The horizontal transfer of the bacterium Wolbachia pipientis between invertebrate hosts hinges on the ability of Wolbachia to adapt to new intracellular environments. The experimental transfer of Wolbachia between distantly related host species often results in the loss of infection, presumably due to an inability of Wolbachia to adapt quickly to the new host. To examine the process of adaptation to a novel host, we transferred a life-shortening Wolbachia strain, wMelPop, from the fruit fly Drosophila melanogaster into a cell line derived from the mosquito Aedes albopictus. After long-term ser

Legionella pneumophila is the causative agent of the pneumonia-like Legionnaires' disease. The bacterium's survival and spread depend on the ability to replicate inside eukaryotic phagocytic cells. A particular feature of Legionella is its dual host system allowing the intracellular growth in protozoa like Acanthamoeba castellanii, and during infection in human alveolar macrophages. Genome analysis and comparisons as well as expression profiling of the pathogen and the host helped to identify regulatory circuits mediating adaptation of the L. pneumophila transcriptome to the intracellular envi

BACKGROUND/OBJECTIVES: Whether inflammatory and nutritional parameters shift meaningfully in the first months of systemic treatment - and whether these shifts predict outcomes - has not been adequately addressed. We set out to track these changes from treatment initiation to month 3 in adults with newly diagnosed solid tumors, with progression-free and overall survival as primary endpoints.
METHODS: This was a single-center prospective study running from August 2023 through August 2024. Patients aged ≥ 18 with histologically proven solid tumors who were about to start systemic anticancer ther

BACKGROUND: Although Rousettus aegyptiacus is the primary natural reservoir of Marburg virus (MARV), recent atypical outbreaks and viral genomic diversification suggest a highly complex host-pathogen ecology. Here, we systematically evaluated the molecular homologies between MARV and diverse mammalian taxa to identify putative reservoir hosts and guide targeted surveillance.
METHODS: A multi-dimensional molecular characterization of the Marburg virus (MARV) genome was performed using an integrative suite of computational approaches. Distant sequence homology was interrogated via BLAST in conj

The H7N9 avian influenza virus (AIV) has posed a major global public health concern since its first detection in China in 2013. Transmitted among wild birds and poultry, this virus has crossed the species barrier to infect humans, causing severe respiratory disease and high mortality. Although the widespread use of H7 vaccines has markedly reduced human infections, the ongoing circulation and adaptive evolution of the virus in poultry remain a serious threat. In this study, we analyzed three highly pathogenic H7N9 isolates collected in China in 2022, representing two hemagglutinin (HA) gene ev

UNLABELLED: The aim of this study was to characterize the in vivo evolution of Staphylococcus aureus strains involved in recurrent prosthetic joint infections (PJIs) both phenotypically and genomically. We conducted a monocentric retrospective study in a 1,437-bed French teaching hospital between 2013 and 2021. All patients presenting a recurrent S. aureus-related PJI-defined as at least two strains isolated from distinct clinical samples more than 90 days apart-of the knee, hip, or shoulder were included. Clinical data were reviewed, and all isolates underwent phenotypic characterization, inc

Riemerella anatipestifer (RA) is an economically important avian pathogen that has historically caused significant disease in waterfowls but is increasingly recognized in various poultry production systems and wild bird species, highlighting its expanding host range and adaptive potential. Despite the availability of vaccines and antimicrobial interventions, RA remains a persistent challenge due to serotype diversity, limited cross-protective immunity, evolving antimicrobial resistance patterns, and an incomplete understanding of the host-pathogen interactions that govern disease outcome. Avian influenza A viruses (IAVs) pose a public health threat, as they are capable of triggering pandemics by crossing species barriers. Replication of avian IAVs in mammalian cells is hindered by species-specific variation in acidic nuclear phosphoprotein 32 (ANP32) proteins, which are essential for viral RNA genome replication. Adaptive mutations enable the IAV RNA polymerase (FluPolA) to surmount this barrier. Here, we present cryo-electron microscopy structures of monomeric and dimeric avian H5N1 FluPolA with human ANP32B. ANP32B interacts with the PA subunit of FluPolA in the monomeric for

Influenza A viruses continue to be a serious health risk to people and result in a large-scale socio-economic loss. Avian influenza viruses typically do not replicate efficiently in mammals, but through the accumulation of mutations or genetic reassortment, they can overcome interspecies barriers, adapt to new hosts, and spread among them. Zoonotic influenza A viruses sporadically infect humans and exhibit limited human-to-human transmission. However, further adaptation of these viruses to humans may result in airborne transmissible viruses with pandemic potential. Therefore, we are beginning

BACKGROUND: Divergent selection on host-plants is one of the main evolutionary forces driving ecological speciation in phytophagous insects. The ecological speciation might be challenging in the presence of gene flow and assortative mating because the direction of divergence is not necessarily the same between ecological selection (through host-plant adaptation) and assortative mating. The fall armyworm (FAW), a major lepidopteran pest species, is composed of two sympatric strains, corn and rice strains, named after two of their preferred host-plants. These two strains have been hypothesized t

A type of monophasic group B Salmonella enterica with the antigenic formula 4,12:a:- ("Fulica-like") has been described as associated with harbour porpoises (Phocoena phocoena), most frequently recovered from lung samples. In the present study, lung tissue samples from 47 porpoises found along the Swedish coast or as bycatch in fishing nets were analysed, two of which were positive for S. enterica. Pneumonia due to the infection was considered the likely cause of death for one of the two animals. The recovered isolates were whole genome sequenced and found to belong to sequence type (ST) 416 a

BACKGROUND: Chlamydia are ancient intracellular pathogens with reduced, though strikingly conserved genome. Despite their parasitic lifestyle and isolated intracellular environment, these bacteria managed to avoid accumulation of deleterious mutations leading to subsequent genome degradation characteristic for many parasitic bacteria.
RESULTS: We report pan-genomic analysis of sixteen species from genus Chlamydia including identification and functional annotation of orthologous genes, and characterization of gene gains, losses, and rearrangements. We demonstrate the overall genome stability o

Host manipulation whereby a parasite increases its transmission to a subsequent host by altering the behaviour of its current host is very far spread. It also occurs in host-parasite systems that are widely distributed. This offers the potential for local adaptation. The tapeworm Schistocephalus solidus modifies its first intermediate copepod host's predation susceptibility to suit its own needs by reducing its activity before it becomes infective and increasing it thereafter. To investigate potential differences in host manipulation between different populations and test for potential local a

UNLABELLED: Inositol pyrophosphates (PP-IPs) comprising inositol, phosphate, and pyrophosphate (PP) are essential for multiple functions in eukaryotes. Their role in fungal pathogens has never been addressed. Cryptococcus neoformans is a model pathogenic fungus causing life-threatening meningoencephalitis. We investigate the cryptococcal kinases responsible for the production of PP-IPs (IP7/IP8) and the hierarchy of PP-IP importance in pathogenicity. Using gene deletion and inositol polyphosphate profiling, we identified Kcs1 as the major IP6 kinase (producing IP7) and Asp1 as an IP7 kinase (p

The macaque malaria parasite Plasmodium knowlesi has recently emerged as an important zoonosis in Southeast Asia. Infections are typically mild but can cause severe disease, achieving parasite densities similar to fatal Plasmodium falciparum infections. Here we show that a primate-adapted P. knowlesi parasite proliferates poorly in human blood due to a strong preference for young red blood cells (RBCs). We establish a continuous in vitro culture system by using human blood enriched for young cells. Mathematical modelling predicts that parasite adaptation for invasion of older RBCs is a likely

Neisseria meningitidis (the meningococcus) colonizes the human nasopharynx of about 10% of the human population. However, for reasons that are still mostly unknown meningococci occasionally enter the cerebrospinal fluid leading to often fatal bacterial meningitis especially in children and young adults. The genetic basis for the observed differences in the pathogenic potential of different strains has only partially been unravelled so far. With the advent of whole genome sequencing technologies, complete genome sequences from three pathogenic meningococcal strains have become available and all

BACKGROUND: Pseudomonas aeruginosa is an opportunistic pathogen responsible for causing severe hospital-acquired infections, largely due to its adaptability, diverse range of virulence factors and drug resistance mechanisms. Despite extensive research, a substantial portion of the genome of P. aeruginosa remains functionally uncharacterized. The aim of this study was to identify candidate host-adaptation genes, including potential virulence factors, among these numerous uncharacterized genes.
RESULTS: We performed a comparative genomic analysis of P. aeruginosa strains isolated from human cli

Influenza viruses remain major global health threats due to their rapid evolution and complex interactions with host regulatory systems. While genetic variation is a key driver of viral pathogenicity and host adaptation, growing evidence indicates that post-translational modifications (PTMs) provide an additional regulatory layer that can influence viral replication, immune evasion, and metabolic remodeling. In this review, we synthesize current knowledge on how coordinated PTM networks, including phosphorylation, ubiquitination, SUMOylation, glycosylation, acetylation, lipidation, and RNA met

Tuberculosis is a chronic infectious disease caused by Mycobacterium tuberculosis (Mtb). As primary host cells targeted by Mtb, macrophages play central roles in both innate and adaptive immunity. Accumulating evidence indicates that macrophage polarization is a key determinant of tuberculosis pathogenesis. In response to diverse microenvironmental cues, macrophages adopt functionally distinct polarization states, ranging from pro-inflammatory, microbicidal programs (M1-like) to anti-inflammatory, tissue-reparative programs (M2-like). These states differentially shape tuberculosis progression.

SARS-CoV-2 rapidly adapts to new hosts following cross-species transmission; this is highly relevant as unique within-host variants have emerged following infection of susceptible wild and domestic animal species. Furthermore, SARS-CoV-2 transmission from animals (e.g., white-tailed deer, mink, domestic cats, and others) back to humans has been observed, documenting the potential of animal-derived variants to infect humans. Here, we investigate SARS-CoV-2 evolution and host-specific adaptation during an outbreak in Amur tigers (Panthera tigris altaica), African lions (Panthera leo), and spotte
