BACKGROUND: Chronic obstructive pulmonary disease (COPD) is characterized by persistent airway inflammation, epithelial barrier dysfunction, and progressive structural remodeling. Although glycocalyx impairment has been implicated in COPD pathogenesis, the mechanisms linking glycocalyx degradation to epithelial barrier injury remain incompletely understood. Heparanase (HPSE), the only mammalian endoglycosidase capable of degrading heparan sulfate, may contribute to COPD progression by regulating epithelial glycocalyx integrity.
METHODS: HPSE expression was evaluated in serum samples from pati

Heparanase (HPSE) is a promising therapeutic target, yet current heparin-derived inhibitors are limited by structural heterogeneity, poor selectivity, and off-target anticoagulant effects. Here, we synthesized a library of structurally defined heparan sulfate (HS) oligosaccharides via a robust chemoenzymatic strategy, enabling a systematic investigation of HPSE cleavage specificity. Our data confirm the minimal cleavable tetrasaccharide motif for HPSE and the preference for the endolytic site proximal to the nonreducing end in HS hexasaccharides. We demonstrate that a 2-O-sulfated iduronic aci

Heparanase is an endoglycosidase that specifically cleaves heparan sulfate side chains of heparan sulfate proteoglycans, the major proteoglycans in the extracellular matrix and cell surfaces. Traditionally, heparanase activity was implicated in cellular invasion associated with angiogenesis, inflammation, and cancer metastasis. More recently, heparanase up-regulation was documented in an increasing number of primary human tumors. Iotan this study, we sought to investigate the expression of heparanase messenger RNA (mRNA) in normal cervical tissue and intraepithelial cervical lesion and its cli

Heparan sulfate proteoglycans are essential for biological processes regulated by fibroblast growth factors (FGFs). Heparan sulfate (HS) regulates the activity of FGFs by acting as a coreceptor at the cell surface, enhancing FGF-FGFR affinity, and being a storage reservoir for FGFs in the extracellular matrix (ECM). Here we demonstrate a critical role for heparanase during mouse submandibular gland (SMG) branching morphogenesis. Heparanase, an endoglycosidase, colocalized with perlecan in the basement membrane and in epithelial clefts of SMGs. Inhibition of heparanase activity in organ culture

Chimeric antigen receptor (CAR) T cell therapy has shown promise in solid tumours, but its efficacy is limited by dense extracellular matrix (ECM) that blocks T cell entry. We engineered mesothelin-targeted CAR T cells to express heparanase (HPSE) fused to the truncated hepatitis A virus pX domain (pX-Δ1-30), enabling surface display of HPSE on extracellular vesicles for localized, pH-dependent ECM degradation within the acidic tumour microenvironment while limiting systemic exposure. HPSE-pX-Δ1-30 CAR T (referred to as HPSE CAR T) cells penetrated ECM mimics nearly fourfold more effectively t

BACKGROUND: Heparan sulfate (HS) degradation mediates pulmonary endothelial hyper-permeability and acute pulmonary edema during acute respiratory distress syndrome (ARDS). The aim of this study was to examine whether histone H4 induced HS degradation by activating heparanase (HPSE) in chlorine gas (Cl2)-induced ARDS.
METHODS: Acute lung injury was induced by Cl2 exposure or histone H4 injection in C57BL/6 mice. Histone H4 in bronchoalveolar lavage fluid (BALF) and plasma was measured by ELISA. HS degradation was measured by immunostaining, ELISA, and flow cytometry. HPSE mRNA and protein were

Peritoneal fibrosis (PF) is a serious complication in patients undergoing peritoneal dialysis (PD). Heparanase (HPA) may drive the progression of PF by promoting inflammation and tissue remodeling. This study aimed to evaluate HPA expression in peritoneal dialysate from end-stage renal disease (ESRD) patients receiving PD and to assess the correlation between HPA and fibrosis markers in PF. Peritoneal dialysate samples from ESRD patients were collected to measure HPA, syndecan-1 (SDC-1), and fibrosis markers. To further investigate the regulatory role of HPA in PF, a mouse model was establishe

Depolymerized Fucoidan derivatives (DFd) are emerging anticancer agents, yet their relationship with heparanase (HPSE) remains unclear. H2O2-assisted depolymerization of native fucoidan (Fucus vesiculosus) generates DFd characterized by reduced number- and weight-average molecular masses (Mn and Mw) and degree of sulfation (DS) with oxidative structural modifications, resulting in heterogeneous yet bioactive compounds with reduced anticoagulant activity compared to the native fucoidan. Their inhibitory effect on HPSE in vitro was associated with both Mn and DS, although no dominant factor was

In addition to controlling smooth muscle tone in coronary vessels, endothelial cells also influence subjacent cardiomyocyte growth. Because heparanase, with exclusive expression in endothelial cells, enables extracellular matrix remodeling, angiogenesis, metabolic reprogramming, and cell survival, it is conceivable that it could also encourage development of cardiac hypertrophy. Global heparanase overexpression resulted in physiologic cardiac hypertrophy, likely an outcome of HSPG clustering and activation of hypertrophic signaling. The heparanase autocrine effect of releasing neuregulin-1 cou

Activity of the endo-beta-glucuronidase heparanase, capable of cleaving heparan sulfate (HS), is most often elevated in many types of tumors, associating with increased tumor metastasis and decreased patients' survival. Heparanase is therefore considered to be a valid drug target, and heparanase inhibitors are being evaluated clinically in cancer patients. Heparanase 2 (Hpa2) is a close homolog of heparanase that gained very little attention, likely because it lacks HS-degrading activity typical of heparanase. The role of Hpa2 in cancer was not examined in detail. In head and neck cancer, high

Sepsis is a life-threatening syndrome caused by a pathological host response to an infection that eventually, if uncontrolled, leads to septic shock and ultimately, death. In sepsis, a massive aggregation of pathogen-associated molecular patterns (PAMPs) and damage-associated molecular patterns (DAMPs) cause a cytokine storm. The endothelial glycocalyx (eGC) is a gel like layer on the luminal side of the endothelium that consists of proteoglycans, glycosaminoglycans (GAG) and plasma proteins. It is synthesized by endothelial cells and plays an active role in the regulation of inflammation, per

BACKGROUND: Expression of heparanase (HPSE) in tumor cells is strongly associated with invasion, metastasis and angiogenesis. It also plays a key role during pregnancy, in processes of implantation as well as placentation. Vascular endothelial growth factor (VEGF) and heparin are known to alter HPSE expression, with heparin given prophylactically to women with a history of placenta-mediated complications in subsequent pregnancies.
MATERIALS AND METHODS: We examined the growth-modulatory effects of different concentrations of heparin and VEGF on the choriocarcinoma cell-line JEG-3 and the expr

UNLABELLED: The intraislet heparan sulfate (HS) barrier is essential for β-cell survival, yet its role in type 1 diabetes (T1D) remains incompletely understood. We investigated the expression pattern, pathogenic function, and therapeutic potential of the heparanase (HPSE)-HS axis in T1D by integrating single-cell RNA sequencing data sets from human and mouse islets and peripheral immune cells, together with genetic mouse models and pharmacological intervention. HPSE was selectively enriched in myeloid cells, particularly CD14++ classical monocytes from patients with T1D and NOD mice and was as

Inflammation of the exocrine pancreas accompanies autoimmune diabetes in mouse models and humans. However, the relationship between inflammation in the exocrine and endocrine (islet) compartments has not been explored. To address this issue, we used a transgenic mouse model in which autoimmune diabetes is acutely induced after the transfer of islet beta cell-specific transgenic T cells. Histological analyses demonstrated that inflammation of the exocrine pancreas, which was initially mild, resulted in the transient but widespread disruption of acinar tissue. Islet inflammation preceded exacerb

BACKGROUND: Everolimus (EVE) is a drug widely used in several renal transplant protocols. Although characterized by a relatively low nephrotoxicity, it may induce several adverse effects including severe fibro-interstitial pneumonitis. The exact molecular/biological mechanism associated to these pro-fibrotic effects is unknown, but epithelial to mesenchymal transition (EMT) may have a central role. Additionally, heparanase, an enzyme recently associated with the progression of chronic allograft nephropathy, could contribute to activate this machinery in renal cells.
METHODS: Several biomolecu

Heparanase-1 (HPSE1) is an endo-β-d-glucuronidase that cleaves heparan sulfate proteoglycans into short-chain heparan sulfates (HS). The inhibition of HPSE1 has therapeutic potential for proteinuric diseases such as nephrotic syndrome because increased HPSE1 expression is associated with the loss of HS in the glomerular basement membrane, leading to the development of proteinuria. The present study examined the generation of a lead compound focusing on chemical structures with a sugar moiety, such as glycosides and sugar analogs, taking their physical properties into consideration. Heparanase is an endo-β-glucuronidase that is best known for its pro-cancerous effects but is also implicated in the pathogenesis of various viruses. Activation of heparanase is a common strategy to increase viral spread and trigger the subsequent inflammatory cascade. Using a Single Nucleotide Polymorphisms (SNP)-associated approach we identified enhancer and insulator regions that regulate HPSE expression. Although a role for heparanase in viral infection has been noticed, the impact of HPSE functional SNPs has not been determined. We investigated the effect of cytomegalovirus (CMV) serostat

PURPOSE: Sinusoidal obstruction syndrome (SOS) is a life-threatening early complication after hematopoietic stem cell transplantation (HSCT), and until now, examinations about the influence of genetic risk factors are extremely rare. The purpose of this study was to identify an association between heparanase (HPSE) single nucleotide polymorphisms (SNPs) and SOS in children undergoing allogeneic HSCT.
METHODS: We retrospectively analyzed the distribution of the both HPSE SNPs rs4693608 and rs4364254 and the occurrence of SOS after allogeneic HSCT in 160 children with malignant and non-malignan
