In the present study we report the effect of corticosterone in the regulation of hippocampal muscarinic acetylcholine receptor immunoreactivity (mAChR-ir) expression in rats. Adrenalectomy (ADX) or a single injection of a mineralocorticoid antagonist RU-28318 (1.0 mg/100 g body weight (b.w.)) in adrenally intact rats 24 h prior to sacrifice revealed an increased mAChR-ir in hippocampal CA1 and CA3 areas. Corticosterone replacement (100 microg/100 g b.w.) prevented the increase in mAChR-ir of ADX animals. However, glucocorticoid receptor antagonist (RU38486) treatment in adrenally intact rats f

The fundamental role of the brain-specific myelin transcription factor 1-like (MYT1L) gene in cases of intellectual disability and in the etiology of neurodevelopmental disorders is increasingly recognized. Yet, its function remains under-investigated. Here, we identify a network of helix-loop-helix (HLH) transcriptional regulators controlled by MYT1L, as indicated by our analyses in human neural stem cells and in the human brain. Using cell-based knockdown approaches and microarray analyses we found that (1) MYT1L is required for neuronal differentiation and identified ID1, a HLH inhibitor of

Alzheimer's disease is a rapidly progressive neurodegenerative disease, the development of which is associated with the accumulation of β-amyloid oligomers, dysfunction of the α7-nAChR nicotinic acetylcholine receptor, and activation of inflammation. Previously, we showed that the neuromodulator Lynx1, which belongs to the Ly6/uPAR family, competes with β-amyloid(1-42) for binding to α7-nAChR. In this work, we studied the expression and localization of Ly6/uPAR family proteins in the hippocampus of 2xTg-AD transgenic mice that model AD and demonstrate increased amyloidosis in the brain. BACKGROUND: Post-traumatic stress disorder can be viewed as a memory disorder, with trauma-related flashbacks being a core symptom. Given the central role of the hippocampus in autobiographical memory, surprisingly, there is mixed evidence concerning altered hippocampal functional connectivity in PTSD. We shed light on this discrepancy by considering the distinct roles of the anterior versus posterior hippocampus and examine how this distinction may map onto whole-brain resting-state functional connectivity patterns among those with and without PTSD.
METHODS: We first assessed whole-brain bet

Parkinson's disease (PD) is frequently associated with cognitive impairments, especially deficits in working memory and hippocampus-dependent memory. However, the contribution of dorsal hippocampus (dHipp) Ca2+/calmodulin-dependent protein kinase Ⅱ α (CaMKⅡα)-positive neurons to these deficits remains unclear. In this study, we investigated the impacts of chemogenetic modulation of dHipp CaMKⅡα-positive neurons on cognitive function, hippocampal theta rhythm, and monoamine levels in both sham-operated and unilateral 6-hydroxydopamine (6-OHDA)-lesioned rats. 6-OHDA lesions of the medial forebra

PURPOSE: To construct nomograms for both sides of the fetal fornix-hippocampus complex (FHC) length and hippocampus height between 18 + 0 and 23 + 6 weeks of gestation using two-dimensional sonography. Additionally, we evaluated the effects of laterality, fetal sex, and maternal iron deficiency anemia on these measurements.
METHODS: This was a prospective cross-sectional study of 223 singleton pregnancies between 18 + 0 and 23 + 6 weeks of gestation. The FHC length and hippocampus height were measured on both sides in the parasagittal cranial plane of the fetus by 2D ultrasonography. Measurem

Hippocampal pattern separation is a neural operation supporting mnemonic discrimination of highly similar memories by orthogonalizing neural inputs into nonoverlapping representations. In humans, pattern separation is thought to support the specificity of episodic memories. Recent neuroimaging studies suggest that the hippocampus can also access and manipulate semantic information. However, it is unclear whether the hippocampus can pattern separate highly similar semantic representations. Here, we tested multiple neural and behavioral signatures of pattern separation using high-resolution func

The encoding, storage, and updating of memories in cortical networks are poorly understood. In retrosplenial cortex (RSC), cells respond to the animal's position as it traverses a real or virtual (VR) linear track. Most position correlated cells (PCCs) in RSC require an intact hippocampus to form, but survive subsequent hippocampal damage. To examine whether RSC and hippocampal PCCs undergo remapping and spatial tuning development in parallel or sequentially, neuronal activity in RSC or CA1 was recorded using two-photon calcium imaging in mice running on VR tracks. RSC PCC activity underwent g

Wolfram syndrome is caused by mutations in the WFS1 gene. WFS1 protein dysfunction results in a range of neuroendocrine syndromes and is mostly characterized by juvenile-onset diabetes mellitus and optic atrophy. WFS1 has been shown to participate in membrane trafficking, protein processing and Ca2+ homeostasis in the endoplasmic reticulum. Aim of the present study was to find the transcriptomic changes influenced by WFS1 in the hypothalamus and hippocampus using RNA-sequencing. The WFS1-deficient mice were used as a model system to analyze the changes in transcriptional networks. The number o

Melatonin concentration in plasma reaches high levels during the night and synchronizes body rhythms with the photoperiod. Previous evidence obtained in cultured cells suggests that melatonin synchronizes cytoskeletal re-arrangements at nocturnal plasma concentration. In this study, we determined the amount of microtubules and microfilaments in the rat hippocampus as an index of cytoskeletal organization in rats submitted to a photoperiodic regime. Additionally, these parameters were determined in control rats, sham rats, pinealectomized rats, and rats that were pinealectomized and treated wit

Prenatal hypoxia ischemia is a major cause of neurodevelopmental impairment in the newborn, associated with risk for motor, behavioral and cognitive impaired outcomes. We used an established mouse model of maternal hypoxia to examine the immediate molecular responses of signaling pathways associated with both cell death and neurogenesis. We also characterized responses to maternal pre-treatment with MgSO(4). Maternal hypoxia at embryonic day 17 (E17) failed to trigger inflammation or cell death in fetal brain at 24 h after hypoxia. However, maternal hypoxia decreased levels of neuronal migrati

UNLABELLED: We investigated the contribution of bicarbonate ion, gamma-aminobutyric acid-A (GABA(A)) receptors, and N-methyl-D-aspartate (NMDA) receptors to pentobarbital-induced enhancement of excitatory synaptic transmission in the hippocampal slice. Transverse hippocampal slices (400 microm thick) were prepared from 20- to 30-day-old Sprague-Dawley rats and maintained in an interface chamber perfused with warmed (35 degrees C) oxygenated artificial cerebrospinal fluid. Extracellular field potentials, evoked by orthodromic paired-pulse stimulation of the Schaffer collateral CA1 pathway, were

Dorsal hippocampal (DH) lesions produce a severe deficit in recently, but not remotely, acquired contextual fear without impairing memory of discrete training stimuli, i.e., DH lesions produce an anterograde and time-limited retrograde amnesia specific to contextual memory. These data are consistent with the standard model which posits temporary involvement of the hippocampus in recent memory maintenance. However, three recent controversies apparently weaken the case for a selective mnemonic role for the hippocampus in contextual fear. First, although retrograde amnesia (from posttraining lesi

Regional cerebral blood flow (rCBF) data from two PET-15O water schizophrenia studies were analyzed using individually placed, magnetic resonance (MR)-guided hippocampal volumes of interest (VOI). In one study, normal (N = 10) and schizophrenic (N = 18) volunteers performed an overlearned auditory discrimination task in rest, control, and decision conditions. In the other study, schizophrenic and normal volunteers received the noncompetitive NMDA receptor antagonist ketamine and placebo and had sequential rCBF evaluations. Moreover, the schizophrenic volunteers were off drug in one study and o

The critical, fundamental mechanisms that determine the emergence of status epilepticus from a single seizure and the prolonged duration of status epilepticus are uncertain. However, several general concepts of the pathophysiology of status epilepticus have emerged: (a) the hippocampus is consistently activated during status epilepticus; (b) loss of GABA-mediated inhibitory synaptic transmission in the hippocampus is critical for emergence of status epilepticus; and, finally (c) glutamatergic excitatory synaptic transmission is important in sustaining status epilepticus. This review focuses on

The adrenal gland releases mineralocorticoids (MCs) and glucocorticoids (GCs) in response to a variety of stimuli, including stress. Once released, these adrenal steroids mediate a plethora of physiological responses in both the periphery and the central nervous system. The collective actions of GCs in the brain are paradoxical, however, in that basal levels of GCs are essential for neuronal development, plasticity and survival, while stress levels of GCs produce neuronal loss. Aging represents another contradictory function of GCs in the brain, since lifelong exposure to GCs has been implicat

Cognitive impairment is one of the common non-motor symptoms of Parkinson's disease (PD). Although the mechanism is still unclear, emerging research suggests that the dorsal hippocampus (dHIPP) and astrocyte (AS) may be involved in the cognitive dysfunction in PD. Thus, the effects of Gq or Gi mediated designer receptors exclusively activated by designer drugs (DREADD) manipulation of dHIPP AS on working and hippocampus-dependent memory were investigated in the Morris water maze and hole-board test in sham-operated and unilateral 6-hydroxydopamine (6-OHDA) lesioned rats. The results showed tha

OBJECTIVE: Temporal lobe epilepsy (TLE) is associated with disrupted functional integrity in the amygdala-hippocampus complex. Cortico-cortical evoked potentials (CCEPs) can characterize this disruption and have been proposed as biomarkers of the epileptogenic zone (EZ), but their study is typically limited by the spatial sampling bias inherent to whole-brain intracranial EEG. We investigated how epileptogenicity shapes effective connectivity in the amygdala-hippocampus complex, whether structural connectivity underlies it, and ultimately derived a multimodal EZ biomarker.
METHODS: We retrosp

Hippocampal theta oscillations coordinate computations underlying learning and memory. The frequency of theta varies with factors such as locomotion and anxiety, but the effect of an animal's sleep history on theta frequency remains unknown. Using long-duration CA1 recordings in rats, we found that awake theta frequency progressively decreases following sleep but remains elevated during sleep deprivation. These changes were not accounted for by movement but were predicted by the proportion of sleep in the preceding 30 min. In contrast, theta frequency remained relatively stable during rapid ey

Complement C3 plays important roles in neuroinflammation and is significantly upregulated in hippocampal tissues from patients with mesial temporal lobe epilepsy (mTLE) that is characterized by neuronal loss and gliosis. However, the temporal and spatial expression of complement C3 in the epileptic brain remain unclear. In this study, we first confirmed the upregulation of C3 in resected hippocampal tissues encompassing the CA1-CA4 regions from patients with mTLE with hippocampal sclerosis (mTLE-HS) using RNA sequencing, and in an mTLE mouse model using microarray data from the GEO database.
