Distinct Effects of Type I and III Interferons on Enteric Viruses.

Interferons (IFNs) are key host cytokines in the innate immune response to viral infection, and recent work has identified unique roles for IFN subtypes in regulating different aspects of infection. Currently emerging is a common theme that type III IFNs are critical in localized control of infection at mucosal barrier sites, while type I IFNs are important for broad systemic control of infections. The intestine is a particular site of interest for exploring these effects, as in addition to being the port of entry for a multitude of pathogens, it is a complex tissue with a variety of cell type

Species-independent bioassay for sensitive quantification of antiviral type I interferons.

BACKGROUND: Studies of the host response to infection often require quantitative measurement of the antiviral type I interferons (IFN-alpha/beta) in biological samples. The amount of IFN is either determined via its ability to suppress a sensitive indicator virus, by an IFN-responding reporter cell line, or by ELISA. These assays however are either time-consuming and lack convenient readouts, or they are rather insensitive and restricted to IFN from a particular host species. RESULTS: An IFN-sensitive, Renilla luciferase-expressing Rift Valley fever virus (RVFV-Ren) was generated using revers

Effect of interferons on hormone-receptor levels of endometrial cancer-cells.

Endometrial cancer is a hormone sensitive tumor. Hormone receptor positive tumors respond better to progestins than hormone receptor negative tumors. Interferon has been shown to increase the hormone receptor level of melanocytes, breast and endometrial cancer. We have previously shown that interferons enhance the progesterone receptor level of AE-7 endometrial cancer cell line, which has a considerably high baseline level of progesterone receptors (201+/-19.7 fmole/mg of proteins). In this study the effect of interferons of two other endometrial cancer cell lines (HEC-1A and HEC-1B), with a l

[Possibilities and limits of the use of interferons in the clinic].

Only the recent production of large amounts of highly purified recombinant interferons has made it possible to elucidate precisely the in vitro and in vivo effect of alpha- and gamma-interferon. Interferon-alpha has significantly widened the treatment modalities for some rare tumors such as hairy cell leukemia and chronic myelogenous leukemia. Although treatment results in solid tumors are disappointing, tumor regression greater than 50% is achieved in 15-25% of patients with hypernephroma or melanoma, cancers highly resistant to cytotoxic drugs. The solid tumors must be treated with high dose

[Therapy of AIDS-induced Kaposi sarcoma and malignant melanoma with interferons].

Since 1980, disseminated Kaposi's sarcoma has been occurring in new epidemic proportions with a rapid clinical course in risk populations. Sixteen cases were under therapy and close surveillance from 1982 to 1986. Eight are still under therapy. In disseminated Kaposi's sarcoma with acquired immune deficiency syndrome (AIDS) our experience was encouraging. Following systemic, long-term treatment with recombinant alpha 2a interferon we observed complete remission of the lesions in 2 cases, partial remission and stabilization of the disease in 3 cases, at least temporary stabilization of the dise

Autoantibodies neutralizing type I interferons in older twin pairs: Low concordance.

Autoantibodies (auto-Abs) neutralizing type I interferons (IFN-I) underlie various life-threatening viral infections, and the prevalence of these auto-Abs rises sharply between 65 and 70 years of age. We investigated the familial aggregation of neutralizing IFN-I auto-Abs by assessing concordance rates in Danish older twins, including 3,270 twins aged 65-96 years in a sample comprising 1,318 intact twin pairs (2,636 individuals) and 634 twins from broken twin pairs (634 individuals). Neutralizing IFN-I auto-Abs were detected in 1.5% (50/3,270) of all twins tested, including 1.4% (37/2,636) in

Type-1 interferons associated with microglial-mediated neuroinflammation in Alzheimer's disease.

Type-1 interferons (IFN-1) are a pleiotropic group of cytokines initially known for their antiviral activity, but recent studies highlight a vital role of IFN-1 in central nervous system (CNS) homeostasis, brain aging, and neurodegeneration. Alzheimer’s disease (AD) is an age-related neurodegenerative disorder with a complex etiology. The hallmark signs of AD that appear in the brain are amyloid-β (Aβ) plaques and intracellular neurofibrillary tangles (NFTs) composed of hyperphosphorylated tau. Neuroinflammation facilitated primarily by microglia is known to drive amyloid and tau pathology, wi

Interferons in HIV-1 infection: mechanisms, antiviral potentials, and therapeutic challenges.

Type I interferons (IFNs), particularly IFN-α, occupy a central paradox in HIV-1 infection: they provide an essential early antiviral barrier that limits initial dissemination, yet their sustained activation contributes to chronic immune activation, CD4+ T-cell dysfunction, and incomplete viral control. This duality-protective in acute infection but pathogenic during chronic disease-remains a major unresolved challenge for interferon-based therapeutic strategies in HIV-1. Recent advances in ISG functional profiling, IFN-α subtype-specific antiviral potency, and the development of targeted inna

Downregulation of type III interferons in patients with severe COVID-19.

Coronavirus disease 2019 (COVID-19) is globally rampant, and to curb the growing burden of this disease, in-depth knowledge about its pathophysiology is needed. This was an observational study conducted at a single center to investigate serum cytokine and chemokine levels of COVID-19 patients, based on disease severity. We included 72 consecutive COVID-19 patients admitted to our hospital from March 21 to August 31, 2020. Patients were divided into Mild-Moderate I (mild) and Moderate II-Severe (severe) groups based on the COVID-19 severity classification developed by the Ministry of Health, La

Type III interferons: Balancing tissue tolerance and resistance to pathogen invasion.

Type III IFNs, or IFN-λ, are the newest members of the IFN family and were long believed to play roles that were redundant with those of type I IFNs. However, IFN-λ displays unique traits that delineate them as primary protectors of barrier integrity at mucosal sites. This unique role stems both from the restricted expression of IFN-λ receptor, confined to epithelial cells and to a limited pool of immune cells, and from unique immunomodulatory properties of IFN-λ. Here, we discuss recent findings that establish the unique capacity of IFN-λ to act at the barriers of the host to balance tissue t

Type III interferons disrupt the lung epithelial barrier upon viral recognition.

Viral infections of the lower respiratory tract are a leading cause of mortality. Mounting evidence indicates that most severe cases are characterized by aberrant immune responses and do not depend on viral burden. In this study, we assessed how type III interferons (IFN-λ) contribute to the pathogenesis induced by RNA viruses. We report that IFN-λ is present in the lower, but not upper, airways of patients with coronavirus disease 2019 (COVID-19). In mice, we demonstrate that IFN-λ produced by lung dendritic cells in response to a synthetic viral RNA induces barrier damage, causing susceptibi

[The decisive importance of constitutional interferons in human and animal ontogenesis].

Biomedical publications contain little information on the constitutional (endogenous) interferons (IFNs) produced by different cells without prior exposure to viruses and oncogenic factors. Literature analysis has provided a generalized concept that these interferons play different functional roles according to ontogenetic stages. The maximum production of high-molecular- weight immature forms of IFNα/β was detected in the embryos of mice, Syrian hamsters, and humans and in the syncytiotrophoblast of placental villi. This is due to the direct involvement of IFNs in embryogenesis and fetogenesi

Type I interferons in the pathogenesis and treatment of canine diseases.

Type I interferons (IFNs) such as IFN-α, IFN-β, IFN-ε, IFN-κ, and IFN-ω represent cytokines, which are deeply involved in the regulation and activation of innate and adaptive immune responses. They possess strong antiviral, antiproliferative, and immunomodulatory activities allowing their use in the therapy of different viral diseases, neoplasms, and immune-mediated disorders, respectively. Initially, treatment strategies were based on nonspecific inducers of type I IFNs, which were soon replaced by different recombinant proteins. Drugs with type I IFNs as active agents are currently used in t

The role of type I interferons and other cytokines in dermatomyositis.

Much work has been done to unveil the mechanisms behind the pathogenesis of dermatomyositis (DM) - mainly those involving certain pathogenic cytokines, termed "pathokines" as the principal cytokines involved. Recently, it has become clear that a group of cytokines known as type I interferons (IFN-Is) play a significant role in the development of DM. We review the literature published between 1946 and 2014 using an Ovid Medline database search to provide an update on the role of IFN-Is and other cytokines in the pathogenesis of DM. We provide information about the genes and proteins induced by

Confounding roles for type I interferons during bacterial and viral pathogenesis.

Although type I interferons (IFN-I) were initially defined as potent antiviral agents, they can also cause decreased host resistance to some bacterial and viral infections. The many antiviral functions of the IFN-I include direct suppression of viral replication and activation of the immune response against viruses. In addition to their antiviral effects, IFN-I are also protective against several extracellular bacterial infections, in part, by promoting the induction of TNF-α and nitric oxide. In contrast, there is a negative effect of IFN-I on host resistance during chronic infection with lym

Development of pegylated interferons for the treatment of chronic hepatitis C.

The chemical attachment of poly(ethylene glycol) [PEG] to therapeutic proteins produces several benefits, including enhanced plasma half-life, lower toxicity, and increased drug stability and solubility. In certain instances, pegylation of a protein can increase its therapeutic efficacy by reducing the ability of the immune system to detect and mount an attack on the compound. A PEG-protein conjugate is formed by first activating the PEG moiety so that it will react with, and couple to, the protein. PEG moieties vary considerably in molecular weight and conformation, with the early moieties (m

Biology and therapeutic uses of myeloid hematopoietic growth factors and interferons.

Recent advances in basic science and clinical trials have demonstrated that IFNs and myeloid hematopoietins play crucial roles in host defense against pathogens and immune surveillance. Here we have reviewed the biologic functions of GM-CSF, G-CSF, IFN-alpha and IFN-gamma. For patients with neutropenia resulting from cytotoxic chemotherapy, bone marrow transplantation, congenital agranulocytosis and cyclic neutropenia, therapeutic uses of GM-CSF and G-CSF were reviewed. Application of these growth factors to patient management represents a major contribution of biotechnology to a difficult are

The role of biotherapies (interleukins, interferons and erythropoietin) in multiple myeloma.

During the last decade, the availability of large numbers of cytokines and growth factors has greatly favoured the use of biotherapies in several haematological disease. For MM, the majority of clinical studies have dealt with the use of IFN-alpha. From these studies it appears that IFN-alpha has a definite role in the treatment of MM especially in the setting of minimal residual disease, as maintenance therapy after response to conventional therapies or HDC followed by BMT procedures or PBSCI. Data on the use of EPO have consistently demonstrated the role of this growth factor in ameliorating
