Chemical ecology and convergent evolution of natural hallucinogens: From ecological defense to conserved neural targets.

Natural hallucinogenic compounds have arisen independently across plants, fungi, and animals, evolving into a diverse chemical arsenal that includes phenethylamines, indolealkylamines, and terpenoid scaffolds. Beyond clinical and cultural frameworks, their ecological origins and evolutionary trajectories may help explain why such potent modulators of perception, emotion, and cognition persist in nature. Here, integrating chemical ecology, comparative genomics, biosynthetic logic, and evolutionary biology, we propose that these molecules may function as defensive agents or symbiosis-associated

The Use of Classic Hallucinogens/Psychedelics in a Therapeutic Context: Healthcare Policy Opportunities and Challenges.

Psychedelics or serotonergic hallucinogens are a group of substances that share the agonism of serotonergic 5-HT2A receptors as their main mechanism of action. Its main effects include changes in perception, cognitive process, and mood. Despite being used for centuries by different cultures in ritual contexts, these substances have currently aroused the interest of science and industry for their promising antidepressant, anxiolytic, and anti-addictive effects. Considering this evidence, this article aims to explore some of the possible health policy challenges to integrate these therapeutic to

Dissociations between the effects of hallucinogens on behavior and raphe unit activity in behaving cats.

The hypothesis that hallucinogenic drugs exert their behavioral effects by an action at pre- or postsynaptic serotonin receptors was evaluated by co-administering various drugs that possess either serotonin agonist or antagonist properties, while concurrently monitoring behavior and the electrophysiological activity of serotonin-containing dorsal and median raphe neurons in freely moving cats. Co-administration of the serotonin receptor blockers, metergoline or mianserin, with lysergic acid diethylamide (LSD) produced no change in the inhibitory effects of LSD on raphe neurons, but produced a

[Hallucinogenic fungi (psilocybe). Part II. Identification of Psilocybe semilanceata by PCR].

Psilocybe semilanceata belongs to the genus of Psilocybe; it is a hallucinogenic fungus, frequently found in regions with high rainfall, in the grass and moss. There are estimated to be as many as eight Psilocybe hallucinogenic species containing psychoactive alkaloids, such as psilocybin and psilocin, which may cause visual, auditory and other hallucinations, as well as profound changes in the perception of time and space. Since identification of P semilanceata in a form different than a fresh mushroom is almost impossible, the authors made an attempt at developing a method of fungi identific

[Hallucinogen induced psychic disorders. Subjective experiences, psychopathology and differential diagnosis].

The concepts of intoxication, intoxication psychosis, and drug-induced psychosis are frequently used to refer to psychopathological states caused by the intake of hallucinogenic substances. The considerable increase of intoxications with substances of the chemical class of amphetamines (in particular, "Ecstasy") in western countries should cause major concern. Moreover, the number of schizophrenic patients who also use hallucinogenic drugs is increasing. High incidence and comorbidity cause clinical problems and call for detailed knowledge of the drug related acute and chronic effects of the v

Hallucinogenic and neuroleptic drug interactions with potential neurotransmitter receptors in parasitic nematodes.

Receptors potentially involved in neurotransmitting have been characterised in the muscle tissue and in whole worms of the nematodes Ascaris suum and Onchocerca volvulus, respectively. Binding studies revealed a high affinity for LSD with apparent KD values of 94 nM for A. suum and 120 nM for O. volvulus, whereas those of the neuroleptics haloperidol, spiperone and mianserin were found to be in the micromolar range. A variety of neurotransmitter antagonists, known to bind with high affinities either to mammalian D1/2 or to 5-HT1/2 receptors, were tested for their ability to bind to the nematod

Pilocarpine, a non-hallucinogenic cholinergic agonist, elicits limb flicking in cats.

The centrally- and peripherally-acting muscarinic cholinergic agonist pilocarpine (PILO, 0.125-1.0 mg/kg, IP) elicited a significantly increased frequency of occurrence of limb flicking at 0.25-1.0 mg/kg, and significantly increased the frequency of occurrence of other grooming behaviors, in 4-6 cats in the 90 min following its administration. These effects of PILO at 0.5 mg/kg were antagonized by the peripherally-acting antimuscarinic agent, N-methylscopolamine (MESCO, 0.5 mg/kg, IP), when MESCO was administered 15 min before PILO. The same lisuride (LIS, 0.05 mg/kg, IP). These results provid

2,5-dimethoxy-4-methyl-amphetamine (STP): a new hallucinogenic drug.

We have assessed the effects in normal control volunteers of 2,5-dimethoxy-4-methyl-amphetamine, the chemical present in the hallucinogenic drug STP, in two independent trials. In low doses, this compound produces a mild euphoria. Doses greater than 3 milligrams may cause pronounced hallucinogenic effects lasting about 8 hours and similar to those produced by hallucinogenic doses of lysergic acid diethylamide, mescaline, and psilocybin. 2,5-Dimethoxy-4-methyl-amphetamine, which is chemically related to mescaline and amphetamine, is about 100 times more potent as a hallucinogen than mescaline a

Assessment of the reporting of harms in systematic reviews focused on hallucinogens: A cross-sectional study.

OBJECTIVE: To investigate the reporting of harms in systematic reviews (SRs) focused on hallucinogen use. METHODS: A search was conducted in May 2022 using MEDLINE, Embase, Epistemonikos, and Cochrane databases to retrieve SRs focused on the use of hallucinogens. Investigators screened the titles and abstracts from the search for study inclusion in a masked, triplicate fashion. Investigators analyzed the included SRs for reported harms linked to hallucinogen use via a pre-established harms reporting assessment. Methodological quality of SRs was graded using the A MeaSurement Tool to Assess Sy

Simultaneous analysis of stimulants, opioids, gabapentin, xylazine, benzodiazepines, cannabinoids, and novel stimulants/hallucinogens in human urine.

A liquid chromatography-tandem mass spectrometry (LC-MS/MS) method was developed for the determination of 67 drugs including stimulants, opioids, gabapentin, xylazine, benzodiazepines, cannabinoids, novel stimulants/hallucinogens, and their metabolites in urine. Urine samples were deconjugated, diluted, and fortified with isotopically labelled internal standards prior to analysis. The method was linear for all analytes with regression coefficients (R-values) > 0.99. The method limits of quantification were in the range of 0.1-15 ng/mL for all analytes. The recoveries of all drugs fortified in

Double-blind comparison of the two hallucinogens psilocybin and dextromethorphan: similarities and differences in subjective experiences.

RATIONALE: Although psilocybin and dextromethorphan (DXM) are hallucinogens, they have different receptor mechanisms of action and have not been directly compared. OBJECTIVE: This study compared subjective, behavioral, and physiological effects of psilocybin and dextromethorphan under conditions that minimized expectancy effects. METHODS: Single, acute oral doses of psilocybin (10, 20, 30 mg/70 kg), DXM (400 mg/70 kg), and placebo were administered under double-blind conditions to 20 healthy participants with histories of hallucinogen use. Comparative epidemiology of initial drug opportunities and transitions to first use: marijuana, cocaine, hallucinogens and heroin.

The earliest stages of involvement with illicit drugs have been understudied. In a recent report, we examined initial opportunities to try marijuana and transitions from first opportunity to first use of that drug. This report extends that work by investigating early involvement with cocaine, heroin, and hallucinogens as well. We examine sex and race ethnicity differences in estimates of having a drug opportunity, and in the probability of progressing from having an opportunity to try a drug to actually using the drug. Self-report interview data collected for the National Household Surveys on

Enhanced effect of the hallucinogen DOI in L-DOPA receptor Gpr143-deficient mice.

Hallucinogens, such as lysergic acid diethylamide, act primarily through the 5-hydroxytryptamine 2A (5-HT2A) receptor, but their regulatory mechanisms remain unclear. G protein-coupled receptor 143 (GPR143), an L-3,4-dihydroxyphenylalanine (L-DOPA) receptor, modulates specific GPCRs. We examined the role of GPR143 in the action of 5-HT2A receptor agonist 2,5-dimethoxy-4-iodoamphetamine (DOI). DOI-induced hyperlocomotion and c-Fos expression in the nucleus accumbens were enhanced in GPR143 knockout mice. In Chinese hamster ovary cells expressing 5-HT2A receptor, DOI-induced extracellular signal

A fatal case of aspiration due to consumption of the hallucinogenic tryptamine derivative dipropyltryptamine (DPT).

BACKGROUND AND AIM: This case involves a 20-year-old man with prior hallucinogen-use experience, who sniffed an unknown amount of dipropyltryptamine in an apartment. Dipropyltryptamine, a hallucinogenic compound belonging to the tryptamine class is recognized for inducing effects similar to dimethyltryptamine (DMT) but with a longer duration. Ten to fifteen minutes later he experienced visual hallucinations, followed by increasing apathy. Two hours post consumption he developed abdominal pain, leading to collapse, seizure, and vomiting. Despite emergency medical resuscitation on site, transpor

Neurovascular and neuroimaging effects of the hallucinogenic serotonin receptor agonist psilocin in the rat brain.

The development of pharmacological magnetic resonance imaging (phMRI) has presented the opportunity for investigation of the neurophysiological effects of drugs in vivo. Psilocin, a hallucinogen metabolised from psilocybin, was recently reported to evoke brain region-specific, phMRI signal changes in humans. The present study investigated the effects of psilocin in a rat model using phMRI and then probed the relationship between neuronal and haemodynamic responses using a multimodal measurement preparation. Psilocin (2 mg/kg or 0.03 mg/kg i.v.) or vehicle was administered to rats (N=6/group) d

Derived trail making test indices in a sample of hallucinogen abusers: demographic effects.

Derived indices on the Trail Making test (TMT), a test often used for screening for cognitive impairment, were examined in a sample of hallucinogen abusers in drug abuse treatment programs. A mixed race sample (N = 128) was drawn from electronic files of data from the Drug Abuse Treatment Outcome Study (DATOS). The DATOS was a naturalistic, prospective cohort study that collected data from 1991 to 1993 in 96 programs in 11 cities in the United States. Data were analyzed to determine the effects of demographic variables on derived indices created by adding, subtracting, multiplying, and dividin
