Cytokines and local factors which affect osteoclast function.

Bone remodeling is a local phenomenon which occurs in discrete packets throughout the skeleton. The cellular events which comprise the remodeling sequence are controlled by cytokines which are generated in the microenvironment of the bone resorbing pockets. These cytokines are derived from marrow mononuclear cells or from bone cells themselves, or they are incorporated into the bone matrix and released in biologically active form as bone resorbs. Evidence is accumulating that some of these cytokines play an important role not just in physiological bone remodeling, but also in common diseases o

The behaviour of fatigue-induced microdamage in compact bone samples from control and ovariectomised sheep.

This study investigates the effect of microdamage on bone quality in osteoporosis using an ovariectomised (OVX) sheep model of osteoporosis. Thirty-four sheep were divided into an OVX group (n=16) and a control group (n=18). Fluorochromes were administered intravenously at 3 monthly intervals after surgery to label bone turnover. After sacrifice, beams were removed from the metatarsal and tested in three-point bending. Following failure, microcracks were identified and quantified in terms of region, location and interaction with osteons. Number of cycles to failure (Nf) was lower in the OVX gr

[Peri-tumor mandibular demineralization].

This study describes hitherto unpublished observations suggesting mechanism by which a tumor can cause massive resorption of the mandible. Microradiographic analysis of the cancerous areas has shown a mobilization of minerals from compact and spongious bone tissue accompanied by incomplete degradation of collagen fibres. The minerals are deposited in the lumen of the Haversian canals, the medullar cavity and the perimandibular soft tissue, areas which are not normally mineralized. Electronic microprobe and X-ray diffraction analysis revealed that in the mineralized zones undergoing demineraliz

Peripheral Osteoma on the Medial Surface of the Mandibular Ramus: A Case Report.

Peripheral osteomas are uncommon benign osteogenic lesions that arise from the periosteum, with the lingual mandibular surface being a recognized site. However, solitary pedunculated lesions at the posterior medial ramus-body junction are rare. This case report describes a 34-year-old man who presented with mild, intermittent dull pain in the right posterior mandible, exacerbated by mastication for three months, without swelling or paresthesia. Clinical examination revealed tenderness near the ramus-body junction. Cone-beam computed tomography (CBCT) demonstrated a well-circumscribed, peduncul

Spatial orientation of the microscopic elements of cortical repair bone.

This study was designed to assess whether the original orientation of the elements of cortical diaphyseal bone is reestablished during 18 months of repair. Full thickness 3-mm drill holes were created surgically through the tibial and femoral cortices of adult rabbits and studied by light and polarizing microscopic analysis at 1, 4, 8, and 16 weeks and 6, 12, and 18 months after the defects were created. From 16 weeks to 12 months the defects were repaired fully with lamellar bone, while the longitudinal spatial orientation of the repair lamellae was at a distinct angle to that of the original

Ontogenetic and regional morphologic variations in the turkey ulna diaphysis: implications for functional adaptation of cortical bone.

This study examines relationships between bone morphology and mechanically mediated strain/fluid-flow patterns in an avian species. Using mid-diaphyseal transverse sections of domestic turkey ulnae (from 11 subadults and 11 adults), we quantified developmental changes in predominant collagen fiber orientation (CFO), mineral content (%ash), and microstructure in cortical octants or quadrants (i.e., %ash). Geometric parameters were examined using whole mid-diaphyseal cross-sections. The ulna undergoes habitual bending and torsion, and demonstrates nonuniform matrix fluid-flow patterns, and high

Effect of Solute Molecular Weights on Mass Transfer within the Rat Lacunar-Canalicular System under Gravity.

BACKGROUND: The lacunar-canalicular system (LCS) serves as the mechanobiological foundation for bone tissue metabolism, mechanotransduction, and functional adaptation. However, the impact of solutes with varying molecular weights on LCS mass transfer under gravity remains unclear. METHODS: Rhodamine tracers of varying molecular weights were injected into the peritoneal cavity of SD rats and LCS mass transfer experiments were performed under normal and hypergravity conditions. Femurs were extracted from rats and prepared into bone section samples, which were then observed under a laser scannin

Histopathology of subcutaneously preserved autologous bone flap after decompressive craniectomy: a prospective study.

BACKGROUND: Limited reports are available regarding the viability of subcutaneously preserved autologous bone flaps after decompressive craniectomy. The present study was undertaken to evaluate the histopathological changes in these autologous bone flaps. METHODS: Between January 2011 and July 2012, 50 patients were prospectively studied at the time of cranioplasty. Bone flap retrieved from the abdominal wall was subjected to histopathological examination to look for mononuclear cell infiltration into the Haversian system, presence of osteocytes, osteoblastic activity, angiogenesis and new bo

Micro- and nano-structural analyses of damage in bone.

Skeletal fractures represent a significant medical and economic burden for our society. In the US alone, age-related hip, spine, and wrist fractures accounted for more than $17 billion in direct health care costs in 2001. Moreover, skeletal fractures are not limited to the elderly; stress fractures and impact/trauma-related fractures are a significant problem in younger people also. Gaining insight into the mechanisms of fracture and how these mechanisms are modulated by intrinsic as well as extrinsic factors may improve the ability to define fracture risk and develop and assess preventative t

Atypical mature bone in the otosclerotic otic capsule as the differentiated zone of an invasive osseous neoplasm.

CONCLUSION: A large proportion of the mature otic capsule bone in cases of otosclerosis lies in plaques in direct contiguity with active otosclerosis and, because it shows significant structural defects, it should be regarded as part of the otosclerotic process. These appearances support our previously described suggestion that otosclerosis is an invasive osseous neoplasm, the mature atypical bone representing differentiation of earlier-formed invasive neoplastic osseous tissue. OBJECTIVES: We sought structural features in differentiated bone within the otic capsules of cases of otosclerosis

Maxillary sinus floor augmentation using a nano-crystalline hydroxyapatite silica gel: case series and 3-month preliminary histological results.

The aim of this case series is to histologically examine a new hydroxyapatite in sinus lift procedure after 3 months. Ten 2-stage sinus lifts were performed in 10 healthy patients having initial bone height of 1-2mm and bone width of 5mm, asking for a fixed implant-supported rehabilitation. After graft material augmentation, a rough-surfaced mini-implant was inserted to maintain stability of the sinus widow. A bioptical core containing a mini-implant was retrieved 3 months after maxillary sinus augmentation with NanoBone(®) and processed for undecalcified histology. From the histomorphometric

Evaluating bone mass and bone quality in patients with breast cancer.

Bone remodeling is a process by which bone renews itself focally in distinct areas on cancellous (ie, trabecular) bone and/or in the Haversian systems of cortical (or compact) bone. Normal bone turnover involves the ordered metabolism of bone-resorbing cells (osteoclasts) and bone-forming cells (osteoblasts). Estrogen exerts a multitude of actions on bone tissues and is integral to bone health, and estrogen deprivation leads to accelerated bone loss. Bone strength reflects the integration of bone density and bone quality. Methods to assess bone strength fall into 3 categories: radiologic (ie,

Site-specific alterations of bone matrix mineralization, micro-hardness, and density of mineralized osteocyte lacunae in men with alcohol-associated liver disease: implications for vertebral fracture predilection.

Low bone quality has been reported as a source of bone fragility in individuals with alcohol-associated liver disease (AALD). Alterations of the bone matrix, including the spatial BMD distribution and osteocyte lacunar network characteristics, have not been comprehensively assessed in common fracture sites of individuals with AALD. Considering the clinical relevance of vertebral and femoral fragility fractures among these individuals, this cross-sectional study aimed to analyze site-specific BMD distribution, bone mechanical properties, osteocyte lacunar network, and Haversian system character

The cast imaging of the osteon lacunar-canalicular system and the implications with functional models of intracanalicular flow.

A casting technique with methyl-methacrylate (MMA) was applied to the study of the osteon lacunar-canalicular network of human and rabbit cortical bone. The MMA monomer infiltration inside the vascular canals and from these into the lacunar-canalicular system was driven by capillarity, helped by evaporation and the resulting negative pressure in a system of small pipes. There was uniform, centrifugal penetration of the resin inside some osteons, but this was limited to a depth of four to five layers of lacunae. Moreover, not all of the osteon population was infiltrated. This failure can be the

Fourier transform infrared spectroscopic imaging parameters describing acid phosphate substitution in biologic hydroxyapatite.

Acid phosphate substitution into mineralized tissues is an important determinant of their mechanical properties and their response to treatment. This study identifies and validates Fourier transform infrared spectroscopic imaging (FTIRI) spectral parameters that provide information on the acid phosphate (HPO4) substitution into hydroxyapatite in developing mineralized tissues. Curve fitting and Fourier self-deconvolution were used to identify subband positions in model compounds (with and without HPO4). The intensity of subbands at 1127 and 1110 cm(-1) correlated with the acid phosphate conten
