This study evaluated the efficacy of four different culture media in cultivating Mycobacterium avium subspecies paratuberculosis (MAP). The aim of this study was to compare ELISA, immunodiffusion and immunoblot for the detection of anti-Jo-1 antibodies, and to investigate the association of the results with clinical manifestations. In two medical centres for rheumatology and one for pulmonology, all patients with suspected connective tissue disease were screened over a 5-year period for anti-Jo-1 antibodies by ELISA. Indirect immunofluorescence and simple immunodiffusion for trypanosomiasis were normalized. A high percent of patients were found to be positive, both by simple immunodiffusion and indirect immunofluorescence. The use of these techniques is recommended in epidemiological studies.

Antibodies to saline-extractable nuclear antigens are hallmarks of autoimmune diseases including systemic lupus erythematosus (SLE), mixed connective tissue disease, progressive systemic sclerosis and Sjogren's syndrome. Swine brucellosis caused by Brucella suis biovar 2 is an emerging disease in Europe. 30 vaginal secreta of women affected by acute and chronic trichomoniasis from the Gynaecological Hospital S. Anna of Turin were examined. In the same secreta the specific IgA anti Trichomonas vaginalis were detected using indirect immunofluorescence and titered on LC-Partigen IgA immunodiffusion plates. Only in 30% of these cases a correlation between immunodiffusion and fluorescence test resulted.

Three agar immunoprecipitin techniques (double immunodiffusion, immunoelectrophoresis and immunoelectroosmophoresis-immunodiffusion) made with paracoccidioidin and serum of a paracoccidioidomycosis patient's wife, permitted us to observe a specific band in 6 successive samples of blood taken from the wife over a period of 21 months. The case represents an example of a subclinical paracoccidioidomycosis infection that is usually diagnosed in its disseminated, progressive form. Three possible sources of infection are discussed. But an interhuman contagion appears to be the most feasible.

Plasma retinol-binding protein (RBP) concentrations have been suggested as surrogate indicators for plasma retinol concentrations in the assessment of vitamin A status in less technologically developed settings. The cerebrospinal fluid of 589 subjects, 78 of whom were suffering from a purulent meningitis were examined. Human chorionic prealbumin (HCPA) was immunochemically identified and isolated in pure form from immature placenta, particularly, from chorion. Comparative immunodiffusion analysis shows that HCPA differs from the known placenta-specific proteins. HCPA is a glycoprotein with MW about 100 KD determined by gel-filtration and 45 KD by SDS polyacrylamide gel electrophoresis. By immunodiffusion HCPA was not found in fetal and adult tissues and in biological fluids. HCPA was discovered in patients with trophoblastic diseases but was not revealed in other oncological situations.
