Hydrostatic pressure of the renal pelvis as a radiation-free alternative to fluoroscopic nephrostogram following percutaneous nephrolithotomy.

BACKGROUND: We evaluated the hydrostatic pressure of the renal pelvis (RPP) as a radiation-free alternative to fluoroscopic nephrostogram to assess ureteral patency after percutaneous nephrolithotomy (PCNL). METHODS: Retrospective non-inferiority study analyzing 248 PCNL-patients (86 female (35%) and 162 males (65%)) between 2007 and 2015. Postoperatively, RPP was measured using a central venous pressure manometer in cmH2O. The primary endpoint was to assess RPP depending on the patency of the ureter and the nephrostomy tube removal. Secondary, the upper limit of normal RPP of [Formula: see t

Analysis of HIC and Hydrostatic Pressure in the Human Head during NOCSAE Tests of American Football Helmets.

Brain damage is a serious economic and social burden. Contact sports such as American football, are one of the most common sources of concussions. The biomechanical response of the head-helmet system caused by dynamic loading plays a major role. The literature has focused on measuring the resultant kinematics that act on the head and helmet during tackles. However, few studies have focused on helmet validation tests, supported by recent findings and emerging numerical approaches. The future of helmet standards could benefit from insights at the level of injury mechanisms, using numerical tools

Blood pressure measurement and nocturnal dipping patterns are heavily affected by body posture through changes in hydrostatic pressure between the arm and the heart.

Nocturnal blood pressure (BP) shows the highest predictive power for cardiovascular events. However, there is a poor reproducibility of personalized dipping patterns in single individuals. We hypothesize that changes in body position during sleep cause variations in hydrostatic pressure, leading to incorrect BP values and dipping classifications. 26 subjects aged 18-30 years, as well as 25 participants aged 50 years and older underwent ambulatory BP measurements on the left arm, as well as determination of the hydrostatic pressure difference between the cuff and heart level during BP measuremen

Cellular behavior and extracellular matrix turnover in bovine annulus fibrosus cells under hydrostatic pressure and deviatoric strain.

Intervertebral disc herniation is a common spinal disorder that is often treated with discectomy when conservative measures fail. To devise therapeutic strategies for tears in the annulus fibrosus (AF), the regenerative capability of AF cells under spinal loading needs to be addressed. We hypothesized that the compressive loading associated with deformation in AF cells reduces synthetic and degradative activities in extracellular matrix and cell proliferation. We evaluated expression of key matrix molecules and cell proliferation by RT-PCR and immunohistochemistry by inner and outer bovine AF

Physiological cyclic hydrostatic pressure induces osteogenic lineage commitment of human bone marrow stem cells: a systematic study.

BACKGROUND: Physical loading is necessary to maintain bone tissue integrity. Loading-induced fluid shear is recognised as one of the most potent bone micromechanical cues and has been shown to direct stem cell osteogenesis. However, the effect of pressure transients, which drive fluid flow, on human bone marrow stem cell (hBMSC) osteogenesis is undetermined. Therefore, the objective of the study is to employ a systematic analysis of cyclic hydrostatic pressure (CHP) parameters predicted to occur in vivo on early hBMSC osteogenic responses and late-stage osteogenic lineage commitment. METHODS:

Stability of different influenza subtypes: How can high hydrostatic pressure be a useful tool for vaccine development?

BACKGROUND: Avian influenza A viruses can cross naturally into mammals and cause severe diseases, as observed for H5N1. The high lethality of human infections causes major concerns about the real risk of a possible pandemic of severe diseases to which human susceptibility may be high and universal. High hydrostatic pressure (HHP) is a valuable tool for studies regarding the folding of proteins and the assembly of macromolecular structures such as viruses; furthermore, HHP has already been demonstrated to promote viral inactivation. METHODS: Here, we investigated the structural stability of av

Biological functions at high pressure: transcriptome response of Shewanella oneidensis MR-1 to hydrostatic pressure relevant to Titan and other icy ocean worlds.

High hydrostatic pressure (HHP) is a key driver of life's evolution and diversification on Earth. Icy moons such as Titan, Europa, and Enceladus harbor potentially habitable high-pressure environments within their subsurface oceans. Titan, in particular, is modeled to have subsurface ocean pressures ≥ 150 MPa, which are above the highest pressures known to support life on Earth in natural ecosystems. Piezophiles are organisms that grow optimally at pressures higher than atmospheric (0.1 MPa) pressure and have specialized adaptations to the physical constraints of high-pressure environments - u

High and intermittent hydrostatic pressure promotes maturation of engineered cardiac tissues derived from human pluripotent stem cells.

The immaturity of human pluripotent stem cell-derived cardiomyocytes (hPSC-CMs) and engineered cardiac tissues (ECTs) limits their use in regenerative therapies. Ventricular loading pressure plays a vital role in cardiac repair, prompting the hypothesis that hydrostatic pressure could enhance ECT maturation. ECTs were created by co-culturing hPSC-derived cardiovascular cells with extracellular matrix proteins and subjecting them to intermittent hydrostatic pressure (1 h/day at 50 kPa for 3 days after 2 weeks of pre-culture). This protocol improved cardiomyocyte alignment, mitochondrial content

Gas permeability-dependent plastron longevity on superhydrophobic surfaces: opposite trends under hydrostatic pressure and degassed water conditions.

HYPOTHESIS: The plastron trapped on an underwater superhydrophobic surface plays a crucial role in various applications, such as drag reduction, but its longevity may be limited due to gas diffusion. A gas permeable substrate may significantly affect plastron longevity due to its capability to store or release gas. However, the effect of gas permeability on plastron longevity has received little attentions. EXPERIMENTS: In this work, surfaces with varying gas permeabilities from ∼0.05 to ∼1600 Barrers were fabricated using polydimethylsiloxane (PDMS) and polyepoxide (Epoxy), as well as by var

High hydrostatic pressure induced gastrointestinal digestion behaviors of quercetin-loaded casein delivery systems under different calcium concentration.

Casein micelle has a structure of outer hydrophilicity and inner hydrophobicity, its typical digestion characteristic is gastric coagulation. Based on calcium content as the key factor to control this process, high hydrostatic pressure (HHP) was firstly used to modify the micelle structure by mediating the tight connection between casein molecules themselves and with colloidal calcium, then the quercetin-loaded delivery systems were prepared. And in order to investigate the effect of exogenous calcium, calcium chloride was added for digestion. The results indicated that HHP broke the limitatio

Monocyte Chemotactic Protein-1 Regulates Proliferation and Contractility of Human Bladder Smooth Muscle Cells Under Hydrostatic Pressure.

Bladder growth and development require continuous mechanical stimuli; however, abnormal mechanical conditions caused by partial bladder outlet obstruction (PBOO) result in detrusor smooth muscle hypertrophy and extracellular matrix accumulation. Our previous study demonstrated that PBOO induced an increase in monocyte chemotactic protein-1 (MCP-1) expression. To extensively illustrate whether and how MCP-1 participates in the remodeling of PBOO, we assess the pivotal effect of MCP-1 and its specific receptor chemokine (C-C motif) receptor 2 (CCR2) on the proliferation and contraction of human

β-Adrenoceptor signaling regulates proliferation and contraction of human bladder smooth muscle cells under pathological hydrostatic pressure.

BACKGROUND: Partial bladder outlet obstruction (PBOO) promotes bladder detrusor hyperplasia, increases bladder pressure, and decreases bladder compliance. To extensively explore its underlying mechanism, our study aimed to investigate the effect of pathological hydrostatic pressure on human bladder smooth muscle cell (hBSMC) proliferation and contraction through β-adrenoceptor (ADRB) signaling in vitro. METHODS: hBSMCs were subjected to pathological hydrostatic pressure (100 cm H2 O) to investigate the effect of ADRBs on the proliferation and contraction of hBSMCs treated with its agonists an

The independent effects of hydrostatic pressure and hypercapnic breathing during water immersion on ventilatory sensitivity and cerebrovascular reactivity.

Head-out water immersion (HOWI) induces ventilatory and hemodynamic changes, which may be a result of hydrostatic pressure, augmented arterial CO2 tension, or a combination of both. We hypothesized that the hydrostatic pressure and elevated CO2 tension that occur during HOWI will contribute to an augmented ventilatory sensitivity to CO2 and an attenuated cerebrovascular reactivity to CO2 during water immersion. Twelve subjects [age: 24 ± 3 yr, body mass index (BMI): 25 ± 3 kg/m2] completed HOWI, waist water immersion with CO2 (WWI + CO2), and WWI, where a rebreathing test was conducted at base

Hydrostatic pressure and muscarinic receptors are involved in the release of inflammatory cytokines in human bladder smooth muscle cells.

AIMS: Abnormal intravesical pressure results in a series of pathological changes. We investigated the effects of hydrostatic pressure and muscarinic receptors on the release of inflammatory cytokines in rat and human bladder smooth muscle cells (HBSMCs). METHODS: Animal model of bladder outlet obstruction was induced by urethra ligation. HBSMCs were subjected to elevated hydrostatic pressure and/or acetylcholine (Ach). Macrophage infiltration in the bladder wall was determined by immunohistochemical staining. The expression of inflammatory genes was measured by RT-PCR, ELISA and immunofluores

High hydrostatic pressure upon the vasa vasorum of the greater saphenous and splenic vein walls: a comparative study.

AIMS: Hypoxia and high hydrostatic pressure can induce an increase in the thickness of the tunica media and intima; secondary vasa vasorum (VV) increase to fit the remodeling of the vessel wall. We aimed to investigate the impact of high hydrostatic pressure on VV in the varicose greater saphenous veins (VGSVs) and diseased splenic veins (DSVs). METHODS: We collected 34 VGSVs and DSVs. Thirty-four normal greater saphenous veins (GSVs) and splenic veins (SVs) were also collected (control group). Samples were cut into slices, and observed under both light and electron microscopy. Blood pressure variation in response to changing arm cuff height cannot be explained solely by the hydrostatic effect.

OBJECTIVE: To challenge the widely accepted explanation of hydrostatic effect for blood pressure (BP) variation in response to arm elevation by testing the hypothesis that SBP and DBP measurements in the same individual vary differently with cuff height, and to evaluate the potential deviation of these measurements from the value predicted by the hydrostatic theory. METHODS: Arm BP was measured in 37 individuals from the general population in the sitting position using a digital BP monitor. Measurements included nine consecutive BP readings at four cuff heights. The height dependence of BP wa

Insight into the mechanism of high hydrostatic pressure effect on inhibitory efficiency of three natural inhibitors on polyphenol oxidase.

The development of natural inhibitors of polyphenol oxidase (PPO) is crucial in the prevention of enzymatic browning in fresh foods. However, few studies have focused on the effect of subsequent sterilization on their inhibition efficiency. This study investigated the influence and mechanism of high hydrostatic pressure (HHP) on the inhibition of PPO by epigallocatechin gallate (EGCG), cyanidin-3-O-glucoside (C3G), and ferulic acid. Results showed that under the conditions of 550 MPa/30 min, the activity of EGCG-PPO decreased to 55.92%, C3G-PPO decreased to 81.80%, whereas the activity of FA-P

Metabolic hormones in human breast milk are preserved by high hydrostatic pressure processing but reduced by Holder pasteurization.

In human milk banks (HMBs), donor milk (DM) is commonly sterilized by Holder pasteurization (HoP). High hydrostatic pressure (HHP) processing is an innovative, alternative method for DM sterilization. We evaluated the impact of HHP processing on the concentration of seven metabolic milk hormones. Eight samples of raw DM were aliquoted. One aliquot was sterilized by HoP (62 °C for 30 min), and another was processed by HHP (350 MPa at 38 °C). Compared with raw DM, HoP milk displayed reduced concentrations of insulin, nesfatin-1, cortisol, leptin, apelin and GLP-1, though adiponectin levels were

In vivo methods for testing allergenicity show that high hydrostatic pressure hydrolysates of β-lactoglobulin are immunologically inert.

The major milk allergen β-lactoglobulin (β-LG) exhibits an enhanced susceptibility to proteolysis under high hydrostatic pressure and this may be an efficient method to produce hypoallergenic hydrolysates. The aim of this work was to evaluate the in vivo allergenicity of 3 β-LG hydrolysates produced under atmospheric pressure or high-pressure conditions. Hydrolysates were chosen based on previous experiments that showed that they provide a complete removal of intact β-LG but differed in vitro IgE-binding properties that could be traced to the peptide pattern.
