Development of portable and highly sensitive electrochemical sensor for rapid on-site analysis of levofloxacin residue in pork samples.

Levofloxacin is widely utilized as a livestock feed additive, yet its residues in animal-derived foods pose risks to human health. Herein, a portable and highly sensitive electrochemical sensor was developed for on-site levofloxacin detection in food of animal origin. The sensor employs facile electrochemical polymerization of 2-hydroxypropyl-β-cyclodextrin (HP-β-CD) on electrodes, leveraging its dual recognition: hydrophobic cavities form complexes with levofloxacin, while hydroxypropyl groups enable hydrogen bonding to enhance the specificity. The optimized parameters endowed the sensors wit

3D-QSAR Studies on Barbituric Acid Derivatives as Urease Inhibitors and the Effect of Charges on the Quality of a Model.

Urease enzyme (EC 3.5.1.5) has been determined as a virulence factor in pathogenic microorganisms that are accountable for the development of different diseases in humans and animals. In continuance of our earlier study on the helicobacter pylori urease inhibition by barbituric acid derivatives, 3D-QSAR (three dimensional quantitative structural activity relationship) advance studies were performed by Comparative Molecular Field Analysis (CoMFA) and Comparative Molecular Similarity Indices Analysis (CoMSIA) methods. Different partial charges were calculated to examine their consequences on the

In Silico Characterization of Conserved Epitopes in Alphavirus E2 Proteins: A Promising Approach for Pan-vaccine Design.

Alphaviruses infect a wide range of hosts, including humans and domestic animals, and they represent an increasing public health concern. Among them, arthritogenic Chikungunya virus (CHIKV) and encephalitogenic Eastern equine encephalitis virus (EEEV) stand out for their epidemic potential and clinical severity. Developing effective and licensed vaccine models against these viruses remains a significant challenge. Rational epitope design, supported by immunoinformatics, offers a promising route for next-generation effective vaccine development. In this study, we utilized the POA pipeline to as

PTSD assistance dogs: concerns for animal well-being, rights, and justice.

PTSD assistance dogs are service dogs trained to assist individuals living with PTSD. A growing body of research links the use of PTSD assistance dogs with substantial benefits for their human partners, including significant reductions in PTSD symptoms, and improvements in family functioning, social integration, and quality of life. However, research on the effect of assistance work on PTSD assistance dogs themselves is notably lacking. This paper aims to address this gap by mapping potential animal welfare and ethical concerns associated with PTSD assistance dog interventions. Moreover, a rig

Therapeutics for Inflammatory-Related Diseases Based on Plasmon-Activated Water: A Review.

It is recognized that the properties of liquid water can be markedly different from those of bulk one when it is in contact with hydrophobic surfaces or is confined in nano-environments. Because our knowledge regarding water structure on the molecular level of dynamic equilibrium within a picosecond time scale is far from completeness all of water's conventionally known properties are based on inert "bulk liquid water" with a tetrahedral hydrogen-bonded structure. Actually, the strength of water's hydrogen bonds (HBs) decides its properties and activities. In this review, an innovative idea on

The periplasmic disulfide oxidoreductase DsbA contributes to Haemophilus influenzae pathogenesis.

Haemophilus influenzae is an obligate human pathogen that persistently colonizes the nasopharynx and causes disease when it invades the bloodstream, lungs, or middle ear. Proteins that mediate critical interactions with the host during invasive disease are likely to be secreted. Many secreted proteins require addition of disulfide bonds by the DsbA disulfide oxidoreductase for activity or stability. In this study, we evaluated the role in H. influenzae pathogenesis of DsbA, as well as HbpA, a substrate of DsbA. Mutants of H. influenzae Rd and type b strain Eagan having nonpolar deletions of ds

Investigating cerebral blood flow in anxious dogs: a 99mTc-HMPAO SPECT imaging study.

Anxiety-related disorders have a significant effect on the welfare of pet dogs, often leading to behavioral problems and straining the human-animal bond. Conventional treatments sometimes prove inadequate, highlighting the need for a more objective understanding of the neurobiological pathways underlying canine anxiety disorders. Therefore, this study aimed to investigate anxiety disorders in dogs using 99mTc-HMPAO single photon emission computed tomography (SPECT), focusing on the left frontal region, subcortical region, and cerebellum. While a frequentist approach found no significant differ

Population medicine and control of epidemics.

Population medicine is an important component of veterinary care in livestock (farm animals) and companion animals (pets). This chapter covers some of the chemotherapeutic approaches undertaken at population level to control infectious diseases in domestic animals. Optimisation of health, productivity and welfare in livestock commonly entails implementation of whole-herd or whole-flock strategies to effectively counter the negative impact of infectious diseases. Gastro-intestinal and liver parasites of grazing cattle and sheep are endemic in most parts of the world and can result in significan

GILT is a critical host factor for Listeria monocytogenes infection.

Listeria monocytogenes is a gram-positive, intracellular, food-borne pathogen that can cause severe illness in humans and animals. On infection, it is actively phagocytosed by macrophages; it then escapes from the phagosome, replicates in the cytosol, and subsequently spreads from cell to cell by a non-lytic mechanism driven by actin polymerization. Penetration of the phagosomal membrane is initiated by the secreted haemolysin listeriolysin O (LLO), which is essential for vacuolar escape in vitro and for virulence in animal models of infection. Reduction is required to activate the lytic activ

Amino Acid Nutrition for Optimum Growth, Development, Reproduction, and Health of Zoo Animals.

Proteins are large polymers of amino acids (AAs) linked via peptide bonds, and major components for the growth and development of tissues in zoo animals (including mammals, birds, and fish). The proteinogenic AAs are alanine, arginine, aspartate, asparagine, cysteine, glutamate, glutamine, glycine, histidine, isoleucine, leucine, lysine, methionine, phenylalanine, proline, serine, threonine, tryptophan, tyrosine, and valine. Except for glycine, they are all present in the L-isoform. Some carnivores may also need taurine (a nonproteinogenic AA) in their diet. Adequate dietary intakes of AAs are

Selenium-centered radicals: generation, detection and their relevance in chemistry and biology.

A shift in perception of selenium from a metabolic toxin to an essential trace element for humans and animals has a profound implication on various scientific disciplines. Selenium is vital for maintaining cellular redox homeostasis, preserving structural integrity, and regulating immune defense and cancer prevention strategies. These biological impacts are driven by its unique properties including redox sensitivity, high nucleophilicity and multiple oxidation states. Transitions between different oxidation states occur via multi-step electron transfers that often involve transient selenium-ce

Structure-Activity Relationship of Chlorotoxin-Like Peptides.

Animal venom (e.g., scorpion) is a rich source of various protein and peptide toxins with diverse physio-/pharmaco-logical activities, which generally exert their action via target-specific modulation of different ion channel functions. Scorpion venoms are among the most widely-known source of peptidyl neurotoxins used for callipering different ion channels, such as; Na⁺, K⁺, Ca⁺, Cl(-), etc. A new peptide of the chlorotoxin family (i.e., Bs-Tx7) has been isolated, sequenced and synthesized from scorpion Buthus sindicus (family Buthidae) venom. This peptide demonstrates 66% with chlorotoxin (C

Salivary amylase - The enzyme of unspecialized euryphagous animals.

INTRODUCTION: Alpha-amylase (EC 3.2.1.1) is the most abundant enzyme in the saliva of man and of several vertebrates. In humans, salivary amylase is mainly formed in the parotid gland; its activity is of high inter-individual and intra-individual variability. The physiological functions of α-amylase have not yet been explored completely. It is well known that the enzyme cleaves the α-(1,4)-glycosidic bonds of polysaccharides. Furthermore it plays an important role in initial bioadhesion in man, facilitating carbohydrate metabolism and bacterial adherence at the tooth surface and therewith cari

Mutation of a conserved lysine in the RdRp fingers domain broadly attenuates orthobunyaviruses.

Orthobunyaviruses, comprising more than 130 species, constitute a significant threat to human and animal health, yet no effective broad-spectrum treatments or vaccines are currently available. Here, we initially identified an evolutionarily constrained lysine residue, highly conserved across all 22 Orthobunyavirus serogroups, within the α30 helix of the RNA-dependent RNA polymerase (RdRp). Functional interrogation of this site by charge-reversal substitution (K-to-E) substantially impaired polymerase activity and led to marked attenuation of Ebinur Lake virus (EBIV), as evidenced by impaired r

Evolutionary Adaptability Coupled with Computation-Driven Engineering for Thermostability Enhancement of a Deoxynivalenol-Detoxifying Fusion Enzyme.

Deoxynivalenol (DON), a trichothecene mycotoxin commonly found in cereal grains and their derived products, poses significant risks to human and animal health. In previous work, a fusion enzyme composed of the dehydrogenase DADH and the aldo-keto reductase AKR13B3 was engineered to convert DON into the non-toxic 3-epi-DON in a single step. However, the poor thermal stability of this fusion enzyme limited its industrial application. In this study, EVcouplings and the GRAPE-WEB platform were utilized to identify key amino acid residues governing the thermal stability of the fusion enzyme AKR13B3

Hydration and hydrolysis define antibiotic resistance conferred by macrolide esterases.

Macrolides are an antibiotic class widely used in both human and veterinary medicine, and function by interfering with protein synthesis. Regrettably, numerous strategies for evading the antibiotic properties of macrolides have been found in bacteria, including enzyme-mediated inactivation. These mechanisms are now widely disseminated among pathogenic, animal-associated, and environmental bacteria making them a One Health issue. Macrolide esterases, which hydrolyze the macrolactone's ester bond, confer one such resistance mechanism. Two types of macrolide esterases have thus far been identifie

Multilevel dynamics of the brain, hormones, mind, and behavior in social human-robot interaction.

As robots enter homes, workplaces, and health care settings, sustaining trust during social interaction becomes a central challenge for human-robot interaction. However, relatively little is known about how humans integrate signals across the brain, hormones, mind, and behavior when robots violate expectations or display social expressiveness. Addressing this gap, we examined how robot performance (congruent versus erroneous) and expressiveness (animated versus stationary) shape multilevel human responses during face-to-face decision-making with an embodied humanoid robot. Participants engaged

Essential role of docosahexaenoic acid towards development of a smarter brain.

Evolution of the high order brain function in humans can be attributed to intake of poly unsaturated fatty acids (PUFAs) of which the ω-3 fatty acid, docosahexaenoic acid (DHA) has special significance. DHA is abundantly present in the human brain and is an essential requirement in every step of brain development like neural cell proliferation, migration, differentiation, synaptogenesis etc. The multiple double bonds and unique structure allow DHA to impart special membrane characteristics for effective cell signaling. Evidences indicate that DHA accumulate in areas of the brain associated wit

Psycho-Spiritual Reflections on the Horse/Human Relationship.

This essay offers a psycho-spiritual and psychoanalytic meditation on the horse-human relationship as a nonverbal site of attunement, regulation, and meaning-making. Drawing on autobiographical clinical narrative, it explores how horseback riding became a medium for experiencing and reflecting upon embodied connection, mutual responsiveness, and contemplative presence. Encounters with horses illuminate forms of reciprocity that preceded and exceed speech, while also opening reflection on maternal attachment, separation, loss, and the emergence of selfhood. In dialogue with Freud, Winnicott, Pi
