Status of vaccine research and development of vaccines for Nipah virus.

Nipah virus (NiV) is a highly pathogenic, recently emerged paramyxovirus that has been responsible for sporadic outbreaks of respiratory and encephalitic disease in Southeast Asia. High case fatality rates have also been associated with recent outbreaks in Malaysia and Bangladesh. Although over two billion people currently live in regions in which NiV is endemic or in which the Pteropus fruit bat reservoir is commonly found, there is no approved vaccine to protect against NiV disease. This report examines the feasibility and current efforts to develop a NiV vaccine including potential hurdles

Emerging tropical diseases in Australia. Part 5. Hendra virus.

Hendra virus (HeV) was first isolated in 1994, from a disease outbreak involving at least 21 horses and two humans in the Brisbane suburb of Hendra, Australia. The affected horses and humans all developed a severe but unidentified respiratory disease that resulted in the deaths of one of the human cases and the deaths or putting down of 14 of the horses. The virus, isolated by culture from a horse and the kidney of the fatal human case, was initially characterised as a new member of the genus Morbillivirus in the family Paramyxoviridae. Comparative sequence analysis of part of the matrix prote

Genomic conservation and CNS-associated intra-host variation of Nipah virus during the 1998-1999 Malaysian outbreak.

OBJECTIVES: Nipah virus (NiV) is a highly pathogenic henipavirus responsible for recurrent outbreaks of severe encephalitis. This study aimed to address gaps in historical genomic data by characterizing NiV genomes recovered from archived central nervous system (CNS) tissues from the 1998 to 1999 Malaysian outbreak and evaluating viral evolution and intra-host genetic variation. METHODS: We analyzed 18 NiV genomes obtained from archived CNS tissues. Whole-genome sequencing was performed, followed by phylogenetic reconstruction, comparative genomic analysis, and intra-host single nucleotide va

Infectome Landscape of Rodents and Shrews in Guangdong Province Reveals Diverse Pathogens with Zoonotic Potential in Wildlife.

Rodents and shrews are important reservoir hosts due to their close association with human activities and their role in carrying various zoonotic pathogens. Recently, meta-transcriptomic sequencing has become a powerful tool for surveilling and screening novel pathogens from wild animals. However, many of these studies focused only on the diversity and genetic evolution of viruses from wildlife, while ignoring non-viral pathogens such as bacterial and eukaryotic microorganisms. Here, we performed a comprehensive infectome analysis of 227 tissue samples collected from 42 rodents and 16 shrews a

RNA Sensors as a Mechanism of Innate Immune Evasion among SARSCoV2, HIV and Nipah Viruses.

Innate immunity is the first line of defence elicited by the host immune system to fight against invading pathogens such as viruses and bacteria. From this elementary immune response, the more complex antigen-specific adaptive responses are recruited to provide a long-lasting memory against the pathogens. Innate immunity gets activated when the host cell utilizes a diverse set of receptors known as pattern recognition receptors (PRR) to recognize the viruses that have penetrated the host and responds with cellular processes like complement system, phagocytosis, cytokine release and inflammatio

Persistent T Cell Immunity Following Nipah Virus Infection: Evidence From Malaysian Survivors.

Nipah virus (NiV) induces strong humoral immunity, but the durability of NiV-specific memory T-cell responses remains unclear. We characterized cellular immunity approximately 25 years after infection in 4 survivors of the 1998 Malaysian outbreak. PBMCs were stimulated with overlapping peptide mini-pools spanning the NiV fusion (NiV-F) and attachment (NiV-G) glycoproteins, and responses were assessed using enzyme-linked immune absorbent spot and intracellular cytokine staining. Durable cytokine-producing T-cell responses were detected, with immunodominant epitopes in the NiV-G ectodomain. Surv

The Nipah virus threat in India: Epidemiological trends, risk drivers, and lessons for pandemic preparedness.

Nipah virus (NiV), a WHO-priority zoonotic pathogen, poses a recurrent public health threat in India. Since its initial detection in 2001, India has encountered multiple NiV spillover events, including repeated outbreaks in Kerala and recent cases reported in West Bengal in 2026. Although the pig-amplified epidemic in Malaysia (1998-1999) and the seasonal outbreaks associated with NiV-contaminated date palm sap consumption in Bangladesh are extensively documented, India represents a unique and relatively understudied epidemiological setting. NiV outbreaks in India are primarily linked to the N

Establishment of a Pseudovirus-Based Golden Hamster Model for the Attachment and Entry Stages of Hendra Virus Infection and Evaluation of Protective Immunity.

OBJECTIVE: Establish an in vivo evaluation model focused on the attachment and entry stages of Hendra virus infection for protective immunity assessment. METHODS: A golden hamster infection model based on recombinant Hendra-F/G pseudovirus was developed, and a luciferase luminescence assay was used to assess the optimal pseudoviral challenge in terms of route of infection, dose and detection time. The biodistribution of the pseudovirus in infected organs was evaluated using the IVIS spectral CT system. The protective effect of antibody prophylaxis was evaluated by measuring the luminescence i

Immunological correlates of protection afforded by PHV02 live, attenuated recombinant vesicular stomatitis virus vector vaccine against Nipah virus disease.

INTRODUCTION: Immune correlates of protection afforded by PHV02, a recombinant vesicular stomatitis (rVSV) vector vaccine against Nipah virus (NiV) disease, were investigated in the African green monkey (AGM) model. Neutralizing antibody to NiV has been proposed as the principal mediator of protection against future NiV infection. METHODS: Two approaches were used to determine the correlation between neutralizing antibody levels and outcomes following a severe (1,000 median lethal doses) intranasal/intratracheal (IN/IT) challenge with NiV (Bangladesh): (1) reduction in vaccine dose given 28 d

Detailed analysis of the pathologic hallmarks of Nipah virus (Malaysia) disease in the African green monkey infected by the intratracheal route.

Disease associated with Nipah virus infection causes a devastating and often fatal spectrum of syndromes predominated by both respiratory and neurologic conditions. Additionally, neurologic sequelae may manifest months to years later after virus exposure or apparent recovery. In the two decades since this disease emerged, much work has been completed in an attempt to understand the pathogenesis and facilitate development of medical countermeasures. Here we provide detailed organ system-specific pathologic findings following exposure of four African green monkeys to 2.41×105 pfu of the Malaysia

Case fatality rate and risk factors for Nipah virus encephalitis: A systematic review and meta-analysis.

BACKGROUND: A wide range of Nipah virus (NiV) encephalitis case fatality rates (CFR) have been reported. Data on the involvement of several potential risk factors in Nipah virus transmission remain controversial. We performed a systematic review and meta-analysis to estimate the pooled CFR of NiV encephalitis and to assess the risk factors for NiV infection. METHODS: Articles published up to the 27thof November 2018 in MedLine, Embase and Web of knowledge databases were considered for this study. We included cross-sectional, cohort, and case-control studies that have reported NiV CFR and/or r

The Importance of Lipid Conjugation on Anti-Fusion Peptides against Nipah Virus.

Nipah virus (NiV) is a recently emerging zoonotic virus that belongs to the Paramyxoviridae family and the Henipavirus genus. It causes a range of conditions, from asymptomatic infection to acute respiratory illness and fatal encephalitis. The high mortality rate of 40 to 90% ranks these viruses among the deadliest viruses known to infect humans. Currently, there is no antiviral drug available for Nipah virus disease and treatment is only supportive. Thus, there is an urgent demand for efficient antiviral therapies. NiV F protein, which catalyzes fusion between the viral and host membranes, is

The role of Denisovan paleohabitats in shaping modern human genetic resistance to viral, bacterial, and parasitic infections.

Denisovans contributed notably to the genomes of present-day East and Southeast Asians. However, the relationship between the inhabited paleohabitats and the adaptive genetic traits related to infections in modern humans remains underexplored. This study uses geospatial techniques to analyze climatic factors associated with three Denisovan archaeological sites linked to nine specimens. Additionally, past and present climates and biomes, as well as the geographic distributions of eight infectious agents and disease vector groups, were modeled and compared with the modern genetic heritage of Den

Biophysical Elucidation of the Distinct Roles the Nipah Virus Attachment and Fusion Glycoproteins Play in the Entry Process.

Nipah virus (NiV), a World Health Organization priority pathogen, is a lethal enveloped RNA virus within the Paramyxoviridae family. NiV attachment G and fusion F glycoproteins execute viral-cell or cell-cell membrane fusion during viral entry and syncytia formation, respectively. Despite past studies, several aspects of the molecular-level orchestration underlying this process, such as how F, G, and their interactions regulate fusion, remain poorly understood, obscured by challenges in decoupling fusion determinants in complex cellular environments. Here, we report a reconstitution-based appr

Multi-epitope vaccine design using in silico analysis of glycoprotein and nucleocapsid of NIPAH virus.

According to the 2018 WHO R&D Blueprint, Nipah virus (NiV) is a priority disease, and the development of a vaccine against NiV is strongly encouraged. According to criteria used to categorize zoonotic diseases, NiV is a stage III disease that can spread to people and cause unpredictable outbreaks. Since 2001, the NiV virus has caused annual outbreaks in Bangladesh, while in India it has caused occasional outbreaks. According to estimates, the mortality rate for infected individuals ranges from 70 to 91%. Using immunoinformatic approaches to anticipate the epitopes of the MHC-I, MHC-II, and B-c

Advances in diagnostics, vaccines and therapeutics for Nipah virus.

Nipah virus is an emerging zoonotic paramyxovirus that causes severe and often fatal respiratory and neurological disease in humans. The virus was first discovered after an outbreak of encephalitis in pig farmers in Malaysia and Singapore with subsequent outbreaks in Bangladesh or India occurring almost annually. Due to the highly pathogenic nature of NiV, its pandemic potential, and the lack of licensed vaccines or therapeutics, there is a requirement for research and development into highly sensitive and specific diagnostic tools as well as antivirals and vaccines to help prevent and control

Epidemiology, surveillance and control of Nipah virus infections in Malaysia.

The outbreak of Nipah virus, affecting pigs and pig-farm workers, was first noted in September 1998 in the north-western part of peninsular Malaysia. By March 1999, the outbreak had spread to other pig-farming areas of the country, inclusive of the neighbouring country, Singapore. A total of 283 human cases of viral encephalitis with 109 deaths were recorded in Malaysia from 29 September 1998 to December 1999. During the outbreak period, a number of surveillances under three broad groups; Surveillance in Human Health Sector, Surveillance in Animal Health Sector, and Surveillance for the Reserv
