Depression exacerbates AD pathology through lactate-dependent activation of microglial Kv1.3 to promote Aβ-containing exosome spreading.

Depression has been widely recognized as an important accelerating factor contributing to the aggravation of cognitive decline in Alzheimer's disease (AD) patients. Previous studies show that microglia-mediated neuroinflammation is a common and critical event in the etiology of both depression and dementia, but whether and how microglia participate in the process of depression-exacerbating AD pathology is largely unknown. By establishing the learned helplessness depression model on 5×FAD mice, we confirmed that depression can indeed promote Aβ plaque deposition and deteriorate the cognitive pe

Deep-brain magnetic stimulation promotes adult hippocampal neurogenesis and alleviates stress-related behaviors in mouse models for neuropsychiatric disorders.

BACKGROUND: Repetitive Transcranial Magnetic Stimulation (rTMS)/ Deep-brain Magnetic Stimulation (DMS) is an effective therapy for various neuropsychiatric disorders including major depression disorder. The molecular and cellular mechanisms underlying the impacts of rTMS/DMS on the brain are not yet fully understood. RESULTS: Here we studied the effects of deep-brain magnetic stimulation to brain on the molecular and cellular level. We examined the adult hippocampal neurogenesis and hippocampal synaptic plasticity of rodent under stress conditions with deep-brain magnetic stimulation treatmen

A NOS-III haplotype that includes functional polymorphisms is associated with bipolar disorder.

The pleiotropic messenger molecule nitric oxide (NO) has been implicated in a variety of higher CNS functions, including learning, memory, and emotionality. In the human brain, NO is predominantly formed by neuronal NO synthase (NOS-I), while the so-called 'endothelial' isoform NOS-III also contributes to NO generation. We recently reported that NOS-III knockout mice display decreased adult neurogenesis and reduced responsiveness in a learned helplessness paradigm. To examine whether NOS-III plays a role in affective disorders as well, we tested a NOS-III gene haplotype, consisting of three fu

Oversampled and undersolved: Depressive rumination from an active inference perspective.

Rumination is a widely recognized cognitive deviation in depression. Despite the recognition, researchers have struggled to explain why patients cannot disengage from the process, although it depresses their mood and fails to lead to effective problem-solving. We rethink rumination as repetitive but unsuccessful problem-solving attempts. Appealing to an active inference account, we suggest that adaptive problem-solving is based on the generation, evaluation, and performance of candidate policies that increase an organism's knowledge of its environment. We argue that the problem-solving process

Proteomic Analysis of the Hippocampus in Mouse Models of Trigeminal Neuralgia and Inescapable Shock-Induced Depression.

To investigate the behavioral and biomolecular similarity between neuralgia and depression, a trigeminal neuralgia (TN) mouse model was established by constriction of the infraorbital nerve (CION) to mimic clinical trigeminal neuropathic pain. A mouse learned helplessness (LH) model was developed to investigate inescapable foot-shock-induced psychiatric disorders like depression in humans. Mass spectrometry was used to assess changes in the biomolecules and signaling pathways in the hippocampus from TN or LH mice. TN mice developed not only significant mechanical allodynia but also depressive-

Phosphodiesterase-1b deletion confers depression-like behavioral resistance separate from stress-related effects in mice.

Phosphodiesterase-1b (Pde1b) is highly expressed in striatum, dentate gyrus, CA3 and substantia nigra. In a new Floxed Pde1b × CreCMV global knockout (KO) mouse model, we show an immobility-resistance phenotype that recapitulates that found in constitutive Pde1b KO mice. We use this new mouse model to show that the resistance to acute stress-induced depression-like phenotype is not the product of changes in locomotor activity or reactivity to other stressors (learned helplessness, novelty suppressed feeding or dexamethasone suppression), and is not associated with anhedonia using the sucrose p

Metabotropic glutamate receptor 5 antagonist 2-methyl-6-(phenylethynyl)pyridine produces antidepressant effects in rats: role of brain-derived neurotrophic factor.

Recent studies highlight that the brain glutamate system is involved in the etiology of depression and glutamatergic-targeting drugs are currently being explored as novel antidepressant medications. Previous studies reveal that the selective metabotropic glutamate receptor 5 antagonist 2-methyl-6-(phenylethynyl)pyridine (MPEP) produces antidepressant-like effects in behavioral despair and olfactory bulbectomy models. The current study aimed to further explore its behavioral actions in additional animal models of depression (forced swimming test (FST) and learned helplessness (LH) test) and its

Lesion of the medial prefrontal cortex and the subthalamic nucleus selectively affect depression-like behavior in rats.

Major depression (MD) has been projected to be the first leading cause of disability worldwide. Still, the pathophysiological processes and factors leading to depression remain largely unknown. The present study investigated the differential effects of selective medial prefrontal cortex (mPFC) and subthalamic nucleus (STN) lesions on depression-like and depression-associated behavior in rats, using the following behavioral paradigms: (i) learned helplessness model testing depressive behavior, (ii) elevated plus-maze testing anxious behavior, (iii) 8-arm radial maze testing cognitive performanc

Intimacy and depression in older women.

This study examines the effects of the qualities of intimate relationships on depression in older women. The data are from a study of women over 50, randomly selected from five census tracts in Madison, Wisconsin, who were given questionnaires about depression and the quality of their intimate relationships on two occasions (summer of 1978 and summer of 1979). The results showed that some dimensions of the relationships that the women described with their most significant others predicted depression. The more depressed the women were, the more they felt that (1) the relationship was less frien

A review of the demographic, clinical and psychosocial correlates of perceived control in three chronic motor illnesses.

PURPOSE: To review the correlates of measures of control in people with multiple sclerosis (MS), Parkinson's disease (PD) and motor neurone disease (MND). METHOD: Studies on the relationship between aspects of control and demographic, clinical and psychological factors were collated and reviewed using a narrative synthesis. RESULTS: Forty-four papers were found which examined different types of both disease and life control. PD studies had not examined self-efficacy or helplessness and only locus of control was used in MND studies. Age, gender and disease duration were not consistently relat

Long-term effects of controllability or the lack of it on coping abilities and stress resilience in the rat.

Findings suggest that stress-induced impaired learning and coping abilities may be attributed more to the psychological nature of the stressor, rather than its physical properties. It has been proposed that establishing controllability over stressors can ameliorate some of its effects on cognition and behavior. Gaining controllability was suggested to be associated with the development of stress resilience. Based on repeated exposure to the two-way shuttle avoidance task, we previously developed and validated a behavioral task that leads to a strict dissociation between gaining controllability

'I accept it [staff assistance]; no choice': an ethnographic study of residents' attitudes towards mobility within nursing homes.

BACKGROUND: Mobility contributes to the quality of life and independence of residents in nursing homes. To perform resident-centred mobility care, staff need to understand residents' physical capacity and perspectives of care. AIMS AND OBJECTIVES: The aim of the study was to explore residents' perspectives of intrinsic factors influencing their mobility and associations between these factors with a view to informing resident mobility care practice improvements. DESIGN: The study was part of a larger ethnographic project exploring safe resident mobility care in nursing homes. METHODS: Semi-s

Gender and victimization by intimates.

Recent data demonstrate that, although gender has an impact upon the experience of being a victim of an intimate's violence, there is no particular personality pattern that leads one to become a victim. Rather, women--who are socialized to adapt and submit, and who are likely to become victims of men's sexual violence or physical abuse--may not develop adequate self-protection skills as children, especially if they come from childhood homes in which females are victimized, leading to a later vulnerability to physical and sexual abuse. Men, however, socialized to express anger and aggression in

Animal models of depression and neuroplasticity: assessing drug action in relation to behavior and neurogenesis.

Depression is among the most prevalent forms of mental illness and a major cause of morbidity worldwide. Diagnosis of depression is mainly based on symptomatic criteria, and the heterogeneity of the disease suggests that multiple different biological mechanisms may underlie its etiology. Animal models have been important for recent advances in experimental neuroscience, including modeling of human mood disorders, such as depression and anxiety. Over the past few decades, a number of stress and neurobiochemical models have been developed as primary efficacy measures in depression trials, which

Hypnosis in treating symptoms and risk factors of major depression.

This article summarizes aspects of effective psychotherapy for major depression and describes how hypnosis can further enhance therapeutic effectiveness. Hypnosis is helpful in reducing common symptoms of major depression such as agitation and rumination and thereby may decrease a client' sense of helplessness and hopelessness. Hypnosis is also effective in facilitating the learning of new skills, a core component of all empirically supported treatments for major depression. The acquisition of such skills has also been shown to not only reduce depression, but also the likelihood of relapses, t

Psychometric Validation of the STEM Disciplines Attitude Scale (SDAS): Investigating Measurement Invariance and Psychological Correlates in Italian High School Students.

Promoting positive student attitudes toward STEM is essential for fostering innovation; however, many existing measures do not adequately distinguish attitudes across specific disciplines, particularly within hard sciences such as physics and chemistry. This study presents the development and validation of the STEM Disciplines Attitude Scale (SDAS). Across three studies involving 2577 high school students, Exploratory and Confirmatory Factor Analyses supported a 25-item, five-factor structure assessing attitudes toward Mathematics, Physics, Chemistry, Engineering, and Science and Technology. Mice heterozygous for cathepsin D deficiency exhibit mania-related behavior and stress-induced depression.

Mutations in cathepsin D (CTSD), an aspartic protease in the endosomal-lysosomal system, underlie congenital neuronal ceroid-lipofuscinosis (cNCL, also known as CLN10), a devastating neurodegenerative disease. CLN10 patients die within the first few days of life, and in the few patients who live into adulthood psychopathological symptoms have not been reported. Extensive neuropathology and altered neurotransmission have been reported in CTSD-deficient mice; however signs of neuropsychiatric behavior in these mice are not well characterized due to the severe movement disorder and premature deat
