BACKGROUND: The absence of representative Epstein-Barr virus-associated intrahepatic cholangiocarcinoma (EBVaICC) cell lines has limited our understanding of the molecular and immunological characteristics of this cancer subtype.
METHODS: We reviewed patients with metastatic cholangiocarcinoma at Sun Yat-sen University Cancer Center from January 2015 to August 2023. Among them, 22 patients with EBVaICC and 66 patients with non-EBVaICC who received anti-PD1 treatment were included. Additionally, 2 EBV-positive ICC cell lines, RBE-EBV and HuH28-EBV, were developed through cell-to-cell infection

Peritoneal carcinomatosis (PC) is a common metastatic cancer with limited treatment options. Herein, we present a novel strategy for the combined treatment of PC involving plasma-activated saline (PAS) and hyperthermic intraperitoneal perfusion. PAS revealed a strong cytotoxic effect because of reactive oxygen species (ROS) in two-dimensional cultures and three-dimensional tumor spheroids of PC-related cell lines. Notably, PAS induced Gasdermin E (GSDME)-dependent pyroptosis and immunogenic cell death in vitro. PAS-enhanced hyperthermic intraperitoneal perfusion (PE-HIP) increased the number o

BACKGROUND: Adoptive cell therapy (ACT) utilizing tumor-infiltrating lymphocytes (TIL) is a promising immunotherapeutic approach for disseminated malignancies. However, ex vivo-expanded tumor-infiltrating lymphocytes (TIL) often rapidly progress to a state of functional exhaustion or suppression within the tumor microenvironment. Interleukin-7(IL-7) not only supports the survival of T-lymphocyte in vivo but also induces vigorous expansion of naïve and memory T lymphocytes in vitro. Upregulating the expression of interleukin-7 receptor alpha (IL-7Rα) helps T cells restore their responsiveness t

Efficient cancer immunotherapy depends on selective targeting of high bioactivity therapeutic agents to the tumours. However, delivering exogenous medication might prove difficult in clinical practice. Here we report a cooperative Nano-CRISPR scaffold (Nano-CD) that utilizes a specific sgRNA, selected from a functional screen for triggering endogenous GDSME expression, while releasing cisplatin to initiate immunologic cell death. Mechanistically, cascade-amplification of the antitumor immune response is prompted by the adjuvantic properties of the lytic intracellular content and enhanced by th

High expression of programmed death ligand 1 (PD-L1) and strong immune evasion ability of the tumor microenvironment (TME) are maintained through mutual regulation between different immune and stromal cells, which causes obstructions for cancer immunotherapy, especially immunosuppressive M2-like phenotype tumor-associated macrophages (TAMs). Repolarization of TAMs to the M1-like phenotype could secrete proinflammatory cytokines and reverse the immunosuppressive state of the TME. However, we found that reactive oxygen species (ROS) generated by repolarized TAMs could be a double-edged sword: RO

Murine studies have shown that immunologic targeting of the tumor vasculature, a key element of the tumor stroma, can lead to protective immunity in the absence of significant pathology. In the current study, we expand the scope of stroma-targeted immunotherapy to antigens expressed in tumor-associated fibroblasts, the predominant component of the stroma in most types of cancer. Mice were immunized against fibroblast activation protein (FAP), a product up-regulated in tumor-associated fibroblasts, using dendritic cells transfected with FAP mRNA. Using melanoma, carcinoma, and lymphoma models,

BACKGROUND: As in vitro diagnosis of wasp venom sensitization by specific serum IgE has a sensitivity of only 60-80%, additional in vitro tests are desirable. Basophil activation is associated with the expression of CD63 and its measurement has been proposed as a novel in vitro test for immediate-type allergy. Furthermore, to date, no in vitro test exists to monitor successful specific immunotherapy (SIT) with wasp venom. Therefore, the potentially harmful sting challenge is still recommended.
OBJECTIVE: We compared the CD63-based basophil activation test (BAT) in the diagnosis of wasp venom

X-ray triggered local activation of prodrugs is promising for tumor radio-chemo-immunotherapy. However, the insufficient active drugs within tumors under prescribed X-ray doses lead to poor immunogenic effects. Here, we proposed to develop ClO--responsive nanoprodrug (DOX/S-M) that could persistently release DOX in response to inflammation induced by single-dose X-ray irradiation, thereby maintaining high levels of active DOX in tumors. DOX/S-M, prepared by the self-assembly of DOX-S-mPEG2000 and tumor-targeting amphiphilic polymers (DSPE-PEG-Folate), were stable in normal environments. After

Anti-neutrophil cytoplasmic antibody (ANCA)-associated vasculitis (AAV) is a systemic autoimmune disease characterized by necrotizing small-vessel inflammation, in which dysregulated adaptive immune responses play a central pathogenic role. Beyond the well-established contribution of ANCAs and innate immune activation, increasing evidence highlights profound alterations in T-cell regulation that drive persistent inflammation, autoantibody production, and organ damage. Immune checkpoints (ICs)-a network of co-stimulatory and co-inhibitory pathways that fine-tune lymphocyte activation and mainta

The stimulator of interferon genes (STING) pathway is a promising immuno-oncology target. Despite their potential, STING agonists have yielded underwhelming results in clinical trials due to pharmacological barriers that limit their safety and efficacy. Herein, we describe the design and pre-clinical evaluation of a polymeric dimeric amidobenzimidazole (diABZI) STING agonist delivery platform for cancer immunotherapy. Central to our technology is a stimuli-responsive diABZI-functionalized reversible addition-fragmentation chain transfer (RAFT) polymerization chain transfer agent (CTA) that all

Stimulator of interferon genes (STING) agonists have shown limited antitumor efficacy, in part because STING activation is not preferentially focused on dendritic cells (DCs), which specialize in cross-priming. We present a DC-centric strategy that synergistically licenses DCs by coordinating CD47-SIRPα checkpoint relief and STING activation via dual ultra-pH-sensitive gating. Mild tumor acidity first unmasks the αCD47 cue to prime antigen acquisition. Following DC-biased uptake, a second acidic gate releases cGAMP to engage STING in antigen-bearing DCs. Functionally, efficacy requires Batf3-d

Resistance to immune checkpoint blockade in gastric cancer often arises from defective antigen presentation due to MHC-I downregulation, yet strategies to restore MHC-I expression remain limited. Here, using the natural alkaloid koumine as a chemical probe, we identified IGF2BP3 as a druggable regulator of MHC-I antigen presentation. Network pharmacology, proteomics, surface plasmon resonance, and cellular thermal shift assays were used to identify and validate koumine's target. Genetic rescue experiments, loss-of-function studies, in vitro T-cell co-culture assays, and in vivo gastric cancer

Single-cell transcriptomics identifies a convergent activation state of conventional dendritic cells (cDCs) shared by type 1 and type 2 cDCs (cDC1s and cDC2s). These activated DCs (actDCs) are characterized by co-expression of T cell-stimulating and inhibitory molecules. Here, we examined the functional contribution of actDCs to anti-tumor immunity by developing mouse models that leverage CCR7 expression to conditionally label or ablate actDCs. The capacity of cDCs to stimulate tumor-specific cytotoxic T lymphocytes (CTLs) was restricted to the actDC state. cDC1- and cDC2-derived actDCs suppor

Malignant tumors remain one of the most serious challenges to global health. Although chemotherapy and targeted therapy are available treatment options, their effectiveness is often limited by the complexity of the tumor microenvironment (TME). In recent years, immunotherapy has demonstrated significant potential in harnessing the immune system to combat cancer. Polymeric nanoparticles (PNPs) have emerged as versatile platforms for cancer immunotherapy, offering favorable biocompatibility, tunable size, and surface functionalization for targeted delivery. In this review, we critically evaluate

Messenger RNA (mRNA)-based vaccines have great potential in cancer treatment. However, poor lymphatic transport, insufficient targeted delivery, intracellular degradation and insufficient immune response without adjuvants limit the application of mRNA vaccines. Herein, a novel mRNA nanovaccine (HM@Mn3O4-mRNA) was constructed by ovalbumin (OVA) mRNA-loaded Mn3O4 encapsulation with a hybrid membrane (HM) of dendritic cells (DCs) and bacterial membrane for enhancing cancer immunotherapy. In vitro results indicated that HM@Mn3O4-mRNA nanovaccine could target DC2.4 cells, achieve lysosomal escape t

Gastric cancer (GC) is characterised by a dense stromal microenvironment, lack of therapeutic targets, and limited effective treatment options, collectively leading to a poor prognosis. Here, we identify UL16 binding protein 2 (ULBP2) as a potential therapeutic target in GC. Mechanistically, ULBP2 overexpression activates the TGF-β signalling pathway, promoting the activation of cancer-associated fibroblasts (CAFs) and tumor progression in GC. Furthermore, we developed ULBP2 CAR-T cells and assessed their therapeutic potential in GC cell lines, organoids, cell line-derived xenograft (CDX) and

Tumor senescence, a double-edged sword, can suppress tumor growth but also promote immune evasion if not properly cleared. Herein, a cell membrane-coated ZIF-8@MnOx nanoplatform co-loaded with doxorubicin (DOX) and piperlongumine (PL), termed mPDZM, is developed to remodel the senescence-mediated immune response in hepatocellular carcinoma. PL synergizes with DOX to amplify intracellular oxidative stress, which promotes both the killing of tumor cells and the clearance of senescent cells. The biomimetic ZIF-8@MnOx nanoplatform potentiates the efficacy of DOX and PL by integrating targeted deli

As natural agonists of the stimulator of interferon genes (STING) protein, cyclic dinucleotides (CDNs) can activate the STING pathway, leading to the expression of type I interferons and various cytokines. Efficient activation of the STING pathway in antigen-presenting cells (APCs) and tumor cells is crucial for antitumor immune response. Tumor-derived exosomes can be effectively internalized by APCs and tumor cells and have excellent potential to deliver CDNs to the cytoplasm of APCs and tumor cells. Here, we leverage tumor exosomes as a delivery platform, designing an EXOTLR1/2-STING loaded

Advances in adaptive immunity have greatly contributed to the development of cancer immunotherapy. However, its over-low efficacy and insufficient invasion of immune cells in the tumor tissue, and safety problems caused by cytokine storm, have seriously impeded further clinical application for solid tumor immunotherapy. Notably, the immune microenvironment of the lungs is naturally enriched with alveolar macrophages (AMs). Herein, we introduce a novel nebulized magnetothermal immunotherapy strategy to treat orthotopic lung cancer by using magnetothermal nanomaterial (Zn-CoFe2O4@Zn-MnFe2O4-PEG,
