Induction of opposite response of hindlimb vasculature to adrenergic agents in hypothyroid rats.

In order to investigate whether hypothyroidism induces some functional change in the vascular smooth muscle, the responsiveness of the femoral vascular bed of hypothyroid rats to vasoactive agents was compared with that of euthyroid rats. The vascular beds were perfused by using the constant flow method. Ad libitum feeding of 0.15% 6-propylthiouracil to young male Wistar rats (120 g) over 3 months leads to hypothyroidism. Hypothyroidism did not induce any changes in the Ach- or papaverine-induced vasodilation and 5-HT-induced vasoconstriction. In contrast to euthyroidism, hypothyroidism induce

Effects of transient ischemia on nutrient flow and arteriovenous shunting in canine hindlimb.

A canine model was developed to simulate use of a pneumatic tourniquet in the clinical setting in order to study the acute and delayed effects of transient ischemia on limb and tissue blood flow, using radioactive microspheres and electromagnetic flow probes. Experimental femoral artery flow rose markedly after tourniquet ischemia, and remained significantly elevated for 24 hours (p less than 0.01). Blood flow to the rectus femoris and anterior tibial muscles rose significantly (p less than 0.05) immediately after tourniquet ischemia, and the latter remained significantly elevated at 24 hours

Evaluation of Prophylactic Negative Pressure Therapy in the Murine Hindlimb Lymphedema Model.

INTRODUCTION: Lymphedema is limb swelling secondary to lymphatic injury. Utilization of negative pressure therapy (NPT) has been shown to stimulate lymphatic vessel density, but the effect of NPT on secondary lymphedema is unknown. The purpose of this study is to evaluate the role of prophylactic negative pressure therapy in murine hindlimb lymphedema model. METHODS: The murine hindlimb model of lymphedema was used. Mice underwent 20-Gy irradiation of one hindlimb. One week post-radiation, an incision in the proximal hindlimb was created to disrupt lymphatic vessels and remove popliteal lymph

Kaempferol Protects Against Hindlimb Ischemia-Reperfusion Injury via Activation of PPARα Signaling.

Limb ischemia-reperfusion (I/R) injury is a life-threatening complication of acute limb ischemia that can result in severe skeletal muscle damage and limb loss, yet effective pharmacological therapies remain limited. Kaempferol (KAE), a naturally occurring dietary flavonoid with well-documented anti-inflammatory, antioxidant, and anti-apoptotic properties, has been shown to confer protection in myocardial and cerebral I/R models. Nevertheless, its therapeutic potential and molecular mechanisms in limb I/R injury have not yet been elucidated. In this study, we systematically investigated the pr

Hindlimb biomechanics of Lagosuchus talampayensis (Archosauria, Dinosauriformes), with comments on skeletal morphology.

Lagosuchus talampayensis is a small-bodied (~0.5 m long) Late Triassic dinosauriform archosaur from Argentina. Lagosuchus long has been a pivotal taxon for reconstructing the evolution of form and function on the dinosaur lineage. This importance is because it has a mix of ancestral archosaurian traits, such as a small pelvis with a mostly closed acetabulum lacking prominences that would restrict hip mobility much, with derived "dinosaurian" traits such as bipedalism, proximally shifted thigh muscle insertions, elongate hindlimbs, "advanced mesotarsal" ankle joints and digitigrade feet. Here,

HUVECs-derived exosomes increase neovascularization and decrease limb necrosis in hindlimb ischemia.

Chronic limb-threatening ischemia (CLTI) is the most severe manifestation of peripheral arterial disease (PAD) and imposes a significantly high burden due to its high risk of mortality and amputation. Revascularization is the first-line treatment for CLTI; however, the amputation rate remains high, and approximately one-third of patients are not eligible for this treatment. Therefore, there is an urgent need for more effective therapeutic strategies. The aim of this study was to investigate the effects and mechanisms of human umbilical vein endothelial cells (HUVECs)-derived exosomes on neovas

Mechanical Effect of Performance Pressure Boots on Cadaveric Equine Hindlimb Fetlock Biomechanics.

Pressure boots are applied to hind limbs of showjumping horses with the intent to enhance jumping form. Manufacturers claim acupressure points enhance proprioception of hind limbs. With this increased awareness, horses are expected to retract their hind limbs to clear jump rails. This research aimed to investigate a more direct, mechanical effect of pressure boots on hind limb biomechanics. Cadaveric hind limbs (n = 6) were mechanically loaded in axial compression (3 cycles at 0.25 Hz, displacement control ~3300 N) with (2 trials) and without (2 trials) a pressure boot applied. During mechanic

Dexamethasone Protects Against Tourniquet-Induced Acute Ischemia-Reperfusion Injury in Mouse Hindlimb.

Extremity injuries with hemorrhage have been a significant cause of death in civilian medicine and on the battlefield. The use of a tourniquet as an intervention is necessary for treatment to an injured limb; however, the tourniquet and subsequent release results in serious acute ischemia-reperfusion (IR) injury in the skeletal muscle and neuromuscular junction (NMJ). Much evidence demonstrates that inflammation is an important factor to cause acute IR injury. To find effective therapeutic interventions for tourniquet-induced acute IR injuries, our current study investigated effect of dexameth

Voluntary running protects against neuromuscular dysfunction following hindlimb ischemia-reperfusion in mice.

Ischemia-reperfusion (IR) due to temporary restriction of blood flow causes tissue/organ damages under various disease conditions, including stroke, myocardial infarction, trauma, and orthopedic surgery. In the limbs, IR injury to motor nerves and muscle fibers causes reduced mobility and quality of life. Endurance exercise training has been shown to increase tissue resistance to numerous pathological insults. To elucidate the impact of endurance exercise training on IR injury in skeletal muscle, sedentary and exercise-trained mice (5 wk of voluntary running) were subjected to ischemia by unil

Fetal serine fluxes across fetal liver, hindlimb, and placenta in late gestation.

Eleven studies of fetal serine fluxes were performed in chronically catheterized fetal lambs by continuous infusion of [1-13C]- and [U-14C]serine into a fetal brachial vein. At tracer serine steady state, samples were collected from the fetal abdominal aorta, umbilical vein, fetal hepatic vein, and fetal femoral vein and from the maternal femoral artery and uterine vein. Analyses were performed for plasma serine and glycine concentration, for serine and glycine 13C mole percent enrichment, and for whole blood 14CO2 and O2 concentrations. Uterine and umbilical blood flows were also measured. Th

Fatigue and contractile responses of rat hindlimb muscles following excessive dexamethasone administration.

Muscle weight, protein content and contractile performance (tetanic tension, fatigue and recovery) of extensor digitorum longus and soleus were investigated in rat following systemic administration of Dexamethasone (DX), 5 mg/kg/day for ten days. These animals showed marked reduction in food intake during the course of DX treatment. As a control, a group of food restricted (FR) rats receiving equal amount of food consumed by the DX treated rats was also studied along with the saline control group, to differentiate the effect of DX on muscle from that of dietary deficiency. There was a greater

Establishment of a long-term secondary lymphedema animal model in the rodent hindlimb.

INTRODUCTION: Secondary lymphedema is a chronic swelling of the extremities caused by physical disruption of the lymphatic system, leading to impaired lymph drainage. It frequently develops in cancer patients after surgical removal of lymph nodes and vessels during tumor resection, when reconnection of lymphatic pathways fails. Current clinical therapies are mainly palliative or conservative, with limited therapeutic effects. Therefore, an animal model that closely mimics the chronic and severe characteristics of secondary lymphedema in patients is required to enable mechanistic and therapeuti

Effects of spinal transection and locomotor speed on muscle synergies of the cat hindlimb.

It has been suggested that during locomotion, the nervous system controls movement by activating groups of muscles, or muscle synergies. Analysis of muscle synergies can reveal the organization of spinal locomotor networks and how it depends on the state of the nervous system, such as before and after spinal cord injury, and on different locomotor conditions, including a change in speed. The goal of this study was to investigate the effects of spinal transection and locomotor speed on hindlimb muscle synergies and their time-dependent activity patterns in adult cats. EMG activities of 15 hindl

Comparative muscle anatomy of the anuran pelvis and hindlimb in relation to locomotor mode.

Frogs have a highly conserved body plan, yet they employ a diverse array of locomotor modes, making them ideal organisms for investigating the relationships between morphology and locomotor function, in particular whether anatomical complexity is a prerequisite for functional complexity. We use diffusible iodine contrast-enhanced microCT (diceCT) imaging to digitally dissect the gross muscle anatomy of the pelvis and hindlimbs for 30 species of frogs representing five primary locomotor modes, including the first known detailed dissection for some of the world's smallest frogs, forming the larg

Effects of Hindlimb Suspension on the Development of Hip Bone Morphologies in Growing Rats.

Abnormal hip bone morphologies are associated with various diseases of the hip joint. Weight bearing, especially during growth, may be important to achieve normal acetabulum development. This study aimed to investigate whether hip bone morphologies were affected by hindlimb suspension (HS) in 4 week-old rats. In HS groups, tail suspension was applied for 0, 2, 4, and 8 weeks. Age-matched rats were used as controls. The complex of hip bones with lumbar and sacral vertebrae were assessed based on morphological indexes using three-dimensional reconstructed images from X-ray computed tomography. A

Proteomic Profiling of Hindlimb Skeletal Muscle Disuse in a Murine Model of Sepsis.

CONTEXT: Sepsis leads to multiple organ dysfunction and negatively impacts patient outcomes. Skeletal muscle disuse is a significant comorbidity in septic patients during their ICU stay due to prolonged immobilization. HYPOTHESIS: Combination of sepsis and muscle disuse will promote a unique proteomic signature in skeletal muscle in comparison to disuse and sepsis separately. METHODS AND MODELS: Following cecal ligation and puncture (CLP) or Sham surgeries, mice were subjected to hindlimb suspension (HLS) or maintained normal ambulation (NA).
