Neutrophils recruited to the myocardium after acute experimental myocardial infarct generate hypochlorous acid that oxidizes cardiac myoglobin.

Myocardial inflammation following acute myocardial infarct (AMI) is associated with risk of congestive heart failure. Pro-inflammatory neutrophils were recruited to the damaged myocardium 24 h after permanent coronary ligation in rats to induce AMI as judged by the presence of immune-positive myeloperoxidase (MPO) in the tissues; MPO generates the oxidant hypochlorous acid (HOCl). Neutrophils were absent in hearts from Control (untreated) and surgical Sham. Similarly, rats exposed to 1 h coronary ligation (Ischemia) showed no neutrophil infiltrate. Concomitantly, MPO activity increased in left

Identification of Nrf2-dependent airway epithelial adaptive response to proinflammatory oxidant-hypochlorous acid challenge by transcription profiling.

In inflammatory diseases of the airway, a high level (estimated to be as high as 8 mM) of HOCl can be generated through a reaction catalyzed by the leukocyte granule enzyme myeloperoxidase (MPO). HOCl, a potent oxidative agent, causes extensive tissue injury through its reaction with various cellular substances, including thiols, nucleotides, and amines. In addition to its physiological source, HOCl can also be generated by chlorine gas inhalation from an accident or a potential terrorist attack. Despite the important role of HOCl-induced airway epithelial injury, the underlying molecular mech

Inactivation Effects of Hypochlorous Acid, Chlorine Dioxide, and Ozone on Airborne SARS-CoV-2 and Influenza A Virus.

Severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) and influenza A virus are primarily transmitted through droplets or aerosols from patients. The inactivation effects of existing virus control techniques may vary depending on the environmental factors. Therefore, it is important to establish a suitable evaluation system for assessing virus control techniques against airborne viruses for further real-world implementation. This study aimed to assess the inactivating effects of chemical substances on SARS-CoV-2 and influenza A virus in the air using an established evaluation system. A

Rationally constructed nanoprobe based on the molecular docking strategy for rapid hypochlorous acid detection in foods and biosystems.

Hypochlorous acid (HClO) is not only often used as an effective preservative in the food industry, but also is an important reactive oxygen species in organisms. Herein, a series of Near-infrared (NIR) small-molecule HClO fluorescent probes based on dicyanoisophorone fluorophore were constructed by reasonable design, and docking with bovine serum albumin was carried out, and the best docking effect was screened to prepare a fluorescent nanoprobe (DBH-1) for HClO detection via the self-assembly of host and guest. This nanoprobe features outstanding water solubility, good biocompatibility, and h

A near-infrared AND logic fluorescent probe that simultaneously responds to Cu2+ and hypochlorous acid for the imaging of Wilson's disease.

Wilson's disease (WD) features aberrant hepatic copper accumulation with oxidative and inflammatory injury. Single-target probes responsive to Cu2+ or reactive oxygen species alone have limited specificity for WD imaging in complex biological environments. Here, we report a near-infrared AND logic fluorescent probe, namely DHU-LP, for the simultaneous detection of Cu2+ and hypochlorous acid (HOCl) to enable specific WD imaging. DHU-LP, constructed by integrating an Oxazine 1 fluorophore with a bis(pyridin-2-ylmethyl)amine Cu2+-binding unit, exhibited a pronounced turn-on response at 669 nm onl

Bactericidal Efficacy and Mechanisms of Non-Electrolytic Slightly Acidic Hypochlorous Water on Pseudomonas fragi and Pseudomonas fluorescens.

Chilled pork is frequently contaminated with Pseudomonas fragi and Pseudomonas fluorescens. In this study, the bactericidal efficacy and mechanisms of non-electrolytic slightly acidic hypochlorous water (NE-SAHW) against two strains of these two species were evaluated. The results showed that the antibacterial efficacy of NE-SAHW was positively correlated with the concentration level of NE-SAHW and negatively correlated with the initial populations of the strains. The strains of small populations were completely inhibited when provided with each level of NE-SAHW. The killed cells of P. fragi w

Fabrication of CdS quantum dots with egg white and application in the assay of hypochlorous acid and myeloperoxidase activity and inhibition.

The myeloperoxidase (MPO)/H2O2-Cl- enzymatic reaction system and its product hypochlorous acid (HOCl) are closely related to many disease processes, and new methods to detect the levels of HOCl and MPO are being focused on. MPO is the only known enzyme for the catalytic production of HOCl in biological systems; therefore, monitoring the HOCl levels is a selective and direct readout of MPO activity. This study reported a simple and efficient fluorescence assay of HOCl and MPO activity and inhibition. Highly fluorescent CdS quantum dots (CdS QDs) were prepared in one pot where NaOH-pretreated eg

A near-infrared fluorescent probe for hypochlorous acid enables high-content screening of natural products for hypertension intervention.

Hypertension is accompanied by persistent oxidative stress and inflammatory activation, which contribute to vascular dysfunction and target-organ injury. Hypochlorous acid (HClO), a highly reactive oxidant generated through the myeloperoxidase pathway, may serve as a molecular indicator of hypertension-associated oxidative and inflammatory damage. Herein, we developed OX-MT, an HClO-responsive near-infrared fluorescent probe, and integrated it with high-content imaging for the screening of natural products with potential antihypertensive activity. OX-MT enabled rapid (10 s), selective, and sen

Dose-dependent transitions in Nrf2-mediated adaptive response and related stress responses to hypochlorous acid in mouse macrophages.

Hypochlorous acid (HOCl) is potentially an important source of cellular oxidative stress. Human HOCl exposure can occur from chlorine gas inhalation or from endogenous sources of HOCl, such as respiratory burst by phagocytes. Transcription factor Nrf2 is a key regulator of cellular redox status and serves as a primary source of defense against oxidative stress. We recently demonstrated that HOCl activates Nrf2-mediated antioxidant response in cultured mouse macrophages in a biphasic manner. In an effort to determine whether Nrf2 pathways overlap with other stress pathways, gene expression prof

Detection of hypochlorous acid fluctuation via a near-infrared fluorescent probe in Parkinson's disease cells and mouse models.

Parkinson's disease (PD) is a neurodegenerative disorder caused by excessive reactive halogen species leading to the death of dopaminergic (DA) neurons, which disrupts the coordination of normal physiological structures and functions. Hypochlorous acid (HOCl) is a reactive halogen species whose overproduction is associated with the death of DA neurons. Herein, overproduction of HOCl may be a neurotoxin substance in the pathogenesis of PD. Therefore, it is essential to understand the disease of HOCl in PD model. However, early detection HOCl in PD model remains lacking of effective methods. In

Evaluation of 415-nm Blue Light and Hypochlorous Acid on the Rhesus Macaque (Macaca mulatta) Cranial Implant Microbiota.

Cranially-implanted macaques are a crucial model for neuroscience research. Implant complications include abscesses, meningoencephalitis, and implant-tissue margin infections. Antimicrobial overuse has increased bacterial resistance, risking macaque health and complicating treatment of infections. This project aimed to assess the antimicrobial effects of blue light (∼415 nm) and hypochlorous acid (HOCl) treatment of cranial implant margins. Blue light exerts antibacterial effects via the induction of reactive oxygen species. We hypothesized that exposing cranial implant margins to a commercial

Histones in neutrophil extracellular traps (NETs) contain oxidative post-translational modifications induced by the myeloperoxidase oxidant hypochlorous acid.

Extracellular traps (NETs) released by neutrophils during inflammation play a role in clearing infection but also contribute to disease pathology. NETs consist of a DNA backbone containing histones, anti-microbial granule proteins, such as myeloperoxidase (MPO), and other proteins. MPO remains enzymatically active and generates hypochlorous acid (HOCl) to kill pathogens. However, HOCl also readily reacts with proteins, but whether histones and other NET proteins are modified by this oxidant is unknown. This is significant as post-translational modification of histones alters their intracellula
