A uniquely leptin sensitive hypothalamic neuron population limits hyperphagia and weight gain in diet-induced obesity.

Despite widespread loss of leptin responsiveness in obesity, endogenous leptin continues to restrain feeding, yet the neural substrates that remain sensitive and mediate this effect are unknown. Combining spatial transcriptomics with single-nucleus RNA sequencing in mice with diet-induced obesity (DIO), we show that while most hypothalamic leptin receptor (Lepr) neurons minimally respond to elevated leptin, a single population defined by glucagon-like peptide-1 receptor (Glp1r) co-expression retains robust leptin sensitivity. These Lepr/Glp1r neurons project onto and restrain orexigenic Agrp n

Hyperphagia in craniopharyngioma- a real-world study from the international hypothalamic-pituitary brain tumors patient registry.

We previously showed that hyperphagia affects quality of life of craniopharyngioma (CP) survivors. Characteristics of hyperphagia are unknown. This study aimed to characterize and quantify hyperphagia in CP survivors with and without hypothalamic obesity (HO). Forty caregivers responded to online questionnaires including the Hyperphagia Questionnaire for Clinical Trials (HQ-CT) and Food Safe Zone (FSZ). Caregivers reported significantly more comorbidities in survivors with HO, with 82% presenting with hypothalamic syndrome. Survivors with HO had higher hunger, lower satiety, and higher food pr

Promotion of obesity by fibroblast growth factor 21-oxytocin system dysfunction due to sugar-specific hyperphagia.

Fibroblast growth factor (FGF) 21 activates oxytocin (OXT) neurons in the hypothalamus and suppresses simple sugar preference; however, alterations in the FGF21-OXT system in obesity remain unclear. In this study, we examined alterations in FGF21 secretion to systemic circulation and FGF21 sensitivity of OXT neurons in obesity, and the effects of FGF21-OXT dysfunction on feeding and body weight regulation. High-fat high-sucrose diet (HFHSD) feeding promoted hypersecretion of FGF21. The administration of recombinant FGF21 to normal diet-fed mice significantly activated OXT neurons in the parave

Psychometric evaluation of novel hyperphagia questionnaires in a real-world setting for patients with a rare MC4R pathway disease.

BACKGROUND: There are no validated measures to assess hyperphagia associated with rare MC4R pathway diseases, such as Bardet-Biedl Syndrome (BBS). Symptoms of Hyperphagia© (SoH) and Impacts of Hyperphagia© (IoH) are novel questionnaires designed to assess signs and symptoms of hyperphagia and their impacts on patients and caregivers. We evaluated the psychometric performance of the caregiver-versions of the SoH: Caregiver (Observer-reported) and IoH: Caregiver (Observer-reported and Self-reported subscales). RESULTS: Reliability and validity were evaluated using data from a multi-country cros

Gsα deficiency in the dorsomedial hypothalamus leads to obesity, hyperphagia, and reduced thermogenesis associated with impaired leptin signaling.

OBJECTIVE: Gsα couples multiple receptors, including the melanocortin 4 receptor (MC4R), to intracellular cAMP generation. Germline inactivating Gsα mutations lead to obesity in humans and mice. Mice with brain-specific Gsα deficiency also develop obesity with reduced energy expenditure and locomotor activity, and impaired adaptive thermogenesis, but the underlying mechanisms remain unclear. METHODS: We created mice (DMHGsKO) with Gsα deficiency limited to the dorsomedial hypothalamus (DMH) and examined the effects on energy balance and thermogenesis. RESULTS: DMHGsKO mice developed severe,

A new stress-related syndrome of growth failure and hyperphagia in children, associated with reversibility of growth-hormone insufficiency.

BACKGROUND: Growth failure without organic aetiology but associated with behavioural disturbance and psychosocial stress has been termed psychosocial short stature. This condition is not a valid diagnostic entity, but encompasses failure to thrive, stunting secondary to chronic malnutrition, and idiopathic hypopituitarism. Some children show spontaneous catch-up growth when removed from the source of stress, without further treatment, but until now precise definition of this subgroup for the purpose of clinical identification has not been possible. METHODS: Hospital-referred children with gro

Leptin reverses hyperglycemia and hyperphagia in insulin deficient diabetic rats by pituitary-independent central nervous system actions.

The hypothalamic-pituitary-adrenal (HPA) axis has been postulated to play a major role in mediating the antidiabetic effects of leptin. We tested if the pituitary is essential for the chronic central nervous system mediated actions of leptin on metabolic and cardiovascular function in insulin-dependent diabetic and non-diabetic rats. Male 12-week-old hypophysectomized Sprague-Dawley rats (Hypo, n = 5) were instrumented with telemetry probes for determination of mean arterial pressure (MAP) and heart rate (HR) 24-hrs/day and an intracerebroventricular (ICV) cannula was placed into the brain lat

Increased sugar intake as a form of compensatory hyperphagia in patients with type 2 diabetes under dapagliflozin treatment.

AIMS: Sodium-glucose cotransporter 2 inhibitors (SGLT2i) cause substantially less weight loss than would be expected based on their caloric deficits, probably due to enhanced appetite regulation known as "compensatory hyperphagia," which occurs to offset the negative energy balance caused by increased glycosuria. We examined whether any specific nutrients contributed to the compensatory hyperphagia in diabetic patients taking SGLT2i. METHODS: Sixteen patients with type 2 diabetes were newly administered dapagliflozin 5 mg daily as the experimental SGLT2i group. Sixteen age-, sex- and BMI-matc

Stimulation of dopamine D4 receptors in the paraventricular nucleus of the hypothalamus of male rats induces hyperphagia: involvement of glutamate.

Obesity is a serious worldwide health problem, affecting 20-40% of the population in several countries. According to animal models, obesity is related to changes in the expression of proteins that control energy homeostasis and in neurotransmission associated to regulation of food intake. For example, it has been reported that diet-induced obesity produces overexpression of dopamine D4 receptor (D4R) mRNA in the ventromedial hypothalamic nucleus (VMH) of mice. Neurons in the VMH send dense glutamatergic projections to other hypothalamic regions as the paraventricular nucleus (PVN), where multi

Relationship between Aberrant Behavior Checklist (ABC) scores, hyperphagia, and quality of life in youth with Prader-Willi syndrome.

BACKGROUND: Patients with Prader-Willi syndrome (PWS) can experience a complex array of symptoms including hyperphagia and behavioral disturbances. The Aberrant Behavior Checklist (ABC) is a standardized rating scale for the assessment of maladaptive behaviors with established use in populations with developmental disabilities. Our study aimed to describe ABC scores in youths with PWS and how they relate to changes in hyperphagia and quality of life with age. METHODS: Families seen at the Seattle Children's Hospital Prader-Willi syndrome (SCH PWS) clinic filled out questionnaires (ABC, PWS Hy

Programmed hyperphagia in offspring of obese dams: Altered expression of hypothalamic nutrient sensors, neurogenic factors and epigenetic modulators.

Maternal overnutrition results in programmed offspring obesity, mediated in part, by hyperphagia. This is remarkably similar to the effects of maternal undernutrition on offspring hyperphagia and obesity. In view of the marked differences in the energy environment of the over and under-nutrition exposures, we studied the expression of select epigenetic modifiers associated with energy imbalance including neurogenic factors and appetite/satiety neuropeptides which are indicative of neurogenic differentiation. HF offspring were exposed to maternal overnutrition (high fat diet; HF) during pregnan

Liraglutide attenuates nicotine self-administration as well as nicotine seeking and hyperphagia during withdrawal in male and female rats.

RATIONALE: Nicotine cessation is associated with increased consumption of highly palatable foods and body weight gain in most smokers. Concerns about body weight gain are a major barrier to maintaining long-term smoking abstinence, and current treatments for nicotine use disorder (NUD) delay, but do not prevent, body weight gain during abstinence. Glucagon-like peptide-1 receptor (GLP-1R) agonists reduce food intake and are FDA-approved for treating obesity. However, the effects of GLP-1R agonist monotherapy on nicotine seeking and withdrawal-induced hyperphagia are unknown. OBJECTIVES: We sc

Hypothalamic neuropeptide Y (NPY) and the attenuation of hyperphagia in streptozotocin diabetic rats treated with dopamine D1/D2 agonists.

1. Dopamine is an appetite suppressant, while neuropeptide Y (NPY), an appetite stimulant in the brain, is reported to be involved in anorectic action induced by a combined administration of D1/D2 agonists in normal rats. In diabetic rats, however, these factors have not been studied. 2. Rats (including normal, diabetic and insulin-treated diabetic rats) were given daily injections of saline or D1/D2 agonists for 6 days. Changes in food intake and hypothalamic NPY content of these rats were assessed and compared. 3. The D1/D2 agonist-induced anorectic responses were altered in diabetic rats co

Reduction of normal food intake in rats and dogs and inhibition of experimentally induced hyperphagia in rats by CM 57373 and fenfluramine.

The anorectic effect of CM 57373 in dogs and in rats food-deprived or with experimentally induced hyperphagia (cafeteria-diet hyperphagia and insulin hyperphagia) was compared to the effect of serotoninergic anorectic drug dl-fenfluramine. CM 57373 and dl-fenfluramine administered orally caused a dose-related reduction of food consumption by food-deprived rats (ID50 = 7.4 mg/kg and 2.5 mg/kg respectively). The oral ID50 in dogs was 2.4 mg/kg for CM 57373 and 1.1 mg/kg for dl-fenfluramine. This animal species tolerated CM 57373 better than dl-fenfluramine. The latter induced mydriasis, dyskines

Pituitary adenylate cyclase-activating polypeptide modulates circadian feeding and emotional behavior in mice independent of stress-induced hyperphagia.

BACKGROUND AND AIM: Pituitary adenylate cyclase-activating polypeptide (PACAP) is a neuropeptide widely implicated in stress responses, appetite regulation, and emotional behavior. While PACAP has been linked to stress-induced appetite suppression, its role in circadian feeding and stress-related hyperphagia remains poorly defined. This study aimed to clarify the contribution of PACAP to circadian feeding behavior, chronic stress-induced hyperphagia, and emotional regulation in mice. MATERIALS AND METHODS: Experiments were conducted using PACAP knockout (KO) and wild-type (WT) CD-1 mice. Acut
