Cardiovascular Drugs and Osteoarthritis: Effects of Targeting Ion Channels.

Osteoarthritis (OA) and cardiovascular diseases (CVD) share many similar features, including similar risk factors and molecular mechanisms. A great number of cardiovascular drugs act via different ion channels and change ion balance, thus modulating cell metabolism, osmotic responses, turnover of cartilage extracellular matrix and inflammation. These drugs are consumed by patients with CVD for many years; however, information about their effects on the joint tissues has not been fully clarified. Nevertheless, it is becoming increasingly likely that different cardiovascular drugs may have an im

Ion Channels in the Eye: Involvement in Ocular Pathologies.

The eye is the sensory organ of vision. There, the retina transforms photons into electrical signals that are sent to higher brain areas to produce visual sensations. In the light path to the retina, different types of cells and tissues are involved in maintaining the transparency of avascular structures like the cornea or lens, while others, like the retinal pigment epithelium, have a critical role in the maintenance of photoreceptor function by regenerating the visual pigment. Here, we have reviewed the roles of different ion channels expressed in ocular tissues (cornea, conjunctiva and neur

Ion channels and drug transporters as targets for anthelmintics.

Infections with parasitic helminths such as schistosomes and soil-transmitted nematodes are hugely prevalent and responsible for a major portion of the global health and economic burdens associated with neglected tropical diseases. In addition, many of these parasites infect livestock and plants used in agriculture, resulting in further impoverishment. Treatment and control of these pathogens rely on anthelmintic drugs, which are few in number, and against which drug resistance can develop rapidly. The neuromuscular system of the parasite, and in particular, the ion channels and associated rec

The structure and regulation of magnesium selective ion channels.

The magnesium ion (Mg(2+)) is the most abundant divalent cation within cells. In man, Mg(2+)-deficiency is associated with diseases affecting the heart, muscle, bone, immune, and nervous systems. Despite its impact on human health, little is known about the molecular mechanisms that regulate magnesium transport and storage. Complete structural information on eukaryotic Mg(2+)-transport proteins is currently lacking due to associated technical challenges. The prokaryotic MgtE and CorA magnesium transport systems have recently succumbed to structure determination by X-ray crystallography, provid

Therapeutic potential of peptide toxins that target ion channels.

Traditional healthcare systems in China, India, Greece and the Middle East have for centuries exploited venomous creatures as a resource for medicines. This review focuses on one class of pharmacologically active compounds from venom, namely peptide toxins that target ion channels. We highlight their therapeutic potential and the specific channels they target. The field of therapeutic application is vast, including pain, inflammation, cancer, neurological disorders, cardioprotection, and autoimmune diseases. One of these peptides is in clinical use, and many others are in various stages of pre

Ion channels and the control of myometrial electrical activity.

Understanding the role of ion channels in the generation of slow waves and action potentials in the myometrium is critical in designing strategies to regulate uterine contractile activity. The development of the patch clamp technique has allowed the identification of specific types of channels in the myometrium and provided insights into their regulation by hormones and drugs. Specifically, new studies suggest that KATP and KCa channel openers could be important tools in the management of inappropriate uterine contractions, but peripheral effects will have to be controlled. Conversely, blocker

Mechanosensitive ion channels of E. coli activated by amphipaths.

Mechanosensitive channels have been found in more than 30 cell types, including bacterial, yeast, plant and animal cells. Whether tension is transferred to the channel through the lipid bilayer and/or underlying cytoskeleton is not clear. Using the patch-clamp method, we found that amphipathic compounds, which are molecules having hydrophobic and hydrophilic character with positive, negative or no net electric charge at pH 7, could slowly activate the mechanosensitive channels of giant Escherichia coli spheroplasts, with effectiveness proportional to their lipid solubility. The cationic or ani

Abnormal regulation of ion channels in cystic fibrosis epithelia.

Cystic fibrosis (CF), the most common lethal genetic disease in Caucasians, is characterized by defective electrolyte transport in several epithelia. In sweat duct, pancreatic, intestinal, and airway epithelia, abnormalities in transepithelial ion transport may account for the manifestations of the disease. A Cl- impermeable apical cell membrane is a common feature in these CF epithelia. The rate of transepithelial Cl- transport is controlled in part by hormonally regulated apical membrane Cl- channels; in CF epithelia, Cl- channels are present but their regulation is defective. Most regulatio

Tetrahydroaminoacridine blocks voltage-dependent ion channels in hippocampal neurons.

Tetrahydroaminoacridine (THA) is a centrally active anticholinesterase that may produce functional improvement in patients with Alzheimer's disease. Because of its structural similarity to the potassium channel (A-current) blocking drug 4-aminopyridine (4-AP), it has been speculated that some of the therapeutic benefit of THA might result from inhibition of potassium currents. In the present study, THA produced a dose-dependent, reversible block of the A-current in cultured hippocampal neurons (IC50, 30 microM). However, at similar concentrations it also inhibited other voltage-dependent curre

delta-Philanthotoxin, a semi-irreversible blocker of ion-channels.

1. The digger wasp, Philanthus triangulum, which preys on honeybees, produces a paralysing venom possessing a wide variety of activities. 2. In insects, the venom has a central as well as a peripheral effect; the latter effect consists of a presynaptic as well as a postsynaptic block of the skeletal neuromuscular transmission. 3. The presynaptic block is probably caused by an inhibition of the re-uptake of the transmitter. The postsynaptic effect probably consists of a block of open ion channels. 4. The venom contains at least four active toxins called alpha-, beta-, gamma- and delta-philantho

Sequence alignment of viral channel proteins with cellular ion channels.

Sequence alignment is an important tool for identifying regions of similarities among proteins and for, thus, establishing functional and structural relationships between different proteins. Here, alignments of transmembrane domains (TMDs) of viral channel forming proteins with host ion channels and toxins are evaluated. The following representatives of polytopic viral channel proteins are chosen: (i) p7 of HCV and 2B of Polio virus (two TMDs) and (ii) 3a of SARS-CoV (three TMDs). Using ClustalW2, each of the TMDs of the viral channels is aligned, and the overlap is mapped onto structural mode

Supraventricular tachycardias in ion channel diseases.

Supraventricular tachyarrhythmias are a clinical finding that is seen to a varying extent in patients with ion channel diseases depending on the underlying genetic disease. In addition to the classical symptoms that they cause, they may trigger ventricular tachyarrhythmias or induce inappropriate implantable cardioverter-defibrillator (ICD) therapies and, thus, cause psychological harm. Antiarrhythmic drugs can be used in some diseases, but are contraindicated in a number of genetic conditions. In these patients, catheter ablation is a good alternative and sometimes the only treatment choice.

Ion channel regulation of gut immunity.

Mounting evidence indicates that gastrointestinal (GI) homeostasis hinges on communications among many cellular networks including the intestinal epithelium, the immune system, and both intrinsic and extrinsic nerves innervating the gut. The GI tract, especially the colon, is the home base for gut microbiome which dynamically regulates immune function. The gut's immune system also provides an effective defense against harmful pathogens entering the GI tract while maintaining immune homeostasis to avoid exaggerated immune reaction to innocuous food and commensal antigens which are important cau

Ion Channel Pharmacology for Pain Modulation.

A large series of different ion channels have been identified and investigated as potential targets for new medicines for the treatment of a variety of human diseases, including pain. Among these channels, the voltage gated calcium channels (VGCC) are inhibited by drugs for the treatment of migraine, neuropathic pain or intractable pain. Transient receptor potential (TRP) channels are emerging as important pain transducers as they sense low pH media or oxidative stress and other mediators and are abundantly found at sites of inflammation or tissue injury. Low pH may also activate acid sensing

Ion channel defects in idiopathic epilepsies.

Idiopathic epilepsies are genetically determined diseases of the central nervous system characterized by typical epileptic seizures and EEG abnormalities but not associated with structural brain lesions. In recent years, an increasing number of mutations associated with idiopathic epilepsy syndromes were identified in genes encoding subunits of voltage- or ligand-gated ion channels. These encouraging results provide a plausible pathophysiological concept, since ion channels form the basis for neuronal excitability and are the major targets for anticonvulsive pharmacotherapy. The first epilepsy

[Ion-channel related muscular diseases].

INTRODUCTION: Though ion channel-related muscular disorders were described long ago, better understanding of their underlying mechanisms has been more recently achieved. These mechanisms include myotonic syndromes that may be caused by mutations in sodium and chloride channels, as well as periodic paralysis which is due to mutations in sodium and calcium channels. CURRENT KNOWLEDGE AND KEY POINTS: Knowledge of the involved pathophysiological mechanisms has led to better clinical description of these disorders, as well as more efficacious treatment. In some cases, it is now possible to establi

Therapeutic approaches to ion channel diseases.

More than 400 genes are known that encode ion channel subunits. In addition, alternative splicing and heteromeric assembly of different subunits increase tremendously the variety of ion channels. Such many channels are needed to accomplish very complex cellular functions, whereas dysfunction of ion channels are key events in many pathological processes. The recent discovery of ion channelopathies, which, in its more stringent definition, encloses monogenic disorders due to mutations in ion channel genes, has largely contributed to our understanding of the function of the various channel subtyp

Cardiac ion channel effects of tolterodine.

Tolterodine is a muscarinic antagonist widely used in the treatment of urinary incontinence. Although tolterodine has not been reported to alter cardiac repolarization, it is chemically related to other muscarinic antagonists known to prolong cardiac repolarization. For this reason, we studied the effects of tolterodine on cardiac ion channels and action potential recordings. Using patch-clamp electrophysiology, we found that tolterodine was a potent antagonist of the human ether-a-go-go-related gene (HERG) K(+) channel, displaying an IC(50) value of 17 nM. This potency was similar to that obs

[GABA receptor chloride ion channel].

Gamma-Aminobutyric acid(GABA) has been established as a major inhibitory neurotransmitter in the brain. GABAA receptors are coupled to benzodiazepine receptors and barbiturate binding site. They are ligand-gated Cl- channels that consist of a heteromeric mixture of protein subunits forming a pentameric structure. On the other hand, GABAC receptors appear to be relatively simple ligand-gated Cl- channels with a distinctive pharmacology, in that they are not blocked by bicuculline and not modulated by barbiturates, benzodiazepines or neuroactive steroids. Therefore, it is suggested that these mu
