Suppression of TNF-alpha gene expression by hemin: implications for the role of iron homeostasis in host inflammatory responses.

Tumor necrosis factor alpha (TNF-alpha) has multiple effects on iron homeostasis and erythropoiesis and has been implicated in the pathogenesis of the anemia of inflammation. We postulated that intracellular iron in turn may regulate the expression of TNF-alpha. In the human monocytic leukemia cell line, THP-1, low basal TNF-alpha message levels were stimulated (sevenfold) when serum was excluded from the culture medium. Addition of hemin completely suppressed TNF-alpha expression. Similarly, hemin suppressed lipopolysaccharide-induced expression of TNF-alpha. Sn-protoporphyrin, an inhibitor o

Development of refractoriness of induced human heme oxygenase-1 gene expression to reinduction by UVA irradiation and hemin.

Using primary human fibroblasts we have observed the existence of an acquired refractoriness of the heme oxygenase-1 gene to induction by a second dose of UVA (320-380 nm) radiation. We studied the kinetics of development of refractoriness over a time interval of up to 72 h between the first inducing event and the second (challenge) dose. Complete refractoriness was observed at 48 h. We also studied development of refractoriness after UVA, sodium arsenite and H2O2 treatment in all possible combinations and demonstrated that only UVA led to refractoriness. Ultraviolet radiation induced partial

Hemin-induced reactive oxygen species triggers autophagy-dependent macrophage differentiation and pro-inflammatory responses in THP-1 cells.

The toxic effect of oxidized-heme, also known as hemin, is implicated in developing adverse clinical outcome in various hematolytic diseases. To simulate and reconstruct the molecular events associated with hemin exposure on circulating monocytes, we employed a THP-1 cell line based in vitro model. Flow cytometry and Western blot analyses were subsequently applied. Hemin-treated THP-1 produced ROS in a dose-dependent manner which resulted in 10-30 % of cell death primarily through apoptosis. Surviving cells induced autophagy which too was ROS-dependent, as revealed by application of N-acetyl-L

5-Aminolevulinate dehydratase porphyria: Update on hepatic 5-aminolevulinic acid synthase induction and long-term response to hemin.

BACKGROUND: 5-Aminolevulinic acid dehydratase (ALAD) porphyria (ADP) is an ultrarare autosomal recessive disease, with only eight documented cases, all of whom were males. Although classified as an acute hepatic porphyria (AHP), induction of the rate limiting hepatic enzyme 5-aminolevulinic acid synthase-1 (ALAS1) has not been demonstrated, and the marrow may also contribute excess 5-aminolevulinic acid (ALA). Two patients have died and reported follow up for the others is limited, so the natural history of this disease is poorly understood and treatment experience limited. METHODS: We report

Efficient double-quenching of electrochemiluminescence from CdS:Eu QDs by hemin-graphene-Au nanorods ternary composite for ultrasensitive immunoassay.

A novel ternary composite of hemin-graphene-Au nanorods (H-RGO-Au NRs) with high electrocatalytic activity was synthesized by a simple method. And this ternary composite was firstly used in construction of electrochemiluminescence (ECL) immunosensor due to its double-quenching effect of quantum dots (QDs). Based on the high electrocatalytic activity of ternary complexes for the reduction of H2O2 which acted as the coreactant of QDs-based ECL, as a result, the ECL intensity of QDs decreased. Besides, due to the ECL resonance energy transfer (ECL-RET) strategy between the large amount of Au nano

Intravenous hemin for prevention of frequent attacks of acute hepatic porphyria - a double-blind placebo controlled pilot study.

INTRODUCTION: Hepatic heme production is deficient in the acute hepatic porphyrias (AHPs) due to partial deficiencies of specific enzymes in the heme biosynthetic pathway. Attacks of severe neurological symptoms of AHP may recur frequently during adult life and are accompanied by induction of hepatic δ-aminolevulinic acid (ALA) synthase 1 (ALAS1, the initial and rate-limiting enzyme of this pathway) and marked elevations of ALA, porphobilinogen (PBG) and porphyrins. These levels decrease rapidly after intravenous heme replenishment, but verification of clinical benefit of this treatment approa

Ultrasensitive determination of aflatoxin B1 by flow injection chemiluminescence immunoassay based on hemin-integrated Tb-MOF(Fe) nanozyme.

Nanozymes are promising alternatives to natural enzymes in biosensing applications. To address the critical need for sensitive detection of the food contaminant aflatoxin B1 (AFB1), this study developed a chemiluminescence immunosensor utilizing a novel Tb-MOF(Fe) nanozyme. The Tb3+-based metal-organic framework (MOF) was synthesized via a facile one-pot method employing 4,4'-oxybisbenzoic acid (OBBA) and catalytic hemin as ligands. Encapsulation of hemin within the MOF matrix markedly enhanced the stability and peroxidase-like activity of the material. Featuring an abundant porous structure a

Hemin loaded Zn-N-C single-atom nanozymes for assay of propyl gallate and formaldehyde in food samples.

Single-atom nanozymes (SAzymes) show distinct advantages in catalytic activity and selectivity owing to their stability and special characteristic of maximum atomic utilization. Inspired by the structure of natural horseradish peroxidase (HRP), we developed a simple method for specific determination of both propyl gallate (PG) and formaldehyde (HCHO) by utilizing the intrinsic peroxidase mimics activity of hemin (hem) loaded Zn-nitrogen-carbon single-atom nanozymes (Zn-N-C@hem SAzymes). Zn-N-C@hem was prepared via a salt-template strategy and self-assembly, where hemin exhibits enhancing perox

Hemin enhances the cardioprotective effects of mesenchymal stem cell-derived exosomes against infarction via amelioration of cardiomyocyte senescence.

BACKGROUND: Application of mesenchymal stem cell-derived exosomes (MSC-EXO) has emerged as a novel therapeutic strategy for myocardial infarction (MI). Our previous study showed that pretreatment with hemin, a potent heme oxygenase-1 (HO-1) inducer, enhanced the cardioprotective effects of MSCs in a mouse model of MI. This study aimed to investigate the therapeutic effects of EXO derived from hemin-pretreated MSCs (Hemin-MSC-EXO) in MI and explore the potential mechanisms. METHODS: MSC-EXO and Hemin-MSC-EXO were collected and characterized. MSC-EXO and Hemin-MSC-EXO were intramuscularly injec

Hemin Reduces HMGB1 Release by UVB in an AMPK/HO-1-dependent Pathway in Human Keratinocytes HaCaT Cells.

BACKGROUND AND AIMS: High mobility group box 1 (HMGB1) plays an important role as a pro-inflammatory cytokine that regulates inflammation in various diseases. We hypothesized that hemin might reduce HMGB1 release through the induction of HO-1 in UVB-induced HaCaTs. METHODS: The effects of hemin on the release of HMGB1 in UVB exposure were evaluated. The mechanisms were investigated using various signal inhibitors and small interfering RNA techniques. RESULTS: Treatment with hemin inhibited reactive oxygen species (ROS) in UVB-induced HaCaTs in a dose-dependent manner. HMGB1 release by UVB wa

Upregulation of heme oxygenase-1 with hemin prevents D-galactosamine and lipopolysaccharide-induced acute hepatic injury in rats.

Heme oxygenase-1 (HO-1), the rate-limiting enzyme in heme catabolism, has been shown to be induced during oxidative injury, and its induction acts as an important cellular defense mechanism against such injuries. In this study, we examined the functional roles of HO-1 induction in a rat model of d-galactosamine (GalN) and lipopolysaccharide (LPS)-induced liver injury. We found that GalN/LPS treatment of rats produced severe hepatic injury, whereas upregulation of HO-1 by hemin pretreatment prevented rats from liver damage, as evidenced by decreased serum ALT, AST levels and ameliorated histolo

Multiple roles of haem in cystathionine β-synthase activity: implications for hemin and other therapies of acute hepatic porphyria.

The role of haem in the activity of cystathionine β-synthase (CBS) is reviewed and a hypothesis postulating multiple effects of haem on enzyme activity under conditions of haem excess or deficiency is proposed, with implications for some therapies of acute hepatic porphyrias. CBS utilises both haem and pyridoxal 5'-phosphate (PLP) as cofactors. Although haem does not participate directly in the catalytic process, it is vital for PLP binding to the enzyme and potentially also for CBS stability. Haem deficiency can therefore undermine CBS activity by impairing PLP binding and facilitating CBS de

Pharmacokinetics study of hemin in rats by applying (58)Fe-extrinsically labeling techniques in combination with ICP-MS method.

Iron is a challenging element due to its high background in various matrixes including blood, tissues even in the air and it is urgent to develop a method for the accurate determination of iron in bio-samples. After optimization of mass spectrometric conditions using collision cell technology and compensating for interference using a mathematical correction equation, an inductively coupled plasma mass spectrometry (ICP-MS) method for the quantitative determination of (58)Fe originating from hemin extrinsically labeled avoiding endogenous interference was developed. After a single step of dilut

Congo red binding phenotype is associated with hemin binding and increased infectivity of Shigella flexneri in the HeLa cell model.

Wild-type isolates of Shigella flexneri bind the dye Congo red from solid media, thus producing red (Crb+) colonies. Mutants which fail to bind the dye produce white colonies (Crb-) and are avirulent in a variety of systems. In S. flexneri the ability to bind Congo red correlates with the ability to bind hemin and protoporphyrin IX. Binding of hemin by Crb+ S. flexneri was observed both in solid media and in liquid assays. Results of competition experiments suggest that Congo red and hemin bind to the same site on the bacterial cell and are retained on the cell surface. Binding of hemin by Crb

Heme oxygenase-1 induction by hemin: a novel approach to enhance mitochondrial turnover and mitigate diet-induced liver damage.

Metabolic dysfunction-associated steatohepatitis (MASH) is characterized by lipid accumulation and inflammatory injury. Kupffer cells (KC) play a key role in the immune response and metabolic homeostasis in the liver. We evaluated the impact of hemin treatment on the phenotype of KC and on metabolism and mitochondrial dynamics of hepatocytes in a rat model of early-stage MASH induced by a high-carbohydrate diet. In the liver, HO-1 induction by hemin was associated with reduced tissue injury and apoptosis, restored oxidative balance, attenuated UPR activation, reinstated autophagic flux and imp

Hemin activates interleukin-17 signaling and CCAAT/enhancer-binding protein beta to promote neuroinflammation and blood-brain barrier disruption.

Heme overload, a feature of hemolytic conditions and cellular injury, can initiate oxidative stress and inflammatory cascades that perturb neurovascular homeostasis. However, the molecular mechanisms by which peripheral heme exposure disrupts blood-brain barrier (BBB) integrity remain incompletely defined. Here, we investigated key molecular mediators linking peripheral hemin challenge to neurovascular injury. Intraperitoneal administration of hemin induced marked BBB permeability in mice, accompanied by immune cell infiltration into the brain parenchyma, microglial pro-inflammatory activation

Employing aqueous CdTe quantum dots with diversified surface functionalities to discriminate between heme (Fe(II)) and hemin (Fe(III)).

The discrimination of ferrous and ferric states in the human body is one of the basic issues for disease control and prevention because Fe(II) and Fe(III) are a crucial redox pair during the process of material and energy metabolism. Herein, aqueous CdTe quantum dots (QDs) with diversified surface functionalities are applied to discriminate between heme (Fe(II)) and hemin (Fe(III)) by virtue of their difference in quenching QD fluorescence. In aqueous media, the interaction between QDs and heme/hemin mainly involves electrostatic interaction, which is greatly determined by the surface function
