First-void urine: A potential biomarker source for triage of high-risk human papillomavirus infected women.

Great interest has been directed towards the use of first-void urine as a liquid biopsy for high-risk human papillomavirus DNA testing. Despite the high correlations established between urinary and cervical infections, human papillomavirus testing is unable to distinguish between productive and transforming high-risk infections that have the tendency to progress to cervical cancer. Thus far, investigations have been primarily confined to the identification of biomarkers for triage of high-risk human papillomavirus-positive women in cervicovaginal specimens and tissue biopsies. This paper revie

Unique human papillomavirus-type distribution in South African women with invasive cervical cancer and the effect of human immunodeficiency virus infection.

OBJECTIVES: Cervical cancer is the most common cause of cancer-related deaths among South African women. Viral types associated with cervical cancer may differ not only between countries and regions, but possibly also between human immunodeficiency virus (HIV)-infected and noninfected women. METHODS: In a population with high HIV prevalence, human papillomavirus (HPV)-type infections detected with DNA analyses were reported in a cohort of 299 women diagnosed with invasive cervical cancer. RESULTS: One hundred fifty-four women tested HIV negative, 77 tested HIV positive, and HIV status was un

Quick assessment of DNA damage in cervical epithelial cells using a chromatin dispersion test.

PURPOSE: This study was aimed to quantify genomic DNA breakages in the cervical epithelium cells of patients diagnosed with different grades of cervical lesions using a quick test based on chromatin dispersion after controlled protein depletion. The association between the progressive stages of cervical dysplasia and the levels of DNA damage, taking into account the presence of papillomavirus human (HPV) infection, was investigated. METHODS: A hospital-based unmatched case-control study was conducted during 2018 with a sample of 78 women grouped according to histological diagnosis as follows:

Use of the NucliSENS EasyQ HPV assay in the management of cervical intraepithelial neoplasia.

Persistent infection by high-risk human papillomavirus is a necessary cause for cervical cancer. DNA-based human papillomavirus (HPV) assays show high sensitivity but poor specificity in detecting high-grade cervical lesions. Assays detecting mRNA of the oncoproteins E6 and E7 show higher specificity but lack either detection of all high-risk genotypes or the ability to specify the detected genotypes. The aim of this study was to evaluate the clinical performance of the NucliSENS EasyQ HPV assay in comparison with the Hybrid Capture 2 test (HC2) and the CLART Human Papillomavirus 2 assay (CLAR

HPV-DNA testing from self-sampled menstrual blood using M-strip: A proof-of-concept study on feasibility & acceptance of a novel biosampling method.

Background & objectives Screening for cervical cancer by self-sampling appears more acceptable to women and has the potential to boost screening uptake, which is dismal at present in India. Studies have shown that menstrual blood (MB) provides equivalent results to cervical smear for Human Papillomavirus (HPV)-Deoxyribonucleic acid (DNA) testing, but sample collection needs standardization. This study explored the feasibility and acceptance of self-sampling using 'M-strip' for high-risk HPV DNA (hr-HPV DNA) testing from MB. Methods One hundred and eleven women aged 30-50 yr without a previous

Association of Oral Papivir/Pavirona® Supplementation with HPV DNA Clearance.

BACKGROUND: Persistent cervical human papillomavirus (Human papillomavirus) infection remains a significant public health concern, as it is the primary etiological factor in the development of cervical cancer and its precursor lesions. While prophylactic vaccination and standard screening programs are cornerstones of prevention, a substantial proportion of women with established infection are managed conservatively, often with prolonged follow-up and associated psychological burden. Interest has therefore grown in supportive interventions that may facilitate viral clearance during routine clin

Pseudobowenoid change of the vulva: a histologic variant of untreated condylata acuminatum.

Vulvar condylomata are distinguished from vulvar intraepithelial neoplasia by orderly maturation, a low mitotic index, and the absence of nuclear atypia in the lower epithelial layers. Podophyllin therapy might produce changes in condylomata mimicking vulvar intraepithelial neoplasia but can be distinguished, particularly with a recent history of podophyllin use. This report describes five patients with vulvar condylomata that morphologically resemble both podophyllin effect and vulvar intraepithelial neoplasia. The mean age was 30.5 years. All lesions exhibited superficial keratinocytes in va

International Papillomavirus Society policy statement on human papillomavirus-based cervical cancer screening for women living with HIV.

OBJECTIVES: Women living with HIV (WLWH) are among those at highest risk for cervical cancer development, thereby making this a key population for primary and secondary prevention. Human papillomavirus (HPV) nucleic acid (DNA or RNA) tests, which have been clinically validated, are supported by the World Health Organization (WHO) as the preferred method for cervical screening for all women, although HPV DNA is preferred in WLWH until further evidence demonstrates that HPV RNA is equivalent. We sought to describe current guidelines for HPV-based cervical cancer screening for WLWH. METHODS: Thi

Failure to detect human papillomavirus sequences at the 3p21 rearrangement site in pleomorphic adenomas.

Eighteen salivary gland pleomorphic adenomas, including nine with a t(3;8)(p21;q12) as the sole chromosome abnormality, were tested for the presence of human papillomavirus (HPV) DNA and for rearrangements of the cellular flanking regions of a HPV 16 integration site mapping to 3p21, as previously described in a cervical carcinoma. In none of the tumors could evidence for either HPV DNA or related sequences, or a rearranged allele of the integration site, be found.

Biological activities of p53 mutants in Burkitt's lymphoma cells.

Wild-type human p53 and a series of p53 point mutants isolated from Burkitt's lymphoma (BL) cell lines were tested for their ability to inhibit DNA synthesis in a p53-negative BL cell line and to bind and be degraded by the human papillomavirus type 16 E6 protein. All the mutants lost the wild-type ability to inhibit DNA synthesis, demonstrating that they are all functionally altered. Binding to E6 and consequent degradation of the p53 mutants frequently correlated with changed suppressor properties in BL cells.

Oral lesions mimicking hairy leukoplakia: a diagnostic dilemma.

Oral hairy leukoplakia is seen in immunosuppressed persons infected with the human immunodeficiency virus and is a predictor of the development of acquired immunodeficiency syndrome in that population. Over the past 3 years we have seen 16 examples of a lesion that histologically resembles hairy leukoplakia but is found in patients who are not in risk groups for acquired immunodeficiency syndrome. All these specimens tested negative for Epstein-Barr virus DNA and for human papillomavirus antigen. Sera from five of the 16 patients were tested for antibodies to human immunodeficiency virus, and

Human papillomavirus DNA remains detectable longer than related cervical cytologic abnormalities.

Cervical human papillomavirus (HPV) infections are at high risk of neoplastic progression if they persist. Persistence can be measured by repeated HPV DNA tests or by cytologic testing. Thus, it is useful to understand the relationship between these 2 measurements. To explore the relative timing of HPV DNA clearance versus cytologic regression, data were analyzed from 840 study participants who were followed-up by repeat thin-layer cytology and HPV testing by a hybrid capture test at 6-month intervals for 2 years. On average, HPV DNA detection persisted longer than related cytologic abnormalit

A pilot analytic study of a research-level, lower-cost human papillomavirus 16, 18, and 45 test.

The analytic performance of a low-cost, research-stage DNA test for the most carcinogenic human papillomavirus (HPV) genotypes (HPV16, HPV18, and HPV45) in aggregate was evaluated among carcinogenic HPV-positive women, which might be used to decide who needs immediate colposcopy in low-resource settings ("triage test"). We found that HPV16/18/45 test agreed well with two DNA tests, a GP5+/6+ genotyping assay (Kappa = 0.77) and a quantitative PCR assay (at a cutpoint of 5000 viral copies) (Kappa = 0.87). DNA sequencing on a subset of 16 HPV16/18/45 positive and 16 HPV16/18/45 negative verified

Non-16/18 high-risk HPV infection predicts disease persistence and progression in women with an initial interpretation of LSIL.

BACKGROUND: The current management strategy for women with low-grade squamous intraepithelial lesions (LSILs) is inefficient and expensive because of the inability to identify patients at high risk for disease progression. The current study was designed to determine the genotypic patterns of human papillomavirus (HPV) associated with the persistence and progression of cervical lesions in women with an initial interpretation of LSIL. METHODS: One hundred sixty-seven women with LSIL on Papanicolaou tests collected between December 1, 2009 and March 30, 2011 were studied. HPV DNA was extracted f

Human papillomavirus typing and viral gene expression analysis for the triage of women with abnormal results from papanicolaou test smears to colposcopy.

CONTEXT: A cascade of molecular tests for human papillomavirus (HPV), as a follow-up to Papanicolaou test screening, could eliminate unnecessary colposcopy. Tests based on detection of HPV E6 messenger RNA (mRNA) are already being used as screening tools, but there is a good biological rationale for expecting that an increase in the relative amounts of HPV E6 mRNA in cervical samples may better predict cancerous transformation. OBJECTIVE: To compare some of the available diagnostic methods and our novel method of relative quantification (RQ) of HPV gene expression for the effective triage of

A papillomavirus DNA from a cervical carcinoma and its prevalence in cancer biopsy samples from different geographic regions.

DNA from one biopsy sample of invasive cancer of the cervix contained sequences hybridizing with human papillomavirus (HPV) type 11 DNA only under nonstringent conditions. This DNA was molecularly cloned in lambda phage. Under stringent conditions of hybridization it cross-hybridized to a minor extent (less than 0.1%) with HPV types 10, 14, and 15 and showed no homology with DNA of other human HPV types. We therefore propose to designate it tentatively as HPV 16. HPV 16 DNA was used as a probe to test additional cancer biopsy samples from cervical, vulval, and penile cancer, as well as benign

Dual genitotropic human papillomavirus infections in genital warts.

BACKGROUND AND METHODS: We have carried out a prospective study of dual genitotropic human papillomavirus (HPV) infections by means of two different DNA detection methods in biopsy specimens obtained from patients who were examined for genital warts at the STD clinic of the School of Medicine in Seville, between January 1990 and December 1991. RESULTS: 100 patients with a clinical diagnosis of condilomata acuminata were seen during the study period. DNA of the genitotropic HPV 6/11, 16/18 and 31/33/35 was detected by an in situ hybridisation method in 75 (77%) of the 98 evaluable samples; one
