Impact of transcutaneous vagus nerve stimulation on healthy cognitive and brain aging.

Evidence for clinically meaningful benefits of transcutaneous vagus nerve stimulation (VNS) has been rapidly accumulating over the past 15  years. This relatively novel non-invasive brain stimulation technique has been applied to a wide range of neuropsychiatric disorders including schizophrenia, obsessive compulsive disorder, panic disorder, post-traumatic stress disorder, bipolar disorder, and Alzheimer's disease. More recently, non-invasive forms of VNS have allowed for investigations within healthy aging populations. These results offer insight into protocol considerations specific to olde

Healthy brain aging assessed with [18F]FDG and [11C]UCB-J PET.

BACKGROUND: The average human lifespan has increased dramatically over the past century. However, molecular and physiological alterations of the healthy brain during aging remain incompletely understood. Generalized synaptic restructuring may contribute to healthy aging and the reduced metabolism observed in the aged brain. The aim of this study was to assess healthy brain aging using [18F]FDG as a measure of cerebral glucose consumption and [11C]UCB-J PET as an indicator of synaptic density. METHOD: Using in vivo PET imaging and the novel synaptic-vesicle-glycoprotein 2A (SV2A) radioligand [

Respiratory-related evoked potentials in chronic obstructive pulmonary disease and healthy aging.

Altered neural processing and increased respiratory sensations have been reported in chronic obstructive pulmonary disease (COPD) as larger respiratory-related evoked potentials (RREPs), but the effect of healthy-aging has not been considered adequately. We tested RREPs evoked by brief airway occlusions in 10 participants with moderate-to-severe COPD, 11 age-matched controls (AMC) and 14 young controls (YC), with similar airway occlusion pressure stimuli across groups. Mean age was 76 years for COPD and AMC groups, and 30 years for the YC group. Occlusion intensity and unpleasantness was rated

Physiological and metabolic effects of healthy female aging on exhaled breath biomarkers.

Healthy aging driven physio-metabolic events in females hold the key to complex in vivo mechanistic links and systemic cross talks. Effects from basic changes at genome, proteome, metabolome, and lipidome levels are often reflected at the upstream phenome (e.g., breath volatome) cascades. Here, we have analyzed exhaled volatile metabolites (measured via real time mass spectrometry based breathomics) data from 204 healthy females, aged between 07 and 80 years. Age related substance-specific differences were observed in breath biomarkers. Exhalation of blood-borne endogenous organosulfur, short-

Relationship between brain AD biomarkers and episodic memory performance in healthy aging.

The presence of brain biomarkers can be observed decades before the first clinical symptoms of Alzheimer's disease (AD). We aimed to determine whether associations between biomarkers and episodic memory performance already exist in a healthy late middle-aged population or only in participants over 60 years old. Performance at the Free and Cued Selective Reminding Test [FCSRT], the Logical Memory test and the Mnemonic Similarity Task [MST] was determined in sixty healthy participants (50-70 y.) with a negative status for amyloid-beta (Aβ) biomarker. We assessed Aβ cortical level and tau/neuroin

Pleiotropic Effects of French Maritime Pine Bark Extract to Promote Healthy Aging.

Extension of the healthy life span is of primary importance for the aging society. Among exercise, healthy nutrition, and mental training, food supplements are widely used as preventive measures to postpone the diverse symptoms of aging. The extract from the bark of the French maritime pine, Pycnogenol, rich on flavonoids, has anti-inflammatory and antioxidative property, proven in in vivo studies. The extract reduces oxidative stress and improves endothelial health. Its antithrombotic properties are based on inhibition of platelet aggregation. In double-blind, placebo-controlled clinical stud

Nutritional Considerations for Healthy Aging and Reduction in Age-Related Chronic Disease.

A projected doubling in the global population of people aged ≥60 y by the year 2050 has major health and economic implications, especially in developing regions. Burdens of unhealthy aging associated with chronic noncommunicable and other age-related diseases may be largely preventable with lifestyle modification, including diet. However, as adults age they become at risk of "nutritional frailty," which can compromise their ability to meet nutritional requirements at a time when specific nutrient needs may be high. This review highlights the role of nutrition science in promoting healthy aging

Gender-Specific Differences in Skeletal Muscle 11β-HSD1 Expression Across Healthy Aging.

CONTEXT: Cushing's syndrome is characterized by marked changes in body composition (sarcopenia, obesity, and osteoporosis) that have similarities with those seen in aging. 11β-hydroxysteroid dehydrogenase type 1 (11β-HSD1) converts glucocorticoids to their active form (cortisone to cortisol in humans), resulting in local tissue amplification of effect. OBJECTIVE: To evaluate 11β-HSD1 expression and activity with age, specifically in muscle. To determine putative causes for increased activity with age and its consequences upon phenotypic markers of adverse aging. DESIGN: Cross-sectional obser

Healthy aging and preclinical dementia: the United States-Israel Longitudinal Database project.

This article proposes the establishment of a United States-Israel Longitudinal Database for Healthy Aging and Preclinical Dementia as a prototype model for the eventual creation of an international database. It is envisioned that such a comprehensive international database, as a shared research resource, will provide the foundation for a systems approach to solve the dual public health problems of: (1) Early detection of individuals at an elevated risk of developing Alzheimer's disease, and (2) Developing interventions to delay onset of, or prevent, chronic brain disorders later in life.

Six Bioactive Small Molecules from Tremella fuciformis with Potential for Healthy Aging.

Six representative small-molecule constituents were isolated and structurally characterized from the edible mushroom Tremella fuciformis: cis-oleic acid (1), 1-oleoylglycerol (2), cerevisterol (3), a cerebroside (4), trans-homoaconitic acid (5), and uridine (6). Compound 5, a tricarboxylic acid derivative, is reported for the first time as abundantly accumulated in this fungus. Aging-relevant assays showed that 1 potently inhibited α-glucosidase (IC50 = 0.02 mM, surpassing acarbose) and moderately inhibited acetylcholinesterase (IC50 = 0.29 mM); 5 competitively inhibited alkaline phosphatase (

Sensorimotor Adaptation to a Nonuniform Formant Perturbation Is Preserved in Healthy Aging.

PURPOSE: Young adult speakers can be induced to increase the acoustic contrast between vowels through exposure to a nonuniform auditory "centralization" feedback perturbation that makes vowels sound less distinct from one another. While this result suggests that sensorimotor adaptation is a promising learning mechanism to drive behaviorally relevant changes related to speech intelligibility, existing res sensorimotor adaptation in speech has largely focused on younger adults. Given established age-related deficits in speech motor performance, it is possible that healthy aging itself negatively

Organ-Specific Proteomic Aging Clocks Reveal Complexities Underlying the Healthy Drinking Paradox.

The "healthy drinking" paradox refers to situations where alcohol consumption is misleadingly associated with better health outcomes, a pattern frequently observed in observational studies that are prone to a range of confounding factors. Some recent studies developing organ-specific proteomic aging clocks report that alcohol consumption is linked to slower aging in select organs, even as it accelerates aging in others, thus echoing this paradox, with substantial discrepancies across such studies. In this Perspective, I summarize findings to date, discuss potential reasons for these contradict

Immunofitness in the elderly: The role of vaccination in promoting healthy aging.

Aging reshapes immunity through immunosenescence and inflammaging, increasing susceptibility to infection, exacerbating chronic conditions, and blunting vaccine responses. This review frames "immunofitness" as a practical goal of healthy aging and examines how adult vaccination builds immune resilience. Vaccination strengthens adaptive memory, leverages adjuvants to optimize antigen presentation, and can reprogramme innate cells (trained immunity), yielding heterologous benefits beyond target pathogens. We integrate evidence in older adults for influenza, respiratory syncytial virus, pneumococ

Neural and motor mechanisms of handwriting: from healthy aging to neurodegenerative disorders.

Handwriting is a complex cognitive and motor skill supported by a distributed brain network involving cortical, subcortical, and cerebellar regions responsible for planning, execution, and sensorimotor integration. Beyond its communicative role, handwriting provides biologically meaningful information about brain function and motor control, serving as a sensitive marker of both normal and pathological changes. Age-related alterations, such as reduced fine motor precision, impaired sensory feedback, and cognitive slowing, contribute to the progressive decline in handwriting fluency and legibili

Multimodal neuroimaging insights into the neurobiology of healthy aging across the lifespan.

PURPOSE: The purpose of this study was to advance our understanding of the neurobiology of healthy aging, which is crucial for improving quality of life and preventing age-related diseases. Despite its importance, a comprehensive investigation of this process has yet to be fully characterized. METHODS: We used a hybrid PET/MRI scanner to assess neurophysiological parameters in 80 healthy individuals aged 20-78. Cerebral amyloid-beta (Aβ) deposition and glucose metabolism were assessed using PET scans, while participants underwent simultaneous MRI scans. RESULTS: We found a positive correlati

Current Strategies for Healthy Aging: The Interplay Between Nutrition, Metabolism, and Exercise.

Healthy behaviors, such as a balanced diet, physical activity, and health care, have favored an increase in life expectancy. However, in Western society and some developing nations, a sedentary lifestyle and poor nutritional choices are more common, increasing the risk of developing disease in old age. We propose to characterize the interplay between nutrition, metabolism, and exercise for better and worse: its contribution to a pathological state when unbalanced, while integrated dietary interventions and exercise promote metabolic health and healthy aging. Caloric-restricted diets or diets r

Changes in Naturalistic Viewing in Healthy Aging and Amnestic Mild Cognitive Impairment.

Visual exploration-where the eyes move and when-is guided by prior experiences. Memory-guided viewing behavior is altered in healthy aging and is further disrupted in amnestic mild cognitive impairment (aMCI), a condition in which there is accelerated structural and functional decline of the hippocampus and associated medial temporal lobe structures (HC/MTL). Computational modeling has demonstrated the potential for rapid information flow from the HC/MTL to regions responsible for the cognitive control of eye movements, such that visual exploration behavior could be impacted in the moment and

Frontal GABA levels associate with musical rhythm production in healthy aging adults.

Changes in neuronal inhibition have been implicated in age-related declines in sensorimotor performance. While indirect evidence suggests that inhibitory mechanisms are also involved in rhythm entrainment, this association has not been tested. Using magnetic resonance spectroscopy, we tested the association between dorsomedial frontal GABA+/H2O concentrations and musical rhythm production in healthy younger (n = 14; 18-35) and older (n = 12; 55-79) adults, hypothesizing that lower GABA+/H2O concentrations would be associated with increased timing error, particularly on more difficult exercises

Addressing Healthy Aging: Time to Stop a Tsunami of Rising Alzheimer's Disease.

Alzheimer's disease [AD] disproportionately affects our seniors, diminishing their health and life expectancy. As the world population grows older, the collective burden of AD has become unsustainable. Globally, there were 43.8 million patients in 2016, with a projection of affecting 152 million by 2050. Recent discoveries have shown that molecular changes characteristic to AD manifested 20 years before discernable neurological phenotypes emerge. It is feasible to halt or reverse this pathological process before reaching an irremediable stage. To take advantage of this treatment window, we nee

Study protocol of the Hungarian Longitudinal Study of Healthy Brain Aging (HuBA).

BACKGROUND AND PURPOSE: Neurocognitive aging and the associated brain diseases impose a major social and economic burden. Therefore, substantial efforts have been put into revealing the lifestyle, the neurobiological and the genetic underpinnings of healthy neurocognitive aging. However, these studies take place almost exclusively in a limited number of highly-developed countries. Thus, it is an important open question to what extent their findings may generalize to neurocognitive aging in other, not yet investigated regions. The purpose of the Hungarian Longitudinal Study of Healthy Brain Agi
