Salivary innate immune peptides are associated with dental caries experience in young adults.

BACKGROUND: Saliva has traditionally been studied in relation to dental caries through parameters such as flow rate, pH, and buffering capacity. However, additional components, particularly proteins of the innate immune system, may also contribute to caries development or prevention. This study evaluated whether antimicrobial peptides (AMPs) in saliva are associated with caries status in young adults. METHODS: Forty healthy volunteers (18-35 years) attending the University Dental Clinics provided stimulated saliva samples and were assigned to either a low-caries group or a high caries experie

Antimicrobial peptides and proteins in Alzheimer's and Parkinson's diseases: implications for biomarker exploration.

Biomarkers are necessary tools to validate the diagnosis of Alzheimer's and Parkinson's diseases (AD and PD), especially when clinical symptoms are less apparent in the early stages of diseases. Current biomarkers used in clinical practice rely on brain imaging and cerebrospinal fluid (CSF) levels of amyloid-β (Aβ) peptides and α-synuclein (α-syn) for AD and PD, respectively. However, these diagnostic techniques are not only highly invasive and costly, but they also face limitations in diagnostic accuracy, particularly for preclinical or early stages of diseases. Thus, alternative biomarkers h

Proteomic signature of the acquired salivary pellicle on zirconia and its role in modulating oral biofilms formation.

Although zirconia is widely used, the proteomic composition of its salivary pellicle and its impact on biofilm formation remain poorly understood. Therefore, this study aimed to analyze the protein composition of the salivary pellicle and evaluate its impact on microbial adhesion and biofilm formation. Surface roughness and wettability of zirconia and hydroxyapatite (HA) discs were assessed. Discs were incubated with pooled parotid saliva collected from 10 healthy adults using a Carlson-Crittenden device. Adsorbed proteins were eluted, quantified, digested, and identified via Liquid Chromatogr

Identification of Lys-Pro-Gln as a novel cleavage site specificity of saliva-associated proteases.

The nonsterile environment of the oral cavity facilitates substantial proteolytic processing, not only of resident salivary proteins but also of dietary proteins. To gain insight into whole saliva enzymatic processes, the in vivo generated peptides in this oral fluid were subjected to nano-flow liquid chromatography electrospray ionization tandem mass spectrometry. The 182 peptides identified were predominantly derived from acidic and basic proline-rich proteins, statherin, and histatins. The proteolytic cleavages in the basic proline-rich proteins occurred preferentially after a Gln residue w

Temporary modification of salivary protein profile and individual responses to repeated phenolic astringent stimuli.

The extent of the change in salivary protein characteristics after repeated stimulations was shown to be correlated to differences in perceived astringency. Salivary characteristics of 77 subjects were compared after masticatory (S1) and taste/masticatory (S2) stimulations. The variations (S2 minus S1) of protein concentration and saliva haze-forming capacity (HFC) were used to define 3 subject groups: low responding (LR, n = 20), medium responding (MR, n = 37), and high responding (HR, n = 20). Salivary protein concentration did not change in LR subjects; decreased a little, but significantly

Aging envisage imbalance of the periodontium: A keystone in oral disease and systemic health.

Aging is a gradual and progressive deterioration of integrity across multiple organ systems that negatively affects gingival wound healing. The cellular responses associated with wound healing, such as collagen synthesis, cell migration, proliferation, and collagen contraction, have been shown to be lower in gingival fibroblasts (the most abundant cells from the connective gingival tissue) in aged donors than young donors. Cellular senescence is one of the hallmarks of aging, which is characterized by the acquisition of a senescence-associated secretory phenotype that is characterized by the r

Functional roles of purified yapsins from Candida glabrata (Nakaseomyces glabratus) in immune modulation and cross-species biofilm formation.

Candida glabrata (currently classified as Nakaseomyces glabratus) is an opportunistic yeast-like fungus that causes infections in humans, with limited treatment options due to resistance to antifungal drugs. In contrast to C. albicans, which produces secreted aspartic proteases (Saps) involved in pathogenicity, C. glabrata expresses a distinct group of cell surface-associated aspartic proteases known as yapsins (Yps). While YPS gene deletion mutants have proposed roles in cellular homeostasis, their precise contribution to fungal virulence and host interactions remains unclear. Herein, we pres

Interaction of a Histidine-Rich Antimicrobial Saliva Peptide with Model Cell Membranes: The Role of Histidines.

Histatin 5 is a histidine-rich, intrinsically disordered, multifunctional saliva protein known to act as a first line of defense against oral candidiasis caused by Candida albicans. An earlier study showed that, upon interaction with a common model bilayer, a protein cushion spontaneously forms underneath the bilayer. Our hypothesis is that this effect is of electrostatic origin and that the observed behavior is due to proton charge fluctuations of the histidines, promoting attractive electrostatic interactions between the positively charged proteins and the anionic surfaces, with concomitant

Utilisation of peptides against microbial infections - a review.

The emergence of resistance in microorganisms on a global scale has made it necessary to search for new antimicrobial factors. Antimicrobial peptides (AMPs) seem to meet these expectations. AMPs are produced by bacteria, viruses, plants, and animals, and may be considered as a new class of drugs intended for the prophylaxis and treatment of both systemic and topical infections. The aim of this study is to review the results of studies on the use of peptides to combat infections in vivo. Antimicrobial peptides may be applied topically and systemically. Among the peptides used topically, a very

Rational Design Yields Molecular Insights on Leaf-Binding of Anchor Peptides.

In times of a constantly growing world population and increasing demand for food, sustainable agriculture is crucial. The rainfastness of plant protection agents is of pivotal importance to reduce the amount of applied nutrients, herbicides, and fungicides. As a result of protective agent wash-off, plant protection is lost, and soils and groundwater are severely polluted. To date, rainfastness of plant protection products has been achieved by adding polymeric adjuvants to the agrochemicals. However, polymeric adjuvants will be regarded as microplastics in the future, and environmentally friend

Characterization, expression profiling, and functional analysis of a Populus trichocarpa defensin gene and its potential as an anti-Agrobacterium rooting medium additive.

The diverse antimicrobial properties of defensins have attracted wide scientific interest in recent years. Also, antimicrobial peptides (AMPs), including cecropins, histatins, defensins, and cathelicidins, have recently become an antimicrobial research hotspot for their broad-spectrum antibacterial and antifungal activities. In addition, defensins play important roles in plant growth, development, and physiological metabolism, and demonstrate tissue specificity and regulation in response to pathogen attack or abiotic stress. In this study, we performed molecular cloning, characterization, expr

Significant modifications of the salivary proteome potentially associated with complications of Down syndrome revealed by top-down proteomics.

People with Down syndrome, a frequent genetic disorder in humans, have increased risk of health problems associated with this condition. One clinical feature of Down syndrome is the increased prevalence and severity of periodontal disease in comparison with the general population. Because saliva plays an important role in maintaining oral health, in the present study the salivary proteome of Down syndrome subjects was investigated to explore modifications with respect to healthy subjects. Whole saliva of 36 Down syndrome subjects, divided in the age groups 10-17 yr and 18-50 yr, was analyzed b

LL-37: Structures, Antimicrobial Activity, and Influence on Amyloid-Related Diseases.

Antimicrobial peptides (AMPs), as well as host defense peptides (HDPs), constitute the first line of defense as part of the innate immune system. Humans are known to express antimicrobial precursor proteins, which are further processed to generate AMPs, including several types of α/β defensins, histatins, and cathelicidin-derived AMPs like LL37. The broad-spectrum activity of AMPs is crucial to defend against infections caused by pathogenic bacteria, viruses, fungi, and parasites. The emergence of multi-drug resistant pathogenic bacteria is of global concern for public health. The prospects of

A novel antimicrobial peptide AH-12 attenuates mitochondrial response to inflammatory stimuli and prevents periodontitis via antibacterial and anti-inflammatory effects.

Antibiotic-assisted periodontal therapy remains a primary clinical strategy, yet the rising prevalence of bacterial resistance severely compromises its therapeutic efficacy. To address this challenge, antimicrobial peptides (AMPs) have emerged as promising alternatives owing to their distinct antibacterial mechanisms. Here, we developed a novel antimicrobial peptide, AH-12, by systematically optimizing P-113-the minimal inhibitory fragment of human salivary Histatin 5-through N-terminal modification, amidation, and acetylation. In vitro evaluations demonstrated potent inhibitory effects of AH-
