AhlD, an N-acylhomoserine lactonase in Arthrobacter sp., and predicted homologues in other bacteria.

Quorum sensing is a signalling mechanism that controls diverse biological functions, including virulence, via N-acylhomoserine lactone (AHL) signal molecules in Gram-negative bacteria. With the aim of isolating strains or enzymes capable of blocking quorum sensing by inactivating AHL, bacteria were screened for AHL degradation by their ability to utilize N-3-oxohexanoyl-L-homoserine lactone (OHHL) as the sole carbon source. Among four isolates, strain IBN110, identified as Arthrobacter sp., was found to grow rapidly on OHHL, and to degrade various AHLs with different lengths and acyl side-chai

Synthesis of Mixed Phosphonate Esters and Amino Acid-Based Phosphonamidates, and Their Screening as Herbicides.

While organophosphorus chemistry is gaining attention in a variety of fields, the synthesis of the phosphorus derivatives of amino acids remains a challenging task. Previously reported methods require the deprotonation of the nucleophile, complex reagents or hydrolysis of the phosphonate ester. In this paper, we demonstrate how to avoid these issues by employing phosphonylaminium salts for the synthesis of novel mixed n-alkylphosphonate diesters or amino acid-derived n-alkylphosphonamidates. We successfully applied this methodology for the synthesis of novel N-acyl homoserine lactone analogues

Quorum-sensing, microbiome interactions, and emerging artificial intelligence-assisted anti-virulence strategies in Salmonella Typhi: a critical review of translational opportunities and challenges.

Typhoid fever, caused by Salmonella enterica subsp. enterica serovar Typhi (Salmonella Typhi), remains a significant global health challenge that is increasingly complicated by the emergence and spread of multidrug-resistant (MDR) and extensively drug-resistant strains. Growing limitations of antibiotic-centered treatment strategies have stimulated interest in anti-virulence approaches targeting bacterial regulatory networks rather than viability alone. Among these, quorum-sensing (QS), particularly the LuxS-mediated autoinducer-2 (AI-2) signaling system, has emerged as a potential regulator o

Mechanism insights into the regulation of the LuxS/AI-2 quorum sensing system on the formation of viable but nonculturable state in biofilm cells of beer-spoilage Lactiplantibacillus plantarum.

INTRODUCTION: Beer spoilage Lactiplantibacillus plantarum poses a significant challenge in brewing due to biofilm formation and entry into the viable but nonculturable (VBNC) state, evading detection and resisting sterilization. The LuxS/AI-2 quorum sensing (QS) system is implicated in stress adaptation, yet its regulatory role in biofilm-associated VBNC transition remains poorly understood. OBJECTIVES: This study aims to elucidate how the luxS gene regulates VBNC-state formation in biofilm-embedded L. plantarum under hop-derived iso-α-acid stress and to identify molecular mechanisms underlyi

The AI-2/LuxS quorum sensing system enhances stress tolerance and enological performance of Lactiplantibacillus plantarum during malolactic fermentation.

Although Lactiplantibacillus plantarum shows potential for malolactic fermentation (MLF), its stress sensitivity compared to Oenococcus oeni limits its enological applications. This study reveals the critical role of the AI-2/LuxS quorum sensing (QS) system in the wine stress adaptation of L. plantarum XJ25 through targeted luxS gene manipulation. Deletion of luxS did not impact growth or acid production of L. plantarum XJ25 in optimal conditions (no stress) but significantly decreased AI-2 signal molecule activity. However, under simulated wine stress conditions, including low pH and high eth

Magnetic nanofactories: localized synthesis and delivery of quorum-sensing signaling molecule autoinducer-2 to bacterial cell surfaces.

Magnetic 'nanofactories', for localized manufacture and signal-guided delivery of small molecules to targeted cell surfaces, are demonstrated. They recruit nearby raw materials for synthesis, employ magnetic mobility for capture and localization of target cells, and deliver molecules to cells triggering their native phenotypic response, but with user-specified control. Our nanofactories, which synthesize and deliver the "universal" bacterial quorum-sensing signal molecule, autoinducer AI-2, to the surface of Escherichia coli, are assembled by first co-precipitating nanoparticles of iron salts

Pathogenesis and Sustainable Management of Pseudomonas syringae: Emerging Roles of Microbial Consortia and Responsive Nanomaterials.

Pseudomonas syringae remains a globally devastating phytopathogen threatening over 500 species. This review delineates the biochemical coevolution between P. syringae and hosts, focusing on Type III secretion system (T3SS) effectors and phytotoxins (e.g., coronatine) that enzymatically reprogram plant hormone signaling. To circumvent the limitations of traditional bactericides, we propose integrated green strategies. In biocontrol, we advocate for rationally engineered Synthetic Microbial Consortia (SynComs) and the biochemical quenching of N-acyl homoserine lactones (AHLs) to disrupt quorum-s

Linking biosynthetic genes to natural products using inverse stable isotopic labeling (InverSIL).

The sequencing of microbial genomes has far outpaced their functional annotation. Stable isotopic labeling can be used to link biosynthetic genes with their natural products; however, the availability of the required isotopically substituted precursors can limit the accessibility of this approach. Here, we describe a method for using inverse stable isotopic labeling (InverSIL) to link biosynthetic genes with their natural products. With InverSIL, a microbe is grown on an isotopically substituted medium to create a fully substituted culture, and subsequently, the incorporation of precursors of

CytoSorb in burn patients with septic shock and acute kidney injury on continuous kidney replacement therapy efficiently reduces the levels of Gram-negative-derived quorum-sensing molecules associated with lactate increases.

INTRODUCTION: Quorum-sensing (QS) molecules produced by Gram-negative bacteria regulate virulence and host-pathogen interactions, but their systemic role in human septic shock remains largely unexplored. We hypothesized that QS molecules are detectable in the circulation of burn patients with septic-shock-associated acute kidney injury (SA-AKI), are associated with disease severity, and are amenable to removal by extracorporeal treatment. METHODS: In this observational pilot study, plasma concentrations of eight Gram-negative-derived QS molecules were quantified by HPLC-MS/MS in 12 burn patie

Small molecule communication of Legionella: the ins and outs of autoinducer and nitric oxide signaling.

SUMMARYLegionella pneumophila is a Gram-negative environmental bacterium, which survives in planktonic form, colonizes biofilms, and infects protozoa. Upon inhalation of Legionella-contaminated aerosols, the opportunistic pathogen replicates within and destroys alveolar macrophages, thereby causing a severe pneumonia termed Legionnaires' disease. Gram-negative bacteria employ low molecular weight organic compounds as well as the inorganic gas nitric oxide (NO) for cell-cell communication. L. pneumophila produces, secretes, and detects the α-hydroxyketone compound Legionella autoinducer-1 (LAI-

Vibrio cholerae strains with inactivated cqsS gene overproduce autoinducer-2 which enhances resuscitation of dormant environmental V. cholerae.

BACKGROUND: Toxigenic Vibrio cholerae resides in aquatic reservoirs of cholera-endemic areas mostly in a dormant form known as conditionally viable environmental cells (CVEC) in which the bacteria remain embedded in an exopolysaccharide matrix, and fail to grow in routine bacteriological culture. The CVEC can be resuscitated by supplementing culture media with either of two autoinducers CAI-1 and AI-2, which are signal molecules controlling quorum sensing, a regulatory network of bacterial gene expression dependent on cell density. This study investigated possible existence of variant strains

Interaction between Cervical Microbiota and Host Gene Regulation in Caesarean Section Scar Diverticulum.

Cesarean section scar diverticulum (CSD) has become a formidable obstacle preventing women receiving CS from reproducing. However, the pathogenesis of CSD remains unexplored. In this study, we characterized the cervical microbiota, metabolome, and endometrial transcriptome of women with CSD. Based on the 16s rRNA results of cervical microbes, the microbial diversity in the CSD group was higher than that in the control group. Lactobacillus were significantly decreased in the CSD group and were mutually exclusive with potentially harmful species (Sphingomonas, Sediminbacterium, and Ralstonia) ab

Anti-quorum-sensing activity of Plectranthus aliciae in multispecies Cutibacterium acnes-Staphylococcus epidermidis systems.

BACKGROUND: Quorum sensing (QS) is a bacterial communication process mediated by the production, release, and detection of autoinducers, regulating gene expression linked to virulence and biofilm formation. Cutibacterium acnes and Staphylococcus epidermidis exhibit opportunistic pathogenic behavior on the skin, complicating treatment strategies for inflammatory skin conditions such as acne vulgaris and wound-associated infections. The interspecies relationship between C. acnes and S. epidermidis, particularly within multispecies biofilms and under different oxygen conditions, remains insuffici

RhlR quorum-sensing receptor ligand sensitivity regulates the differential expression of phenazine genes in Pseudomonas aeruginosa.

Quorum sensing enables bacteria to coordinate gene expression in response to population density through the detection of small-molecule signals. In gram-negative bacteria, LuxR-type transcription factors bind acyl-homoserine lactones to regulate collective behaviors, yet how ligand sensitivity is tuned to shape transcriptional outcomes remains poorly defined. In Pseudomonas aeruginosa, the quorum-sensing receptor RhlR responds to N-butyryl-L-homoserine lactone (C4HSL) and controls late-stage quorum-sensing behaviors, including phenazine biosynthesis. Here, we use a chemical-genetic and structu

Acinetobacter quorum sensing contributes to inflammation-induced inhibition of orthopaedic implant osseointegration.

Implant infection impairs osseointegration of orthopaedic implants by inducing inflammation. Acinetobacter spp. are increasingly prevalent multi-drug resistant bacteria that can cause osteomyelitis. Acinetobacter spp. can also cause inflammation and thereby inhibit osseointegration in mice. The purpose of the present study was to investigate the role of quorum sensing in this context. Therefore, wild-type bacteria were compared with an isogenic abaI mutant defective in quorum sensing in a murine osseointegration model. The abaI quorum- sensing mutant affected significantly less osseointegratio

Selective oxidation of methionine residues in proteins.

Methionine residues in peptides and proteins were oxidized to methionine sulfoxides by mild oxidizing reagents such as chloramine-T and N-chlorosuccinimide at neutral and slightly alkaline pH. With chloramine-T cysteine was also oxidized to cystine but no other amino acid was modified; with N-chlorosuccinimide tryptophans were oxidized as well. In peptides and denaturated proteins all methionine residues were quantitatively oxidized, while in native proteins only exposed methionine residues could be modified. Extent of oxidation of methionine residues was determined by quantitative modificatio

Interplay between chemotaxis, quorum sensing, and metabolism regulates Escherichia coli-Salmonella Typhimurium interactions in vivo.

Motile bacteria use chemotaxis to navigate complex environments like the mammalian gut. These bacteria sense a range of chemoeffector molecules, which can either be of nutritional value or provide a cue for the niche best suited for their survival and growth. One such cue molecule is the intra- and interspecies quorum sensing signaling molecule, autoinducer-2 (AI-2). Apart from controlling collective behavior of Escherichia coli, chemotaxis towards AI-2 contributes to its ability to colonize the murine gut. However, the impact of AI-2-dependent niche occupation by E. coli on interspecies inter

Deciphering bacterial universal language by detecting the quorum sensing signal, autoinducer-2, with a whole-cell sensing system.

Bacteria communicate with neighboring bacteria of the same species or of other species by means of chemical signaling molecules. The concentration of such signaling molecules is proportional to the bacterial population size; upon reaching a threshold concentration, corresponding to a threshold cell density, certain specialized genes are expressed. This system of communication among bacteria is known as quorum sensing (QS). QS regulates diverse behaviors, such as formation of biofilms and production of pathogenic factors. Autoinducer-2 (AI-2) is a QS signaling molecule that is used for interspe

LuxS-mediated quorum sensing in Borrelia burgdorferi, the lyme disease spirochete.

The establishment of Borrelia burgdorferi infection involves numerous interactions between the bacteria and a variety of vertebrate host and arthropod vector tissues. This complex process requires regulated synthesis of many bacterial proteins. We now demonstrate that these spirochetes utilize a LuxS/autoinducer-2 (AI-2)-based quorum-sensing mechanism to regulate protein expression, the first system of cell-cell communication to be described in a spirochete. The luxS gene of B. burgdorferi was identified and demonstrated to encode a functional enzyme by complementation of an Escherichia coli l

The regulatory network of Salmonella biofilm formation: the role of CsgD on autoinducer-2 activity and expression of quorum-sensing-related genes in Salmonella Typhimurium.

Salmonella biofilms are mainly formed by cellulose and curli fimbriae under the control of CsgD. Biofilm regulation also involves small RNAs and LuxS-mediated AI-2 quorum sensing. This study aimed to investigate the relationship between CsgD and quorum-sensing regulation in Salmonella Typhimurium. By modulating csgD expression, the impact on the expression levels of MicA, CsrB, CsrC, and RyeE sRNAs-sRNAs implicated in QS-linked regulatory pathways-was assessed. Quantitative real-time PCR (qRT-PCR) analyses demonstrated that the expression of these sRNAs was increased following perturbation of
