Halotolerant microbial consortia able to degrade highly recalcitrant plant biomass substrate.

The microbial degradation of plant-derived compounds under salinity stress remains largely underexplored. The pretreatment of lignocellulose material, which is often needed to improve the production of lignocellulose monomers, leads to high salt levels, generating a saline environment that raises technical considerations that influence subsequent downstream processes. Here, we constructed halotolerant lignocellulose degrading microbial consortia by enriching a salt marsh soil microbiome on a recalcitrant carbon and energy source, i.e., wheat straw. The consortia were obtained after six cycles

Bacterial microbiome and flavor metabolome shifts in traditional Hinggan league sauerkraut fermentation.

Microbial diversity is crucial for the flavor and quality of fermented vegetables. The unique geography and fermentation techniques of Hinggan League, Inner Mongolia, endow local sauerkraut with distinctive characteristics; however, the association between its microbiota and metabolites, as well as the underlying flavor formation mechanism, remains unclear, representing a gap in current research. Therefore, this study systematically investigated the interactions between microbial communities and metabolites during traditional sauerkraut fermentation in Hinggan League, Inner Mongolia, before an

Multi-omics uncovers nutrient stress-driven interactions in a synthetic Prymnesium parvum holobiont, with vitamin B12-limitation revealing mutualism.

Microalgal-bacterial interactions are central to nutrient cycling and ecosystem functioning in marine habitats, yet the mechanisms structuring these associations under defined nutrient constraints remain poorly resolved. Using a synthetic 15-member bacterial community (SynCom) and controlled nitrogen (N), phosphorus (P), and vitamin B12-limitation, we investigated how nutrient scarcity shapes the physiology, metabolism, and transcriptional activity of the harmful alga Prymnesium parvum and its associated microbiota. Under N- and P-limitation, the SynCom had minimal impact on algal growth despi

Microbial community succession during tobacco fermentation reveals a flavor-improving mechanism.

INTRODUCTION: Flue-cured tobacco (FCT) requires fermentation to increase quality, with microorganisms playing a key role. However, microbial succession and functions during long-term fermentation remain unclear. Artificial microbial fermentation, which is more controllable and efficient, focuses on mining functional strains to optimize the process. METHODS: In this study, the microbial community structure and function of FCT fermentated for 0-4 years were analyzed, and the changes of metabolites in tobacco leaves in different years were analyzed. Functional microorganisms were screened, and t

Characterization of proteins from the 3N5M family reveals an operationally stable amine transaminase.

Amine transaminases (ATA) convert ketones into optically active amines and are used to prepare active pharmaceutical ingredients and building blocks. Novel ATA can be identified in protein databases due to the extensive knowledge of sequence-function relationships. However, predicting thermo- and operational stability from the amino acid sequence is a persisting challenge and a vital step towards identifying efficient ATA biocatalysts for industrial applications. In this study, we performed a database mining and characterized selected putative enzymes of the β-alanine:pyruvate transaminase clu

Bacterial diversity in gut compartments of Tomicus brevipilosus: from shoot-boring preconditioning of Pinus yunnanensis to trunk-boring reproduction.

BACKGROUND: Gut bacteria are critically important to bark beetles, especially under environmental fluctuations. Tomicus brevipilosus undergoes a niche shift from shoot-boring (which weakens Pinus yunnanensis) to trunk-boring for reproduction. The relationship between this transition and the gut microbiota remains to be elucidated. METHODS: Using 16S rRNA sequencing, we characterized gut bacterial communities across adult feeding phase, sexes, and intestinal compartments. RESULTS: The alimentary canal was composed of six morphologically distinguishable regions, to which Malpighian tubules wer

Alterations in gut microbiota and metabolic profiling are associated with papillary thyroid cancer and BRAFV600E mutation.

PURPOSE: Emerging evidence indicates that gut microbial dysbiosis contributes to cancer development through metabolic regulation. The gut microbial and metabolic characteristics of papillary thyroid cancer (PTC) remain incompletely characterized, and the role of microbial dysbiosis in PTC tumorigenesis is not well understood. METHODS: This study recruited 70 PTC patients and 70 healthy controls (HCs) and performed 16S rRNA sequencing and LC-MS/MS metabolomic analysis of fecal samples. RESULTS: The PTC group demonstrated significantly greater microbial diversity and richness compared to the HC

Understanding phage dynamics and their potential roles during soy sauce fermentation using metagenome-assembled genomes.

The composition and functional roles of phages in fermented foods have been gaining increasing attention. However, their ecological functions and underlying mechanisms in high-salt soy sauce fermentation remain largely unexplored. In this study, we investigated phage communities, their potential functions, phage-host interactions, and host defense mechanisms in two different soy sauce fermentation processes (Cantonese-type process, CP; Japanese-type process, JP) using shotgun metagenomics. A total of 823 phage species (viral operational taxonomic units, vOTUs) were identified, with the majorit

Analysis of pharyngeal microbiome characteristics in HIV-infected individuals: correlation between the degree of immunosuppression and microbial dysbiosis.

OBJECTIVES: Despite the significant improvement in survival rates due to antiretroviral therapy, opportunistic infections continue to pose a major health risk for individuals living with HIV. As a key anatomical intersection of the upper respiratory and digestive tracts, the pharyngeal microbiome during HIV infection remains poorly characterized. This study aimed to compare the composition of bacterial and fungal communities in the pharynx between HIV-infected individuals and healthy controls, and to investigate their association with host immune status. METHODS: Throat swab samples were colle

Genomic insights into the metabolic complementarity of a synthetic marine consortium for hydrocarbon degradation in coastal sand microcosms.

Microbial consortia are essential for the degradation of complex hydrocarbon mixtures in coastal environments, yet the functional basis of consortium performance rarely resolved at strain level. In this study, we employed a Plackett-Burman experimental design to rationally assemble and optimize a synthetic consortium for the degradation of medium-grade crude oil in a sand-seawater microcosm. Six strains were selected: Stutzerimonas stutzeri CBC1 and CBC2, S. balearica GOM11, Pseudomonas sp. CBC3, Halomonas litopenaei GOM23 and Enterobacter kobei GOM24. After 45 days, the consortium removed 15.

Systemic lupus erythematosus patients have a distinct structural and functional skin microbiota compared with controls.

OBJECTIVE: The skin is the second most affected organ after articular involvement in systemic lupus erythematosus (SLE) patients. Cutaneous involvement occurs in approximately 80% of patients during the course of SLE. Interaction between the host and skin microorganism is a complex process. There are few studies on the diversity of skin microbes in SLE patients. Therefore, this study aims to explore the relationship between skin microorganisms and SLE. METHODS: A total of 20 SLE patients, 20 controls with rosacea and 20 healthy controls were selected as study subjects. Both the skin microbiot

A multi-omics investigation of the lung injury induced by PM2.5 at environmental levels via the lung-gut axis.

Long-term exposure to fine particulate matter (PM2.5) posed injury for gastrointestinal and respiratory systems, ascribing with the lung-gut axis. However, the cross-talk mechanisms remain unclear. Here, we attempted to establish the response networks of lung-gut axis in mice exposed to PM2.5 at environmental levels. Male Balb/c mice were exposed to PM2.5 (dose of 0.1, 0.5, and 1.0 mg/kg) collected from Chengdu, China for 10 weeks, through intratracheally instillation, and examined the effect of PM2.5 on lung functions of mice. The changes of lung and gut microbiota and metabolic profiles of m

Pretty in pink? Complementary strategies for analysing pink biofilms on historical buildings.

Salt-weathering is a deterioration mechanism affecting building materials that results from repetitive cycles of salt crystallisation-dissolution in the porous mineral network under changing environmental conditions, causing damage to surfaces. However, an additional biodeterioration phenomenon frequently associated with salt efflorescence is the appearance of coloured biofilms, comprising halotolerant/halophilic microorganisms, containing carotenoid pigments that cause pinkish patinas. In this work, two Austrian historical salt-weathered buildings showing pink biofilms, the St. Virgil's Chape

Chronic Intermittent Hypoxia Participates in the Pathogenesis of Atherosclerosis and Perturbs the Formation of Intestinal Microbiota.

Chronic intermittent hypoxia (CIH) is the prominent signature of highly prevalent obstructive sleep apnea (OSA) pathophysiology, which leads to increased risk and aggravation of atherosclerotic cardiovascular diseases. However, whether intestinal microbiota is implicated in the mechanisms linking CIH to arteriosclerosis (AS) pathogenesis remains unclear. The association of CIH with the development of altered gut microbiota (GM) may provide the opportunity to develop preventive strategies for atherosclerotic cardiovascular risk reduction. Animal models of apolipoprotein E-deficient (apoE-/-) mi

Heat stress during late gestation disrupts maternal microbial transmission with altered offspring's gut microbial colonization and serum metabolites in a pig model.

Heat stress (HS) during gestation has been associated with negative outcomes, such as preterm birth or postnatal metabolic syndromes. The intestinal microbiota is a unique ecosystem playing an essential role in mediating the metabolism and health of mammals. Here we hypothesize late gestational HS alters maternal microbial transmission and structures offspring's intestinal microbiota and serum metabolic profiles. Our results show maternal HS alters bacterial β-diversity and composition in sows and their piglets. In the maternal intestine, genera Ruminococcaceae UCG-005, [Eubacterium] coprostan

Salinity-adapted and core microbiota along the soil salinity gradients encircling the Tarim Basin.

BACKGROUND: Soil salinization affects nearly one-tenth of global drylands and imposes strong environmental filters on microbial communities. The Tarim Basin in Xinjiang, China, is one of the world's largest inland basins and an extreme saline-arid ecosystem, yet a basin-scale understanding of its microbiota remains lacking. In this study, we collected 132 soil samples along a salinity gradient spanning more than 3,000 km encircling the Tarim Basin. By combining amplicon sequencing with measurements of soil physicochemical properties, we aimed to unravel the salinity-adapted microbiota and core
