidant properties, represent a promising sustainable alternative for parasite control. In this study, silver nanoparticles (AgNPs) were synthesized using an aqueous extract of A. cina to evaluate their anthelmintic activity against infective larvae (L3) of H. contortus and their ef

Breeding options for nematode resistance in Lacaune dairy sheep.

Due to progressing anthelmintic resistance of gastrointestinal nematodes (GIN), supportive measures are needed to control these parasites. In sheep, it has been shown that selection towards an increased nematode resistance is feasible and that faecal egg count (FEC) is the generally acknowledged trait for selection. However, a selection based on FEC would come with certain costs, therefore auxiliary, cheaper resistance traits would be most welcome. FAMACHA©, a colour classification of the eyelid, usually used to determine the manifestation of an infection with Haemonchus contortus, could serve

Rapid, automated quantification of Haemonchus contortus ova in sheep faecal samples.

Haemonchus contortus is one of the most pathogenic nematodes affecting small ruminants globally and is responsible for large economic losses in the sheep and goat industry. Anthelmintic resistance is rampant in this parasite and thus parasite control programs must account for drug efficacy on individual farms and, sometimes, whether H. contortus is the most prevalent trichostrongylid. Historically, coproculture has been the main way to determine the prevalence of H. contortus in faecal samples due to the inability to morphologically differentiate between trichostrongylid egg types, but this pr

Effects of herbal nutraceuticals and/or zinc against Haemonchus contortus in lambs experimentally infected.

BACKGROUND: The gastrointestinal parasitic nematode Haemonchus contortus is a pathogenic organism resistant to several anthelmintics. This study assessed the efficacy of a medicinal herbal mixture (Herbmix) and organic zinc, as an essential trace element for the proper functioning of both unspecific and specific immune defensive mechanisms, against experimental infections with H. contortus in lambs. All lambs were infected orally with approximately 5000 third-stage larvae of a strain of H. contortus susceptible to anthelmintics (MHco1). Twenty-four female lambs 3-4 months of age were divided i

Development of a faecal occult blood test to determine the severity of Haemonchus contortus infections in sheep.

Haemonchus contortus commences feeding on host blood by day 11 of infection, which leads to the presence of blood in the host's faeces. This study examined the capacity for a faecal occult blood (FOB) test to determine the severity of H. contortus infection in sheep at pasture, and to predict a rise in worm egg count (WEC) as infection matures. Diluted faeces were assayed with Bayer Hemastix and the change in colour of the reagent patch was scored on a 9-point scale from 1 (negative) to 5 in half unit increments. Performance of the test was compared with four benchmarks for severe infection: (
