Investigating the Role of Known Arabidopsis Iron Genes in a Stress Resilient Soybean Line.

Genes involved in iron deficiency responses have been well characterized in Arabidopsis thaliana, but their roles in crop species have not been well explored. Reliance on model species may fail to identify novel iron stress mechanisms present within crop species, likely selected by hundreds of years of selection. Fiskeby III (PI 438471) is a soybean line from Sweden that demonstrates high levels of resilience to numerous stresses. Earlier Fiskeby III studies have identified a suite of genes responding to iron deficiency stress in Fiskeby III that are also associated with Arabidopsis iron defic

Not All Maca Is Created Equal: A Review of Colors, Nutrition, Phytochemicals, and Clinical Uses.

Maca (Lepidium meyenii, Lepidium peruvianum) is part of the Brassicaceae family and grows at high altitudes in the Peruvian Andes mountain range (3500-5000 m). Historically, it has been used as a nutrient-dense food and for its medicinal properties, primarily in enhancing energy and fertility. Scientific research has validated these traditional uses and other clinical applications by elucidating maca's mechanisms of action, nutrition, and phytochemical content. However, research over the last twenty years has identified up to seventeen different colors (phenotypes) of maca. The color, hypocoty

[Improved protoplast-derived plants of Astragalus adsurgens through somatic embryogenesis].

Embryogenic callus was obtained only from hypocotyl explants of Astragalus adsurgens and light inhibited the formation of embryogenic callus. A high yield (1.2 x 10(6)/g F. Wt.) of protoplasts with high viability (over 80%) could be isolated from 10-day-old embryogenic callus. Protoplasts were induced to undergo sustained division and to form cell colonies when cultured in agarose-solidified medium (KMP) containing 1/4 strength of mineral salts and supplemented with 1.5 mg/L 2, 4-D, 0.5 mg/L BA and 0.5 mol/L glucose at a plating density of 1.0 x 10(5)mL, where the plating efficinency was 16.8%

Water stress enhances beta-amylase activity in cucumber cotyledons.

Cotyledons detached from 4-d-old cucumber (Cucumis sativus L.) seedlings were subjected to water stress (air-drying or PEG-treatment) to examine the effects of the stress on carbohydrate metabolism. Amylolytic activity in the cotyledon was increased about 6-fold by water stress within 1 d. The substrate specificity and the action pattern indicated that beta-amylase is responsible for the activity. Activities of azocaseinase, malate dehydrogenase and triose-phosphate isomerase were not affected by water stress, indicating that the effect of the stress on beta-amylase is rather specific. Cyclohe

Transformation of opium poppy (Papaver somniferum L.) with Agrobacterium rhizogenes MAFF 03-01724.

Transformed cultures of opium poppy (Papaver somniferum L.) were established by infecting hypocotyl segments with Agrobacterium rhizogenes MAFF 03-01724. Undifferentiated calli formed on the infected site grew satisfactorily on phytohormone-free solid medium in the dark and produced opine, mikimopine, which could not be detected in a normal culture. Numerous adventitious shoots developed from transformed calli during subculture. The transformed shoots separated individually were cultured on phytohormone-free MS solid medium at 22 ° C under 14 h/day light. They displayed wider leaves and longer

Expansion and Nitrate-Responsive Expression of NRT3 Transport Regulators in Maritime Pine.

Nitrate uptake in plants is mediated by coordinated transporter systems, which include NPF, NRT2 and NRT3 proteins. While these families have been extensively studied in angiosperms, their evolution and regulation in conifers are still not well understood. In this work, we examined the NRT3 family in maritime pine (Pinus pinaster) and in representative plant lineages. Phylogenetic analyses of nucleotide sequences and the NRT3 protein revealed a broad expansion in gymnosperms, particularly in conifers, while copy number increases among angiosperms appeared to be more lineage-specific. In additi

Effects of Atmospheric-Pressure N2, He, Air, and O2 Microplasmas on Mung Bean Seed Germination and Seedling Growth.

Atmospheric-pressure N2, He, air, and O2 microplasma arrays have been used to investigate the effects of plasma treatment on seed germination and seedling growth of mung bean in aqueous solution. Seed germination and growth of mung bean were found to strongly depend on the feed gases used to generate plasma and plasma treatment time. Compared to the treatment with atmospheric-pressure O2, N2 and He microplasma arrays, treatment with air microplasma arrays was shown to be more efficient in improving both the seed germination rate and seedling growth, the effect attributed to solution acidificat

Integrative modeling of cold and light signaling in tomato: unraveling the transcriptional network governing cold acclimation.

The integration of light and cold signaling is critical for cold acclimation in plants, but the regulatory complexity of the underlying transcriptional network remains poorly understood. To systematically dissect this interplay in tomato (Solanum lycopersicum), we constructed a computational model that merges the core cold-responsive inducer of CBF expression 1 (ICE1)-C-repeat binding factor (CBF)-cold regulated factors (COR) pathway with light-sensitive modules, including the constitutive photomorphogenic 1 (COP1)-elongated hypocotyl 5 (HY5)-MYB domain protein 15 (MYB15) cascade and the phyto

Tomato drought-responsive transcription factor TINY1 suppresses embryonic growth.

Dehydration-responsive element-binding (DREB) transcription factors play an important role in plant responses to drought. DREB subfamily A4 contains a subgroup named TINY. Previous studies in Arabidopsis suggest that TINYs suppress plant growth and mediate abscisic acid (ABA)-induced stomatal closure. In this study, we investigated the function of the tomato drought-induced TINY1. Under drought conditions, the tiny1 mutant lost turgor and wilted more rapidly than control M82 plants. However, this sensitivity was attributed to its larger leaf area, rather than intrinsic differences in drought r

Ultraviolet-B-induced OsKOL4 promotes ABA accumulation by inhibiting OsABA8ox1 and OsABA8ox2 expression.

Ultraviolet-B (UV-B) light is a component of sunlight that influences plant survival and adaptation. UV-B induces plants to regulate their phenotypes and metabolism to increase resistance to UV-B and associated stresses. Abscisic acid (ABA) metabolism and signaling are important for plant responses to UV-B. However, the mechanisms underlying UV-B-induced ABA accumulation and signaling in rice remain poorly understood. Here, we report that ENT-KAURENE OXIDASE LIKE 4 (OsKOL4) regulates UV-B-induced responses and ABA biosynthesis. UV-B activates OsKOL4 expression via OsbZIP48, an ELONGATED HYPOCO

Fusariumic Acids I and J, Two New Phytotoxic Isocassadiene-Type Diterpenoids from Tomato Fusarium Crown and Root Rot Pathogen Fusarium oxysporum f. sp. radicis-lycopersici.

Fusarium oxysporum f. sp. radicis-lycopersici (Forl) is the etiological agent of tomato Fusarium crown and root rot (FCRR), a devastating soil-borne disease that severely compromises global tomato production. The pathogenicity of Forl has been increasingly linked to its capacity to produce phytotoxic isocassadiene-type diterpenoids. In this study, Forl was cultured in rice medium to obtain Forl cultures, which were used for the separation and identification of secondary metabolites. After removing the known metabolites, two new isocassadiene-type diterpenoid compounds, namely fusariumic acids

HY5: a transcription factor integrating environmental and endogenous signals in plant development and acclimation.

As a master bZIP transcription factor, ELONGATED HYPOCOTYL 5 (HY5) coordinates plant growth with environmental acclimation across developmental stages. Beyond its role in photomorphogenesis, HY5 integrates diverse environmental and endogenous signals, linking environmental perception with developmental and metabolic programs to ensure coordinated physiological adaptation. This review synthesizes current understanding of the multilayered regulatory networks controlling HY5, ranging from constitutive photomorphogenic 1/suppressor of phytochrome A (COP1/SPA)-mediated ubiquitin-dependent degradati

The circadian clock regulates receptor-mediated immune responses to an herbivore-associated molecular pattern.

Plants activate induced defenses through the recognition of molecular patterns. Like pathogen-associated molecular patterns (PAMPs), herbivore-associated molecular patterns (HAMPs) can be recognized by cell surface pattern recognition receptors leading to defensive transcriptional changes in host plants. Herbivore-induced defensive outputs are regulated by the circadian clock, but the underlying molecular mechanisms remain unknown. To investigate how the plant circadian clock regulates transcriptional reprogramming of a specific HAMP-induced pathway, we characterized the daytime and nighttime

Light signaling-dependent regulation of plastid RNA processing in Arabidopsis.

Light is a vital environmental signal that regulates the expression of plastid genes. Plastids are crucial organelles that respond to light, but the effects of light on plastid RNA processing following transcription remain unclear. In this study, we systematically examined the influence of light exposure on plastid RNA processing, focusing on RNA splicing and RNA editing. We demonstrated that light promotes the splicing of transcripts from the plastid genes rps12, ndhA, atpF, petB, and rpl2. Additionally, light increased the editing rate of the accD transcript at nucleotide 794 (accD-794) and

Identification and functional characterization of a MAX2 ortholog from switchgrass (Panicum virgatum L.).

Switchgrass (Panicum virgatum L.) is a sustainable cellulosic energy crop with high biomass yield on marginal soils. Tillering, an important agronomic characteristic related to biomass production in gramineous plants, is regulated by complex interacting factors, such as plant hormones. Strigolactones (SLs) comprise a novel class of plant hormones that inhibit shoot branching. The MORE AXILLARY GROWTH2 (MAX2)/DWARF 3 (D3)/RAMOSUS (RMS4) genes encode proteins involved in the SL signaling pathway in various plants. The switchgrass tetraploid genome likely contains two high-similarity MAX2 homolog

A pair of light signaling factors FHY3 and FAR1 regulates plant immunity by modulating chlorophyll biosynthesis.

Light and chloroplast function is known to affect the plant immune response; however, the underlying mechanism remains elusive. We previously demonstrated that two light signaling factors, FAR-RED ELONGATED HYPOCOTYL 3 (FHY3) and FAR-RED IMPAIRED RESPONSE 1 (FAR1), regulate chlorophyll biosynthesis and seedling growth via controlling HEMB1 expression in Arabidopsis thaliana. In this study, we reveal that FHY3 and FAR1 are involved in modulating plant immunity. We showed that the fhy3 far1 double null mutant displayed high levels of reactive oxygen species and salicylic acid (SA) and increased

Mutations in the Pectin Methyltransferase QUASIMODO2 Influence Cellulose Biosynthesis and Wall Integrity in Arabidopsis.

Pectins are abundant in the cell walls of dicotyledonous plants, but how they interact with other wall polymers and influence wall integrity and cell growth has remained mysterious. Here, we verified that QUASIMODO2 (QUA2) is a pectin methyltransferase and determined that QUA2 is required for normal pectin biosynthesis. To gain further insight into how pectin affects wall assembly and integrity maintenance, we investigated cellulose biosynthesis, cellulose organization, cortical microtubules, and wall integrity signaling in two mutant alleles of Arabidopsis (Arabidopsis thaliana) QUA2, qua2 an

AtOFPs regulate cell elongation by modulating microtubule orientation via direct interaction with TONNEAU2.

As a group of plant-specific proteins, OVATE family protein (OFP) members have been shown to function as transcriptional repressors and involve in plant growth regulation in Arabidopsis and rice. It has also been shown that OFPs can interact with TONNEAU1 Recruiting Motif (TRM) proteins to regulate tomato fruit shape. In this study, we show that mutant plants with knock-down expression of OFP1, OFP2, OFP3, and OFP5 exhibit longer hypocotyls and cotyledons due to enhanced cell elongation. Overexpression of OFPs disturb the arrangement of cortical microtubule arrays in pavement cells and promote

Utilization of a response-surface technique in the study of plant responses to ozone and sulfur dioxide mixtures.

A second order rotatable design was used to obtain polynomial equations describing the effects of combinations of sulfur dioxide (SO(2)) and ozone (O(3)) on foliar injury and plant growth. The response surfaces derived from these equations were displayed as contour or isometric (3-dimensional) plots. The contour plots aided in the interpretation of the pollutant interactions and were judged easier to use than the isometric plots. Plants of ;Grand Rapids' lettuce (Lactuca sativa L.), ;Cherry Belle' radish (Raphanus sativus L.), and ;Alsweet' pea (Pisum sativum L.) were grown in a controlled env
